Description

Antibacterial composition against Malassezia patchermatis

[0001] It relates to an antibacterial composition against Malassezia pachydermatis.

[0002] Malassezia is a genus of fungi belonging to the Basidiomycota, and 15 species have been identified to date. Of these, 10 species (M. restricta, M. globosa, M. furfur, M. sympodialis, M. obtusa, M. slooffiae, M. japonica, M. yamatoensis, M. dermatis, M. arunalokei) are considered to be human-friendly. In addition to the above species, 5 species (M. pachydermatis, M. cuniculi, M. caprae, M. equine, M. nana) are mainly isolated from livestock or companion animals. In particular, M. pachydermatis is mainly observed in companion dogs and cats, and is also known as a fungus that causes dermatitis and otitis externa in companion dogs. Malassezia patchydermatis lives on the skin of dogs and cats. While it doesn't pose a significant problem when your pet is healthy, it can cause skin problems when the fungus proliferates in humid environments, due to allergies or a weakened immune system.

[0003] Therefore, in order to prevent or improve skin diseases in companion animals, it is necessary to develop an antibacterial agent against Malassezia patchdermatis.

[0004] One aspect is to provide an antibacterial composition against Malassezia pachydermatis, comprising two or more selected from 1,2-hexanediol, ethylhexylglycerin, 1,3-propanediol, hydroxyacetophenone, and caprylyl glycol.

[0005] Another aspect is to provide a cosmetic composition comprising the above antibacterial composition.

[0006] Another aspect is to provide a pharmaceutical composition for preventing or improving a disease caused by Malassezia pachydermatis, comprising the above antibacterial composition.

[0007] Another aspect provides a method for preventing or ameliorating a disease caused by Malassezia patchdermatis, comprising administering to a subject an effective amount of the antimicrobial composition.

[0008] Another aspect provides a use of the antimicrobial composition for preparing a medicament for preventing or ameliorating a disease caused by Malassezia patchdermatis.

[0009] One aspect provides an antibacterial composition against Malassezia pachydermatis, comprising two or more selected from 1,2-hexanediol, ethylhexylglycerin, 1,3-propanediol, hydroxyacetophenone, and caprylyl glycol.

[0010] An antimicrobial composition according to one aspect may exhibit a synergistic antimicrobial effect against Malassezia pachydermatis by including a combination of specific ingredients.

[0011] The above antibacterial composition may include two or three selected from 1,2-hexanediol, ethylhexylglycerin, 1,3-propanediol, hydroxyacetophenone, and caprylyl glycol.

[0012] In one embodiment, the antimicrobial composition may comprise a combination of ingredients selected from the following:

[0013] (1) 1,2-hexanediol and ethylhexylglycerin;

[0014] (2) 1,3-propanediol and hydroxyacetophenone;

[0015] (3) 1,3-propanediol and ethylhexylglycerin;

[0016] (4) Caprylyl glycol and hydroxyacetophenone;

[0017] (5) Caprylyl glycol and ethylhexylglycerin;

[0018] (6) 1,2-hexanediol, 1,3-propanediol, and ethylhexylglycerin;

[0019] (7) caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone; and

[0020] (8) Caprylyl glycol, 1,3-propanediol, and ethylhexylglycerin.

[0021] In the above combination (1), the concentration ratio of 1,2-hexanediol and ethylhexylglycerin may be 1 to 32:1, 1 to 8:1, 1 to 4:1, or 1 to 2:1. In a specific embodiment, the concentration ratio of 1,2-hexanediol and ethylhexylglycerin may be any range or any value selected from the range of 1 to 32:1. For example, the concentration ratio of 1,2-hexanediol and ethylhexylglycerin may be 1:1, 2:1, 4:1, 8:1, or 32:1. In the above concentration ratio range, 1,2-hexanediol and ethylhexylglycerin may exhibit a partial antibacterial synergistic effect against Malassezia patchdermatis. When the concentration ratio of 1,2-hexanediol and ethylhexylglycerin is more than 32:1, the FICI judgment may be 'no antibacterial synergy effect' due to the increase in the 1,2-hexanediol content; when it is less than 1:1, there may be no antibacterial effect due to the decrease in the 1,2-hexanediol content and the proliferation of microorganisms.

[0022] In the above combination (2), the concentration ratio of 1,3-propanediol and hydroxyacetophenone may be 0.25 to 4:1, 1 to 4:1 to 4, 1:1 to 4, 1 to 4:1, 2 to 4:1, 1:2 to 4, 1:1 to 2, or 1 to 2:1. In a specific embodiment, the concentration ratio of 1,3-propanediol and hydroxyacetophenone may be any range or any value selected from the range of 1 to 4:1 to 4. In certain embodiments, the concentration ratio of 1,3-propanediol and hydroxyacetophenone can be any range or any value selected from the range of 0.25 to 4:1. For example, the concentration ratio of 1,3-propanediol and hydroxyacetophenone can be 4:1, 2:1, 1:1, 1:2 (0.5:1), or 1:4 (0.25:1). In the concentration ratio range of 2 to 4:1, 1,3-propanediol and hydroxyacetophenone can exhibit partial antibacterial synergistic effect against Malassezia patchdermatis. In the concentration ratio range of 1:1 to 4, 1,3-propanediol and hydroxyacetophenone can exhibit antibacterial synergistic effect against Malassezia patchdermatis. When the concentration ratio of 1,3-propanediol and hydroxyacetophenone is more than 4:1, the FICI judgment may be 'no antibacterial synergy effect' due to the increase in 1,3-propanediol content; when it is less than 1:4 (less than 0.25:1), there may be no antibacterial effect due to the decrease in 1,3-propanediol content and microbial growth.

[0023] In the above combination (3), the concentration ratio of 1,3-propanediol and ethylhexylglycerin may be 2 to 8:1, 2 to 4:1, or 4 to 8:1. In a specific embodiment, the concentration ratio of 1,3-propanediol and ethylhexylglycerin may be any range or any value selected from the range of 2 to 8:1. For example, the concentration ratio of 1,3-propanediol and ethylhexylglycerin may be 2:1 or 8:1. In the above concentration ratio range, 1,3-propanediol and ethylhexylglycerin may exhibit a partial antibacterial synergistic effect against Malassezia patchdermatis. When the concentration ratio of 1,3-propanediol and ethylhexylglycerin is more than 8:1, the FICI judgment may be 'no antibacterial synergy effect' due to the increase in 1,3-propanediol content; when it is less than 2:1, there may be no antibacterial effect due to the decrease in 1,3-propanediol content and microbial growth.

[0024] In the above combination (4), the concentration ratio of caprylyl glycol and hydroxyacetophenone may be 1 to 4:1, 1 to 3:1, 1 to 2.5:1, 1 to 2:1, or 1 to 1.5:1. In a specific embodiment, the concentration ratio of caprylyl glycol and hydroxyacetophenone may be any range or any value selected from the range of 1 to 2:1. For example, the concentration ratio of caprylyl glycol and hydroxyacetophenone may be 1:1 or 2:1. In the above concentration ratio range, caprylyl glycol and hydroxyacetophenone may exhibit a partial antibacterial synergistic effect against Malassezia patchdermatis. When the concentration ratio of caprylyl glycol and hydroxyacetophenone is more than 4:1, there may be no antibacterial effect due to a decrease in the content of hydroxyacetophenone and microbial growth; when it is less than 1:1, the FICI judgment may be 'no antibacterial synergy effect' due to an increase in the content of hydroxyacetophenone.

[0025] In the above combination (5), the concentration ratio of caprylyl glycol and ethylhexylglycerin may be 0.25 to 4:1, 0.25 to 2:1, 1 to 2:1 to 4, 1 to 2:1, 1:1 to 4, or 1:2 to 4. In a specific embodiment, the concentration ratio of caprylyl glycol and ethylhexylglycerin may be any range or any value selected from the range of 1 to 2:1 to 4. In a specific embodiment, the concentration ratio of caprylyl glycol and ethylhexylglycerin may be any range or any value selected from the range of 0.25 to 2:1. For example, the concentration ratio of caprylyl glycol and ethylhexylglycerin may be 2:1, 1:2 (0.5:1), or 1:4 (0.25:1). Within the above concentration ratio range, caprylyl glycol and ethylhexylglycerin may exhibit a partial antibacterial synergistic effect against Malassezia patchdermatis. When the concentration ratio of caprylyl glycol and ethylhexylglycerin is greater than 4:1, the FICI evaluation may be 'no antibacterial synergistic effect' due to the increase in the caprylyl glycol content; and when it is less than 1:4 (less than 0.25:1), the antibacterial effect may be absent due to the decrease in the caprylyl glycol content and the proliferation of microorganisms.

[0026] In the above combination (6), the concentration ratio of 1,2-hexanediol, 1,3-propanediol, and ethylhexylglycerin may be 8 to 32:2 to 8:1, 10 to 30:3 to 7:1, 10 to 20:3 to 6:1, 14 to 18:3 to 5:1, or 15 to 17:3 to 5:1. In a specific embodiment, the concentration ratio of 1,2-hexanediol, 1,3-propanediol, and ethylhexylglycerin may be any range or any value selected from the range of 8 to 32:2 to 8:1. For example, the concentration ratio of 1,2-hexanediol, 1,3-propanediol, and ethylhexylglycerin may be 16:4:1. Within the above concentration range, 1,2-hexanediol, 1,3-propanediol, and ethylhexylglycerin may exhibit partial antibacterial synergistic effects against Malassezia patchdermatis. Beyond the above concentration range, the FICI may determine "no antibacterial synergistic effect" or "antagonistic effect," or the antibacterial effect may be absent due to microbial proliferation.

[0027] In the above combination (7), the concentration ratio of caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone may be 1:0.5 to 16:1 to 8, 1 to 2:1 to 16:1 to 8, 1:1 to 16:1 to 8, 1 to 2:1:2 to 4, 1:2 to 4:1, or 1:1 to 2:1. In a specific embodiment, the concentration ratio of caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone may be any range or any value selected from the range of 1 to 2:1 to 16:1 to 8. In certain embodiments, the concentration ratio of caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone can be any range or any value selected from the range of 1:0.5 to 16:1 to 8. For example, the concentration ratio of caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone can be 1:4:8, 1:4:2, 1:2:2, 1:1:2, 2:1:4 (1:0.5:2), 1:4:1, 1:16:2, or 1:2:1. Caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone can exhibit partial antibacterial synergistic effect or antibacterial synergistic effect against Malassezia patchdermatis at a concentration ratio of 1 to 2:1 to 16:1 to 8 or 1:0.5 to 16:1 to 8. Caprylyl glycol, 1,3-propanediol, and hydroxyacetophenone can exhibit antibacterial synergistic effect against Malassezia patchdermatis at a concentration ratio of 1:2:1. When the concentration ratio is out of the above range, the FICI judgment may be 'no antibacterial synergistic effect' or 'antagonistic effect', or there may be no antibacterial effect due to microbial proliferation.

[0028] In the above combination (8), the concentration ratio of caprylyl glycol, 1,3-propanediol, and ethylhexylglycerin may be 2 to 8:8 to 16:1, 2 to 4:8 to 16:1, 4 to 8:8 to 16:1, or 2 to 8:16:1. In a specific embodiment, the concentration ratio of caprylyl glycol, 1,3-propanediol, and ethylhexylglycerin may be any range or any value selected from the range of 2 to 8:8 to 16:1. For example, the concentration ratio of caprylyl glycol, 1,3-propanediol, and ethylhexylglycerin may be 4:8:1, 2:16:1, or 8:16:1. At the above concentration ratio, caprylyl glycol, 1,3-propanediol, and ethylhexylglycerin may exhibit a partial antibacterial synergistic effect against Malassezia patchdermatis. If the concentration ratio is outside the above range, the FICI may determine "no antibacterial synergistic effect" or "antagonistic effect," or the antibacterial effect may be absent due to microbial proliferation.
