AI Technical Summary
Existing automatic analyzers lack miniaturization due to the complexity of nozzle operations, which is not addressed by reducing the number of actuator axes, leading to increased size and operational inefficiencies.
The automatic analyzer incorporates a magnetic separation unit and nozzles that are movable in both vertical and horizontal directions, reducing the number of movable axes by integrating the B/F separation unit, reaction liquid suction nozzle, and substitution liquid discharge nozzle on a turntable that can rotate and move vertically, allowing for efficient operation with reduced axis movement.
This configuration enables the miniaturization of the analyzer, improves temperature control efficiency, reduces costs, and shortens analysis time by minimizing the device size and necessary drive mechanisms, while maintaining efficient operation and analysis capabilities.
Abstract
Description
automatic analyzer
[0001] The present invention relates to an automatic analyzer.
[0002] In an automatic analyzer for analyzing biological components, there is a technique that includes a magnetic separation unit, a nozzle for removing excess labeled substances, and a nozzle for discharging a replacement liquid (see Patent Document 1).
[0003] International Publication No. 2020 / 235162
[0004] According to Patent Document 1, no consideration is given to reducing the number of axes of the actuator corresponding to the complicated movements of the nozzle, thereby realizing a miniaturized automatic analyzer.
[0005] SUMMARY OF THE INVENTION It is therefore an object of the present invention to provide an automatic analyzer that can reduce the number of movable axes.
[0006] An automated analyzer according to one embodiment of the present invention comprises a magnetic separation unit that captures magnetic particles in a first container that contains a reaction liquid in which a reagent and a sample are mixed, a first nozzle that aspirates a first liquid in the first container, and a second nozzle that ejects a second liquid into the first container into which the first liquid has been aspirated, and the magnetic separation unit is configured to be movable in both vertical and horizontal directions.
[0007] According to the present invention, an automatic analyzer capable of reducing the number of movable axes can be provided.
[0008] 1 is a diagram showing a schematic configuration of an automatic analyzer; a top view of an analysis operation unit; a side view of an analysis operation unit; a top view of another outline of the analysis operation unit; a side view of another outline of the actual analysis operation unit; and a top view of yet another outline of the analysis operation unit.
[0009] An embodiment of the automatic analyzer will be described with reference to Figures 1 to 6. In the drawings used in this specification, identical or similar reference numerals are used to designate identical or corresponding components, and repeated description of these components may be omitted.
[0010] First, the overall configuration of the automatic analyzer will be described with reference to Fig. 1. Fig. 1 is a diagram showing the general configuration of the automatic analyzer.
[0011] The automatic analyzer 1 is composed of a control unit 10 and an analysis operation unit 20. This automatic analyzer 1 stores a plurality of reagent containers 31 along the inside of the outer wall of a cylindrical reagent storage 30 that is supported rotatably around a vertical axis, and has the function of aspirating a predetermined amount of a predetermined reagent from each reagent container 31 using a dispensing pipette and supplying it to a biological specimen such as blood or urine dispensed in a reaction container 51 for analysis.
[0012] First, the transport route of the sample to be analyzed will be described.
[0013] The sample to be analyzed is contained in a sample container 40 and moved within the automatic analyzer 1 by a sample transport mechanism such as a belt conveyor or rack handler (not shown for convenience of illustration), and is transported to a sample dispensing mechanism 41 equipped with a dispensing pipette for dispensing the sample, where it is dispensed.
[0014] A plurality of reaction vessels 51 are supplied to the automatic analyzer 1 while being placed in an unused reaction vessel storage unit 50 .
[0015] The reaction vessels 51 are grasped one by one from the unused reaction vessel storage unit 50 by the reaction vessel transport mechanism 52, which then rises and moves them to the buffer 42 (the sample discharge position of the sample dispensing mechanism 41), and also moves the reaction vessels 51 above the buffer 42 to the reaction vessel grasping position 43. To enable such movement, the reaction vessel transport mechanism 52 is configured to be movable in the X-axis (left-right direction in FIG. 1), Y-axis (front-back direction in FIG. 1), and Z-axis (up-down direction in FIG. 1) directions.
[0016] The disk-shaped incubator 25, which is supported so as to be freely rotatable around a vertical central axis, has multiple reaction vessel mounting sections 26 for securing multiple reaction vessels 51 on the circumference near the outer periphery, and by rotating the incubator 25, each reaction vessel 51 can be moved to a predetermined position.
[0017] Next, the specimen dispensing mechanism 41 moves to an area above the specimen, aspirates the specimen, and then moves to an area above the reaction vessel 51 above the buffer 42, dispensing the specimen into the reaction vessel 51. After this, the specimen dispensing mechanism 41 cleans the nozzle of the dispensing pipette using a cleaning mechanism (not shown). If an exchangeable tip is used instead of the nozzle, the tip may be exchanged using an exchange mechanism (not shown).
[0018] Next, the transport path of the reagent to be added to the sample in the reaction vessel 51 will be described.
[0019] The transport mechanism 60 is configured to be able to move circularly within the range of the buffer 42, the reaction vessel gripping position 43, the reagent discharge position 33 of the reagent dispensing mechanism 32, the reaction vessel installation section 26 on the incubator 25, and the B / F (Bound / Free) separation section 104 of the analysis section 100, and is configured to be rotatable to transport the reaction vessel 51 to these positions.
[0020] The cylindrical, hollow reagent storage 30 is rotatably supported around a vertical central axis and forms a reagent holder 34 that holds multiple reagent containers 31 radially along the hollow housing 38. Some of the reagent containers 31 contain reagents containing a large number of magnetic particles for stirring. The structure will be described in detail later.
[0021] The reagent dispensing mechanism 32 is configured to be movable so that it can aspirate the reagent in the reagent container 31 and move to a predetermined position. First, the reagent dispensing mechanism 32 moves to an area above a predetermined type of reagent in the reagent storage 30 and aspirates a predetermined amount of reagent, and then moves to an area above a reaction container 51 placed at a reagent discharge position 33 set around the incubator 25 and discharges the reagent into the reaction container 51.
[0022] A reagent stirring mechanism (not shown) is provided above the reagent storage 30. This stirring mechanism is equipped with a magnetic particle stirring arm (also called a stirrer) that can rotate around a vertical axis. The magnetic particle stirring arm moves to an upper region of the reagent container 31 containing the reagent to be stirred, which contains magnetic particles. The magnetic particle stirring arm lowers a paddle-shaped or spiral-shaped magnetic particle stirring mechanism, for example, provided at the lower end of the magnetic particle stirring arm, into the reagent. The magnetic particle stirring mechanism rotates to stir the magnetic particle solution. To prevent spontaneous precipitation of the magnetic particles in the solution, the magnetic particle stirring arm stirs the magnetic particles just before the reagent is dispensed. After stirring, the magnetic particle stirring arm rises to the top of the reagent container 31, moves to an upper region of the washing mechanism containing the washing solution, and then lowers into the washing solution. The magnetic particle stirring mechanism then rotates to remove magnetic particles adhering to the stirring mechanism.
[0023] After a predetermined reaction time has elapsed since the sample and the predetermined reagent were dispensed, a reaction solution is formed. The reaction vessel 51 containing this reaction solution is moved by the transport mechanism 60 to the B / F separation unit 104 of the analysis unit 100.
[0024] The B / F separation unit 104 is a part that captures magnetic particles in a reaction vessel 51 that contains a reaction liquid in which a reagent and a sample are mixed.More specifically, it is a mechanism that separates the reaction liquid that does not contain magnetic particles from the magnetic particles by magnetically attracting magnetic particles containing substances that have immunologically bound to the object to be measured that are present in the reaction liquid in the reaction vessel 51 transported by the transport mechanism 60 to the inner wall of the reaction vessel 51 using a magnet 105.
[0025] The reaction liquid suction nozzle 106 is used to suction the reaction liquid in the reaction vessel 51 and is configured to be able to rotate and move up and down.It moves above and down to the reaction vessel 51 after a predetermined time has passed on the B / F separation section 104, and suctions the reaction liquid in the reaction vessel 51 that does not contain magnetic particles.
[0026] The replacement liquid discharge nozzle 108 discharges the replacement liquid into the reaction vessel 51 into which the reaction liquid has been aspirated, and is configured to be able to rotate and move up and down. Above the B / F separation section 104, it moves above and down the reaction vessel 51 into which the reaction liquid not containing magnetic particles has been aspirated, and discharges the buffer solution into the reaction vessel 51.
[0027] The stirring mechanism (not shown) mixes the magnetic particles and buffer solution in the reaction vessel 51 by applying a rotational motion to the reaction vessel 51. After mixing, the reaction vessel 51 is transported below the measurement flow channel introduction nozzle 130 by the rotation of the turntable 102.
[0028] The measurement flow path introduction nozzle 130 is rotatable and movable up and down, and is a nozzle for sucking the reaction liquid in the reaction vessel 51 and introducing it into the photometry unit 134 .
[0029] The photometry unit 134 detects the concentration of the detection target substance in the reaction liquid sucked and delivered from the measurement flow path introduction nozzle 130 .
[0030] Next, the reaction vessel 51 holding the analyzed reaction liquid is moved to the upper area of the reaction vessel disposal port (not shown) by the reaction vessel transport mechanism 52, and the reaction vessel 51 is disposed of through the reaction vessel disposal port into the reaction vessel disposal section (not shown).
[0031] The automatic analyzer 1 shown in FIG. 1 can efficiently analyze a plurality of specimens for a plurality of analysis items by combining or repeating the above operations.
[0032] The mechanism described above in the automatic analyzer 1 is referred to as an analysis operation unit 20 .
[0033] Furthermore, in addition to the analysis operation unit 20, the automatic analyzer 1 includes a control unit 10 that controls the operation of each device within the automatic analyzer 1. The control unit 10 includes an operation unit 11, a processing unit 12, and a memory unit 13.
[0034] The processing unit 12 is composed of, for example, a hardware board and a computer, and includes a storage unit 13 such as a hard disk.
[0035] The storage unit 13 stores, for example, control parameters corresponding to each unit and sample information relating to various samples.
[0036] The processing unit 12 may be configured as hardware using a dedicated circuit board, or may be configured as software executed by a computer. When configured as hardware, it can be realized by integrating multiple arithmetic units that execute the processing on a wiring board, or in a semiconductor chip or package. When configured as software, it can be realized by installing a high-speed general-purpose CPU in a computer and executing a program that executes the desired arithmetic processing. It is also possible to upgrade existing devices using a recording medium on which this program is recorded. Furthermore, these devices, circuits, and computers are connected by a wired or wireless network, and data is sent and received as appropriate.
[0037] The operation unit 11 is composed of a display unit, which is a display, and input units such as a mouse, a keyboard, etc. The display unit displays various information related to the automatic analyzer 1 that is stored in the storage unit 13.
[0038] The above is the configuration of the automatic analyzer 1 of this embodiment.
[0039] Although the configuration of the automatic analyzer has been described as being equipped with a photometric unit 134 that performs immune system analysis as shown in Figure 1, the configuration of the automatic analyzer to which the present invention is applied is not limited to this, and the present invention can also be applied to automatic analyzers that are equipped with, for example, a biochemical detection unit, or a biochemical detection unit and an immune system detection unit, or even detection units for other analysis items, in addition to or instead of the biochemical detection unit.
[0040] Furthermore, the automated analyzer is not limited to a single analysis module configuration as shown in Figure 1, but can be configured to connect two or more analysis modules capable of measuring various identical or different analysis items and preprocessing modules that perform preprocessing via a transport device.
[0041] Next, the details of the analysis unit 100 in the automatic analyzer 1 will be described with reference to Figures 2 to 6. Figures 2 and 4 are top views of the analysis operation unit, and Figures 3 and 5 are side views of the outline of the analysis operation unit. Figure 6 is a top view of another type of analysis operation unit.
[0042] As shown in Figures 2 to 5, the analysis unit 100 includes a turntable 102, a B / F separation unit 104, a reaction solution suction nozzle 106, a replacement solution discharge nozzle 108, a photometry unit 134, a measurement flow path introduction nozzle 130, a cleaning solution storage container 122, a buffer solution storage container 120, an LFC (Liquid Flow Cleaning) container 124, a measurement flow path pump 136, a reaction solution suction / replacement solution discharge pump 140, a nozzle rotation motor 144, a rotation shaft 146, a turntable rotation motor 150, a turntable up / down motor 152, a rotation shaft 154, and the like.
[0043] The buffer solution reservoir 120 is provided on the turntable 102 and is a container for storing a buffer solution containing a luminescent substrate.
[0044] The cleaning liquid storage container 122 is provided on the turntable 102 and is a container for storing a cleaning liquid for cleaning the flow path from the measurement flow path introduction nozzle 130 to the photometry unit 134 .
[0045] The LFC container 124 is provided on the turntable 102 and is a container for storing a flow path cleaning liquid for cleaning the reaction liquid suction nozzle 106 and the measurement flow path introduction nozzle 130. The LFC container 124 is provided with a partition 124a that separates the flow path cleaning liquid into one for cleaning the reaction liquid suction nozzle 106 and one for cleaning the measurement flow path introduction nozzle 130.
[0046] In this embodiment, the B / F separation unit 104 is mounted on the turntable 102 on which a reaction vessel installation unit 110 for installing a reaction vessel 51 on the turntable 102, a buffer solution storage container 120, a cleaning liquid storage container 122, an LFC container 124, etc. are placed, and is configured to be movable vertically and horizontally together with these reaction vessel installation units 110, etc.
[0047] Also, as shown in Figures 2 and 4, on the turntable 102, the reaction vessel installation section 110, the washing liquid storage vessel 122, the buffer storage vessel 120, the LFC vessel 124, and the B / F separation section 104 are arranged radially around the rotation axis 154 of the turntable 102.
[0048] Furthermore, the distance between the measurement flow path introduction nozzle 130 and the transport mechanism 60 is such that when the measurement flow path introduction nozzle 130 is positioned vertically above the buffer solution storage container 120, the transport mechanism 60 is positioned vertically above the B / F separation unit 104. Then, when the measurement flow path introduction nozzle 130 aspirates the buffer solution from the buffer solution storage container 120, the operation of the transport mechanism 60 is controlled so that the reaction vessel 51 is placed on the B / F separation unit 104 or the reaction vessel 51 is recovered from the B / F separation unit 104.
[0049] Furthermore, the turntable 102 is arranged so that the trajectory of the B / F separator 104 caused by the rotation of the turntable 102 intersects with the trajectory of the reaction liquid suction nozzle 106 and the trajectory of the replacement liquid discharge nozzle 108 .
[0050] Next, the operation of the B / F separation cycle in the automatic analyzer 1 will be described.
[0051] 2, the reaction vessel 51 is placed in the B / F separation unit 104 by the transport mechanism 60. Since the B / F separation unit 104 is equipped with a magnet 105, the magnetic particles in the reaction solution held in the reaction vessel 51 are captured by the magnet 105, and the magnetic particles are separated from other substances.
[0052] Thereafter, as shown in FIG. 4, the turntable rotation motor 150 is driven to rotate the turntable 102 around the rotation axis 154 , and the B / F separation unit 104 moves below the reaction liquid suction nozzle 106 .
[0053] The turntable 102 is raised by driving the turntable up / down motor 152, and the reaction liquid suction nozzle 106 is inserted into the reaction vessel 51 of the B / F separation section 104 as shown in FIG.
[0054] Thereafter, the reaction liquid suction / replacement liquid discharge pump 140 is driven to suction the reaction liquid that has not been captured in the B / F separation section 104 .
[0055] Next, the turntable 102 is lowered by driving the turntable up / down motor 152, and then the nozzle rotation motor 144 is driven to rotate the replacement liquid discharge nozzle 108 around the rotation axis 146 above the B / F separation section 104.
[0056] Next, the turntable 102 is raised again by driving the turntable up / down motor 152 , and the replacement liquid discharge nozzle 108 is inserted into the reaction vessel 51 of the B / F separation section 104 .
[0057] Thereafter, the substitution liquid pumped up from the substitution liquid bottle 142 using the reaction liquid suction / substitution liquid discharge pump 140 is discharged into the reaction vessel 51 of the B / F separation section 104 .
[0058] Next, the overlap of the detection cycle and the B / F separation cycle in the automatic analyzer 1 will be described.
[0059] In the measurement cycle, the turntable rotation motor 150 is driven to rotate the turntable 102 around the rotation axis 154, and the reaction vessel installation part 110 moves directly below the measurement flow path introduction nozzle 130, as shown in Figures 2 and 3.
[0060] Thereafter, the measurement flow channel pump 136 is driven to suck the reaction solution placed in the reaction container placement section 110 and introduce it into the measurement flow channel 132 .
[0061] Thereafter, the turntable 102 is raised by driving the turntable up / down motor 152, and the measurement flow path introduction nozzle 130 is inserted into the buffer solution storage container 120 to aspirate the buffer solution.
[0062] Finally, the turntable 102 is raised by driving the turntable up / down motor 152 , and while the buffer solution is being sucked, the reaction vessel 51 is placed in and collected from the B / F separation section 104 by the transport mechanism 60 .
[0063] 2 to 5, the turntable 102 is capable of rotating and moving up and down, and the cleaning liquid reservoir 122, buffer solution reservoir 120, and B / F separation unit 104 are not limited to being arranged radially around the rotation axis 154 of the turntable 102. As shown in FIG. 6, the table 102A may be capable of moving horizontally and up and down, and the cleaning liquid reservoir 122, buffer solution reservoir 120, and B / F separation unit 104 may be arranged along the horizontal movement direction of the table 102A.
[0064] Next, the effects of this embodiment will be described.
[0065] The above-described automatic analyzer 1 includes a B / F separator 104 that captures magnetic particles in a reaction vessel 51 that contains a reaction liquid obtained by mixing a reagent and a specimen, a reaction liquid suction nozzle 106 that aspirates the reaction liquid (first liquid) in the reaction vessel 51, and a replacement liquid discharge nozzle 108 that discharges a replacement liquid (second liquid) into the reaction vessel 51 from which the reaction liquid has been aspirated. The B / F separator 104 is configured to be movable in vertical and horizontal directions. Alternatively, the B / F separator 104 that captures magnetic particles in a reaction vessel 51 that contains a reaction liquid obtained by mixing a reagent and a specimen, and a replacement liquid discharge nozzle 108 that discharges a replacement liquid (second liquid) into the reaction vessel 51 from which the reaction liquid has been aspirated. the reaction vessel 51 into which the reaction liquid has been aspirated; a reaction liquid suction nozzle 106 that aspirates the reaction liquid; a replacement liquid discharge nozzle 108 that discharges a replacement liquid into the reaction vessel 51 into which the reaction liquid has been aspirated; a photometry unit 134; a measurement flow path introduction nozzle 130 that introduces the reaction liquid into the photometry unit 134; a cleaning liquid storage container 122 that stores a cleaning liquid for cleaning the flow path from the measurement flow path introduction nozzle 130 to the photometry unit 134; and a buffer solution storage container 120 that stores a buffer solution containing a luminescent substrate; and the B / F separation unit 104, the cleaning liquid storage container 122, and the buffer solution storage container 120 are mounted on a turntable 102.
[0066] Therefore, during B / F separation, the reaction liquid suction nozzle 106 and the replacement liquid discharge nozzle 108 do not need to be moved up and down, and the turntable 102 can be moved up and down to perform both functions, thereby reducing the number of axes. This effect makes it possible to reduce the temperature control space by reducing the size of the device, thereby improving temperature control efficiency and reducing costs.
[0067] Furthermore, the turntable 102 can rotate and move up and down, and the cleaning solution storage container 122, the buffer solution storage container 120, and the B / F separation section 104 are arranged radially around the rotation axis 154 of the turntable 102, so that these components can be accessed simply by rotating the turntable 102, and the size of the turntable 102 can be prevented from becoming too large.
[0068] Furthermore, the table 102A can rotate and move up and down, and the cleaning solution storage container 122, the buffer solution storage container 120, and the B / F separation section 104 are arranged along the axial direction of the horizontal movement of the table 102A, so that these components can be accessed with horizontal movement only and the size of the table 102A can be prevented from becoming larger.
[0069] Furthermore, by having the trajectory of the B / F separation section 104 intersect with the trajectory of the reaction liquid suction nozzle 106 and the trajectory of the replacement liquid discharge nozzle 108, the necessary operations can be achieved with the minimum necessary driving mechanisms and movable shafts, thereby shortening the analytical operations and further miniaturizing the device.
[0070] Furthermore, by further providing an LFC container 124 that is provided on the turntable 102 and stores a flow path cleaning solution for cleaning the reaction liquid suction nozzle 106 and the measurement flow path introduction nozzle 130, the components required for immunoassay items can be efficiently arranged in an even smaller space.
[0071] Furthermore, the LFC container 124 is provided with a partition 124a that separates the flow path cleaning liquid into one for cleaning the reaction liquid suction nozzle 106 and one for cleaning the measurement flow path introduction nozzle 130, thereby enabling the necessary components to be efficiently arranged in a small space.
[0072] Furthermore, a transport mechanism 60 is further provided for transporting the reaction vessel 51 to the B / F separation section 104, and when the measurement flow path introduction nozzle 130 is positioned vertically above the buffer solution storage container 120, the transport mechanism 60 is positioned vertically above the B / F separation section 104; alternatively, a transport mechanism 60 is further provided for transporting the reaction vessel 51 to the B / F separation section 104, and when the measurement flow path introduction nozzle 130 aspirates the buffer solution from the buffer solution storage container 120, the transport mechanism 60 is controlled to place the reaction vessel 51 above the B / F separation section 104 or to recover the reaction vessel 51 from the B / F separation section 104. This allows the detection cycle and the B / F separation cycle to partially overlap, and makes it possible to shorten the cycle time even when both the up and down operations are used.
[0073] <Others> The present invention is not limited to the above-described examples, and includes various modifications. The above-described examples have been described in detail to clearly explain the present invention, and the present invention is not necessarily limited to those having all of the described configurations.
[0074] It is also possible to replace part of the configuration of one embodiment with the configuration of another embodiment, or to add the configuration of one embodiment to the configuration of another embodiment. It is also possible to add, delete, or replace part of the configuration of each embodiment with the configuration of another embodiment.
[0075] For example, there is no limit to the number of B / F separation units 104, reaction liquid suction nozzles 106, replacement liquid discharge nozzles 108, photometry units 134, measurement flow path introduction nozzles 130, washing liquid storage containers 122, buffer solution storage containers 120, LFC containers 124, etc. For example, one reaction liquid suction nozzle 106 or one replacement liquid discharge nozzle 108 may be provided in the middle of two B / F separation units 104.
