SgRNA sequence specifically targeting chicken satb1 gene and immunization application thereof

By targeting the sgRNA sequence of the chicken SATB1 gene and editing it using the CRISPR-Cas9 system, SATB1 gene mutant cells were constructed, solving the problem of insufficient immune response in chickens during intensive farming. This achieved an effective approach to immune regulation and breeding, enhancing the disease resistance of chickens.

Abstract

The application discloses an sgRNA sequence targeting a chicken SATB1 gene, and a CRISPR-Cas9 system using the sgRNA can effectively mutate the SATB1 gene in chicken macrophages, so that the expression level of the SATB1 is reduced. The application also discloses application of the SATB1 gene in regulating immune responses in chicken macrophages. When the chicken macrophages with the mutated SATB1 gene are subjected to a lipopolysaccharide stimulation, the expression of key genes of immune responses is significantly inhibited, which indicates that the SATB1 positively regulates the immune response capacity of the chicken macrophages. Therefore, the application provides the sgRNA for effectively editing the chicken SATB1 gene, and discloses a new function of the SATB1 gene, which provides a new model for research on a chicken immune response mechanism, and has a good application prospect in regulating chicken immune responses and cultivating superior breeds.
