Description

Technical Field

[0001] The present invention belongs to the field of medicine and biotechnology engineering, and specifically relates to a class of novel fluorine-substituted vancomycin analogs and a preparation method and use thereof. Background Art

[0002] Vancomycin is an important glycopeptide antibiotic used to prevent and treat Gram-positive bacterial infections. Traditionally, vancomycin is known as the "last line of defense" in clinical practice and is used to treat serious infections that are ineffective against all antibiotics. However, with the abuse of antibiotics, more and more vancomycin-resistant bacteria have emerged. Introducing new pharmacophores and expanding the structure of new vancomycin analogs are important means to solve the problem of vancomycin resistance.

[0003] Fluorine is an important pharmacophore. Introducing fluorine-containing functional groups into drug molecules can usually change the lipophilicity, metabolic stability and pharmacokinetic properties of candidate drugs, and plays an irreplaceable role in the field of medicinal chemistry. Currently, more than 300 fluorine-containing drugs have been approved by the FDA, accounting for more than 20% of all drugs. Therefore, introducing fluorine-containing functional groups into vancomycin, changing the biological activity of vancomycin or changing the potential target of action, and obtaining new vancomycin analog drugs based on this is an important breakthrough.

[0004] Although the art has been trying to introduce fluorine-containing functional groups into drug molecules using various methods, for highly functionalized complex compounds such as vancomycin, no efficient and highly selective methods for introducing fluorine-containing functional groups have been successfully developed so far, nor have fluorine-substituted vancomycin analogs with advantages such as high activity or high reactivity been developed.
