Abstract
Description
[Technical field]
[0001] The present invention relates to a bactericidal / virucidal composition and a method for producing the same, and more particularly to a bactericidal / virucidal composition which is particularly effective in inactivating norovirus and has excellent stability in a dilution solution, and a method for producing the same. [Background technology]
[0002] Norovirus, which has become a problem in recent years, is the cause of many food poisoning incidents every year, but it is difficult to take food hygiene measures against it because it has low drug sensitivity and is difficult to inactivate. Since norovirus is a non-enveloped virus and has a protein shell called a capsid, the inactivation effect of alcohol or cationic surfactants is not sufficient, and treatment with sodium hypochlorite is recommended. However, sodium hypochlorite has the drawback of being difficult to use because it is corrosive to metals and has a pungent odor.
[0003] As an example of such a disinfectant that can be used in place of sodium hypochlorite, there is disclosed an acidic hydrogen peroxide solution having a concentration of about 0.5% when used, which further contains 0.1 to 5.0% of at least one acid compound, such as phosphoric acid and / or a phosphonate having 1 to 5 phosphonic acid groups, and 0.02 to 5% of at least one anionic surfactant (Patent Document 1). [Prior art documents] [Patent documents]
[0004] [Patent Document 1] Patent No. 3350526 Summary of the Invention [Problem to be solved by the invention]
[0005] However, although the hydrogen peroxide solution in Patent Document 1 requires an anionic surfactant as an essential component, the examples only use alkylbenzenesulfonic acid. Alkylbenzenesulfonic acid is a strong acid with a pKa of -0.5, and may corrode metal manufacturing equipment. In addition, it is a component that is a concern for the safety of workers during manufacturing, as it is included in the list of hazardous substances to be designated in the future. If it becomes a hazardous substance, it will have to be stored in a locked storage facility, making it difficult for users to manage. Therefore, it is preferable to use a neutralized anionic surfactant.
[0006] On the other hand, in light of recent considerations of the impact on the environment, it is desirable to dilute concentrated chemicals before use in order to reduce transportation energy and containers, and it is also desirable to minimize the amount of chemicals released into the environment.
[0007] Under these circumstances, an object of the present invention is to provide a bactericidal / virucidal composition that has excellent virus inactivation effect and also has excellent stability in a dilution solution, and a method for producing the same. [Means for solving the problem]
[0008] However, in light of these circumstances, the inventors conducted extensive research and discovered that by using an alkane sulfonate in combination with a surfactant having an alkylene oxide structure, it is possible to prepare a hydrogen peroxide preparation that has an inactivating effect particularly on norovirus, and that remains stable even when diluted to low concentrations, suppressing cloudiness and separation.
[0009] That is, the present invention has the following aspects. [1] A bactericidal / virucidal composition comprising the following components (A) to (E): (A) Hydrogen peroxide 0.005 to 6% by mass (B) Alkanesulfonate: 0.01 to 30% by mass (C) 0.01 to 20% by mass of a surfactant having an alkylene oxide structure (D) Acid agent 0.01~20% by mass (E)Water [2] The bactericidal / virucidal composition according to [1], wherein the virus in the bactericidal / virucidal composition comprises a norovirus-surrogate virus of both feline calicivirus and murine norovirus. [3] The fungicidal / virucidal composition according to [1] or [2], wherein when the concentration of (B) when diluted for use is less than 0.15% by mass, the mass ratio of (B) / (C) is 1 to 30. [4] The fungicidal / virucidal composition according to any one of [1] to [3], wherein the mass ratio of (B) / (D) is 5 or less. [5] The fungicidal / virucidal composition according to any one of [1] to [4], further comprising a chelating agent in an amount of 0.01 to 5 mass %. [6] The bactericidal / virucidal composition according to any one of [1] to [5], further comprising a corrosion inhibitor. [7] The bactericidal / virucidal composition according to any one of [1] to [6], which has a pH of 5 or less. [8] A method for producing the fungicidal / virucidal composition according to any one of [1] to [7], comprising the steps of: A step of preparing a composition precursor containing the (A) hydrogen peroxide, the (B) alkane sulfonate, the (C) surfactant having an alkylene oxide structure, and the (D) acid agent; and causing the composition precursor to contain the (E) water. Effect of the Invention
[0010] The bactericidal / virucidal composition of the present invention has excellent virus inactivation effect even when diluted, and also has excellent stability in the diluted solution. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0011] The present invention will be described below based on examples of the mode for carrying out the present invention. However, the present invention is not limited to the embodiment described below.
[0012] In the present invention, when expressed as "X to Y" (X and Y are any numbers), unless otherwise specified, it includes the meaning of "X or more and Y or less", as well as "preferably larger than X" or "preferably smaller than Y". Furthermore, when it is expressed as "X or more" (X is any number) or "Y or less" (Y is any number), it also includes the meaning that "it is preferably greater than X" or "it is preferably less than Y". Furthermore, "x and / or y (x and y are optional configurations)" means at least one of x and y, and means the following three cases: x only, y only, and x and y.
[0013] A fungicidal / virucidal composition according to one embodiment of the present invention contains the following components (A) to (E): (A) Hydrogen peroxide 0.005 to 6% by mass (B) Alkanesulfonate: 0.01 to 30% by mass (C) 0.01 to 20% by mass of a surfactant having an alkylene oxide structure (D) Acid agent 0.01~20% by mass (E)Water
[0014] The bactericidal / virucide composition of this embodiment is a composition comprising components constituting a bactericide and / or a virucide. A bactericide means an agent having an effect of killing bacteria (bactericidal), and a virucide means an agent having an effect of killing viruses (virucide). Such a composition is particularly excellent at killing norovirus, which is difficult to inactivate.
[0015] In this specification, "sterilization" includes not only the effect of killing bacteria (sterilization), but also the effect of removing bacteria (bactericidal) and the effect of suppressing bacterial growth (antibacterial). Furthermore, "viricidal" includes not only the effect of killing viruses (viricidal), but also the effect of removing viruses (viricidal) and the effect of suppressing viral growth (antiviral).
[0016] As for human norovirus (hNoV), which is a type of norovirus, it is still impossible to artificially culture it, so we must rely on evaluation using alternative viruses that belong to the same Caliciviridae family and can be cultured. Representative alternative viruses to norovirus are feline calicivirus (FCV) and murine norovirus (MNV). The former has been used as an alternative virus since the 1990s, and although it is considered to have a single serotype (two types in recent years), like hNoV, it has been found to have diversity in its structural proteins, and its sensitivity to general-purpose disinfectants is not consistent. On the other hand, MNV was first reported in 2003, and many MNVs have tended to be detected from mice that have been kept under special environments, such as in laboratory animal facilities, and the MNVs that have been isolated and reported so far have low molecular biological diversity.
[0017] Furthermore, inactivation tests of various disinfectants using FCV or MNV with infectivity titers as an indicator, evaluation tests of survival in the environment, and tests that quantitatively evaluate the genetic content of these tests, it has been shown that the results of surrogate viruses and hNoV do not necessarily match. Therefore, since it is desirable to conduct evaluation tests using both FCV and MNV viruses rather than conducting evaluation tests using "one or the other" of the viruses, in this embodiment, evaluation tests of norovirus were conducted using surrogate viruses for both FCV and MNV, and the virus inactivation effect was obtained.
[0018] Each of the raw material components of the fungicidal / virucidal composition of this embodiment (hereinafter, may be referred to as "the composition") will be described below.
[0019] <(A) Hydrogen peroxide> The hydrogen peroxide of the component (A) is used to maintain the performance of sterilization and / or viricide (hereinafter, sometimes referred to as "viricide, etc."). The content is 0.005 to 6 mass% of the entire composition, preferably 0.01 to 6 mass%, and more preferably 0.01 to 5.5 mass%. That is, in this embodiment, sufficient performance of viricide, etc. can be exhibited with an amount smaller than the amount conventionally required for exhibiting performance of viricide, etc. If the content is too low, the performance of viricide, etc. tends to decrease, and conversely, if the content is too high, it is difficult to obtain the effect of corrosion inhibition performance on metals and a pungent odor tends to be generated.
[0020] <(B) Alkanesulfonate> The alkanesulfonate salt of component (B) is used to obtain an inactivating effect against both FCV and MNV at a practical concentration. The content is 0.01 to 30% by mass, preferably 0.1 to 20% by mass, and more preferably 0.1 to 15% by mass, based on the total mass of the composition. If the content of component (B) is too high, the composition tends to be sticky, and conversely, if the content of component (B) is too low, the effect against viruses and bacteria tends to be inferior.
[0021] As mentioned above, since it is almost impossible to artificially culture human norovirus (hNoV) even now, it is desirable to carry out evaluation using both feline calicivirus (FCV) and murine norovirus (MNV), which are alternative viruses that can be artificially cultured. Therefore, when using a neutralized anionic surfactant, the present inventors first evaluated a wide variety of anionic surfactants that have been proposed in the past against both FCV and MNV. As a result, they found that only a limited number of surfactants are effective at practical concentrations, and that only the alkane sulfonate salt of component (B), alkylbenzene sulfonic acid or its salt, and alkyl sulfate salt are effective. Among them, they found that the alkane sulfonate salt of component (B) can solve the problem of the present invention, especially dilution stability.
[0022] Specifically, when the present composition is diluted and used, the anionic surfactant tends to precipitate calcium or magnesium salts of the anionic surfactant due to the hardness components of tap water, resulting in precipitation or separation. Compounds having alkylene oxides are often used as precipitation inhibitors (solubilizers) for this purpose. However, it is known that the anionic surfactant forms mixed micelles with these compounds. The formation of mixed micelles suppresses the precipitation of the anionic surfactant against the hardness components, but on the other hand, the virucidal and bactericidal effects are reduced. Since this effect is more pronounced at lower anionic surfactant concentrations, in order to use the composition at low drug concentrations, it is preferable to use an anionic surfactant that is easily solubilized by these compounds as solubilizers and whose virucidal and bactericidal effects are less likely to be reduced. For example, anionic surfactants having an alkylene oxide structure have a dilution stabilizing effect, so that no cloudiness or separation occurred during dilution even without the use of other surfactants having an alkylene oxide structure, but they have no effect on norovirus.
[0023] Therefore, the inventors have found that, among the anionic surfactants that are effective against both FCV and MNV at practical concentrations, alkanesulfonates are the most excellent anionic surfactants that maintain their effectiveness against both FCV and MNV at low anionic surfactant concentrations while suppressing precipitation upon dilution with a small amount of a surfactant having an alkylene oxide structure.
[0024] <(C) Surfactants with an alkylene oxide structure> Component (C) is used to improve the problem that the alkane sulfonate of component (B) becomes cloudy when diluted with water (particularly water containing hardness components such as tap water). Although compounds having an ethylene oxide structure are known for this purpose, the present inventors have found that a combination of an alkane sulfonate as component (B) and a surfactant having an alkylene oxide structure as component (C) maintains virucidal and bactericidal effects and is less likely to become cloudy when diluted with water.
[0025] The surfactant having an alkylene oxide structure, component (C), is not particularly limited to being nonionic or ionic, since a dilution stabilizing effect can be obtained regardless of whether it is nonionic or ionic. However, a nonionic surfactant or an anionic surfactant is preferable as a component that does not form a complex with component (B).
[0026] Examples of the nonionic surfactant having an alkylene oxide structure of component (C) include polyoxyalkylene alkyl ether, polyoxyethylene alkenyl ether, polyoxyethylene alkylphenyl ether, polyoxyalkylene alkenyl ether, alkylamine alkylene oxide adduct, polyoxyalkylene block copolymer, polyoxyethylene polyoxypropylene glycol, polyoxyalkylene alkyl ether phosphoric acid, polyoxyalkylene alkyl ether acetate, polyoxyalkylene alkyl sulfate, and salts thereof. Among them, polyoxyethylene alkyl ether, polyoxyethylene alkyl ether phosphoric acid, polyoxyethylene alkyl ether acetate, and polyoxyethylene lauryl ether sulfate are preferred. These can be used alone or in combination of two or more.
[0027] As for the alkylene oxide structure of component (C), ethylene oxide is more soluble than other alkylene oxides such as propylene oxide, and therefore it is preferable from the standpoint of dilution stability that the alkyl oxide in the composition consists solely of ethylene oxide.
[0028] The content of component (C) is from 0.01 to 20 mass %, preferably from 0.05 to 15 mass %, and more preferably from 0.05 to 10 mass %, based on the total mass of the composition.
[0029] As mentioned above, the usable concentration of the (C) component is limited. If the content is too low, it tends to be difficult to suppress the clouding that occurs when a high-concentration composition precursor is mixed with water to prepare a composition (undiluted solution), and conversely, if the content of the (C) component is too high, the effect of the (B) component, such as virucidal effect, tends to be too weak. This is more noticeable when the concentration of the (B) component during use is low, and when the concentration of the (B) component during use is less than 0.15% by mass, it is preferable to use the (B) / (C) ratio in the range of 1 to 30.
[0030] <(D) Acid Agents> The acid agent of component (D) is used to enhance the bactericidal and virucidal activity of component (B). Component (D) is not particularly limited, but examples thereof include phosphoric acid (H3PO4) and phosphonates (phosphonic acids) having 1 to 5 phosphonic acid groups, among which phosphoric acid, citric acid, and sulfamic acid are preferred, and phosphoric acid is more preferred. These may be used alone or in combination of two or more kinds.
[0031] The content of component (D) is not particularly limited, but is preferably 0.01 to 20% by mass, more preferably 0.1 to 15% by mass, and even more preferably 0.1 to 10% by mass, based on the total mass of the composition. If the content of component (D) is too low, the effect against viruses and bacteria tends to be weakened, and conversely, if the content of component (D) is too high, corrosion of metals tends to occur. It is preferable to use the component (B) / (D) in the range of 5 or less.
[0032] <(E)Water> Examples of the water of the component (E) include pure water, ion-exchanged water, soft water, hard water, distilled water, tap water, etc. These can be used alone or in combination of two or more. In particular, when the water contains hardness components such as tap water, the dilution stability, which is one of the effects of the present invention, is more effectively exhibited. The present composition is obtained by adding water to the composition precursor, and the addition of water reduces the effect on metals, and since the amount of active ingredients is small, it is easy to wipe off after application to the surface to be disinfected.
[0033] The above "water" refers to the total amount of water contained in the form of water of crystallization or an aqueous solution derived from each component constituting the composition, and water added from the outside, and is contained in such a way that the total amount of the composition is 100% by mass.
[0034] <Optional ingredients> The present composition is prepared using the above components (A) to (E) as essential components, but may further contain various optional components as appropriate. Examples of such optional components include chelating agents, solvents other than water, corrosion inhibitors, pH adjusters, fragrances, thickeners, etc. These may be used alone or in combination of two or more. The content of such optional components may be contained within a range that does not impair the effects of the present invention, and is, for example, preferably 15% by mass or less, more preferably 10% by mass or less, and even more preferably 5% by mass or less, and may not be contained.
[0035] The chelating agent can be used as a stabilizer for hydrogen peroxide, which is the component (A). Examples of the chelating agent include 1-hydroxyethane-1,1-diphosphonic acid, salicylic acid, sodium aminotrimethylene phosphonate, potassium aminotrimethylene phosphonate, sodium 1-hydroxyethylidene-1,1-diphosphonate, potassium 1-hydroxyethylidene-1,1-diphosphonate, sodium 2-phosphonobutane-1,2,4-tricarboxylate, potassium 2-phosphonobutane-1,2,4-tricarboxylate, sodium ethylenediaminetetramethylene phosphonate, potassium ethylenediaminetetramethylene phosphonate, sodium diethylenetriaminepentamethylene phosphonate, potassium diethylenetriaminepentamethylene phosphonate, trisodium nitrilotriacetate, tripotassium nitrilotriacetate, and hyaluronate. Examples of the hydroxyethyliminodiacetate include disodium hydroxyethyliminodiacetate, dipotassium hydroxyethyliminodiacetate, trisodium methylglycinediacetate, tripotassium methylglycinediacetate, tetrasodium ethylenediaminetetraacetate, tetrapotassium ethylenediaminetetraacetate, trisodium hydroxyethylethylenediaminetriacetate, tripotassium hydroxyethylethylenediaminetriacetate, tetrasodium L-glutamate-N,N-2acetate, tetrapotassium L-glutamate-N,N-2acetate, tetrasodium L-aspartate-N,N-2acetate, tetrapotassium L-aspartate-N,N-2acetate, pentasodium diethylenetriaminepentaacetate, pentpotassium diethylenetriaminepentaacetate, disodium tartrate, dipotassium tartrate, sodium potassium tartrate, etc. Among these, 1-hydroxyethane-1,1-diphosphonic acid is preferred from the viewpoints of economy and stability. These can be used alone or in combination of two or more.
[0036] In addition, since the present composition has stability in the original solution and in the diluted solution, it does not need to contain a chelating agent, but it may contain a chelating agent from the viewpoint of further stabilizing the component (A). When a chelating agent is contained, the content of the chelating agent in the entire present composition is preferably 0.01 to 10 mass%, more preferably 0.05 to 5 mass%, and even more preferably 0.05 to 3 mass%.
[0037] Examples of the solvent other than water include lower alcohols having 1 to 5 carbon atoms, such as ethanol and isopropanol; lower aliphatic ketones, such as acetone and methyl ethyl ketone; polyhydric alcohols having 2 to 5 carbon atoms, such as 1,3-butylene glycol, propylene glycol, triethylene glycol, hexylene glycol, and glycerin; ethylene glycol ethers, such as ethylene glycol monomethyl ether, ethylene glycol monoethyl ether, ethylene glycol monopropyl ether, and ethylene glycol monobutyl ether; diethylene glycol ethers, such as diethylene glycol monomethyl ether, diethylene glycol monoethyl ether, diethylene glycol monopropyl ether, and diethylene glycol monobutyl ether; glycol ethers, such as propylene glycol monomethyl ether, propylene glycol monopropyl ether, and dipropylene glycol monomethyl ether. Among these, propylene glycol monopropyl ether is preferred. These can be used alone or in combination of two or more. A mixed solvent of the organic solvent and the water can also be used.
[0038] The corrosion inhibitor is not particularly limited, but is preferably a corrosion inhibitor for copper or copper alloys, and is preferably at least one selected from the group consisting of triazole or its derivatives, benzimidazole or its derivatives, and benzothiazole or its derivatives. Examples of such corrosion inhibitors include benzotriazole, tolyltriazole, 5-dodecylbenzotriazole, 5-octylbenzotriazole, 5-hexylbenzotriazole, 5-butylbenzotriazole, 5-methylbenzotriazole, carboxybenzotriazole (4-carboxybenzotriazole, 5-carboxybenzotriazole), carboxybenzotriazole methyl ester, carboxybenzotriazole butyl ester, carboxybenzotriazole hexyl ester, benzotriazole octyl ester, dicarboxypropylbenzotriazole, benzimidazole, 2-mercaptobenzimidazole, thiazolylbenzimidazole, thiabendazole, benzothiazole, 2-methylbenzothiazole, 2-aminobenzothiazole, and 2-mercaptobenzothiazole. Benzotriazole is particularly preferred. These can be used alone or in combination of two or more.
[0039] When the corrosion inhibitor is used, its content is preferably 0.001 to 5 mass % based on the total mass of the composition, more preferably 1 mass % or less, and further preferably none is contained. If the content of the corrosion inhibitor is too high, the balance with other components becomes poor, and the stability during storage tends to deteriorate.
[0040] <Bactericidal / virucidal agent composition> This composition (bactericidal / virucidal composition) can be prepared by using the above-mentioned essential components (A) to (E) and optional components contained as necessary, according to a normal method for preparing a bactericide. Alternatively, a composition precursor can be prepared using components (A) to (D) and, if necessary, optional components, and then the composition (undiluted solution) can be prepared by adding water (E) to the precursor. The composition (undiluted solution) can then be diluted usually 1 to 200 times, preferably 10 to 150 times, and more preferably 30 to 100 times to obtain the composition (diluted solution), from which a bactericide and / or virucidal agent can be produced.
[0041] In order to prevent decomposition of (A) hydrogen peroxide, the composition thus obtained preferably has a pH of 5 or less at 25° C., more preferably a pH of 1.0 to 4.5, and even more preferably a pH of 1.5 to 4. If the pH is too low, metal corrosion tends to occur, and if the pH is close to neutral, stability decreases.
[0042] The pH of the composition precursor at 25°C is preferably 5 or less, more preferably pH 0.5 to 4.5, and even more preferably pH 1.0 to 3.
[0043] The resulting composition, whether neat or diluted, can be used as a bactericide and / or virucidal agent (such as a virucide).
[0044] This composition (and the viricide, etc.) does not have a pungent odor like peracetic acid or sodium hypochlorite, and has little effect on plastics such as acrylic and polycarbonate, so it can be used to wipe down target surfaces.
[0045] Since the composition (and the viricide, etc.) contains a small amount of active ingredient, the composition can be easily wiped off after being applied or sprayed onto a target surface.
[0046] Furthermore, the composition (and viricide, etc.) is effective in inactivating non-enveloped viruses even though it contains a small amount of active ingredient and is at a low concentration, and furthermore, is capable of inactivating non-enveloped viruses even in the presence of organic soil.
[0047] Disinfection of target surfaces using the composition (and viricides, etc.) can be carried out by the following method.
[0048] (1) The composition (and the virucide, etc.) can be impregnated into a nonwoven fabric to make a sanitary material, which can be wiped onto a target surface to disinfect it. Examples of places where disinfection can be performed include medical facilities, nursing care facilities, food factories, coffee shops, restaurants, hotels, pubs, schools (school lunches), central kitchens, and supermarket backyards. Examples of surfaces to be disinfected include countertops, refrigerators, and storage units, as well as tables, desks, chairs, handrails on stairs and corridors, doorknobs at entrances and exits, toilets and toilet seats, water faucets, floors, walls, and other hard surfaces. In addition, sanitary materials using the composition (and the virucide, etc.) can also be used as hand towels, wet tissues, etc.
[0049] Examples of raw materials for the nonwoven fabrics constituting the sanitary materials include cellulosic materials such as cotton, protein-based materials such as wool or silk, and chemically polymerized materials such as rayon, polyester, and acrylic. Among these, cellulosic materials and chemically polymerized materials are preferred.
[0050] (2) The present composition (and the bactericide / virucide) can be used in any suitable manner other than as described above. For example, it is sprayed onto the target surface with a spray or the like, and after a specified time, it is left to stand for about 30 seconds to 5 minutes, and then allowed to dry naturally or, if necessary, is rinsed with water and then dried. Specifically, the present composition (and the virucide, etc.) is dispensed at about 6 to 12 mL / m each time it is used using a dedicated dispenser that contains the present composition (and the virucide, etc.). 2 Disinfection can be achieved by spraying the target surface at a rate of
[0051] The composition (and the viricide, etc.) can be provided in a plastic container, a container with a pump, a pouch, a tube, etc. Also, it can be provided in an amount equivalent to that for one-time use, individually packaged for portability. EXAMPLES
[0052] The present invention will be described in more detail below with reference to examples. However, the present invention is not limited to the following examples as long as it does not depart from the gist of the present invention.

[0067] From the evaluation results in Tables 1 and 2 above, it was found that all of Examples 1 to 12 had excellent inactivation effects against both FCV and MNV viruses, and also had excellent dilution solution stability. In contrast, Comparative Examples 10 to 12, which did not use any of the components (B) to (D), were poor in dilution stability. In Comparative Examples 1 to 9, which used an anionic surfactant other than the alkane sulfonate salt (component (B)), it was found that they did not have an inactivating effect on either the FCV or MNV virus, or were poor in dilution stability. In Comparative Example 13, the surfactant not having an alkylene oxide structure did not show a dilution stabilizing effect. Moreover, it has been confirmed that Examples 1 to 12 contain 0.07 to 6% by mass of (A) hydrogen peroxide at the time of use, and therefore have a bactericidal effect against Escherichia coli and Staphylococcus aureus. [Industrial Applicability]
[0068] The fungicidal / virucidal composition of this embodiment not only has excellent FCV inactivation and MNV inactivation effects, but also has excellent dilution solution stability, making it possible to maintain the virus inactivation effect stable over time and to reduce adverse effects on the environment and materials.
