Description

Carriers comprising metal-organic frameworks and lipids and uses thereof

[0001] The present invention relates to a metal-organic framework (MOF)-lipid complex comprising a metal-organic framework (MOF) and a lipid, and uses thereof.

[0002] Even for the same drug, efficacy can vary significantly depending on the delivery method. Thus, a drug delivery system (DDS) is a drug delivery system (DDS) that efficiently delivers the required amount of medication while minimizing side effects and maximizing efficacy and effectiveness. In the pharmaceutical industry, DDS is a high-value, core technology with a high potential for success, generating economic benefits comparable to new drug development.

[0003] Recently, the development of gene therapy using gene drug delivery systems has been expanding among various drug delivery systems. For gene therapy to be successful and safe, it is crucial to deliver genes or gene regulatory factors to the target tissue. Representative examples of these genes or gene regulatory factors include mRNA, siRNA, and miRNA. Nucleic acids such as mRNA function to express specific proteins, allowing them to complement proteins deficient in genetic factors. Furthermore, the use of mRNA to express cancer markers or viral surface proteins enables the development of anticancer drugs and vaccines that activate the immune response in vivo. Furthermore, nucleic acids such as siRNA and miRNA, which can suppress the expression of specific proteins in vivo, are attracting attention as important tools for the treatment of cancer, genetic diseases, infectious diseases, and autoimmune diseases. However, direct delivery of these nucleic acids into cells is difficult and they are easily degraded by enzymes in the blood. Therefore, extensive research is being conducted to overcome this limitation.

[0004] Non-viral gene carriers, such as cationic liposomes and polymers, are attracting attention for the effective delivery of these substances. In particular, lipid nanoparticles have been actively studied as non-viral gene delivery vehicles. Lipid nanoparticles (LNPs) utilize substances existing in the body, such as phospholipids and cholesterol, and are thus particulate drug delivery vehicles with high bioavailability and affinity, as well as the ability to release and control drugs, and high stability against degradation by enzymes.

[0005] Accordingly, the inventors of the present invention confirmed that the delivery efficiency of lipid nanoparticles can be improved by applying a metal-organic framework, thereby improving the delivery efficiency, and completed the present invention.

[0011] The term "metal-organic framework (MOF)" as used herein refers to a porous material in which metal clusters and organic linkers (or organic bridging ligands) are connected by coordination bonds to form a three-dimensional structure, and various MOFs can be created depending on the selection of metal ions and organic ligands. The MOF is characterized by porosity in which empty spaces exist within the structure, and the pore size, porosity, three-dimensional structure, surface area, etc. can be designed in various ways depending on the type and bonding method of the metal ions and organic ligands that make up the MOF. Due to this porosity, MOFs not only have a very large surface area but also have an open pore structure, so they can transport a large amount of molecules or solvents compared to other porous materials known in the art, and when used as a catalyst or gas storage medium, they have the advantage of having many active sites, which can maximize efficiency. In addition, the MOF does not easily deform at high temperatures and has a rigid skeleton, so it has excellent chemical and thermal stability.

Any carrier commonly used in the art and pharmaceutically acceptable for the pharmaceutical composition may be used. Non-limiting examples of the above carriers include lactose, dextrose, maltodextrin, sucrose, sorbitol, mannitol, xylitol, erythritol, maltitol, glycerol, ethanol, starch, acacia gum, alginate, gelatin, calcium phosphate, calcium silicate, cellulose, methyl cellulose, microcrystalline cellulose, polyvinyl pyrrolidone, water, saline solution, sterile water, Ringer's solution, buffered saline, albumin injection solution, methylhydroxybenzoate, propylhydroxybenzoate, talc, magnesium stearate, or mineral oil. [0086] 2) Allow to react at 120℃ for 24 hours.

[0087] 4) The synthesized particles were washed three times each using DMF and ethanol, and dried at 80°C to obtain a powder.

[0137] Experimental Example 2: Cytotoxicity Evaluation of Metal-Organic Frameworks

[0138] To evaluate the cytotoxicity of the metal-organic framework produced in Experimental Example 1 above, the following experiment was performed.

[0139] Specifically, to evaluate the cytotoxicity of the metal-organic framework manufactured / synthesized in Experimental Example 1 above, mouse fibroblasts (L929) were first seeded in a 96-well plate at 1X10 per well. 4Cells were added and cultured for 24 hours in a 37℃, 5% CO2 incubator. After 24 hours, the existing medium in each well was removed, and the metal-organic framework particles to be tested were suspended in the medium at a concentration of 100, 50, 25, 10, or 1 ug / ml to make a test solution, and 100 ul was added to each well. The solution was cultured for 24 hours in a 37℃, 5% CO2 incubator, and the medium was removed. Next, the plate and cells were washed three times with PBS to remove particles attached to the surface. [0144] First, 1.0 g of agarose was added to 100 ml of 1x TAE buffer and melted at high temperature to make a 1% agarose gel. Then, 7 ul of Dyne StainingSTAR (EtBr substitute) was added to the agarose gel solution and mixed sufficiently, and the solution was poured into an electrophoresis gel forming mold and solidified. The solidified agarose gel was placed in an electrophoresis device, and 1x TAE buffer was added so that the gel was sufficiently submerged. The MOF manufactured in Experimental Example 1 was suspended in DW at a concentration of 10 mg / ml, and then diluted to concentrations of 5 mg / ml, 2.5 mg / ml, 1.25 mg / ml, 0.625 mg / ml, and 0.312 mg / ml to prepare (for MOFs that require ligand measurement, the ligand is suspended in DW at a concentration of 1 mg / ml). 200 ng / ul mRNA solution was prepared by adding 80 ul of 10 mM NaOAC to 20 ul of mRNA (1 ug / ul), and 1 ul of mRNA solution and 10 ul of MOF solution were mixed and incubated at room temperature for 10 minutes to induce the formation of mRNA-MOF complexes. In addition, mRNA control was prepared by mixing 1 ul of mRNA solution and 10 ul of DI. After mixing 5 ul of DNA ladder (50 bp) and 1 ul of DNA loading buffer, 5 ul was loaded onto an agarose gel, and 5 ul of mRNA control and 5 ul of mRNA-MOF complex were mixed in 1 ul of DNA loading buffer, respectively, and 5 ul was loaded onto an agarose gel.

[0145] As a result of the above experiment, it was confirmed that the metal-organic framework fabricated above can form a complex with RNA (Fig. 8).

[0146]

[0147] Experimental Example 4: Method for preparing a lipid carrier comprising a metal-organic framework

[0148] In order to manufacture a lipid-based carrier including the metal-organic framework manufactured in Experimental Example 1, the following experiments were performed.

[0149] First, mRNA-loaded lipid nanoparticles were manufactured using the following method.

[0150] 1) Prepare ethanol stocks with concentrations of 40 mM DOTAP (an example of ionized lipid), 25 mM DSPC (an example of phospholipid), 25 mM cholesterol (an example of sterol), and 6.25 mM DMG-PEG (an example of PEG-lipid).

[0151] 2) Take four types of lipids in the amounts shown in Table 1 below, mix them, and add EtOH so that the final volume becomes 25 ul. This is called Lipid-Mix.

[0152] 3) Prepare an mRNA solution with a concentration of 51.23 ug / ml using 10 mM NaOAc buffer.

[0153] 4) Add 25 ul of Lipid-Mix to 75 ul of mRNA solution, vortex for about 30 seconds to prepare mRNA-LNP solution, and incubate at room temperature for 10 minutes.

[0158] Next, a lipid carrier including a metal-organic framework based on the lipid nanoparticles manufactured above was manufactured by the following method.

[0159] 1) Prepare ethanol stocks with concentrations of 80 mM DOTAP (an example of ionized lipid), 25 mM DSPC (an example of phospholipid), 25 mM cholesterol (an example of sterol), and 6.25 mM DMG-PEG (an example of PEG-lipid).

[0167]

[0168] Experimental Example 5: Confirmation of RNA delivery efficiency of lipid carriers containing metal-organic frameworks.

[0169] In order to confirm the RNA delivery efficiency of the lipid carrier (MOF-LNP) containing the metal-organic framework manufactured in Experimental Example 4 above, the following experiment was performed.

[0170]

[0171] 5.1: Confirmation of RNA delivery efficiency according to lipid components of lipid carriers containing metal-organic frameworks

[0172] In relation to the lipid delivery vehicle including the metal-organic framework manufactured in Experimental Example 4 above, the following experiment was performed to compare and analyze the RNA delivery efficiency according to the presence or absence of each lipid component.

[0173] Specifically, HEK293 (human embryonic kidney cell) cells were seeded in 24-well at about 30,000 cells / well and cultured for 24 hours at 37°C, 5% CO2. Thereafter, the cell medium was replaced with 500 μl of Opti-MEM, and the complex volume that would contain 0.1 μg of mRNA contained in the LNP (Example 1) manufactured in Experimental Example 4 and the lipid carrier (LNP-MOF) including various lipid combinations and metal-organic frameworks was calculated, and each volume was added to each well to treat the cells, and incubated at 37°C for 16 hours. After 16 hours, Opti-MEM was removed, 200 ul of lysis buffer was added to each well, incubated for 15 minutes, 50 ul of luciferin and 50 ul of the above sample were added to a 96-well white plate, and luminescence was measured using a microplate reader.

[0174] In the lipid carriers containing the above metal-organic frameworks, MOF-808 or UIO-66-ED were used as examples of the metal-organic frameworks, and the efficiency of delivery of the loaded mRNA was analyzed by confirming the protein expression level of cells according to the use of the carriers of each group. The protein expression level was set to '1' when mRNA was introduced using the carrier of Example 1, which is a lipid nanoparticle that does not contain a metal-organic framework (composed of DOTAP, DSPC, cholesterol, and DMG-PEG - only MOF was excluded from Group 1), and the expression level was expressed based on this.

[0175] As a result, for lipid carriers containing zirconium-based MOF-808 as an example of a metal-organic framework, it was confirmed that the groups excluding PEG-lipid (groups 4, 6, 7, and 8) had significantly higher protein expression levels compared to the other groups (Table 3 and Fig. 9).

[0176] Example Group Composition Protein Expression Level (Compared to Example 1) MOF / mRNA wt ratio MOFDOTAPDSPCCholesterol DMG-PEG2 2 MOF-80 8 OXOO 2.68 13.03 3 OOXO 0.48 13.044 OOX 2 7.85 13.055 OXXO 5.63 13.066 OOX 11.67 13.077 OOX 2 8.31 13.088 OXX 5 0.58 13.0

[0177]

[0178] In addition, in the case of lipid carriers containing zirconium-based UIO-66-ED as another example of metal-organic frameworks, it was confirmed that the groups in which PEG-lipid was excluded (groups 4, 6, 7 and 8) had significantly higher protein expression levels compared to the other groups (Table 4 and Fig. 10).

[0179] Example Group Composition Protein Expression Level (Compared to Example 1) MOF / mRNA wt ratio MOFDOTAPDSPCCholesterol DMG-PEG9 1 UIO-66-ED OOOO 1.03 13.010 2 OX OO 2.05 13.01 13 OO X 0.02 13.01 24 OO X 10.88 13.01 35 OO X 0.11 13.01 46 OX OX 3.98 13.01 57 OO X 5.44 13.01 68 OX XX 4.08 13.0

[0180]

[0181] Based on the above results, it can be seen that in the case of a lipid carrier including a metal-organic framework, the RNA delivery efficiency is significantly improved when PEG-lipid, which is one of the components of the lipid nanoparticle, is excluded.

[0182]

[0183] 5.2: Confirmation of RNA delivery efficiency of lipid carriers containing various types of metal-organic frameworks and excluding PEG-lipids

[0184] In order to confirm whether the RNA delivery efficiency of a lipid carrier containing a metal-organic framework and excluding PEG-lipid, as confirmed in Experimental Example 5.1 above, shows the same trend when using other metal-organic frameworks, the following experiment was performed.

[0185] Specifically, lipid carriers of groups 4, 6, 7 and 8 excluding PEG-lipid were prepared using aluminum-based MIL-101-NH2(AL), magnesium-based Mg-MOF-74, zirconium-based UIO-66 or iron-based MIL-88-NH2(Fe) as metal-organic frameworks, and the RNA delivery efficiency of the carriers was confirmed by the method described in Experimental Example 5.1. Meanwhile, the protein expression level when the carrier of Example 1, which is a lipid nanoparticle without a metal-organic framework, was used was set to '1', and the expression level was expressed based on this.

[0186] The above analysis results confirmed that even when using various types of metal-organic frameworks, the RNA delivery efficiency of the groups excluding PEG-lipid (groups 4, 6, 7, and 8) was significantly increased compared to the control group (Tables 5 to 8).

[0191]

[0192] Based on the above results, it can be seen that the lipid carrier containing a metal-organic framework has significantly improved RNA delivery efficiency compared to lipid nanoparticles not containing a metal-organic framework when PEG-lipid, which is one of the components of the lipid nanoparticle, is excluded.

[0193] 5.3: Confirmation of RNA delivery efficiency of lipid carriers according to metal-organic framework content ratio

[0194] In relation to lipid carriers containing metal-organic frameworks, the following experiments were performed to determine the RNA delivery efficiency of lipid carriers according to the metal-organic framework content ratio.

[0195] Specifically, a lipid carrier including a metal-organic framework was manufactured based on the method for manufacturing group 8 of Experimental Example 4, and the MOF suspension used in the above 'Step 4' was used at various concentrations (2.5 mg / ml, 5 mg / ml, 20 mg / ml, 40 mg / ml, 60 mg / ml, and 80 mg / ml) to control the content ratio of the metal-organic framework, and MOF-808 was used as an example of the metal-organic framework (Table 9).

[0197]

[0198] The RNA delivery efficiency of the lipid carrier comprising the metal-organic framework manufactured above was confirmed, and it was confirmed that the RNA delivery efficiency was excellent when MOF suspensions were used at concentrations of 5 mg / ml (12.5 ug), 20 mg / ml (50 ug), 40 mg / ml (100 ug), and 60 mg / ml (150 ug) (Fig. 11 and Table 9). Therefore, it can be seen that the RNA delivery efficiency of the lipid carrier can be further improved when it is contained in a specific ratio of the metal-organic framework.
