Methods for isolating polynucleotides encoding antibodies against lesional tissues are provided, wherein the methods comprise the steps of: (a) isolating a B cell(s) that infiltrates into a lesional tissue of interest; and (b) obtaining an antibody-encoding polynucleotide from the isolated B cell(s). The lesions may be a cancer tissue or such. Antibody genes can be obtained without depending on B cell cloning. Accordingly, it is also possible to obtain genes encoding human-derived antibodies which are difficult to clone. Genes that encode antibodies against cancer can be obtained using cancer tissues as the lesion.

Non-invasive screening of skin diseases by visible/near-infrared spectroscopy

A non-invasive tool for skin disease diagnosis would be a useful clinical adjunct. The purpose of this study was to determine whether visible / near-infrared spectroscopy can be used to non-invasively characterize skin diseases. In-vivo visible- and near-infrared spectra (400-2500 nm) of skin neoplasms (actinic keratoses, basal cell carcinomata, banal common acquired melanocytic nevi, dysplastic melanocytic nevi, actinic lentigines and seborrheic keratoses) were collected by placing a fiber optic probe on the skin. Paired t-tests, repeated measures analysis of variance and linear discriminant analysis were used to determine whether significant spectral differences existed and whether spectra could be classified according to lesion type. Paired t-tests showed significant differences (p<0.05) between normal skin and skin lesions in several areas of the visible / near-infrared spectrum. In addition, significant differences were found between the lesion groups by analysis of variance. Linear discriminant analysis classified spectra from benign lesions compared to pre-malignant or malignant lesions with high accuracy. Visible / near-infrared spectroscopy is a promising non-invasive technique for the screening of skin diseases.

The lipid casing is prepared from dipalmitoyl phosphoethanolamine (DPPE), distearoyl phosphoethanolamine (DSPE) and polyethylene glycol 1500 which are mixed, and the gas inner core is biological inert gas and further optimized as perfluoropropane gas (C3F8). According to the designed ultrasonic diagnosis target reagent disclosed by the invention, the target microbubble can penetrate through tumor vessels through intravenous injection to enter tumor tissue gaps and be specifically combined with adult vascular endothelial cells and mesenchymal cells; thus, the microbubble can effectively enrich at the tumor position, retention of the microbubble is prolonged, lesion tissues and peripheral normal tissues can be clearly distinguished through gathering imaging, and a non-linear harmonic imaging technology is utilized to achieve target molecular imaging of the liver cancer.
