Abstract
Description
[Technical field]
[0001] CROSS-REFERENCE TO RELATED APPLICATIONS This application claims priority to U.S. Provisional Application No. 63 / 299,678, filed January 14, 2022, the entire contents of which are incorporated by reference in their entirety. [Background technology]
[0002] Prostate cancer is the second leading cause of cancer-related deaths in men in Western countries (Damber, JE and Aus, G. Lancet (2008) 371:1710-1721). Numerous studies have shown that the androgen receptor (AR) is not only important in the development of prostate cancer, but also in the progression of the disease to castration-resistant state (Taplin, ME et al. J. Clin. Oncol. (2003) 21:2673-8, and Tilley, WD et al. Cancer Res. (1994) 54:4096-4102). Therefore, effective inhibition of human AR remains one of the most effective therapeutic approaches to the treatment of advanced metastatic prostate cancer.
[0003] Spinal-bulbar muscular atrophy (SBMA), or Kennedy disease, is an X-linked recessive neuromuscular disorder. The primary symptoms are weakness and atrophy of bulbar and limb muscles due to reduced motor degeneration of the brainstem and spinal cord along with transient muscle involvement. At onset, patients often present with limb weakness, spasms, tremors, and fasciculations, especially prominent in the face and tongue. Dysarthria is also common with hypernasality, laryngospasm, and swallowing dysfunction, often resulting in aspiration pneumonia as the disease progresses. More than half of patients die from respiratory infections. The disease is caused by a CAG expansion in exon 1 of the androgen receptor (AR), and androgens are required for pathogenesis. Currently, there is no cure for Kennedy disease. [Prior art documents] [Non-patent literature]

[0162] The amorphous form was prepared by heat-cool TGA using the free form, which crystallized to form N after exposure to high humidity.
[0163] Table 2 outlines the screening conditions for the polymorphs discussed above. [Table 2]
[0164] Preparation of Form C Form C was prepared using the following procedure: 200 mg of the free form was weighed into a 4 mL glass vial. Under stirring at 25° C., 200 μL of acetone was added to the vial. Approximately 3 mg of Form C seeds were added to the solution. After stirring for 10 days at 25° C., the solids were collected by suction filtration and then dried under vacuum at 25° C. for approximately 4 hours. The dried solids were re-equilibrated in 100 μL of acetone under temperature cycles from 5° C. to 50° C. at a heating / cooling rate of 0.1° C. / min. After three cycles, the solids were collected by suction filtration at 5° C. and then dried under vacuum at 50° C. for approximately 2 hours. 136 mg of Form C was obtained as an off-white solid in 67% yield.

[0166] Polymorphic Interrelationships The interconversion relationships of the identified polymorphs are outlined in FIG.
[0167] The relative stability of anhydrous forms A, C, H, and J was investigated by competitive equilibrium experiments. Form C was obtained in all selected solvents at both 25° C. and 50° C., except for MeOH, where form B was obtained. An intermediate crystalline form, form M, was obtained from methanol as a wet cake and readily further transformed to form B once exposed to air. The presence of intermediate form M influenced the competitive equilibrium experiments, which cannot reflect the actual thermodynamic relationship between anhydrous forms in methanol. In the other selected solvent systems, form C is the most stable form.
[0168] The relative stability of the anhydrate forms C, K, and L was investigated by competitive equilibrium experiments. Form C was obtained in IPAc at both 25° C. and 50° C.
[0169] Thus, Form C is an anhydrate that is thermodynamically stable at both 25°C and 50°C.
[0170] The relative stability of anhydrous form C, hydrate form B, and hydrate form F was investigated by competitive water activity experiments. The relative stability of anhydrous form C and hydrate form N was also investigated by water activity experiments in acetone / water solvent mixture (aw=0.95). The results show that form C is the thermodynamically stable form in the entire range of water activity (0≦aw≦1). Thus, anhydrous form C is the optimal polymorph of 3-(5-(2-hydroxy-2-methylpropoxy)-6-methylpyrazin-2-yl)-1H-indole-7-carbonitrile free form.
[0171] competitive equilibrium To determine the relative stability of the anhydrates, competitive equilibrium experiments were performed in different solvent systems.
[0172] Approximately 2 mg of each anhydride was added to 0.3–0.4 mL of a saturated solution of the selected solvent. The resulting suspensions were stirred at 25 °C for 3–10 days, respectively. The solid portion (wet cake) was isolated by centrifugal filtration and examined by XRPD. The results are shown in Table 3. [Table 3-1] [Table 3-2]
[0173] Physicochemical characteristics The physicochemical characteristics of Form C are summarized in Table 4. [Table 4]
[0174] Bulk Stability The bulk stability of Form C was evaluated over one week at 25° C. / 92% RH in an open container, 40° C. / 75% RH in an open container, and 60° C. in a closed container. Form C is physically and chemically stable after stress under these conditions, as shown in Table 5. [Table 5]
[0175] The stability of Form C was further investigated under the following conditions, and the results are summarized in Tables 5A-5F. [Table 6] [Table 7] [Table 8] [Table 9] [Table 10] [Table 11]
[0176] As can be seen above, Form C is stable under varying conditions of temperature and relative humidity.
[0177] solubility Anhydrous Form C (equivalent to 8 mg of free form) was weighed into a 20 mL glass vial. 4 mL of solubility medium was added. The resulting suspension was stirred at 400 rpm at 37° C. Some of the suspensions were removed at 2 hours and 24 hours, respectively. They were then centrifuged at 14,000 rpm for 5 minutes at 37° C. The supernatant was analyzed for solubility and pH by HPLC and pH meter, respectively. The residual solid (wet cake) was characterized by XRPD to determine the physical form. [Table 12]
[0178] The results of additional solubility studies are shown in Table 7. [Table 13]
[0179] These data indicate that Form C has high solubility in acetone, THF, 2-MeTHF, and MEK, but low solubility in water, n-heptane, and MCH. The use of water and THF was avoided in the following studies, since polymorph screening reports indicate that potential Forms A / H (anhydrate) or Form I (THF solvate) may exist in water or THF systems. To achieve high yields in the crystallization process, additional solubilities were measured in acetone, 2-MeTHF, and MEK in combination with anti-solvents n-hep and MCH at 25° C., as shown in Table 8. [Table 14]
[0180] These data show that in MEK, 2-MeTHF, and acetone solvents, the solubility decreased as more antisolvent was added.
[0181] Additional solubility studies were carried out in 2-MeTHF in combination with the antisolvent n-hep at 50° C., as shown in Table 9. [Table 15]
[0182] Solution state chemical stability was performed in acetone, MEK, and 2-MeTHF solvent systems at 50° C. and 55° C. The results are summarized in Table 10. The data show that the compound was stable after 48 hours of storage in both acetone and 2-MeTHF systems. The purity of the compound decreased from 99.43% to 98.27% after 48 hours of storage at 55° C. in the MEK solvent system. [Table 16]
[0183] 2-MeTHF / n-hep was selected as a potential solvent system for crystallization. Solution state chemical stability was performed at higher temperatures (60°C and 70°C) in the 2-MeTHF solvent system. Data shows that the compound was stable after 48 hours in 2-MeTHF at 60°C and 70°C. Solution state chemical stability was performed at 70°C in the 2-MeTHF / n-hep=1v / 1v solvent system, and data shows that the compound was stable after 48 hours in the system at 70°C. [Table 17]
[0184] Antisolvent crystallization According to the solubility and chemical stability data, anti-solvent crystallization experiments were carried out in acetone / n-hep and 2-MeTHF / n-hep systems as shown below. [0186] Preparative anti-solvent crystallization in acetone / n-hep: 1. Dissolves in 9v acetone at 50℃. 2.Adjust to 45℃. 3. Seeds (10mg) 4. Stir at 45°C for 0.5 hours (thin suspension). 5.Adjust to 42℃. 6. Stir at 42°C for 1 hour (suspension with more solids) 7. Adjust to 25℃ in about 3.5 hours. 8. Take sample for XRPD-1 and PLM-1 analysis. 9. Load 27v n-hep at 25°C for 4.5 hours 10. Stir at 25°C for 12 hours. 11. Take samples for XRPD-2, PLM-2, and Assay-1 analysis. 12. Stir at 25°C for 6 hours. Adjust to 5°C in 13.4 hours. Stir for 14 hours at 14.5°C. 15. Take samples for XRPD-3, PLM-3, and Assay-2 analysis. 16. Filter and wash cake with 2v acetone / n-hep=1v / 3v. 17. Dry the wet cake under vacuum at 50°C. 18. Get materials.
[0187] Prior to the addition of n-hep, the XRPD indicated Form C. After the addition of n-hep, the XRPD reflected Form C even after holding at 25° C. (approximately 17% ML loss). Before filtration, the ML loss was approximately 12%. The dried product had a purity of 99.61% with 0.03% residual acetone and 0.04% residual n-hep.
[0188] Process optimization for 2-MeTHF / n-heptane According to the preliminary anti-solvent crystallization results in 2-MeTHF / n-hep and acetone / n-hep, 2-MeTHF / n-hep system was determined as the main crystallization system. Two use test experiments were carried out, achieving 11 g of crystallization product. The results can be seen in Table 12. [Table 18]
[0189] Using the results of the above experiment, an 11 g scale-up experiment was carried out using the following procedure. 1. Dissolve in 8v 2-MeTHF at 50°C. 2. Load 1.5v n-hep at 50℃ for 1 hour 3. Seeds (110mg) 4. Stir at 50°C for 1.5 hours. 5. Load 22.2v n-hep at 50℃ for 7 hours 6. Take a sample for XRPD-1. 7. Stir at 50°C for 15 hours. 8. Take sample for XRPD-2. 9. [0190] After addition of n-hep, XRPD shows Form C with additional peaks. After holding at 50° C. for 15 and 35 hours, XRPD again showed Form C with additional peaks (approximately 4% ML loss). Since additional peaks were observed in the XRPD, the process was further optimized using the following procedure. 1. Dissolve in 8v 2-MeTHF at 50°C. 2. Load with 2v n-hep at 50℃ for 1-3 hours 3. Seeds (150mg) 4. Stir at 50℃ for 1 to 18 hours (thin suspension) 5. Charge with 2v n-hep at 50°C for 2-3 hours. 6. Take a sample for XRPD analysis. 7. Stir at 50℃ for 2 to 4 hours. 8. Take a sample for XRPD analysis. 9. Charge with 20v n-hep at 50°C for 6-8 hours. 10. Stir at 50°C for 2 hours. Adjust to 25℃ in 11.5 hours 12.Stir at 25°C for 2 hours. 13. Take a sample for XRPD analysis. 14. Stir at 25°C for 6 to 15 hours. 15. Filter and wash the cake with 2v 2-MeTHF / n-hep=1v / 3v. 16. Dry the wet cake under vacuum at 50°C. 17. Materials obtained
[0191] This procedure was performed twice and XRPD both times showed Form C. Purity was measured at 99.24% and 97.86%.
[0192] Hygroscopic The hygroscopicity of Form C was evaluated by dynamic vapor sorption (DVS) test at 25° C. Form C is non-hygroscopic below 90% relative humidity (RH). Form C absorbs about 0.2% water at 95% RH at 25° C. After the DVS test, the obtained sample was still Form C.
[0193] Evaluation of Form C Form C is the optimal polymorph identified. Its stability, solubility, hygroscopicity, and formulation process feasibility were evaluated as described above.
