Description

Technical Field

[0001] Cross-reference This application claims the benefit of U.S. Provisional Application No. 63 / 354,516, filed Jun. 22, 2022, which is hereby incorporated by reference in its entirety.

Background Art

[0002] Washing of hair and the scalp is a common grooming practice that is considered important for optimal hair cleanliness, health, and maintenance. Hair cleansing agents typically remove sebum, sweat, exfoliated cutin, and environmental dirt and help prevent the onset of conditions such as folliculitis and seborrheic dermatitis. Currently commercially available formulations use synthetic detergents, technically referred to as "surfactant systems," formulated in various amounts and ratios to achieve a desired level of detergency. The beauty industry offers consumers numerous options for facilitating normal hair washing, including formulations containing a wide variety of ingredients and compositions.

[0003] In addition to removing sebum, sweat, exfoliated cutin, and environmental dirt, the secondary function of shampoo is to impart a desired sensory effect to the hair. Hair that has been thoroughly cleaned, i.e., hair from which sebum and products have been completely removed, tends to have a texture lacking in smoothness and appear duller, and as a result, is usually not considered cosmetically desirable. One strategy to address this problem is to incorporate a number of conditioners into the cleaning agent. These conditioners typically deposit on the hair surface and only modify the properties of the hair surface (improve the appearance and feel of the hair), and do not address the fundamental damage to the hair structure. When these conditioning agents accumulate excessively, especially in combination with insufficient cleaning, the conditioner may accumulate within the hair, weighing down the hair and making it appear duller. Commercially available products typically focus on providing benefits to either the hair or the scalp. However, since hair health begins at the hair follicle and the treatments and products applied to the hair affect the scalp, it can be said that these two are two sides of the same coin. Products that address the overall health of the hair and scalp structure can provide the most benefits. Therefore, there is a need for a new hair cleaning agent composition that cleans the hair and scalp without excessive deposition of the conditioning agent and provides the desired hair properties. Mode for Carrying Out the Invention

[0021] The fiber structure of human hair consists of the cuticle, cortex, and medulla. The cuticle consists of overlapping thin cell layers like a scale structure and protects the central part inside the hair known as the cortex. Cuticle cells are mechanically strong due to the high isodipeptide bonds and disulfide bonds present in their individual lamellar layers and are resistant to many environmental factors such as the penetration of external molecules. The cuticle accounts for approximately 15% of the weight of a single hair. The cortex is the middle region of the hair and accounts for about 80% of the weight of a single hair. The cortex is composed of a hierarchical structure of elongated fibrous alpha-helical keratin that extends along the fiber axis embedded in an amorphous protein matrix. It is the cortex, which has a structure composed of intertwined protein fibers and a supporting matrix, that gives the hair its tensile properties (strength, stiffness). The cell membrane complex binds cuticle cells to other cuticle cells, cortex cells to other cortex cells, and cuticle and cortex cells. The medulla corresponds to a centrally located, thin cell-connected beam and is absent in some hair fibers.

[0022] Human hair contains approximately 65% - 95% protein. Proteins exhibit their active properties within a specific temperature and pH range, but can unfold beyond these limits. The thermal denaturation of fibrous hard alpha-keratin proteins, i.e., the cortex proteins involved in hair strength and stiffness, shows a significant increase in stability when treated with low pH solutions. This is particularly true for hair damaged by oxidation such as bleaching, and when quantified by DSC, it is proven by a significant increase in the enthalpy and peak temperature of thermal denaturation. Enthalpy is usually understood to reflect the amount and structural integrity of alpha-helical substances within the intermediate diameter filaments (IF) of hair, while the peak temperature reflects the cross-link density of the matrix in which the IF is embedded. This phenomenon is further supported by amino acid analysis, X-ray diffraction, Raman spectroscopy, and observation of tensile strength.

[0023] Alkaline pH creates a favorable environment for the cleavage of cystine (disulfide), hydrolysis of amide bonds, and the formation of lanthionine and lysinoalanine, all of which weaken the protein structure of hair. Although the thermal stability does not change significantly depending on the time hair is exposed to a solution of a specific pH, combined with the fact that the pH of tap water varies widely from neutral to alkaline, it is suggested that negative effects can be experienced during the time hair is reasonably expected to be exposed during standard washing conditions.

[0024] Differences in the thermal stability of proteins in hair exposed to different pH values have been observed, which are due to changes in the environment of the intermediate diameter filaments, i.e., the interface between the IF and the surrounding matrix proteins. The scaffold-like IF surface is created by side-chain interactions that anchor the filaments to the matrix. Strong adsorption of protons to the interface of the IF can help protect this interface, and in addition to disulfide bonds, the electrostatic force further stabilizes this region.

[0025] During washing, the hydrogen bonds that stabilize the protein structure by water molecules are broken in hair, so it becomes structurally fragile. As a result, swelling occurs in specific parts of the cuticle, making the hair fiber more susceptible to damage. Furthermore, it has been established that the swelling behavior of the hair cuticle strongly depends on pH, and the more alkaline the environment, the greater the degree of swelling. Apart from the effect on thermal stability, this modification due to the swelling of the cuticle by pH is closely related to the properties of hair and the tendency of damage directly related to consumers. When the cuticle swells, the friction force between hair fibers increases, resulting in an increase in tangling and a rough feel. Also, when hair is wet, it is in a fragile state, and in wet hair, the combination of an increased unnecessary combing force and a decreased breaking stress can easily lead to peeling of the cuticle, protein loss, and even severe split ends during hair washing or shampooing. The minimal swelling of the cuticle indicated by an acidic pH can help offset this effect.

[0026] The protein in hair is in an optimal state (structurally most elastic) around pH 5. Such a more acidic pH environment can also minimize excessive swelling of the cuticle, reduce protein loss, maintain hair color, and minimize electrostatic repulsion. This is also the ideal pH range for skin with an acidic mantle. Skin with a disrupted pH may experience barrier breakdown, inflammation, and irritation. The pH of tap water varies significantly from 6.5 to 9.53 and may contain metal ions such as divalent calcium and magnesium (hard water minerals) that affect the texture of hair, as well as trace transition metals such as copper, iron, nickel, lead, and others that can contribute to the oxidative degradation of hair fibers.

[0027] Furthermore, the peptides can be used to treat hair damaged during washing. These peptides can interact with hair mainly through covalent (disulfide) bonds, polar bonds, and hydrophobic interactions. However, for the success of the treatment, it is necessary to penetrate the peptides into the cortex of the hair, but penetration can be difficult. Molecular size and polarity play a major role in determining the ability of a molecule to gain access inside the fibrous structure. Furthermore, the presence of sebum or products on the hair hinders the ability of the peptides to move well into the cortex. As noted above, at an appropriate acidic pH, it helps to stabilize the protein structure of the hair, but these peptides can act complementarily and synergistically to provide further reinforcement. Taken together, this structure provides both protection and repair of the protein structure of the hair, which can become brittle during the washing process.

[0028] To optimize the beneficial effects of protein structure reinforcement and strengthening by preventing residues that may impede the penetration of peptides into the cortex from remaining on the hair fibers, it is necessary to use a clarifying shampoo before applying the repair treatment containing peptides. Also, since the accumulation of minerals can also impede the effectiveness of the peptides, the conditioning lotion must be effective enough to remove 1) products, 2) sebum, 3) hard water minerals, and other types of deposits from the hair. Since salicylic acid may be difficult to formulate in a surfactant system, selecting a powerful, acid-compatible surfactant that is ion-resistant and stable in hard water helps to achieve the above three types of deposits. For example, detoxifying agents such as charcoal block metal ions in low amounts without further complicating the formulation with chelating agents that change viscosity. Charcoal has innumerable fine pores, thereby forming a large surface area that can adsorb heavy metals such as iron (Fe), chromium (Cr), cadmium (Cd), and copper (Cu).

[0029] Accordingly, provided herein is a detoxifying hair and scalp cleansing composition having an optimized pH that reduces swelling and protein loss. Formulating the shampoo at a more acidic pH can help neutralize this alkaline pH environment and minimize the net negative charge of hair proteins, thereby minimizing cuticle swelling and reducing the binding sites for dissolved metal cations. The presence of protective agents such as skin softeners, occlusives, cationic agents, and polymers on the hair fiber minimizes friction and associated styling damage and provides a barrier against heat, UV, and contaminants, but if the product accumulates excessively, it can weigh down the hair and inhibit it, or additional products may similarly interfere with its function. The hair composition may include peptides that strengthen the protein structure within the hair cortex and restore the tensile strength of the hair fiber. These peptides may not be compatible with conventional conditioning agents that form a film or coating, preventing the peptides from interacting with hair proteins and, in particular, from entering the cortex.

[0030] In some embodiments, the detoxifying hair and scalp cleansing composition described herein includes low molecular weight peptides that can penetrate the cortex of the hair and improve the mechanical properties of the hair. In some embodiments, the detoxifying hair and scalp cleansing composition described herein provides a long-lasting improved texture and an improved coherence of the hair when dry. In combination, the detoxifying hair and scalp cleansing composition described herein provides the hair with a soft, shiny, undamaged texture and other desired characteristics. The detoxifying hair and scalp cleansing composition described herein reduces protein loss and repairs the underlying damage to the hair without removing any essential oils from the hair, thereby avoiding excessive dependence on conditioners that coat the hair fibers to achieve the desired properties. Further, the detoxifying hair and scalp cleansing composition described herein does not contribute to the excessive accumulation of conditioning agents or other products that can ultimately weigh down the hair due to their limited ingredients.

[0031] In some embodiments, the detoxifying hair and scalp cleansing composition provided herein includes one or more solvents, one or more peptides, one or more hair conditioners, one or more skin conditioners, one or more detergents, and one or more cosmetic or dermatologically acceptable excipients. In some embodiments, the one or more peptides have a peptide sequence of 6 to 15 amino acids. In some embodiments, 2 to 5 of the amino acids of the peptide sequence are cysteine. The peptide composition provides the hair with improved resistance, strength, toughness, and stiffness.

[0032] When attempting to use a minimalist approach of "less is better", selecting multifunctional ingredients is key. Salicylic acid has well-established efficacy in skin care, bringing about desmolase activity and comedolytic activity, promoting the turnover of healthy cells, maintaining the balance of sebum production, providing photoprotection, and having antibacterial and anti-inflammatory effects. Although these properties are expected to be relevant to the scalp, this compound exhibits additional benefits specific to the scalp and is useful when applied topically in treating conditions such as dandruff, ichthyosis, and psoriasis. 31 Salicylic acid has a pKa of approximately 2.8 and, due to the relatively low associated pH, is considered an ideal ingredient for application to hair fibers in addition to the beneficial effects of the aforementioned acidic pH.

[0033] The detoxified hair and scalp cleansing compositions described herein maintain an optimal pH environment for the hair to clean the hair while minimizing fiber swelling and maintaining structural integrity. In addition to this, the hair cleansing composition maintains an optimal pH environment to clean the scalp while avoiding excessive drying of the scalp without disrupting the scalp's natural barrier function. The detoxified hair and scalp cleansing compositions described herein have a pH optimized for cleaning the hair and scalp. In some embodiments, the pH of the detoxified hair and scalp cleansing composition is optimized to promote the efficacy of certain components in the composition. In some embodiments, the pH of the detoxified hair and scalp cleansing composition is optimized to promote the efficacy of salicylic acid. In some embodiments, the pH of the composition is optimized to promote the chelation of certain minerals and metals. In some embodiments, the pH of the composition is optimized to minimize cuticle swelling. In some embodiments, the reduction of cuticle swelling reduces hair tangling. In some embodiments, the pH of the composition is optimized to protect the structural integrity of the hair and reduce the loss of proteins and lipids.

Example

[0098] The following examples are included for illustrative purposes only and are not intended to limit the scope of the present invention.

[0099] Example 1. Preparation of a detoxifying hair and scalp cleanser Exemplary detoxifying hair and scalp cleanser compositions are presented in Tables 2 to 6. The exemplary detoxifying hair and scalp cleanser compositions are manufactured by conventional compounding and mixing techniques or by mixing one or more polyols (such as glycerin), water, and a surfactant with any solid that needs to be heated and melted at a high temperature. These components are thoroughly mixed at a high temperature and then cooled to ambient temperature. Further components are added to the cooled product. Unless otherwise specified, the amounts described are to reflect the weight percentage of the active material.

[0105] Example 2. Evaluation of the Efficacy of a Detoxifying Hair and Scalp Cleanser During and After Washing A detoxifying hair and scalp cleanser composition is prepared according to Example 1. The detoxifying hair and scalp cleanser composition is evaluated using an investigation for determining the sensory effects of the detoxifying hair and scalp cleanser on gloss, texture (i.e., slipperiness), stiffness, and firmness. The sensory effects are evaluated during washing and after drying.

[0106] Gently rinse a human hair bundle with a length of 25 cm, a width of 5.5 cm, and a weight of 10 g in warm water at 40°C. Remove the excess water, and thoroughly lather the hair bundle for about 1 minute using 1.5 g of the detoxifying hair and scalp cleanser composition per 10 g of the hair bundle of the detoxifying hair and scalp cleanser. Conduct a sensory evaluation on the lathered hair and determine its slipperiness. The evaluation is performed by 20 people, and the total value of the evaluation is shown. The evaluation criteria are 4: glide well, 3: glide slightly, 2: do not glide well, and 1: do not glide.

[0107] Gently rinse a human hair bundle with a length of 25 cm, a width of 5.5 cm, and a weight of 10 g in warm water at 40°C. Remove the excess water, and thoroughly lather the hair bundle for about 1 minute using 1.5 g of the detoxifying hair and scalp cleanser composition per 10 g of the hair bundle of the detoxifying hair and scalp cleanser. Rinse the lathered hair with running water at 40°C (2 L / min) for 1 minute. Remove the excess moisture using a towel and air-dry the hair bundle. After drying, conduct a sensory evaluation on the dried hair and determine its gloss and coherence. The evaluation is performed by 20 people, and the total value of the evaluation is shown. The evaluation criteria are 4: good, 3: slightly good, 2: not very good, and 1: not good.

[0108] Thoroughly rinse a human hair bundle with a length of 25 cm, a width of 5.5 cm, and a weight of 10 g in warm water at 40°C. Thoroughly lather the hair bundle with a 5 g or 10 g harmless hair and scalp cleansing composition for about 1 minute. Thoroughly rinse the lathered hair with running water at 40°C (2 L / min). Remove excess moisture using a towel and dry the hair bundle with warm air from a hair dryer. After drying, conduct a sensory evaluation on the dried hair to determine its stiffness. The evaluation is performed by 25 people and shows the total value of the evaluation. The evaluation criteria are as follows: 4: A significant improvement in stiffness is observed; 3: An improvement in stiffness and firmness is observed; 2: A slight improvement in stiffness is observed; 1: No improvement in stiffness; and 0: Stiffness disappears.

[0109] Example 3. Evaluation of the effectiveness of the cleansing agent using FTIR Samples of bleached hair may be collected. A part of the sample of bleached hair may be treated with the harmless hair and scalp cleansing composition described in this specification. For both the treated and untreated samples, cross-sections are prepared using a cryostat and examined using attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR). Using this technique, hyperspectral images are generated and used to visualize the lipid content inside the hair fiber, assess the hair damage (i.e., chemical treatment or UV exposure) inside the hair fiber, identify the modification of the hair structure (i.e., the three-dimensional structure of proteins or the organization of lipids), evaluate the moisture of the hair (i.e., the moisture content), visualize the active penetration inside the hair fiber, and determine the effectiveness of the cleansing agent for removing sebum and deposits from the hair.

[0110] The single fibers of both the treated and untreated samples may be scanned longitudinally using ATR-FTIR, thereby providing an infrared image of approximately 1 to 2 microns outside the fiber surface. Using these hyperspectral images, the lipid content in the cuticle can be determined, hair damage (i.e., chemical treatment or UV exposure) can be examined along the fiber, the deposition of active ingredients can be analyzed along the hair fiber, modifications to the hair structure (i.e., the three-dimensional structure of proteins or the organization of lipids) can be identified, the moisture content of the hair (i.e., the wetness) can be evaluated, and the effectiveness of a detergent for removing sebum and products from the hair can be determined.

[0111] Example 4: Evaluation of Metal Reduction / Chelation Samples of bleached hair may be taken. A portion of the samples of bleached hair may be treated with the hair and scalp detoxifying detergent composition described herein. Both the treated and untreated samples may be rinsed with hard water and examined using time-of-flight secondary ion mass spectrometry (TOF-SIMS). Spectra for calcium, magnesium, copper, aluminum, iron, chromium, zinc, nickel, cadmium, lead, and mercury may be obtained. ***
