[0014] As used herein, "bayberry" refers to an evergreen tall tree belonging to the Myricaceae family. The variety of bayberry is not particularly limited, but examples include black peach, red peach, and white peach. Specific varieties include Zuikou (Kondo peach), Moriguchi, Koutou (Nishimura, Kobashi, etc.), Nakayama Wase, Kogyoku (Hiyama, Hinode), Gozen (Shirodango, Hakusawa), Awanishiki (Oni, Onidango), Kamezo, Ban'inyoubai, Tokai, Seitodo Daiyoubai, Tenga-sen, Yokawachi, Juroku, Taneyoubai, Chubaao, Dingxiangbai, and Daitanbai. Bayberry leaves, for example, are alternate, obovate-oblong or oblanceolate, tapered to a wedge-shaped base, leathery, with small oil spots on the underside, and usually entire. Young trees have sharp sawtooth. For example, the leaves of the Japanese bayberry have short petioles and are about 12 cm long.

[0015] As used herein, "bayberry extract" refers to an extract extracted from a bayberry extraction raw material (hereinafter referred to as the extraction raw material). Examples of extraction raw materials include bayberry leaves, bayberry fruits, bayberry trunks, bayberry branches, bayberry bark, and bayberry roots. Among these, bayberry leaves are suitable as an extraction raw material because they can be harvested regardless of the season and are available in large quantities. Either dried or fresh bayberry leaves may be used. When using bayberry leaves as an extraction raw material, it is preferable to use leaves that have been processed, such as by shredding or crushing. Using bayberry leaves that have been processed in this way can improve extraction efficiency.

[0016] There are no particular limitations on the extraction method for bayberry extract, and extraction can be performed by appropriately selecting a conventionally known extraction method using an extraction raw material and an extraction solvent, etc. For example, a bayberry extract can be obtained by mixing an extraction raw material (e.g., bayberry leaves) with an extraction solvent. [0018] The extraction solvent may be used alone or in combination of two or more. When two or more extraction solvents are used in combination, a mixed solvent of two or more extraction solvents may be used, or a combination of extractions using the extraction solvents may be used. Examples of combinations of extractions using extraction solvents include a combination of water extraction and organic solvent extraction, or a combination of one organic solvent extraction and another organic solvent extraction. As the extraction solvent, water, 1,3-butylene glycol, ethanol, ethyl acetate, and a mixed solvent (mixed solution) containing two or more of these can be suitably used.

[0019] (Extraction Conditions) There are no limitations on the extraction conditions, and they may be appropriately determined depending on the extraction solvent used. [0095] (Production of Bayberry Extract (Bayberry Leaf Extract)) 10 kg of a 30% w / w 1,3-butylene glycol solution prepared from 7 kg of distilled water and 3 kg of 1,3-butylene glycol was heated to 70-80°C, and 100 g of crushed dried Bayberry leaves was added to this solution, followed by stirring for 2 hours while maintaining the heated state at 70-80°C, to obtain an extract. The extract was filtered through 5B filter paper (manufactured by ADVANTEC), and distilled water was added to the filtrate to readjust the volume to 10 kg, to obtain a Bayberry extract (Bayberry Leaf Extract).

[0096] (Evaluation of Gene Expression) NHEK cells (normal human epidermal keratinocytes) were seeded in a 96-well plate, and treatment with bayberry extract (bayberry leaf extract treatment) began the day after seeding. KG2 (Kurabo Industries, Ltd.) medium was used for seeding, and KB2 (Kurabo Industries, Ltd.) was used as the test medium for dissolving the test substance. Filter-sterilized bayberry extract (bayberry leaf extract) prepared as described above was mixed with the medium at 1%, and test media with different concentrations were prepared in a 3-fold dilution series. 30% 1,3-butylene glycol was added to the control and diluted concentration treatment groups to a final concentration of 1%. qPCR was performed to examine gene changes 24 hours after the addition of the test medium.

[0097] To prepare the reverse transcription product for qPCR, a SuperPrep® II Cell Lysis & RT Kit for qPCR (Toyobo Co., Ltd.) was used, and reverse transcription reaction was performed according to the protocol provided by the manufacturer. Using the resulting reverse transcription product as a template and THUNDERBIRD® SYBR qPCR Mix (Toyobo Co., Ltd.) as a qPCR reagent, gene expression analysis of each gene (aquaporin 3, filaggrin, and claudin 1 genes) was performed. The results of the gene expression analysis are shown in Figure 1. Figure 1(A) shows the results of expression analysis of the aquaporin 3 gene, Figure 1(B) shows the results of expression analysis of the filaggrin gene, and Figure 1(C) shows the results of expression analysis of the claudin 1 gene.

[0098] The following primer set was used to amplify the target gene. Hereinafter, Forward Primer is abbreviated as "FP" and Reverse Primer is abbreviated as "RP."・Aquaporin 3 (AQP3) Primer Set "FP" 5'-GCCTTTGGCTTTGCTGTCACTC-3' "RP" 5'-AGCTGGTTGTCGGCGAAGTG-3' ・Filaggrin (FLG) Primer Set "FP" 5'-TCGGCAATCCTGAAGAATCC-3' "RP" 5'-CTGTGCTTTCTGTGCTTGTGTC-3' ・Claudin 1 (CLDN1) Primer Set "FP" 5'-GACGATGAGGTGCAGAAGATGAG-3' "RP" 5'-AGCCAGTGAAGAGAGCCTGAC-3' ・PPIA Primer Set "FP" 5'-TCATCTGCACTGCCAAGACTG-3' "RP" 5'-TGGTGATCTTCTTGCTGGTCTTG-3' The PPIA gene was used as a housekeeping gene for correcting gene expression.

[0099] As shown in Figure 1, it was confirmed that the Bayberry extract increased the expression of the aquaporin 3 gene, filaggrin gene, and claudin 1 gene, which contribute to improving skin function. This demonstrated that the Bayberry extract has the function of improving skin function.

[0100] (Evaluation of Promotion of Tight Junction Formation) Normal human epidermal keratinocytes were seeded onto Lab-Tek II chamber slides (Nunc) using KG2 medium (Kurabo Industries, Ltd.). The following day, the test medium was replaced with KB2 medium (Kurabo Industries, Ltd.) supplemented with the test substance at the designated concentration, and the evaluation was initiated. 24 hours after the test medium was replaced, the cells were fixed using PBS(-) containing 4% formalin and permeabilized with PBS(-) containing 0.1% Triton X-100. Subsequently, the cells were blocked with PBS(-) containing 1% BSA, followed by fluorescent immunostaining. Figure 2 shows fluorescent immunostaining images of claudin-1 and ZO-1.

[0101] For fluorescent immunostaining, mouse anti-CLDN1 (A-9, Santa Cruz) and rabbit anti-ZO-1 (Proteintech) were used as primary antibodies, and Alexa Fluor 488-AffiniPure Goat Anti-Mouse IgG (H + L) and Rhodamine Red-x-AffiniPure Donkey Anti-Rabbit IgG (H + L) (both Jackson ImmunoResearch Laboratories) were used as secondary antibodies. After antibody staining, nuclear staining was performed using VECTASHIELD PLUS Antifade Mounting Medium with DAPI (Vector Laboratories) as a mounting medium. Image data were acquired using a ZOE Fluorescent Cell Imager (Bio-Rad). To eliminate bias, random multiple fields of view were photographed under the same conditions with a fixed exposure time to obtain data suitable for quantitative analysis. Image analysis was performed using ImageJ, an open-source, public-domain image processing software.

[0102] (Evaluation of Claudin 1 Expression Levels) Quantitative comparison was performed using a value obtained by dividing the sum of signal intensities derived from claudin 1 (CLDN1) in the image data representatively shown in Figure 2 by the cell number count using DAPI. Figure 3 shows the fluorescence intensity derived from claudin 1. The fluorescence intensity signal is related to the expression level of the claudin 1 gene, and as shown in Figure 3, it was revealed that the extract of Bayberry can increase the expression of the claudin 1 gene, which contributes to the improvement of skin function. These results also clearly show that the extract of Bayberry has the function of improving skin function.

[0103] (Fluorescence Intensity Values of ZO-1-Derived Signals) The distribution of ZO-1-derived signals on a histogram of the representative image data shown in Figure 2 was divided and quantified by pixel fluorescence intensity. For arbitrary divisions, the histogram values of the control ZO-1 image were used, and the fluorescence intensity values including the bottom 50% and bottom 75% of pixels were used as delimiters. The number of pixels in the low (Low: ∼50%), medium (Mid: 50-75%), and high (High: 75% and above) ranges were divided by the total number of pixels, and the percentage of pixels in each range was quantified. Figure 4 shows the histogram of ZO-1-derived fluorescent signals, and Figure 5 shows the distribution of pixel fluorescence intensity values. Figure 4 confirms that the bayberry extract shifted the ZO-1-derived fluorescent signal histogram to the right. Furthermore, Figure 5 suggests that the bayberry extract locally accumulated ZO-1 expression levels, biasing the fluorescence intensity distribution. This local accumulation of ZO-1 expression is thought to indicate the formation of tight junctions. These results demonstrate that Bayberry extract can promote the formation of tight junctions. These results also demonstrate that Bayberry extract has the function of improving skin function.

[0104] (Evaluation of Skin Function Improvement Function in Human Monitor Test) A human monitor test on skin function improvement function was conducted on 13 healthy Japanese women (aged 29 to 51 years, average age 42.2 years) who gave their consent to the following test.

[0105] (Test conditions) <Sample> Bayberry leaf extract (a compound liquid prepared by diluting the Bayberry leaf extract prepared in the above "(Preparation of Bayberry Extract (Bayberry Leaf Extract))" with pure water (the compound liquid contains 1% of the prepared Bayberry leaf extract and 0.3% of 1,3-butylene glycol at a final concentration) Control (placebo): a compound liquid prepared by diluting 1,3-butylene glycol with pure water (the compound liquid contains 0.3% of 1,3-butylene glycol at a final concentration) <Test site> Designated areas on both left and right arms (inside of upper arms) <Sample application method> Twice a day (morning and night), take one drop of the sample and apply it directly to the test site (designated area) by rubbing it in with your finger

[0106] A comparative test was conducted on test sites before and after single use of the sample and four consecutive weeks of use to evaluate the stratum corneum moisture content. Measurement of stratum corneum moisture content was performed using a Corneometer (Courage+Khazaka electronic GmbH). For single use, stratum corneum moisture content was measured before application and 20 minutes after application. For four consecutive weeks of use, stratum corneum moisture content was measured before the first application (first day) and the day after the end of four weeks of application, at the same time as the measurement before the first application. Measurements were taken three times for each test site on the subject's upper arm, and the average value was used as the stratum corneum moisture content. Figure 6 shows the results of the human monitor test. Bayberry leaf extract demonstrated superior results in terms of stratum corneum moisture content compared to the control (placebo) in both single use and four consecutive weeks of use. This also clearly shows that bayberry extract can increase the skin's moisture retention ability and enhance the skin's moisturizing effect, in other words, it has the function of improving skin function.
