[0057] "Untreated state" refers to an insert which is obtained by cast-molding the polymerizable composition in a mold and which has not been subjected to post-molding processes of extraction and / or hydration (ie has not been in contact with water or any organic solvent or any liquid after molding).
[0058] Generally, the present invention relates to cross-linked materials that are rigid in a dry state at room temperature (from about 22° C. to about 26° C.), have a high oxygen permeability and a high refractive index in a fully hydrated state, and can become softer at temperatures greater than 32° C. Such materials can be used to make inserts in embedded contact lenses to correct corneal astigmatism, presbyopia, and color blindness and to impart photochromic properties to the lenses.
[0059] The present invention is based in part on the discovery that when a polymerizable composition for making an insert comprises as two major components (i.e., in combination constituting at least about 70% by weight relative to the total weight of all polymerizable materials) (1) at least one aromatic acrylic monomer and (2) at least one silicone-containing polymerizable material, and at least one vinyl crosslinker, an insert material having a high oxygen permeability and a high refractive index and being rigid in a dry state (untreated state) at room temperature can be obtained. It is believed that by incorporating a silicone-containing polymerizable component (e.g., a vinyl monomer and / or a crosslinker) into the polymerizable composition for making the insert material (a crosslinked polymer material), the resulting insert material can have a high oxygen permeability. However, such insert materials are relatively soft and tacky at room temperature, resulting in manufacturing and handling problems associated with their softness and tackiness. Opening the mold and removing the cast-molded insert from the mold in its untreated state (i.e., "dry demolding and de-lensing") can be very difficult. It has been discovered that by incorporating an aromatic acrylic monomer and / or a crosslinking agent into the polymerizable composition used to make the insert, the resulting insert material can have a relatively high glass transition temperature (e.g., greater than 32° C.) and thus be rigid in a dry state (i.e., untreated state) at room temperature. Because of its rigid form in a dry state at room temperature, manufacturing and handling problems associated with the softness and stickiness of the insert material can be significantly reduced or eliminated.
[0060] The present invention is also based in part on the discovery that by varying the ratio of the two types of polymerizable components and the amount of the vinyl crosslinker, insert materials can be obtained having a desired set of properties (e.g., oxygen permeability, refractive index, and elastic modulus) suitable for embedded contact lenses used in different applications. The properties of embedded contact lenses can be optimized for specific applications.
[0061] In one aspect, the present invention provides an insert for embedding in a silicone hydrogel contact lens, comprising a cross-linked polymeric material comprising: (1) repeating units of said at least one silicone-containing polymerizable material (or component); (2) repeating units of at least one aromatic acrylic monomer; and (3) repeating units of at least one vinyl cross-linking agent, wherein the sum of the amounts of components (1) and (2) is at least about 70% by weight (preferably from about 75% to about 99% by weight, more preferably from about 80% to about 98% by weight, even more preferably from about 85% to 98% by weight) relative to the total weight of the cross-linked polymeric material, wherein the cross-linked polymeric material comprises: (1) repeating units of said at least one silicone-containing polymerizable material (or component); (2) repeating units of at least one aromatic acrylic monomer; and (3) repeating units of at least one vinyl cross-linking agent, wherein the sum of the amounts of components (1) and (2) is at least about 70% by weight (preferably from about 75% to about 99% by weight, more preferably from about 80% to about 98% by weight, even more preferably from about 85% to 98% by weight) relative to the total weight of the cross-linked polymeric material. The cross-linked polymer material in the dry state has a glass transition temperature of greater than about 28°C (preferably about 30°C or higher, more preferably about 32°C or higher), wherein the cross-linked polymer material has a water content of less than about 5% by weight (preferably about 4% by weight or lower, more preferably about 3% by weight or lower, even more preferably about 2% by weight or lower), an oxygen permeability of at least about 60 barrers (preferably at least about 70 barrers, more preferably at least about 80 barrers, even more preferably at least about 90 barrers), and a refractive index of at least about 1.45 (preferably at least about 1.47, more preferably at least 1.49, even more preferably at least about 1.51).

Examples of suitable methods for forming mold halves are disclosed in U.S. Patent Nos. 4,444,711; 4,460,534; 5,843,346; and 5,894,002.
[0101] Almost all materials known in the art for making molds can be used to make molds for making contact lenses. For example, polymeric materials such as polyethylene, polypropylene, polystyrene, PMMA, COC grade 8007-S10 (a transparent amorphous copolymer of ethylene and norbornene from Ticona GmbH, Frankfurt, Germany and Summit, New Jersey), etc. Other materials that allow UV transmission may be used, such as quartz glass and sapphire.
[0102] According to the present invention, an insert can be placed in a mold and a silicone-hydrogel-lens forming composition can be introduced (dispensed) into the cavity formed by the mold according to any known technique known to those skilled in the art. In a preferred embodiment, the insert is placed at a specified location on the molding surface of the female mold half; and then a specific amount of silicone-hydrogel-lens forming composition is dispensed into the female mold half with the insert thereon by a dispensing device, and then the male mold half is placed and the mold is closed. When the mold is closed, any excess unpolymerized lens forming material is pressed into an overflow area provided on the female mold half (or alternatively, on the male mold half), and the insert is immersed in the silicone-hydrogel-lens forming composition in the mold.
[0103] After the insert of the present invention is placed in the mold and the silicone-hydrogel-lens forming composition is dispensed into the mold, the closed mold containing the silicone-hydrogel-lens forming composition is then thermally or actinically cured (i.e., polymerized) (but preferably thermally initiated) to form an untreated embedded silicone hydrogel contact lens.
[0104] Actinic polymerization of the silicone-hydrogel-lens forming composition in the mold can be performed by irradiating the closed mold having the silicone-hydrogel-lens forming composition therein with UV or visible light according to any technique known to those skilled in the art.
[0105] As is well known to those skilled in the art, thermal polymerization of the silicone-hydrogel-lens-forming composition in the mold can be conveniently carried out in an oven at a temperature of from 25° C. to 120° C., and preferably from 40° C. to 100° C. The reaction time can vary within wide limits, but is suitably, for example, from 1 to 24 hours or preferably from 2 to 12 hours. It is advantageous to pre-degass the silicone-hydrogel-lens-forming composition and to carry out the copolymerization reaction under an inert atmosphere, for example, an N 2 or Ar atmosphere.
[0106] In a preferred embodiment, after the silicone-hydrogel-lens forming composition in the mold is cured in an oven to form an untreated embedded silicone hydrogel contact lens, the temperature of the oven is increased to a post-cure temperature of about 105°C or greater (preferably at least about 110°C, more preferably at least about 115°C, even more preferably at least about 120°C), and the flow rate of nitrogen through the oven is increased to a second flow rate that is at least about 1.5 times (preferably at least about 2.0 times, more preferably at least about 3.0 times, even more preferably at least about 4.0 times) the first flow rate.
[0107] The post-cure treatment step is performed by heating the lens mold having the untreated embedded silicone hydrogel contact lens therein in an oven at the post-cure temperature for at least about 30 minutes (preferably at least about 60 minutes, more preferably at least about 90 minutes, even more preferably at least about 120 minutes) under a nitrogen flow at a second flow rate through the oven.
[0108] Following the curing step and optional post-curing steps, the steps of opening the mold (i.e., separating the male mold half from the female mold half with the untreated embedded silicone hydrogel contact lens attached to one of the male and female mold halves) and delensing (i.e., removing the untreated embedded silicone hydrogel contact lens from the mold half to which the lens is adhered) are performed.
[0109] After delensing the untreated embedded silicone hydrogel contact lens, it is typically extracted with an extraction medium, as is known to those skilled in the art. The extraction liquid medium is any solvent capable of dissolving one or more of the diluent, unpolymerized polymerizable material, and oligomers in the untreated embedded silicone hydrogel contact lens. Water, any organic solvent known to those skilled in the art, or mixtures thereof, may be used in the present invention. Preferably, the organic solvent used in the extraction liquid medium is water, buffered saline, a C1-C3 alkyl alcohol, 1,2-propylene glycol, polyethylene glycol having a number average molecular weight of about 400 Daltons or less, a C1-C6 alkyl alcohol, or a combination thereof.
[0110] The extracted embedded silicone hydrogel contact lens can then be hydrated according to any method known to those skilled in the art.
[0111] The hydrated embedded silicone hydrogel contact lenses can be further subjected to other processes such as, for example, surface treatment, packaging in lens packaging with packaging solutions well known to those skilled in the art; sterilization, such as autoclaving at from 118° C. to 124° C. for at least about 30 minutes; etc.
[0112] Lens packages (or containers) are well known to those skilled in the art for autoclaving and storing soft contact lenses. Any lens package can be used in the present invention. Preferably, the lens package is a blister package comprising a base and a cover, wherein the cover is removably sealed to the base, wherein the base includes a cavity for receiving a sterile packaging solution and the contact lens.
[0113] The lenses are packaged in individual packages, sealed, and sterilized (eg, by autoclaving under pressure at about 120° C. or higher for at least 30 minutes) prior to distribution to users. One skilled in the art will readily understand how to seal and sterilize lens packages.
[0114] In another aspect, the present invention provides an embedded silicone hydrogel contact lens comprising a silicone hydrogel material and an insert of the present invention therein (as described above in this application), wherein the silicone hydrogel material is a cross-linked material having a polymer matrix and comprising repeating units of (a) at least one silicone-containing vinylic monomer and / or at least one silicone-containing vinylic cross-linker and (b) at least one hydrophilic vinylic monomer, wherein the embedded silicone hydrogel contact lens in a fully hydrated state has a water content of from about 15% to about 70% (preferably from about 15% to about 65%, more preferably from about 20% to about 65%, even more preferably from about 25% to about 60%) by weight of water when fully hydrated.
[0115] All of the various embodiments, including preferred embodiments of polymerizable compositions, silicone-containing vinylic monomers, silicone-containing vinylic cross-linkers, hydrophilic vinylic monomers, non-silicone vinylic cross-linkers, hydrophobic vinylic monomers, UV / HEVL absorbing vinylic monomers, blended vinylic monomers, inserts, RGP discs, polymeric non-reactive diluents, water swelling degree of untreated embedded silicone hydrogel contact lenses, and equilibrium water content of embedded silicone hydrogel contact lenses can be incorporated into both aspects of the present invention.

[0207] The above disclosure should enable one of ordinary skill in the art to practice the present invention. Various modifications, variations, and combinations of the various embodiments described herein are possible. To better understand the specific embodiments and advantages thereof, reference is made to the following examples. It is intended that this description and examples be considered illustrative.
[0208] Example 1
[0209] Oxygen permeability coefficient measurement
[0210] Unless otherwise specified, the oxygen permeability (Dk / t), intrinsic (or edge-corrected) oxygen permeability coefficient (Dk i or Dk c ).
[0211] Balanced moisture content
[0212] The equilibrium water content (EWC) of contact lenses was measured as follows.
[0213] The amount of water (expressed as weight percent) present in a fully hydrated hydrogel contact lens equilibrated in saline solution is determined at room temperature. After blotting the lenses with fabric, the lenses are quickly stacked and the lens stack is transferred to an aluminum pan on an analytical balance. The number of lenses used for each sample pan is typically five (5). The hydrated weight of the pan plus the lenses is recorded. Cover the pan with aluminum foil. Place the pan in a laboratory oven at 100°C ± 2°C and dry for 16-18 hours. Remove the pan plus lenses from the oven and cool in a desiccator for at least 30 minutes. Remove the individual pans from the desiccator and remove the aluminum foil. Weigh the pan plus the dried lens sample on an analytical balance. Repeat for all pans. The wet and dry weights of the lens sample can be calculated by subtracting the weight of the empty weighing pan.
[0214] Refractive index
[0215] The refractive index (RI) of the inserts was determined by means of an Abbe transmission laboratory refractometer, Reichert Abbe Mark III, at 25° C. The inserts were fully equilibrated in PBS saline solution before the measurements.
[0216] elastic modulus
[0217] The storage modulus (Young's modulus) of the insert was determined using a TA RSA-G2 DMA (Dynamic Mechanical Analyzer). The insert was cut into 3.08 mm wide strips using a Precision Concept dry lens cutter. Five thickness values were measured over a 6.5 mm gauge length. The strip was mounted on the instrument with a metal clamp. An oscillatory temperature ramp test from 10°C to 50°C was applied to the insert at a linear ramp rate of 2°C / min, and the material response to the temperature increase was monitored at a constant frequency of 1 Hz, a constant amplitude of 0.5% deformation, and a sampling rate of 10.0 pts / s. The storage modulus (E'), loss modulus (E"), and tan δ data were calculated using TRIOS software.
[0218] The elastic modulus of silicone hydrogel materials or contact lenses was determined using an MTS insight instrument. The contact lenses were first cut into 3.12 mm wide strips using a Precision Concept two-stage cutter. Five thickness values were measured over a 6.5 mm gauge length. The strips were mounted on the instrument holder and immersed in PBS (phosphate buffered saline), where the temperature was controlled at 21 ± 2°C. Typically, a 5N load cell was used for testing. A constant force and velocity were applied to the sample until the sample broke. Force and displacement data were collected using TestWorks software. The elastic modulus value was calculated using TestWorks software as the slope or tangent of the stress versus strain curve close to zero elongation in the elastic deformation region.
[0219] Glass transition temperature
[0220] The glass transition temperature (Tg) of the insert was defined as the peak value of tan δ from the dynamic temperature ramp test as described above.
[0221] Layering
[0222] Embedded silicone hydrogel contact lenses were examined for possible delamination using an Optimec instrument or optical coherence tomography (OCT).
[0223] Regardless of the evaluation method, contact lenses were staged at room temperature for at least 12 hours after autoclaving and prior to delamination studies.
[0224] After the desired rest time has been reached, the fully hydrated contact lens is placed in the "V" grid assembly of an Optimec instrument (OPTIMEC, UK, Model JCF). After the contact lens has settled under the influence of gravity, the front view of the contact lens should be carefully examined for any signs of circular patterns. Delamination appears as a circular pattern in the Optimec image.
[0225] OCT (Thorlabs Spectral Domain Optical Coherence Tomography, Model Telesto-II) can also be used to study delamination. OCT allows noninvasive imaging of contact lenses to obtain high-resolution cross-sectional images. For this purpose, after meeting the minimum grading requirements, the contact lenses are removed from their blisters and soaked in PBS solution for at least 30 minutes to reach equilibrium. Then, a cuvette with a "V" block feature is filled approximately 3 / 4 with fresh PBS solution, and the contact lens is transferred to the cuvette using a cotton swab. The lens is allowed to float freely into the "V" shape at the bottom of the cuvette, and the entire contact lens is scanned in 10-degree increments. In the OCT images, delamination appears as air pockets in the spacer surfaces of the insert and carrier.
