Description

Fourier tychographic microscope

[0001] The present invention relates to a microscope, and more particularly, to a Fourier tychographic microscope that guides the path of light toward an object into parallel light to improve resolution even at low magnification.

[0002] The microscope is the most fundamental tool used to observe living organisms and specific objects in all natural sciences, including biology, geology, physics, and chemistry. It is a type of magnifying device that magnifies and allows the visualization of objects invisible to the human eye. The term "microscope" generally refers to an optical microscope, while an optical microscope that uses two block lenses, one for the objective lens and one for the eyepiece, is called a compound microscope.

[0003] Meanwhile, the throughput of a microscope is fundamentally limited by the space-bandwidth product (SBP), making it difficult to secure high resolution and a wide field of view (FOV). To improve the problems of conventional microscopes, Fourier Ptychographic Microscopy (FPM) has recently been proposed. This FPM microscope was developed to expand the spatial bandwidth of a conventional microscope by replacing the LED matrix as a light source. When scanning a low-resolution image, the FPM microscope sequentially turns on the LEDs to measure the image, and then reconstructs a high-resolution image using an iterative phase search algorithm.

[0004] However, FPM microscopes suffer from a loss of resolution when reconstructing measured object images due to the formation of spherical waves around the object. Consequently, there has been a continuous demand for research to improve the performance of microscopes, including FPM microscopes.

[0005] The purpose of the present invention is to provide a Fourier tychographic microscope capable of acquiring high-resolution images by guiding the path of light toward a target object into parallel light to improve the concentration of light.

[0006] In order to achieve the above object, the Fourier-tychographic microscope according to the present invention comprises: a light source unit that irradiates light toward a target object; a guide unit that is located between the light source unit and the target object and guides the light generated from the light source unit and incident on the target object into parallel light; an optical unit that is provided in parallel with the target object and forms an enlarged image of the target object irradiated with the parallel light; a photographing unit that photographs the enlarged image formed by the optical unit; and a control unit that controls the light source unit and the photographing unit to restore an image of the target object irradiated with the light and photographed; and the guide unit includes at least one guide lens that is provided in a path of the light generated from the light source unit and guides the path of the light into parallel light.

[0007] In addition, the light source unit includes a light emitting element array in which a plurality of light emitting elements each irradiating the light toward the target object are provided on a light source substrate, and the control unit can control the plurality of light emitting elements to generate the light simultaneously or sequentially.

[0008] In addition, the guide unit may include a first guide lens that is provided to face the light source unit and guides the direction of the light generated from the light source unit into the parallel light toward the object, and a second guide lens that is provided between the first guide lens and the object and guides the light passing through the first guide lens into the parallel light.

[0009] Additionally, the first and second guide lenses may be provided in a convex shape facing each other.

[0010] In addition, the first guide lens may have an area that can cover the light source unit, and at least one second guide lens may be provided with an area that can cover the object.

[0011] In addition, the second guide lens may be provided such that a first lens having a convex shape toward the first guide lens and a second lens having a convex shape toward the object are adjacent to each other in the direction of propagation of the light.

[0012] Additionally, the first and second guide lenses can guide the path of the light to form a uniform plane wave over the entire area of the object.

[0013] Additionally, the distance between the light source and the target object can be maintained equal to the total focal length of the guide unit.

[0014] In addition, when f1 is the focal length of the first guide lens, f2 is the focal length of the second guide lens, and d is the distance between the first and second guide lenses, f, which is the focal length of the guide part with respect to the light source part, may have a reciprocal value of 1 / f1+1 / f2- d / (f1*f2).

[0015] In addition, the guide unit may further include a lens array including a plurality of micro lenses that are provided between the light source unit and the first guide lens to convert the light into the plane wave and increase the efficiency of the light.

[0016] In addition, the optical unit may include an optical lens including an objective lens through which the light is incident and a tube lens that guides an image formation of the light passing through the optical lens, and the photographing unit may include an image sensor.

[0017] A Fourier-tychographic microscope according to another preferred aspect of the present invention comprises: a light source unit including a plurality of light-emitting elements each irradiating at least one light toward a target object; a guide unit provided between the light source unit and the target object, which guides the light irradiated from the light source unit into parallel light to the target object so as to form a uniform plane wave over the entire area of the target object; an optical unit that forms an enlarged image of the target object irradiated with the parallel light; a photographing unit that photographs the enlarged image formed by the optical unit; and a control unit that controls the light source unit and the photographing unit to restore an image of the target object irradiated with the light and photographed, wherein the guide unit includes at least one guide lens to guide the path of the light in multiple stages.

[0018] Additionally, the control unit can control the plurality of light-emitting elements to generate the light simultaneously or sequentially.

[0019] In addition, the guide unit may include a first guide lens that is provided to face the light source unit and guides the direction of the light generated from the light source unit into the parallel light toward the object, and at least one second guide lens that is provided between the first guide lens and the object and guides the light passing through the first guide lens into the parallel light.

[0020] Additionally, the first and second guide lenses may be provided in a convex shape facing each other.

[0021] In addition, the first guide lens may have an area that can cover the light source unit, and at least one second guide lens may be provided with an area that can cover the object.

[0022] In addition, the second guide lens may be provided such that a first lens having a convex shape toward the first guide lens and a second lens having a convex shape toward the object are adjacent to each other in the direction of propagation of the light.

[0023] Additionally, the distance between the light source and the target object can be maintained equal to the total focal length of the guide unit.

[0024] In addition, when f1 is the focal length of the first guide lens, f2 is the focal length of the second guide lens, and d is the distance between the first and second guide lenses, f, which is the focal length of the guide part with respect to the light source part, may have a reciprocal value of 1 / f1+1 / f2- d / (f1*f2).

[0025] In addition, the guide unit may further include a lens array including a plurality of micro lenses that are provided between the light source unit and the first guide lens to convert the light into the plane wave and increase the efficiency of the light.

[0026] In addition, the optical unit may include an optical lens including an objective lens through which the light is incident and a tube lens that guides an image formation of the light passing through the optical lens, and the photographing unit may include an image sensor.

[0027] According to the present invention having the above configuration, by guiding light incident on an object into parallel light, the light does not spread about the object and a uniform plane wave can be formed over the entire area, thereby further improving the light focusing ability.

[0028] In addition, the resolution of images captured from a target can be improved even at low magnification, enabling high-performance target image capture and restoration with excellent resolution.

[0029] In addition, the path of light guided to the target in multiple stages can be guided in parallel, thereby improving light efficiency compared to light incident as a conventional spherical wave.

[0031] Figure 2 is a schematic enlarged drawing of the light source and guide portion illustrated in Figure 1.

[0032] Figure 3 is a schematic diagram illustrating a Fourier tychographic microscope according to another preferred embodiment of the present invention. And,

[0033] FIG. 4 is a graph schematically comparing the optical efficiency of a Fourier tychographic microscope according to one embodiment of the present invention and another embodiment with that of a conventional microscope.

[0034] Hereinafter, a preferred embodiment of the present invention will be described with reference to the attached drawings. However, the spirit of the present invention is not limited to such an embodiment, and the spirit of the present invention may be proposed differently by adding, changing, or deleting components constituting the embodiment, but this is also included in the spirit of the invention.

[0035]

[0036] FIG. 1 is a schematic drawing of a Fourier tychographic microscope (1) according to a preferred embodiment of the present invention, and FIG. 2 is a schematic enlarged drawing of the light source unit (10) and guide unit (20) illustrated in FIG. 1.

[0037] Referring to FIG. 1, a Fourier tychographic microscope (1) according to a preferred embodiment of the present invention includes a light source unit (10), a guide unit (20), an optical unit (30), a photographing unit (40), and a control unit (50).

[0038]

[0039] The light source unit (10) irradiates light (B) toward the target object (S). Here, the light source unit (10) includes a light emitting element array (Array) in which a plurality of light emitting elements (12) are mounted on a light source substrate (11). Here, the plurality of light emitting elements (12) are positioned at a predetermined distance from each other with respect to the light source substrate (11) and generate first to fourth lights (B1) (B2) (B3) (B4) of different pixels toward the target object (S). At this time, the plurality of light emitting elements (12) irradiate the first to fourth lights (B1) (B2) (B3) (B4) having different incident angles toward the target object (S).

[0040] In addition, the light source substrate (11) is connected to a control unit (50) to be described later, and is controlled so that a plurality of light emitting elements (12) simultaneously or sequentially irradiate light (B) to a target object (S). The technical configuration in which an external signal is applied to the light source substrate (11) of the light source unit (10) so that a plurality of light emitting elements (12) provided on the light source substrate (11) simultaneously or sequentially emit light is not the gist of the present invention, and therefore a detailed description thereof is omitted.

[0041] The guide unit (20) is positioned between the light source unit (10) and the object (S), and guides the angle of light (B) generated from the light source unit (10) and incident on the object (S) into parallel light (B). This guide unit (20) includes a first guide lens (21) and a second guide lens (22), as illustrated in FIG. 2. Here, the first and second guide lenses (21) (22) guide the angle of light (B) to form a uniform plane wave over the entire area of the object (S).

[0042] The first guide lens (21) is provided to face the light source unit (10) and guides the light (B) generated from the light source unit (10) toward the object (S) as parallel light, thereby concentrating the light (B) without dispersing it. The first guide lens (21) converts the light (B) having different incident angles according to the positions of the plurality of light-emitting elements (12) with respect to the object (S) into parallel light, thereby converting the spherical wave into a plane wave. More specifically, the first guide lens (21) converts the direction of propagation of the first to fourth lights (B1), (B2), (B3), and (B4) generated from the plurality of light-emitting elements (12) and irradiated toward the object (S) at different incident angles into a direction toward the object (S), and guides them into parallel light. In summary, the first guide lens (21) guides the light (B) incident on the object (S) to be parallel by changing the angle between the light emitting elements (12) and the object (S) according to the positions of the light emitting elements (12).

[0043] Meanwhile, the first guide lens (21) is provided as a lens having a sufficient area to simultaneously cover and face a plurality of light-emitting elements (12). Therefore, light (B) generated from the plurality of light-emitting elements (12) by the first guide lens (21) can be simultaneously and parallelly incident on the target object (S) without being dispersed.

[0044] The second guide lens (22) is provided between the first guide lens (21) and the object (S), and guides the light (B) passing through the first guide lens (21) into parallel light for the second time. This second guide lens (22) guides the light (B) irradiated as parallel light from the first guide lens (21) toward the object (S) for the second time into parallel light. At this time, the second guide lens (22) has an area that can sufficiently cover the object (S) and faces the object (S), so that the light (B) passing through the first guide lens (21) can be guided to the object (S) without leakage.

[0045] As illustrated in FIG. 2, the first and second guide lenses (21)(22) have a convex lens shape on the surfaces facing each other. In addition, the second guide lens (22) may include a first lens (23) having a convex shape toward the first guide lens (21) and a second lens (24) having a convex shape toward the object (S). Here, the first and second lenses (23)(24) are arranged to be adjacent to each other with respect to the direction in which light (B) travels. That is, the first lens (23) of the second guide lens (22) faces the first guide lens (21), and the second lens (24) faces the object (S) at a position adjacent to the first lens (23).

[0046] The second guide lens (22) additionally guides the light (B) that was initially guided parallel from the first guide lens (21) to a parallel path toward the object (S). That is, the guide unit (20) guides the light (B) into parallel light in multiple stages using the first and second guide lenses (21)(22). Here, the number of second guide lenses (22) provided between the light source unit (10) and the object (S) is exemplified as one in FIG. 1, but two or more may be provided to guide the path of the light (B) in parallel in multiple stages. That is, the number of second guide lenses (22) can be adjusted in consideration of shooting conditions such as the distance between the light source unit (10) and the object (S) and the intensity of the light (B) generated from the light source unit (10).

[0047] In addition, the distance between the light source unit (10) and the target object (S) is maintained equal to the focal length of the entire guide unit (20). Here, when the focal length of the first guide lens (21) is f1, the focal length of the second guide lens (22) is f2, and the distance between the first and second guide lenses (21)(22) is d, the focal length f of the guide unit (20) with respect to the light source unit (10) has a reciprocal value of 1 / f1+1 / f2-d / (f1*f2).

[0048] The optical unit (30) is arranged parallel to the target object (S) and forms an enlarged image of the target object (S) to which light (B) is irradiated. This optical unit (30) includes an optical lens (31) and a tube lens (32).

[0049] The optical lens (31) is an objective lens provided adjacent to the object (S) and onto which light (B) passing through the object (S) is incident. That is, the optical lens (31) is positioned closest to the object (S). The light (B) passing through the object (S) is incident on the optical lens (31), thereby magnifying the image of the object (S). This optical lens (31) may include a plurality of objective lenses, and the number of these optical lenses (31), the distance between them and the object (S), etc. are not limited to the illustrated example.

[0050] Meanwhile, the optical lens (31) may use an objective lens with a relatively low magnification or a relatively low numerical aperture (NA) to enable large-area imaging. Generally, when an objective lens with a low magnification or a low numerical aperture is used, a low-resolution image can be acquired over a large area. Conversely, when a lens with a high magnification or a high numerical aperture is used, a high-resolution image can be acquired, but it is limited to a narrow area and a large-area image cannot be acquired. Accordingly, the present embodiment enables large-area imaging by using an optical lens (31) with a low magnification or a low numerical aperture, and at the same time, a low-resolution image (LRIm) can be restored to a high-resolution image (HRIm) by the control unit (50) at a later time, so that the image resolution can also be acquired at a high level. Accordingly, the low-resolution image (LRIm) may be an image that displays a relatively wide area with a relatively low resolution as a large-area image.

[0051] The tube lens (32) guides the light (B) passing through the optical lens (31) to form an image from the passing light (B). The light (B) guided parallel to the optical lens (31) passes through the optical lens (31) and passes through the tube lens (32) in a parallel light state, thereby forming an image.

[0052] Meanwhile, the optical unit (30) may additionally have at least one filter provided between the optical lens (31) and the tube lens (32). For example, the optical unit (30) may further include at least one of a bandpass filter, a prism, and a polarizing filter, and may have the function of at least one of a fluorescence microscope, a hyperspectral microscope, and a birefringence microscope.

[0053] The photographing unit (40) photographs the enlarged image formed by the optical unit (30) to obtain an enlarged image or video (hereinafter collectively referred to as an image) of the object (S). The photographing unit (40) may include a CCD image sensor that can generate current through a photoelectric element and process the current through a CCD (Charge Coupled Device) or a CMOS (Complementary Metal-Oxide Semiconductor), but is not limited thereto.

[0054] The control unit (50) controls the light source unit (10) and the photographing unit (40) so that light (B) is generated and an image of the target object (S) is restored. The control unit (50) controls a plurality of lights (B) generated from a plurality of light-emitting elements (12) of the light source unit (10), that is, first to fourth lights (B1) (B2) (B3) (B4), to be emitted and irradiated simultaneously or sequentially. In addition, the control unit (50) controls a plurality of first to fourth lights (B1) (B2) (B3) (B4) generated from a plurality of light-emitting elements (12) to pass through the target objects (S) respectively and restore a high-resolution image from the images of the plurality of target objects (S) captured by the photographing unit (40).

[0055] Here, the control unit (50) receives an enlarged low-resolution image (LRIm) of the subject (S) photographed from the photographing unit (40), and restores it to generate a high-resolution image (HRIm) having a higher resolution than the low-resolution image (LRIm). At this time, the low-resolution image (LRIm) is a plurality of images having different spatial frequency information, and the control unit (50) synthesizes the low-resolution images (LRIm) in the frequency domain to restore them to a high-resolution image (HRIm). Since the light (B) generation and image restoration technology of the control unit (50) are not the gist of the present invention, a detailed description thereof will be omitted.

[0056]

[0057] The operation of the Fourier tychographic microscope (1) according to the present invention having the above configuration is described as follows.

[0058] As shown in Fig. 1, a plurality of light emitting elements (12) of a light source unit (10) each generate a plurality of lights (B). Here, the plurality of light emitting elements (12) can generate lights (B) simultaneously or sequentially by a control unit (50), and in this embodiment, the first to fourth lights (B1), (B2), (B3), and (B4) are simultaneously emitted from four light emitting elements (12) and irradiated toward a target object (S) as illustrated and exemplified.

[0059] The first to fourth lights (B1) (B2) (B3) (B4) generated from the plurality of light-emitting elements (12) pass through the guide unit (20), are guided as parallel lights in multiple stages, and are incident on the target object (S). More specifically, as illustrated in FIG. 2, the first to fourth lights (B1) (B2) (B3) (B4) generated from the plurality of light-emitting elements (12) are incident on the first guide lens (21) of the guide unit (20) at different incident angles toward the target object (S). The first to fourth lights (B1) (B2) (B3) (B4) incident on the first guide lens (21) at these different incident angles are converted into parallel lights toward the target object (S) while passing through the first guide lens (21). The first to fourth lights (B1)(B2)(B3)(B4) converted into parallel light are additionally guided in a parallel direction by passing through the second guide lens (22) including the first and second lenses (23)(24) and are incident on the target object (S).

[0060] The first to fourth lights (B1) (B2) (B3) (B4) that are guided as parallel light by sequentially passing through the first and second guide lenses (21) (22) are incident as plane waves on the object (S). The first to fourth lights (B1) (B2) (B3) (B4) that are parallel light incident as plane waves on the object (S) pass through the object (S) and sequentially pass through the optical lens (31) and the tube lens (32) of the optical unit (30), as shown in FIG. 1, thereby capturing an image of the object (S) by the photographing unit (40). The image of the object (S) captured by the photographing unit (40) is restored by the control unit (50), thereby finally obtaining a high-resolution image of the object (S). In this way, by inducing a plane wave over the entire surface of the target object (S), high-resolution image restoration becomes possible, thereby improving the low-resolution problem caused by the existing spherical wave.

[0061]

[0062] Referring to FIG. 3, a Fourier tychographic microscope (100) according to another preferred embodiment of the present invention is schematically illustrated.

[0063] As shown in Fig. 3, a Fourier tychographic microscope (100) according to another embodiment includes a light source unit (110), a guide unit (120), an optical unit (130), a photographing unit (140), and a control unit (150). Here, the configuration of the light source unit (110), the optical unit (130), the photographing unit (140), and the control unit (150) has the same configuration as that of the embodiment described with reference to Fig. 1, and therefore, a detailed description thereof is omitted.

[0064] A guide unit (120) according to another embodiment includes a first guide lens (121), a second guide lens (122), and a lens array (125).

[0065] The first and second guide lenses (121)(122) guide the incident light (B) as parallel light to the target object (S), similar to the first and second guide lenses (21)(22) described in one embodiment. The first and second guide lenses (121)(122) are provided in a lens shape with surfaces facing each other being convex. In addition, the second guide lens (122) may include the first and second lenses (123)(124).

[0066] The lens array (125) is provided between the light source unit (110) and the first guide lens (121), converts light (B) incident from the light source unit (110) into a plane wave, and increases the efficiency of the light (B). To this end, the lens array (125) includes a plurality of micro lenses (125b) (125c) (125d) (125e) provided on a lens substrate (125a). As shown in an enlarged view in FIG. 3, the lens array (125) converts the first to fourth light rays (B1) (B2) (B3) (B4) incident on the plurality of micro lenses (125b) (125c) (125d) (125e) into plane waves with increased efficiency.

[0067]

[0068] The operation of a Fourier tychographic microscope (100) according to another embodiment having the above configuration is described as follows.

[0069] As shown in FIG. 3, light (B) generated from a plurality of light emitting elements (112) provided on a light source substrate (111) of a light source unit (110) is incident on an facing lens array (125), and a plurality of micro lenses (125b)(125c)(125d)(125e) of the lens array (125) convert the incident light (B) into a plane wave and increase the efficiency of the light (B). That is, as shown in an enlarged view in FIG. 3, the plurality of micro lenses (125b)(125c)(125d)(125e) convert the first to fourth lights (B1)(B2)(B3)(B4) into plane waves with superior focusing power and high efficiency and guide them toward the first guide lens (121).

[0070] The first to fourth light beams (B1) (B2) (B3) (B4), which are high-efficiency plane waves incident on the first guide lens (121), sequentially pass through the first and second guide lenses (121) (122), thereby being incident on the object (S) as parallel light beams. Accordingly, the parallel light beams (B) that have passed through the object (S) pass through the optical lens (131) and the tube lens (132) of the optical unit (130) and are captured by the photographing unit (140), thereby being restored as a high-resolution image of the object (S) by the control unit (150).

[0071]

[0072] Referring to FIG. 4, the characteristics of light (B) incident on a target (S) of a Fourier tychographic microscope (1) (100) according to one preferred embodiment and another embodiment of the present invention are schematically compared. FIG. 4 (a) shows the characteristics of light (B) in a conventional Fourier tychographic microscope, and FIG. 4 (b) and (c) show the characteristics of light (B) in a Fourier tychographic microscope (1) (100) according to one preferred embodiment and another embodiment.

[0073] Referring to (a) of Fig. 4, the outputs when the angle ranges of the plurality of lights (B) are 9.45, 10.10, 9.40, and 8.60 degrees are 5.2, 5.2, 4.8, and 4.2 W / sr. In addition, the output intensity of the light (B) is approximately 0.24 W / sr.

[0074] Referring to (b) of FIG. 4, the angular ranges of the plurality of lights (B) in the Fourier-tychographic microscope (1) according to one embodiment are 2.7, 2.2, 2.4, and 2.4 degrees, and the angular distribution range relative to the central angle is reduced to approximately 1 / 3 or less compared to (a) of FIG. 4, thereby confirming that it is close to parallel light. In addition, the intensities of the lights (B) are 4.7, 4.7, 4.6, and 4.2 W / sr, which are similar levels compared to the conventional ones, thereby confirming that the efficiency of the lights (B) is not reduced. The output intensity of the lights (B) in the Fourier-tychographic microscope (1) according to one embodiment is approximately 6.8 W / sr.

[0075] Referring to (c) of FIG. 4, it can be confirmed that the angular ranges of the plurality of lights (B) in the Fourier-tychographic microscope (100) according to another embodiment are 4.68, 4.50, 4.30, and 4.10 degrees, and the angular distribution range relative to the central angle is reduced to about half or less compared to (a) of FIG. 4, thereby approaching parallel light. In addition, it can be confirmed that the intensity of the light (B) is 72.1, 71.2, 66.9, and 59.1 W / sr, which is a similar increase of about 14 times compared to the conventional one. The output intensity of the light (B) in the Fourier-tychographic microscope (1) according to this embodiment is about 30 W / sr.

[0076] As described above, the Fourier tychographic microscope (1) (100) according to the present invention and other embodiments converts light (B) generated from the light source (10) (110) into parallel light and guides it to the target object (S), thereby causing the light (B) to be incident on the target object (S) as a plane wave. As a result, the concentration of light (B) on the target object (S) can be improved, thereby improving the resolution even at low magnification.

[0077]

[0078] As described above, although the present invention has been described with reference to preferred embodiments, it will be understood by those skilled in the art that various modifications and changes can be made to the present invention without departing from the spirit and scope of the present invention as set forth in the claims below.
