Description

[Technical Field]

[0001] The present invention relates to a collection kit and collection method for collecting blood products for testing. [Background technology]

[0002] Blood products include red blood cell products, plasma products, platelet products, and whole blood products. Blood products containing the components needed by patients are used for transfusions. Blood products are stored and transported in medical bags. To ensure the safety of blood products, culture tests may be performed on blood products. Culture tests detect the presence or absence of pathogens by collecting a small sample in a culture bottle and placing the bottle in an environment favorable for bacterial growth.

[0003] Among blood products, platelet products are inspected using the following procedure: A small-capacity collection bag is connected to a medical bag (hereinafter referred to as a platelet bag) that contains the platelet product. A portion of the platelet product in the platelet bag is then transferred to the collection bag. The collection bag is then disconnected from the platelet bag.

[0004] The collection bag is then transferred to a clean bench. In the clean bench, the platelet product in the collection bag is injected into a blood culture bottle. Prior to injection, the tube of the collection bag is connected to a tube extending from a sample collection tube. Using the scale on the sample collection tube as a guide, a predetermined amount of the platelet product is transferred from the collection bag to the sample collection tube.

[0005] The platelet product is then transferred in a specified amount from the sample collection tube to a blood culture bottle, using the scale on the sample collection tube as a guide. Anaerobic and aerobic cultures are performed for the culture test. Therefore, the platelet product is dispensed into a culture bottle for anaerobic culture and a culture bottle for aerobic culture. The blood culture bottle is then placed in an incubator and subjected to the culture test for a specified period.

[0006] For example, Patent Document 1 discloses a collection device for collecting a sample from a medical bag. [Prior art documents] [Patent documents]

[0007] [Patent Document 1] U.S. Patent No. 8,777,921 Summary of the Invention [Problem to be solved by the invention]

[0008] To further improve the safety of platelet preparations, culture testing of all platelet preparations is being considered. Therefore, there is a need to improve the work efficiency of culture testing of platelet preparations.

[0009] However, in conventional testing procedures, the process of dispensing platelet products from a platelet bag to a blood culture bottle requires the attachment and detachment of the collection bag and sample collection tube. This attachment and detachment process requires a sterile joining device that aseptically welds or cuts the tubes together. Multiple attachment and detachment processes using a sterile joining device require a lot of effort and waiting time.

[0010] Furthermore, in conventional testing procedures, the user must manually operate a clamp to measure the amount of sample, relying on the level of the platelet preparation's liquid surface, which is prone to measurement errors.

[0011] Furthermore, the conventional testing procedure results in residual platelet preparations remaining inside the collection bag and tubing, which results in a significant loss of valuable platelet preparations.

[0012] An object of the present invention is to solve the above-mentioned problems. [Means for solving the problem]

[0013] One aspect of the following disclosure is a collection kit comprising: a storage bag for storing a platelet preparation transferred from a platelet bag; a tube line having an upstream portion to which the platelet bag can be connected and a downstream portion connected to the storage bag; a storage container provided midway along the tube line to connect the upstream portion and the downstream portion of the tube line, and for retaining a predetermined amount of the platelet preparation while allowing an amount of the platelet preparation in excess of the predetermined amount to pass; and a holder connected to the tube line and to which a culture bottle is connected, wherein the storage container has an overflow structure that allows an amount of the platelet preparation in excess of the predetermined amount to flow out.

[0014] Another aspect is a collection method for collecting the platelet preparation into the culture bottle using the collection kit of the above aspect, the collection method including the steps of: connecting the platelet bag to the upstream portion of the tube line and connecting the storage bag to the downstream portion of the tube line; transferring the platelet preparation from the platelet bag to the storage bag and storing a predetermined amount of the platelet preparation in the storage container; connecting the culture bottle to the holder; and injecting the predetermined amount of the platelet preparation stored in the storage container into the culture bottle via the tube line. The method for collecting the compound is as follows. [Effects of the Invention]

[0015] The collection kit and collection method of the above aspects reduce the labor required for culture testing, reduce measurement errors, and reduce waste of valuable platelet products. [Brief explanation of the drawings]

[Figure 8] FIG. 8 is a plan view of the collection kit according to the second embodiment. DETAILED DESCRIPTION OF THE INVENTION

[0017] (First embodiment) The collection kit 10 according to the embodiment shown in Figure 1 is used to collect a predetermined amount of platelet preparation for culture testing from a platelet bag 90 (see Figure 3). The platelet bag 90 shown in Figure 3 is a medical bag that contains a platelet preparation obtained by a centrifugation process. The platelet bag 90 is connected during use of the collection kit 10, and is therefore not included in the collection kit 10 in its initial state.

[0018] 1, the collection kit 10 according to this embodiment includes a reservoir 12, a holder 14, a tubing line 16, and a storage bag 18. The reservoir 12, the holder 14, and the storage bag 18 are connected to each other via the tubing line 16.

[0019] The storage bag 18 is a medical bag. The storage bag 18 has a structure in which two resin sheets are overlapped and welded at peripheral portions 181. For example, a flexible resin sheet such as polyvinyl chloride resin can be used for the storage bag 18. The storage bag 18 has a storage space 182 that stores the platelet preparation of the platelet bag 90. The storage space 182 has a capacity sufficient to store the entire amount of the platelet preparation stored in the platelet bag 90. The storage bag 18 has a port 183 at its upper end. The tubing line 16 is connected to the port 183.

[0020] The storage container 12 is a container that temporarily stores a small amount of platelet preparation. The storage container 12 is connected between the upstream end 161 and the downstream end 162 of the tubing line 16. The storage container 12 is connected to the tubing line 16 on the upstream side of the storage bag 18. The collection kit 10 of this embodiment has two storage containers 12. The storage containers 12 include a first storage container 121 and a second storage container 122. The first storage container 121 and the second storage container 122 have the same dimensions and structure.

[0021] As shown in FIG. 2, the storage container 12 has a storage section 123, a first port 124, a second port 125, and an overflow tube 126. The storage section 123 has a cylindrical main body 127. The main body 127 is closed at one end and has an internal space 128 therein. The main body 127 is formed of a transparent or translucent resin material or the like. The transparent or translucent main body 127 allows the liquid level 98 (see FIG. 5) of the stored platelet preparation to be visually confirmed. The first port 124 is formed at one end of the main body 127. The first port 124 communicates with the internal space 128. The first port 124 protrudes from the main body 127. The tubing line 16 is connected to the first port 124.

[0022] The second port 125 is formed at the other end of the main body 127. The second port 125 protrudes from the main body 127. The tubing line 16 is connected to the second port 125. The second port 125 is fluidly connected to the overflow tube 126. The overflow tube 126 protrudes from the other end of the main body 127 toward one end of the main body 127 into the interior of the internal space 128. The tip of the overflow tube 126 opens at an overflow position in the internal space 128. The internal space 128 on the side of the overflow tube 126 contains a specified amount of platelet preparation. The overflow tube 126 discharges any platelet preparation exceeding the specified amount from the reservoir 123.

[0023] The holder 14 can be connected to the opening 82 of the culture bottle 80 shown in FIG. 6. The holder 14 has a structure similar to that of a so-called blood collection holder. The holder 14 shown in FIG. 1 has a cylindrical main body 141. The main body 141 has a lid 143 provided near an opening 144. The lid 143 is rotatably attached to the main body 141 via a hinge 142. The lid 143 covers the opening 144 of the main body 141 until immediately before use, thereby keeping the inside of the main body 141 clean. The holder 14 has a port 146 at one end of the main body 141 to which the tubing line 16 is connected. The holder 14 has a needle 147 inside the main body 141 that protrudes toward the opening 144 along the center of the main body 141. The lumen of the needle 147 is fluidly connected to the port 146. The holder 14 is not limited to the above configuration, and may be a blood collection needle with a cap.

[0024] The tubing line 16 forms a flow path connecting the storage container 12, the holder 14, and the storage bag 18. The tubing line 16 has an upstream section 163, an intermediate section 164, a first branch flow path 165, a second branch flow path 166, and a downstream section 167. The upstream section 163 is the upstream tube of the tubing line 16 and has an upstream end section 161. The collection kit 10 in its initial state has an air vent 22 connected to the upstream end section 161. The air vent 22 allows gas to pass through while preventing liquid from passing through. The air vent 22 is provided to discharge sterilizing gas (e.g., ethylene oxide gas) introduced into the collection kit 10 during the sterilization process of the collection kit 10. The air vent 22 is also connected to the downstream section 167, as described below, and is used to inject the platelet preparation from the storage container 12 into the culture bottle 80.

[0025] The upstream section 163 is connected to a connection tube 92 of a platelet bag 90 shown in FIG. 3. As shown in FIG. 1, the upstream section 163 is connected to the intermediate section 164 via a first branch joint 24. The first branch joint 24 is a Y-shaped joint branched into three branches. A breakable seal connector 26 is connected to the first branch joint 24. The breakable seal connector 26 is a tubular member formed of a bendable flexible material. The breakable seal connector 26 has a breakable sealing member inside. The breakable seal connector 26 can be broken by bending it. The holder 14 is connected to the first branch joint 24 via the breakable seal connector 26.

[0026] The intermediate section 164 connects the first branch joint 24 and the second branch joint 28. The second branch joint 28 is a Y-shaped joint with three branches. The tubing line 16 branches into a first branch flow path 165 and a second branch flow path 166 downstream from the second branch joint 28. The first branch flow path 165 and the second branch flow path 166 are parallel to each other. A first storage container 121 is disposed midway along the first branch flow path 165. The collection kit 10 further includes a first clamp 30, a second clamp 32, a third clamp 34, and a fourth clamp 36. The first clamp 30 is attached to the first branch flow path 165 upstream of the first storage container 121. The second clamp 32 is attached to the first branch flow path 165 downstream of the first storage container 121. The third clamp 34 is attached to the second branch flow path 166 on the upstream side of the second storage container 122. The fourth clamp 36 is attached to the second branch flow path 166 on the downstream side of the second storage container 122.

[0027] The first clamp 30 switches between an open state and a closed state based on the user's operation. In the closed state, the first clamp 30 compresses the attached tubing line 16 to prevent fluid from passing through. In the open state, the first clamp 30 allows fluid to pass through the attached tubing line 16. The second clamp 32, the third clamp 34, and the fourth clamp 36 are configured similarly to the first clamp 30.

[0028] The first branch flow path 165 and the second branch flow path 166 join at a third branch joint 38 on the downstream side. The third branch joint 38 is a Y-shaped joint that branches into three. The third branch joint 38 connects the first branch flow path 165, the second branch flow path 166, and a downstream section 167.

[0029] The downstream portion 167 is located at the most downstream side of the tubing line 16. The downstream portion 167 connects the third branch joint 38 and the storage bag 18. The medical tubing used in the tubing line 16 is a flexible tubing made of, for example, polyvinyl chloride resin or silicone resin.

[0030] The collection kit 10 of this embodiment is configured as described above, and its operation will be described below.

[0031] The first step involves connecting a platelet bag 90 to the collection kit 10, as shown in Figure 3. The platelet bag 90 has a connecting tube 92. The connecting tube 92 is joined to the upstream section 163 of the tubing line 16 using a sterile joining device. The upstream section 163 and the connecting tube 92 are connected via a welded section 94. A portion of the upstream section 163 near its upstream end 161, together with the air vent 22, is separated from the collection kit 10.

[0032] In the next step, as shown in FIG. 4, the platelet product in the platelet bag 90 is transferred to the storage bag 18. This step is performed with the first clamp 30, second clamp 32, third clamp 34, and fourth clamp 36 of the collection kit 10 all in an open state. As shown in the figure, the user uses the stand 96 to position the platelet bag 90 at the top and the storage bag 18 at the bottom. At this time, as shown in FIG. 5, the user positions the first storage container 121 so that the first port 124 of the first storage container 121 is facing up and the second port 125 is facing down, with the longitudinal direction of the storage section 123 facing vertically. The user also positions the second storage container 122 so that the storage section 123 is also facing vertically, similar to the first storage container 121.

[0033] 4, the platelet preparation in the platelet bag 90 flows down the tubing line 16 by its own weight and into the storage bag 18. The platelet preparation flows into a first branch channel 165 and a second branch channel 166 midway through the tubing line 16.

[0034] As shown in Fig. 5, the platelet preparation that has flowed into the first branch flow path 165 flows into the storage section 123 of the first storage container 121. The platelet preparation is stored in the storage section 123 up to the upper end of the overflow tube 126. In the storage section 123, any platelet preparation that exceeds the overflow tube 126 is discharged from the second port 125 via the overflow tube 126. A specified amount of platelet preparation is stored in the storage section 123. The first storage container 121 stores, for example, approximately 8 ml of platelet preparation.

[0035] The platelet preparation also flows into the second storage container 122. As with the first storage container 121, a specified amount (for example, about 8 ml) of platelet preparation is stored therein. Any platelet preparation exceeding the specified amount is discharged through the overflow tube 126 of the second storage container 122. The liquid level 98 of the platelet preparation in the storage section 123 is maintained at the same position as the upper end of the overflow tube 126.

[0036] Almost all of the platelet preparation in the platelet bag 90 is transferred to the storage bag 18, except for droplets adhering to the wall surface of the tubing line 16 and the portion stored in the first storage container 121 and the second storage container 122. When transferring the platelet preparation, the breakable seal connector 26 separates the tubing line 16 from the holder 14. The breakable seal connector 26 prevents contamination of the platelet preparation through the holder 14, where it is difficult to ensure airtightness and sterility.

[0037] After the transfer of the platelet product is completed, the user closes the first clamp 30, the second clamp 32, the third clamp 34, and the fourth clamp 36 shown in FIG. 4. The user then uses a sterile connecting device to detach the storage bag 18 from the collection kit 10. The storage bag 18 is then used to store and use the platelet product. When detaching the storage bag 18, the air vent 22 that was first detached is connected to the downstream portion 167 of the tubing line 16, as shown in FIG. 6.

[0038] In the next step, as shown in FIG. 6, the platelet product stored in the storage container 12 is dispensed into a culture bottle 80. The user lifts the downstream portion 167 of the tubing line 16 and uses the stand 96 (see FIG. 4) to position the collection kit 10 with the storage container 12 facing up and the holder 14 facing down. Next, the user attaches the first culture bottle 80 to the holder 14. The user also seals (closes) and separates the upstream portion 163 toward the platelet bag 90 with a tube sealer or the like, or closes it with a clip 99. Next, the user opens the first clamp 30 and the second clamp 32. As a result, the specified amount of platelet product stored in the first storage container 121 is injected into the culture bottle 80.

[0039] Thereafter, the user attaches the two-sided culture bottle 80 to the holder 14. Next, the user opens the third clamp 34 and the fourth clamp 36. This causes the platelet preparation stored in the second storage container 122 to be injected into the second culture bottle 80. With the above operations, the dispensing of the platelet preparation into the two culture bottles 80 is completed.

[0040] As described above, the collection kit 10 of this embodiment can reduce welding or cutting of the tubing line 16 using a sterile joining device.

[0041] (Modification of the first embodiment) Another exemplary configuration of the storage container 12 will be described below. As shown in FIG. 7 , the storage container 12 according to this modification includes a storage section 123, a first port 124, a second port 125, and a partition wall 129. The first port 124 is disposed in the upper part of the storage section 123. The second port 125 is disposed in the lower part of the storage section 123. The partition wall 129 is formed inside the storage section 123, protruding upward from the lower part. The partition wall 129 divides the internal space 128 into a first section 1281 connected to the second port 125 and a second section 1282 that stores a specified amount of platelet preparation. The platelet preparation that flows into the storage section 123 from the first port 124 flows into the second section 1282. A platelet preparation exceeding the specified amount overflows the partition wall 129 and flows into the first section 1281. The platelet preparation that has overflowed the partition wall 129 flows out of the reservoir 123 through the second port 125 .

[0042] The storage container 12 of this modified example also provides the same effects as the storage container 12 of FIG.

[0043] (Second embodiment) As shown in Figure 8, the collection kit 70 of this embodiment differs from the collection kit 10 shown in Figure 1 in the attachment position of the air vent 22. The collection kit 70 has the same configuration as the collection kit 10 except for the attachment position of the air vent 22. In the collection kit 70, the same configuration as the collection kit 10 is assigned the same reference numeral, and detailed description thereof will be omitted.

[0044] As shown in FIG. 8 , the air vent 22 is connected to the downstream portion 167 of the tubing line 16 via a fourth branch joint 72. A connection tube 74 is connected to the fourth branch joint 72. The air vent 22 is connected to the connection tube 74. Typically, the air vent 22 is connected to the connection tube 74 without a breakable seal connector 76. Note that a breakable seal connector 76 may be connected between the connection tube 74 and the air vent 22. The breakable seal connector 76 is a member similar to the breakable seal connector 26. The breakable seal connector 76 isolates the air vent 22 from the tubing line 16 until immediately before the step of injecting the platelet preparation from the storage container 12 into the culture bottle 80.

[0045] When the breakable seal connector 76 is broken, air flows into the tubing line 16 through the air vent 22. As shown in Figure 6, when the platelet product in the storage container 12 is transferred to the culture bottle 80, air is supplied from upstream through the air vent 22. This allows the user to smoothly inject the platelet product stored in the storage container 12 into the culture bottle 80. Furthermore, the collection kit 70 does not require the step of joining the air vent 22 to the downstream portion 167.

[0046] The collection kits 10, 70 and collection methods according to the above-described embodiments have the following advantages.

[0047] The collection kits 10, 70 each include a storage bag 18 for storing a platelet preparation transferred from a platelet bag 90, a tube line 16 having an upstream section 163 to which the platelet bag 90 can be connected and a downstream section 167 connected to the storage bag 18, a storage container 12 provided midway along the tube line 16 to connect the upstream section 163 and the downstream section 167 of the tube line 16, and which retains a predetermined amount of platelet preparation while allowing an excess of the predetermined amount of platelet preparation to pass through, and a holder 14 connected to the tube line 16 and to which a culture bottle 80 is connected, and the storage container 12 has an overflow structure that allows an excess of the predetermined amount of platelet preparation to flow out.

[0048] The collection kits 10, 70 connect the platelet bag 90 to the tubing line 16 and transfer the platelet product to the storage bag 18. At this time, the storage container 12 can collect a specified amount of platelet product. As described above, the collection kits 10, 70 can reduce the number of times that tubing is connected and disconnected to the tubing line 16 using a sterile connection device, thereby reducing the labor required for culture testing. Furthermore, the collection kits 10, 70 can reduce the waste of platelet products remaining in disconnected tubing. The collection kits 10, 70 collect platelet products using a storage container 12 with an overflow structure, thereby reducing variation in volume.

[0049] In the collection kits 10 and 70, the storage container 12 has a cylindrical storage section 123, a first port 124 arranged at the top of the storage section 123, and a second port 125 arranged at the bottom of the storage section 123, and the overflow structure may have an overflow pipe 126 that communicates with the second port 125 and protrudes a predetermined length from the bottom to the top.

[0050] According to the collection kits 10 and 70, the storage container 12 can store a specified amount of platelet preparation in the storage section 123. The collection kits 10 and 70 facilitate the operation of measuring and collecting a specified amount of platelet preparation.

[0051] In the collection kits 10 and 70, the holder 14 may be connected to the tubing line 16 on the first port 124 side of the storage container 12. The collection kits 10 and 70 can discharge the platelet product collected in the storage container 12 from the first port 124 side. At that time, the platelet product flows out of the storage container 12 without being obstructed by the overflow tube 126.

[0052] In the collection kits 10 and 70, the tubing line 16 may have a plurality of branch flow paths (e.g., a first branch flow path 165 and a second branch flow path 166) that branch off and run in parallel between the upstream section 163 and the downstream section 167, and a storage container 12 may be connected to each of the plurality of branch flow paths. With the collection kits 10 and 70, a predetermined amount of platelet preparation can be measured and collected simultaneously into each of the plurality of storage containers 12. The collection kits 10 and 70 facilitate the dispensing of the platelet preparation into a plurality of culture bottles 80.

[0053] The collection kits 10, 70 described above may have a branching section (e.g., second branch joint 28) disposed between the upstream section 163 and the plurality of storage containers 12, and a junction section (e.g., third branch joint 38) disposed between the downstream section 167 and the plurality of storage containers 12. According to these collection kits 10, 70, even when a plurality of storage containers 12 are provided, they can be connected to the storage bag 18 with a single downstream section 167. Therefore, the collection kits 10, 70 can accommodate a plurality of storage containers 12 without increasing the number of ports on the storage bag 18.

[0054] In the collection kits 10 and 70 described above, the holder 14 may be connected between the upstream section 163 and the branch section (e.g., the second branch joint 28). The collection kits 10 and 70 can allow platelet preparations from multiple storage containers 12 to flow into a common holder 14. Therefore, the collection kits 10 and 70 can dispense platelet preparations from multiple storage containers 12 into culture bottles 80 using a single holder 14.

[0055] The collection kits 10, 70 may include upstream clamps (first clamp 30 and third clamp 34) attached to the tubing line 16 (first branch flow path 165 or second branch flow path 166) near the first port 124 of the storage container 12, and downstream clamps (second clamp 32 and fourth clamp 36) attached to the tubing line 16 (first branch flow path 165 or second branch flow path 166) near the second port 125 of the storage container 12. The collection kits 10, 70 eliminate the need for the user to attach new clamps, thereby reducing the effort required for operation.

[0056] In the collection kits 10, 70, the holder 14 may be sealed from the tubing line 16 via the breakable seal connector 26. The collection kits 10, 70 can increase the safety of the platelet product transferred to the storage bag 18 by reliably isolating the tubing line 16 from the holder 14.

[0057] In the collection kits 10 and 70 described above, the holder 14 may be connected to the tubing line 16 between the upstream end 161 and the storage container 12. With these collection kits 10 and 70, a specified amount of platelet preparation collected in the storage container 12 can be transferred to the culture bottle 80 via the tubing line 16, thereby reducing the effort required for operation.

[0058] The collection kit 70 may have an air vent 22 that is connected to the tubing line 16 between the reservoir 12 and the storage bag 18 and allows air to pass through but prevents liquid from passing through. This collection kit 70 can reduce the effort required to connect the air vent 22 to the tubing line 16.

[0059] The collection kit 10 may have an air vent 22 connected to the upstream portion 163 of the tubing line 16. Since the air vent 22 is already connected to the tubing line 16 in this collection kit 10, it is easy to discharge the sterilizing gas (water vapor or ethylene oxide) introduced into the collection kit 10 during the manufacturing process. The air vent 22 can also be connected to the downstream portion 167. The air vent 22 connected to the downstream portion 167 supplies air to the storage container 12, thereby facilitating the transfer of the platelet preparation to the culture bottle 80.

[0060] The collection method of this embodiment relates to a method for collecting a platelet preparation into a culture bottle 80 using the collection kits 10 and 70. This collection method includes the steps of connecting a platelet bag 90 to the upstream portion 163 of the tubing line 16 and connecting a storage bag 18 to the downstream portion 167 of the tubing line 16, transferring the platelet preparation from the platelet bag 90 to the storage bag 18 and storing a predetermined amount of the platelet preparation in the storage container 12, connecting the culture bottle 80 to the holder 14, and injecting the predetermined amount of the platelet preparation stored in the storage container 12 into the culture bottle 80 via the tubing line 16. This collection method reduces the effort required for culture testing. This collection method also reduces the amount of wasted platelet preparation. Furthermore, this collection method also reduces errors in the amount of platelet preparation collected.

[0061] The present invention is not limited to the above-described embodiment, and various configurations can be adopted without departing from the gist of the present invention. [Explanation of symbols]
