[0003] 002 THE MOISTURIZING CREAM WITH LONG-LASTING ANTISEPTIC PROTECTION FOR HANDS, through the technology employed, aims to be a dermatologically tested moisturizing cream with prolonged action of up to 4 hours of antiseptic protection, making its action more effective by avoiding the need for reapplication in short periods to ensure the protective effect against bacteria and microorganisms that cause diseases such as diphtheria, whooping cough, meningitis, gastroenteritis, urinary infections and cholera, and also antiseptic protection against viruses such as Sars-Cov2 and variants, Influenza, flu, measles, polio, hepatitis A and monkeypox, basically consisting of a cream that has the following formula: Deionized water - 100.00% Qsp (solvent); Polyacrylamide, C13-14 Isoparaffin and Laureth 7 - 1.00 to 3.00% (emulsifier and viscosity agent); Paraffinum liquidum - 0.50 to 3.00% (skin conditioner); Alcohol - 15.00 to 20.00% (solvent);Chemynol F - 0.50 to 1.00% (preservative); PHTALOX® Bioactive functional Phthalocyanine - 0.05% (active), which continuously produces reactive oxygen in the presence of molecular oxygen, with enormous diversification of markets in which the product can be used, such as: in the health area in general clinics, physiotherapy clinics, veterinary clinics, dental clinics, laboratories, hospitals, health care, patients, etc. In the food area in fast food chains, industries, restaurants, catering, food service, gastronomic events, etc. In the retail area in pharmacies, supermarkets, gyms, shopping malls, hotel chains, beauty salons, schools, colleges, public transportation, urban private transportation, bus stations, airports, etc., dealing with a solution of an evolutionary nature, whose practical result obtained reports to a differentiated and effective condition of use.;
[0004] STATE OF THE ART oo3 With the sole and exclusive purpose of providing veracity to the preamble of this description, the applicant will provide in the following paragraphs a brief but consistent explanation of the state of the art for the problems and drawbacks existing in antiseptic moisturizing compositions, where it will be possible for a person skilled in the art to recognize their deficient aspects, in order to later clearly identify the novelty requirement in the invention now claimed, as mandatory in the Industrial Property Law. oo4 The main objective of HPPC (Personal Hygiene, Perfumery and Cosmetics) products is to bring well-being to the general population, providing improvements in health, personal hygiene, oral hygiene, sun protection, treatments and cleansing of skin, hair and others, in addition to assisting in the prevention of diseases, such as Covid-19. (ABIHPEC, 2023. https: / / www.abihpec.org.br / publicacao / panorama-do-setor / ).005 As is generally known, outbreaks and the emergence of a series of diseases transmitted by direct or indirect contact with people suffering from illnesses such as flu, colds, rhinitis, sinusitis, coughs and their respective variations and consequences are increasingly frequent, causing true epidemics and even pandemics. This is the case, for example, with the Sars-Cov2 virus and influenza "A", or swine flu, whose presence has already been detected in all countries, causing a large number of victims, including fatalities. oo6 In this same regard, the agencies responsible for public health and governments in general have been massively disseminating the main precautions that must be taken to prevent the spread and contagion, as well as the measures that must be adopted by all those who suspect or present the symptoms caused by these devastating diseases.oo7 Among the main recommendations are the adoption of wearing a mask and washing your hands thoroughly and frequently, if possible, using products with antiseptic properties that guarantee antimicrobial action. This provides better protection for the individual and also minimizes the spread of pathogens, whether viruses, bacteria, yeasts, or fungi, especially after direct contact with people, objects, and surfaces in general. oo8 Human coronaviruses can survive for more than 28 days at room temperature on surfaces including metal, glass, and vinyl. Hands are a very important vector for the transmission of pathogenic microorganisms if proper hygiene is not maintained. (Saha, 2021 doi: 10.18683 / germs.2021.1278).oo9 However, it is known that it is extremely difficult to change the needs and habits of even almost unconscious actions such as touching or holding various surfaces (doorknobs, handrails, tables, public transportation bars, seats, glasses, cutlery, pens, keyboards, etc.), and greeting people (friends, clients, family, relatives, colleagues, etc.), which makes controlling infections through physical contact very difficult. To further aggravate the situation, it is known that it is not always possible to count on a restroom or similar nearby place with running water and soap to perform the minimum possible hand hygiene more consistently. oio Current methods of inactivating viruses and bacteria basically consist of washing hands with soap and applying alcohol-based hand sanitizer. It is known that 95% of the world's population does not wash their hands frequently.Pathogenic microorganisms are transmitted from one sick person to another through saliva droplets when a person sneezes into their hand, coughs, picks up objects, or touches contaminated surfaces such as cell phones, tables, cutlery, doorknobs, handrails, keyboards, and so on, and then greets others. This initiates the chain of transmission through the hands, mouth, nose, and eyes. Washing hands with soap for up to 10 seconds does not eliminate contamination by potentially pathogenic microorganisms; washing them for 20 seconds with soap and water is necessary for complete hand asepsis, which is not usually the case. There are two groups of hand sanitizers on the market: non-ethyl alcohol-based sanitizers and ethyl alcohol-based sanitizers. The first group has as its most common active ingredient benzalkonium chloride, a quaternary ammonium, widely used as a disinfectant. (Golin, 2020 https: / / doi.Org / 10.1016 / j.(ajic.2020.06.182) Benzalkonium chloride can be irritating to the skin and, in some cases, can cause allergies, i.e., it is a sensitizer, and is also quite irritating to the eyes. Therefore, this compound is not considered harmless due to its irritant properties, even in small concentrations. In addition to problems related to toxicity, the indiscriminate use of benzalkonium chloride leads to the development of resistant strains of microorganisms, as well as cross-resistance to antibiotics, a phenomenon that occurs when exposure to one chemical compound brings adaptive advantages to another chemical compound (Pereira, 2019 https: / / doi.org / 10.1128 / AEM.00377-19).hi2 The use of 70% alcohol gel as a sanitizing product, although relatively effective in inactivating viruses and bacteria, has brought other drawbacks, starting with dry skin and sticky hands due to its gel formulation. This causes discomfort for many people, who may choose not to use this type of product due to its obvious negative consequences. It has also been found that many of these products have flammable characteristics, generating so-called "invisible fire," which poses a risk of fires. This is a major problem, as it jeopardizes the safety of many people, especially children, the physically disabled, and the elderly. Hospitalization may even be necessary if the correct conditions for using this product are not properly observed, leading to serious burns. In addition to the aforementioned drawbacks, the main negative characteristic that many people are unaware of is that alcohol's sanitizing effect is instantaneous, meaning the product needs to be in contact with microorganisms to exert its effect. Therefore, after the alcohol evaporates, which happens very quickly, surfaces are again susceptible to microbiological contamination. Since the antiseptic action of alcohol is not prolonged, the product must be reapplied frequently, especially for healthcare professionals who are in constant contact with contaminating surfaces and substances, which can lead to the negative effects mentioned above. hi5 The skin's main function is to protect the body, acting as a barrier against infectious agents, exogenous substances, mechanical stress, and many other factors. For the skin to perform its function fully, the integrity of the skin barrier and the level of hydration are extremely important factors, as they ensure the maintenance of healthy skin tissue and the protection of the body as a whole (Nolan, 2012. DOI: 10.1111 / Í.1529-8019.2012.01504.x). It is already known that the frequent use of 70% alcohol-based hand sanitizers has very negative consequences for skin tissue, as they are quite aggressive, also causing skin dehydration and, consequently, degrading the skin barrier. It can also cause skin irritation, allergies and exacerbation, as well as poisoning. It is also widely known that the excessive use of ethanol-based hand hygiene products, especially as a preventative method against coronavirus contamination, can increase the risk of infections due to skin problems. These problems are primarily caused by the damage these products cause to the skin, thus decreasing its ability to act as a barrier against potential pathogens (Tachikawa, 2020 https: / / english.kyodonews.net / news / 2020 / 02 / 690b9e1c9b86-focus-overreliance-on-hand-sanitizers-may-increase-risk-of-virus-infection.html). What's the big deal? Furthermore, cutaneous exposure to alcohol on immature skin can lead to local and systemic intoxication reactions, making children the most susceptible to this type of adverse effect. Thus, it can be concluded that prolonged and frequent use of ethanol-based hand sanitizers can result in dermal toxicity, in addition to potentially fatal consequences if accidentally ingested. (Mahmood, 2020 https: / / doi.Org / 10.1016 / j.scitotenv.2020.140561 ).
[0005] 018 The FDA has also issued warnings about possible adverse effects from the use of alcohol-based hand sanitizers, stating that vapors from these products are causing symptoms such as dizziness, malaise, nausea, and headaches, and can also cause serious poisoning, especially in children. These cases of alcohol poisoning are more common when these hand sanitizers are used in enclosed, poorly ventilated areas, such as inside a car. (FDA, 2021 https: / / www.fda.QOv / druQs / fda-druq-safetv-podcasts / fda-warns-vapors-alcohol-based-hand-sanitizers-can-have-side-effects). hi9 Sanitizing agents can come in various forms, including semi-solid, liquid, spray, and foam formulations. Semi-solid formulations include creams, lotions, ointments and gels, intended for topical application, that is, on the skin.Ointments are pharmaceutical forms for external use consisting primarily of fatty excipients or polyethylene glycols. They may be anhydrous or contain a very small amount of water, with a very significant occlusive effect. They are composed of lipophilic substances, consistency-giving excipients, and active ingredients. (Ferraz, https: / / edisciplinas.usp.br / pluginfile.php / 3395742 / mod_resource / con tent / 1 / Texto%20FFP.pdf).
[0006] 020 Gels are oil-free formulations obtained from the hydration of gelling polymers, with a high concentration of water. Their basic composition includes gelling agents, preservatives, humectants, and active ingredients.
[0007] 021 Creams and lotions are emulsions for external use, characterized by a biphasic system of two immiscible liquid phases, in which one of the phases is dispersed, consisting of small droplets, uniformly within the other, and the two phases are stabilized through the use of an emulsifying agent. (Khan, 2011 https: / / www.researchqate.net / publication / 260210690 Basics of pharmaceutical emulsions A review). 022 Emulsions can be oil-in-water (O / W) or water-in-oil (W / O), depending on their composition and the emulsifiers used in the formulation. O / W emulsions, in which the oil is dispersed in the aqueous phase, have a lighter, less occlusive sensory feel, unlike W / O emulsions. In general, creams consist of a phase in which water is present, a chelating agent, a humectant, and, if necessary, a water-soluble thickening agent. The oil phase contains emollients, emulsifying agents, antioxidants, and fat-soluble thickeners. A preservative is also incorporated into the formulation, in addition to the active ingredients, and the timing of inclusion depends on their physical and chemical characteristics.
[0008] 023 Lotions have basically the same composition as creams, but they are emulsions that have a more fluid consistency and can be used on larger areas of the skin, as the consistency of the formulation directly influences the spreadability of the product.
[0009] 024 The prior art reveals Brazilian patent BR 10 2021 003083-6, published on 08 / 30 / 2022, under the title PROLONGED-ACTING ANTISEPTIC PRODUCT FOR SKIN AND OBJECT SURFACES FOR THE PREVENTION OF COVID-19, owned by Jean Marc Millet, which discloses a product for asepsis of hands, or other surfaces. It forms a protective, visible and slightly sticky film in the first 5 minutes after application, ensuring (unlike rapidly evaporating alcohol gels) the minimum contact time with germs necessary for their inactivation (5 to 10 minutes, NT26 / 2020 Anvisa); It has a remaining action thanks to this surface film that becomes invisible, but provides support and protection to the substances, and is also little affected in the presence of organic matter, for a period of 3 to 6 hours during which the minimum contact time is also reached. There is also a synergy in the antiseptic action between the selection of high-concentration surfactants that form the film and the potentially irritating antiseptic substance chlorhexidine, which allows for a reduction in the amount of the latter, while maintaining the same level of antiseptic action. It is available in liquid, spray, gel, lotion, or foam forms. It contains moisturizers such as hyaluronic acid, which, due to its hydrophilic properties, contributes to the physical structure of the film and also has a moisturizing and protective effect on the skin, preventing microcracks, which are entry points for viruses and bacteria into the human body. The subject matter of this patent does not present any characteristics identical or similar to the new composition and improvements proposed in this new application.
[0010] 025 In view of the above in the fundamentals of the technique, with the passing of time and the accumulation of experience, the facts pointed in the direction of the development of a new moisturizing cream with prolonged antiseptic protection, being quite versatile and efficient, where its unique molecular structure eliminates the negative aspects known in the State of the Art, which is presented in this patent.
[0011] PURPOSE OF THE PATENT
[0012] 026 In order to overcome the problems presented, the inventor, through this application, seeks the Invention Patent, entitled MOISTURIZING CREAM WITH LONG-LASTING ANTISEPTIC PROTECTION FOR HANDS, which fundamentally aims to manufacture a moisturizing cream with prolonged antiseptic protection against microorganisms that cause various illnesses, standing out in the consumer market due to the fact that it presents a differentiated product in relation to its use, also obtaining a low-cost and highly efficient product.
[0013] 027 With many years in the market, and always seeking innovations and practical solutions to differentiate his products from other competitors, the inventor developed this simple and innovative product, capable of overcoming all the limitations and drawbacks reported in the State of the Art in an extremely efficient and functional way.

[0016] 030 Another objective is to provide a moisturizing cream that can also be used on dry feet, especially for those with diabetes, as dryness causes cracks in the feet.
[0017] 031 Another advantage is that the new formulation is dermatologically tested, has a moisturizing composition that prevents dry skin, has a low alcohol content, therefore, it is not a flammable product, and is 100% vegan.
[0018] 032 Another great advantage is the enormous diversification of markets in which the product can be used, such as: in the health area in general clinics, physiotherapy clinics, veterinary clinics, dental clinics, laboratories, hospitals, health care, patients, etc. In the food area in fast food chains, industries, restaurants, catering, food service, gastronomic events, etc. In the retail area in pharmacies, supermarkets, gyms, shopping centers, hotel chains, beauty salons, schools, colleges, public transportation, private urban transportation, bus stations, airports, etc.
[0019] 033 It is important to understand that this Invention Patent presented herein is illustrated for demonstrative purposes and is not limitative in its application of the details, dimensions, capabilities, and steps described. It is capable of other embodiments and of being practiced or executed in a variety of ways, without logically departing from the main inventive scope of the subject matter whose protection is claimed.
[0020] 034 The composition of the MOISTURIZING CREAM WITH LONG-ACTING ANTISEPTIC PROTECTION FOR HANDS, which is the subject of this present Invention Patent application as a long-acting antiseptic agent with up to 4 hours of protection, was part of studies carried out by the NB-3 Laboratory of Clinical Molecular Virology (LVCM) in the Department of Microbiology of the Institute of Biomedical Sciences of the University of São Paulo, headquartered at Av. Prof. Lineu Prestes, 1374, Butantã, São Paulo, SP, CEP 05508-900, which follows all the Biosafety regulations of the WHO - World Health Organization and complies with Good Laboratory Practices (GLP).
[0021] 035 In order to carry out tests to prove the effectiveness of the long-acting antiseptic cream, which can be incorporated into pharmaceutical-cosmetic formulations with up to 4 hours of antiseptic protection, the following cream formulation was developed for the tests, consisting of: Deionized water - 100.00% Qsp (solvent); Polyacrylamide, C13-14 Isoparaffin and Laureth 7 - 1.00 to 3.00% (emulsifier and viscosity agent); Paraffinum liquidum - 0.50 to 3.00% (skin conditioner); Alcohol - 15.00 to 20.00% (solvent); Chemynol F - 0.50 to 1.00% (preservative); PHTALOX® Bioactive functional Phthlalocyanine - 0.05% (active), which continuously produces reactive oxygen in the presence of molecular oxygen.
[0022] 036 One of the studies carried out analyzed the virucidal action of samples of PHTALOX® 0.05% moisturizing cream, PHTALOX 0.25% cream gel, control alcohol gel, Ph01 alcohol gel, Ph02 alcohol gel, Ph03 alcohol gel, HM01 alcohol gel, HM02 alcohol gel and HM03 alcohol gel, regarding exposure to the Coronavirus strain Sars-Cov-2 (COVID-19). 037 The experiment used Reconstructed Human Epidermis (SkinEthic™ RHE) from the company EPISKIN; seeded with Vero CCL-81 cell culture with 1x10 5 cells / well in 24-well plate and Sars.CoV2 / human / Bra / SP02cc / 2020 Virus (Genebank accession number MT350282) at a concentration of 1x10 TCID 50 / MI, as per Table 1.
[0023] 038 Another study carried out analyzed the virucidal action of samples of PHTALOX 0.05% moisturizing cream, with regard to exposure to the Influenza A virus strain Influenza A pandemic H1N1 strain ICB - 2022 / 0042, as shown in Table 2 below:
[0024] 039 Based on the observation of cytopathic effect, cytotoxicity and comparison of Real-Time RTPCR results, we obtained the following inhibition percentages of the pandemic Influenza A H1 N1 virus (Influenza A pandemic H1N1 strain ICB - 2022 / 0042), as shown in Table 3. Table 3: Result of the Influenza A H1 N1 viral inhibition test
[0025] 040 Another study carried out analyzed the virucidal and antimicrobial action of the moisturizing cream with moisturizing protection for hands PHTALOX® 0.05%. Virucidal action test against the viruses SARS-Cov-2, Influenza A H1 N1, Monkeypox virus, and bactericidal action, against the microorganisms: Staphylococcus aureus ATCC6538; Pseudomonas aerugionosa ATCC9027; Escherichia coli ATCC8739; Salmonella Choleraesuis ATCC10708 and Leptospira Biflexa (Strain Pathoc 1) as shown in Table 4 below:
[0026] 041 MDCK cell cultures seeded 1x10 were used. 5cells / well in 24-well plate and Influenza A pandemic H1 N1 virus at a concentration of 1x10 3 TCID 50 / mL. Vero CCL-81 cell culture seeded 1x10 5 cells / well in 24-well plate and Monkeypox virus (MPXV - Genbank accession number ON751 962.1) and SARS-Cov-2 / BQ1.1 / human / Bra / SP-HIEA / ICB / 11 / 2022 virus (no Genebank accession number) at a concentration of 1x10 3 TCID 50 / mL.
[0027] 042 Based on the above, the "Virucidal and Antimicrobial Action" of the moisturizing cream with antiseptic protection for hands is 99.78% against bacteria that cause diseases such as Diphtheria, Whooping Cough, Meningitis, Gastroenteritis, Urinary Tract Infections, Cholera, among others, and 100% against viruses that cause diseases such as Influenza, Measles, Rubella, Polio, Hepatitis A, Monkeypox, among others. 043 Another study analyzed the determination of the product's cytotoxicity to estimate the initial doses for oral systemic toxicity tests using the human epidermal culture model in samples of PHTALOX 0.05% moisturizing cream.
[0028] 044 The experiment used in vitro Reconstructed Human Epidermis from human keratinocytes cultured on a polycarbonate membrane in a chemically defined medium (SkinEthicTM RHE) from the company EPISKIN.
[0029] 045 SkinEthic™ RHE cultures were received at the laboratory 24 hours before the experiment. Each transwell was removed from the solid medium, transferred to a 24-well plate in liquid maintenance medium, and maintained in an incubator at 37°C with a 5% CO2 atmosphere.
[0030] 046 PHTALOX® 0.05% moisturizing cream was investigated in 5 different action times (5 minutes, 1 hour, 2 hours, 3 hours and 4 hours).
[0031] 047 The transwells with SkinEthic™ RHE were treated with a layer of the test product PHTALOX® 0.05% moisturizing cream on the surface of the treated SkinEthic™ RHE for 5 min, 1 hour, 2 hours, 3 hours, and 4 hours. After inoculation, the plate was kept in an oven at 37°C with 5% CO2. After this period, the plates were observed under an optical microscope to verify the integrity of the cell mat and determine the cytotoxicity of the tested product. Then, the wells were completed with 600 µl of culture medium (DMEM) supplemented with 2.5% fetal bovine serum. The plate was kept in an oven at 37°C with a 5% CO2 atmosphere for 72 hours in a humid chamber. 048 After 72 hours of incubation, the plates were observed again under an optical microscope to verify the integrity of the cell mat in comparison with the negative control. And for the determination of cytotoxicity (morphological changes in the host cell caused by the Phitta Cream product inoculated in 5 min., 1 hour, 2 hours, 3 hours, and 4 hours), the cell mat was compared with the positive control. Immediately after, microscopic analysis was performed. After incubation, cell viability was analyzed by incorporating neutral red. Subsequently, the 50% cell lethality index (IC50) was calculated, and the LD50 of the product under evaluation was estimated using a mathematical model. Positive control: Sodium Lauryl Sulfate. After collection, the cell plate was fixed and stained with Naphtol Blue Black (Sigma-Aldrich).
[0032] 049 The cytotoxicity and LD50 results are shown in Table 05 below. Table 05: Product action time, sample identification, observation of the cell carpet, observation of cytotoxic effect and LD50.
[0033]
[0034] 050 Based on the observation of cytotoxicity and LD50 (lethal dose for 50% of exposed individuals) for acute oral systemic toxicity and survival percentages to cytotoxic action in human epithelial cells, as per Table 6:
[0035] Table 6: Cytotoxicity test results and LD50
[0036] 051 The object of this present patent application was also part of a study carried out by PróLab - Laboratório de Biotecnologia SS Ltda, headquartered at Rua Angelo Maglio, 65, Vila Yara, Osasco, SP, CEP 06020-020, which follows the analysis methodology in accordance with the Brazilian Pharmacopoeia 6 o edition / 2019 - Microbiological Tests item 5.5.3.1.2 and FB 5.5.3.1.3 and Cosmetic Product Stability Guide, 2004, which analyzed the accelerated stability in cosmetic products of the moisturizing cream with moisturizing protection for hands PHTALOX moisturizing cream 0.05%, as per Table 7 below:
[0037] Table 8 - REPL 067 - Rev. 03 of 10 / 18 / 2022
