Abstract
Description
Use of Compound A for Treating a CancerTechnical FieldThis application relates to use of a KRAS G12C inhibitor, in particular to use of Compound A for treating a cancer. Background ArtRAS gene is the proto-oncogene with the highest mutation frequency, which is found in about 30%of human tumors, and even 100%of pancreatic cancers. Among them, KRAS gene is the subtype with the highest mutation frequency in the RAS gene family, and KRAS gene mutations account for about 83%of the total RAS gene mutations. NRAS and HRAS mutations account for 13%and 4%of total RAS mutations, respectively. The most common mutation mode of KRAS gene is point mutation, and the most frequently mutated sites are codons 12, 13 and 61, of which the mutation in codon 12 is the most common. KRAS gene mutations mainly include: G12D, G12V, G13D, G12C, G12A and G12S, and their mutation frequencies account for 33.4%, 22.8%, 12.4%, 11.3%, 5.3%, and 4.4%of KRAS mutations, respectively. Among them, the KRAS G12C mutation occurs in 13%of lung adenocarcinomas, 3%of colorectal cancers, and 2%of other solid tumors. Mutations in KRAS often interfere with GAP-mediated GTP hydrolysis, thereby allowing these mutant KRASs to remain in a GTP-bound activated state. The intrinsic GTPase activity and GDP-GTP exchange activity of KRAS are different in different KRAS mutations. For example, mutations at codons 12, 13, and 61 generally attenuate the intrinsic GTPase activity of KRAS.Drug development for KRAS has progressed very slowly over the past few decades and is considered an undruggable target. The difficulty of directly targeting KRAS is that the binding of GTP and KRAS is very tight, at the picomolar level, and there is a large amount of GTP in the cell at millimolar level, with which drug molecules cannot directly compete with it. In addition, for drug molecules to act on proteins, they actually need a pocket that interacts with proteins, while the surface of RAS is very smooth, with a nearly spherical structure and no obvious binding sites. It is difficult to synthesize a compound that can directly targets KRAS binding and inhibits its activity.WO2021121367A1 discloses KRAS mutant protein (KRAS p. G12C) inhibitors that can lock KRAS p. G12C in an inactive state and block KRAS signal transduction downstream. Results from preclinical studies showed potent antitumor effects against KRAS p. G12C mutation-bearing lung, pancreatic and colorectal cancers, and favorable pharmacokinetic properties with a very wide therapeutic safety window. There remains a need for new treatment regimens for cancer using a KRAS mutant protein inhibitor. The present disclosure is directed toward this need and others.SummaryProvided herein is a method of treating cancer comprising administering to a subject in need thereof Compound A, a pharmaceutically acceptable salt or a co-crystal thereof at a daily dose of 100 mg to 3600 mg on salt-free basis.Provided herein is Compound A, a pharmaceutically acceptable salt or a co-crystal thereof for use in the treatment of a cancer, wherein, Compound A, a pharmaceutically acceptable salt or a co-crystal thereof is administered at a daily dose of 100 mg to 3600 mg on the salt-free basis.Provided herein is use of Compound A, a pharmaceutically acceptable salt or a co-crystal thereof in the manufacture of a medicament in the treatment of a cancer, wherein, the Compound A, a pharmaceutically acceptable salt, or a co-crystal thereof is administered in a daily dose of 100 mg to 3600 mg on salt-free basis.Detailed DescriptionThe present invention provides:[1] . The X-ray tube voltage and amperage were set to 40 kV and 40 mA respectively. Data was collected using Collection Software (Diffrac Plus XRD Commander) at the Cu Kα radiation with a wavelength of from 3.0 to 40 degrees (2θ) or 3.0 to 30 degrees (2θ) (Hot Stage XRPD) using a increment of 0.02o (2θ) degrees and a scan rate of 0.2 seconds. A typical error associated with measurement can occur as a result of a variety of factors . Therefore peaks are considered to have a typical associated error of ± 0.2° 2θ.The model of the zero background sample holder is 24.6mm diameter x1.0 mm thickness, manufactured by MTI corporation. The manufacturer of the hot stage sample holder is Shanghai Wei Tu Instruments and the sample plate material of hot stage sample holder is Copper. Unless indicated otherwise, the sample hasn’ t been ground before testing and the sample weight >2 mg.Peak selection method:The XRPD patterns collected were imported into MDI Jade. The measured XRPD pattern was aligned to a pattern of a sample with an internal reference to determine the absolute peak positions of the sample. The calibrated substance was Corundum (Al2O3) . The absolute peak position for corundum was calculated based on the corundum cell parameters. All peaks of the sample were extracted in a table with the accurate peak position together with the relative peak intensities. A typical error of ± 0.2°2θ in peak positions applies to this data. The minor error associated with this measurement can occur as a result of a variety of factors, including: (a) Sample preparation (e.g., sample height) ; (b) Instrument; (c) Calibration; (d) Operator (including those errors present when determining the peak locations) , and (e) The nature of the material (e.g. preferred orientation and transparency errors) .
