Description

[0001] DESCRIPTION

[0002] New derivatives useful as antibacterial agents

[0003] The present invention relates to a compound of formula I for the treatment and / or prevention of a disease caused by a bacterium, in particular where the disease is caused by a gram-negative bacterium such as Acinetobacter baumanii and Escherichia coli.

[0004] BACKGROUND OF THE INVENTION

[0005] Gram-negative bacilli are highly efficient at acquiring antimicrobial resistance through genomic, transcriptomic, and proteomic changes (see Kung VL, Ozer EA, Hauser AR. The accessory genome of Pseudomonas aeruginosa. Microbiol Mol Biol Rev. 2010;74(4):621-41 ; and Yelin I, Kishony R. Antibiotic Resistance. Cell. 2018;172(5):1136-1136. e1 ). The problem of antimicrobial resistance is compounded by the immediate threat of a reduction in the discovery and development of new antibiotics, as highlighted by the World Health Organization (WHO) (Tacconelli E, Carrara E, Savoldi A, et al. Discovery, research, and development of new antibiotics: the WHO priority list of antibiotic-resistant bacteria and tuberculosis. Lancet Infect Dis. 2018;18(3):318-327) and other European institutions (see, e.g., Árdal C, Baraldi E, Ciabuschi F, et al. To the G20: incentivizing antibacterial research and development. Lancet Infect Dis. 2017;17(8):799-801 ).Consequently, new solutions are urgently needed in the field of antimicrobial therapy, such as the development of new therapeutic strategies such as the use of non-antibiotic agents and / or the repurposing of drugs for use as monotherapy or in combination with one of the few clinically relevant antibiotics.

[0006] Therefore, it would be desirable to have new antibacterial agents with improved properties for the treatment and / or prevention of a bacterial disease caused by gram-negative bacteria.

[0080] Procedures . Analytical grade solvents were used for all reactions. Argon was used to carry out the reactions in an inert atmosphere. The reagents used for the reactions were purchased from Sigma Aldrich or Fluorochem. The reactions under microwave irradiation were carried out in an InitiatorTM device (Biotage®). Melting points were recorded with a Büchi Melting point M-560 apparatus. The NMR spectra of 1 H and 13C were recorded on a Bruker AVI II 300 MHz BACS60 or Bruker AV 500 MHz spectrometer located at the Research Support Center of the Complutense University of Madrid. The abbreviations used are s = singlet, d = doublet, t = triplet, q = quadruplet, dd = doublet of doublets, m = multiplet. Coupling constants (J) are expressed in Hz. Mass spectra were acquired on a Thermo Mod. Finnigan™ LXQ™ spectrometer coupled to a high performance liquid chromatograph (HPLC), using electrospray ionization (ESI). Pre-coated aluminum foils (ALUGRAM® Xtra SIL G / UV254) were used for thin layer chromatography. Column chromatographies were performed on silica gel 60 (Merk) manually or automatically using the IsoleraOne equipment (Biotage®).The exact molecular mass was determined using an Agilent 1200LC liquid chromatograph coupled to an Agilent 6500 mass spectrometer with an ESI / APCI ionization source and a hybrid quadrupole-time-of-flight (QTOF) analyzer located at the Mass Spectrometry Service of the Institute of General Organic Chemistry (IQOG-CSIC). The purity of the final compounds was verified to be >95% by HPLC analysis. The HPLC conditions for purity assessment were as follows: Surveyor HPLC equipped with a Surveyor PDA plus a UV-VIS detector; ZORBAX® SB-C18 column (3.5 µm, 4.6 mm x 50 mm); H2O / CH3CN gradient elution from 100 / 0 to 0 / 100 for 5 min; flow rate 800 µL / min; UV wavelength 254 nm. All final compounds are >95% pure by HPLC-MS.

[0081] Three different gradient conditions were used:

[0082] Gradient I: 23 eC, flow rate of 0.5 mL / min. Gradient elution with mobile phases (A) H2O containing 0.1% volume / volume (v / v) formic acid and (B) CH3CN containing 0.1% (v / v) formic acid. The gradient conditions were initially 5% B, increasing linearly to 95% B over 5 min, remaining at 95% B for 1.45 min, and then decreasing to 10% B over 0.55 min.

[0083] Gradient II: 23 e C, flow rate of 0.8 mL / min. Gradient elution with mobile phases (A) H2O containing 0.1% volume / volume (v / v) formic acid and (B) CH3CN containing 0.1% (v / v) formic acid. The gradient conditions were initially 5% B, increasing linearly to 100% B over 3 min, remaining at 100% B for 1.45 min, and then decreasing to 5% B over 0.55 min.

[0084] Gradient III: 23 eC, flow rate of 0.8 mL / min. Gradient elution with mobile phases (A) H2O containing 0.1% volume / volume (v / v) formic acid and (B) CH3CN containing 0.1% (v / v) formic acid. The gradient conditions were initially 10% B, increasing linearly to 95% B over 5 min, remaining at 95% B for 4 min, and then decreasing to 0% B over 1 min.

[0208] Example 2. Antibacterial activity in reference strains of A. baumannü and E. coli

[0209] Bacterial strains and growth conditions

[0210] Two gram-negative bacterial reference strains were used: A. baumannü ATCC 17978 and E. coli ATCC 25922. The strains were thawed from a collection stored at -80 e C and grown on sheep blood agar plate (Columbia Agar 5% Sheep Blood, ThermoFisher, Spain) at 37 e C under stirring for 24 hours.

[0211] Microdilution assays

[0212] The MIC, i.e., the minimum concentration of compound necessary to inhibit bacterial growth, was evaluated. For this purpose, microdilution assays were performed in duplicate for the three study strains. This assay was carried out in 96-well U-bottom microplates (Microdur, Spain). The procedure was as follows: first, 100 pL of the study compound was added to the wells of the first column, starting from an initial concentration of 64 or 128 mg / L. Subsequently, 50 pL of Luria Bertani (LB) culture medium was added to the remaining wells, except for those used as sterility and growth controls. After this step, serial dilutions were made.

[0213] Each well was then inoculated with 50 pL of a bacterial culture at a concentration of 1x10 A6 colony-forming units (CFU) / mL, derived from a 0.5 McFarland (McF) suspension. Only 100 pL of medium was added to the sterility control well to ensure it was not contaminated. Bacterial culture, without drug, was added to the growth control well to verify that our strain was growing properly.

[0219] Microdilution assays

[0220] The MIC of 1 1 thiophene derivatives was evaluated. Microdilution assays were performed in duplicate for the 17 study strains. This assay was carried out in 96-well U-bottom microplates.

[0221] The procedure was as follows: first, 100 pL of the test compound was added, starting from an initial concentration of 64 or 128 mg / L, to the wells of the first column. Subsequently, 50 pL of LB culture medium was added to the remaining wells, except for those used as sterility and growth controls. After this step, serial dilutions were made.

[0222] Each well was then inoculated with 50 pL of a bacterial culture at a concentration of 1x10 A6 CFU / mL, which comes from a 0.5 McF suspension. Only 100 pL of medium was added to the sterility control well to ensure it was not contaminated. Bacterial culture, without drug, was added to the growth control well to verify that our strain was growing properly. Results
