Provided is a method for solid-phase peptide synthesis comprising: (a) introducing glutamic acid and aspartic acid derivatives to a peptide backbone on a solid support to obtain a starting peptide; (b) activating the starting peptide with nitrite; (c) coupling the starting peptide with aliphatic chain and functionalities under ultrasound irradiation, or reacting the starting peptide with a reducing agent or an oxidizing agent under ultrasound irradiation to obtain a desired peptide; and (d) cleaving the desired peptide from the solid support.

METHODS FOR SOLID-PHASE PEPTIDE SYNTHESISFIELD OF THE INVENTION

[0001] The present invention relates to a method for solid-phase peptide synthesis, which comprises introducing glutamic acid derivatives or aspartic acid derivatives to a peptide backbone on a solid support and making the glutamic acid derivatives or aspartic acid derivatives for side chain attachment or modification by activation and ultrasound treatment. DESCRIPTION OF PRIOR ART

[0002] Site-selective modification on a peptide is important for the clinical applications and biomedical investigation of a peptide. For various applications, peptides are usually needed to functionalize suitable moieties, such as PEG, fluorescence tags, and contrast agents. However, unspecific modification causes low purity and batch-to-batch inconsistency. Moreover, the biological activity of a peptide is jeopardized once the modification is mistakenly introduced to the residue as a pharmacophore.

[0003] Therefore, a selective modification is needed, especially for biomedical applications. Besides, current peptide functionalization relies mostly on lysine and cystine, which own hydroxy or mercapto groups as nucleophiles on the side chain. Therefore, the introduction of an extra functional group relies on nucleophilic reaction. However, similar modifications relying on electrophilic reactions are rarely reported due to the nature of amino acids. No efficient way to introduce a nucleophile to a peptide limited the synthesis of peptide analogues and their development.DETAILED DESCRIPTION OF THE INVENTION

[0004] For this purpose, 1, 3-diamino-4-nitrobenzene (Dnb) is introduced to aspartic acid and glutamic acid to generate the Fmoc-Glu (Dnb) -OH and Fmoc-Asp (Dnb) -OH. Both residues can be introduced to a peptide through conventional solid-phase peptide synthesis. Dnb is on-bead activated by alkyl nitrite to be benzotriazole (Bt) , which is a good leaving group for the introduction of various functional groups. Regardless, hydrolysis is inevitable while the reaction proceeded at room temperature with a shaker or under microwave irradiation.

[0005] The present invention discovers that the substitution under ultrasonic irradiation will greatly accelerate the reaction and reduce the hydrolysis. Ultrasound irradiation exhibits a facile, efficient route in synthetic chemistry and accomplishes a fast-growing technique, especially for solid-phase peptide synthesis. Ultrasound treatment enhances the thermal heating and mass transfer through the “cavitation effect, ” and improves the solubility of nucleophiles, and increases reaction rates. Armed with ultrasound irradiation, the present invention efficiently suppresses the hydrolysis at the substitution step. To combine the new protecting group and ultrasound irradiation, the present invention demonstrates a “clickable conjugation” strategy to attain the side chain modification, cyclization, and on-bead ligation in minute time. This method is suitable for the modification of aspartic acids, glutamic acids, glutamine, and asparagine.

[0006] Using these two residues, the present invention reports an efficient and selective synthetic method to functionalize the sidechain of aspartic acids, glutamic acids, glutamine, and asparagine with the assistance of ultrasonication.

[0007] While molecules with mono-and multi-nucleophiles are used as nucleophiles, corresponding functional peptides, including amphiphilic peptides, functional peptides, and functional branched peptides, are collected with only filtration. When another peptide is used as nucleophile, isopeptides are collected with simple filtration. While the prepared peptides possessed inherent nucleophiles, side chain-to-side chain, and side chain-to-tail cyclic peptides are collected with only filtration.

[0014] As used herein, the term “solid support” refers to a polymer carrier for use in solid-phase synthesis of polypeptides. The solid support refers to a resin or other polymer substances having porous-rich solid and solubility-poor ability. In one embodiment, the solid support comprises controlled-pore glass, magnetic beads, Rink amide resin, Tentagel resin, Wang resin, Merrifield resin, MBHA resin, PAM resin, PAL resin, Sieber Amide resin, trityl resin, Polyethylene Glycol-Polystyrene resin, Weinreb resin, oxime resin, DHP resin or Safety-catch resin. In a preferred embodiment, the trityl resin comprises trityl chloride resins, 2-chlorotrityl chloride resins, 4-carboxytrityl resins, 4-methyltrityl chloride resins, 2-hydrazine trityl resins, or 4-methoxytrityl chloride resins.

[0015] In the present invention, the compound of formula (I) is prepared by a solid phase peptide synthesis (SPPS) . The SPPS method comprises the steps of coupling of amino acid fragments, deprotection, reprotection, isolation and purification for synthesizing desired peptides. Therefore, the SPPS method is performed by treatment with a coupling reagent and a deprotection solution.

[0048] Example 1: General procedure for Dnb monomer synthesis

[0049] Fig. 1 showed the synthesis of Dnb monomer. The synthetic method comprised the following steps:

[0050] 1. Fmoc-LGlu-OtBu (1) (10 g, 23.50 mmol) , HATU (hexafluorophosphate azabenzotriazole tetramethyl uronium) (1.5 equiv, 13.40 g, 35.25 mmol) , DMF (Dimethylformamide) (100 mL) and NMM (N-methylmorpholine) (2 equiv, 4.759 g, 47 mmol) were sequentially added to the reaction bottle then cooled the reaction mixture to 5℃.

[0051] 2. The mixture was stirred for 15 min, and 1, 2-diamino-4-nitrobenzene (1.2 equiv, 3.96 g, 25.85 mmol) was added to the reaction mixture.

[0052] 3. After removing ice bath, the resulting mixture was additionally stirred for 1 hour.

[0053] 4. The resulting mixture was cooled to 5℃ and was added slowly 0.5 M HCl (aq) (300 mL) .

[0063] 1. Solid-phase peptide synthesis (SPPS) resin (Rink amide resin 0.05 mmol) was pre-swollen in DMF (2 mL) for 1 hour and then filtrated and washed by DMF (2 mL) .

[0064] 2. The Fmoc protecting groups were removed by ultrasound agitation with 20%piperidine in DMF (2 mL ×1 min each) , followed by washing with DMF (6 mL) .

[0065] 3. Fmoc-amino acids (3 equiv) , PyBOP (benzotriazol-1-yl-oxytripyrrolidinophosphonium hexafluorophosphate) (3 equiv) and 5%NMM in DMF (0.5 mL) were sequentially added to the resin and then performed with ultrasound agitation for 5-10 mins.

[0067] 5. Fmoc-Asp (diamino-nitrobenzene) -OH (3 equiv) , HATU (3 equiv) and 5%NMM in DMF (0.5 mL) were sequentially added to the resin and then performed with ultrasound agitation for 5-10 mins.

[0068] 6. In N-terminal capping step, acetic anhydride (4 equiv) and 5%NMM in DMF (4 mL) were sequentially added to the resin, followed by ultrasound irradiation for 5 mins.

[0069] 7. After each coupling, capping and step, the resin was washed with DMF (6 mL) and DCM (Dichloromethane) (6 mL) three times to obtain the product peptide (Ia) .

[0070] Example 3: The side chain modification procedure

[0071] Fig. 3 showed the side chain modification of the peptide (Ia) . The synthetic method comprised the following steps:

[0072] 1. Isoamyl nitrite (60 equiv, 0.4 mL) and AcOH (1.8 equiv, 5 uL) were added to a vial and stirred for 30 mins at 28℃.

[0073] 2. The protected peptide-bounded resin (Ia) was pre-swollen with DMF (3.6 mL) for 30 mins.

[0074] 3. After 30 mins, the resin was treated with isoamyl nitrite solution, followed by mechanical shaking for 15 mins.

[0075] 4. The protected peptide-bounded resin (IIa) was filtrated and washed with DMF (6 mL) and DCM (6 mL) .

[0076] 5. DMF (4 mL) and octylamine (4 equiv) were added to the resin, followed by ultrasound agitation for 1 min.

1.A method for solid-phase peptide synthesis, comprising:(a) providing a solid support loaded with a compound of formula (I) ,wherein,X is -C-, -C-C-,P1 is H or a first amino acid derivative,P2 is OH or a second amino acid derivative, which is linked to the solid support, andY is an electron-withdrawing functional group or an electron-donating functional group;(b) reacting the compound of formula (I) with a nitrite in a polar solution to obtain a compound of formula (II) ,(c) reacting the compound of formula (II) with a third amino acid derivative having a N-terminal protecting group in the polar solution under ultrasound to obtain a compound of formula (III) ,wherein P3 is the third amino acid derivative having the N-terminal protecting group; and(d) deprotecting and cleaving the compound of formula (III) from the solid support.2.The method of claim 1, wherein the first amino acid derivative, the second amino acid derivative and the third amino acid derivative comprise a cyclic peptide, lipopeptide, peptide-drug conjugate, peptide dendron conjugate, isopeptide, branch peptide, or amphiphilic dendrimer.3.The method of claim 1, wherein the electron-withdrawing functional group comprises NO2, halogen, carboxylic acid, and carboxylic acid derivatives.4.The method of claim 1, wherein the electron-donating functional group comprises methyl or methoxy.5.The method of claim 1, wherein the step (c) further comprises adding a base solution in the reaction.6.The method of claim 1, wherein the deprotection of the step (d) is performed by treatment with an acid solution.7.The method of claim 1, wherein the frequence of the ultrasound ranges from 35 kHz-80 kHz.8.A method for solid-phase peptide synthesis, comprising:(i) providing a solid support loaded with a compound of formula (I) ,wherein,X is -C-, -C-C-,P1 is a N-terminal protecting group or a first amino acid derivative having a N-terminal protecting group,P2 is OH or a second amino acid derivative, which is linked to the solid support, and Y is an electron-withdrawing functional group or an electron-donating functional group;(ii) reacting the compound of formula (I) with a nitrite in a polar solution to obtain a compound of formula (II) ,(iii) reacting the compound of formula (II) with a reducing agent in the polar solution under ultrasound to obtain a compound of formula (IV) ,(iv) deprotecting and cleaving the compound from the step (iii) from the solid support.9.The method of claim 8, wherein the first amino acid derivative and the second amino acid derivative comprise a cyclic peptide, lipopeptide, peptide-drug conjugate, peptide dendron conjugate, isopeptide, branch peptide, or amphiphilic dendrimer.10.The method of claim 8, wherein the electron-withdrawing functional group comprises NO2, halogen, carboxylic acid, and carboxylic acid derivatives.11.The method of claim 8, wherein the electron-donating functional group comprises methyl or methoxy.12.The method of claim 8, wherein the reducing agent comprises NaBH4, trimethylamine borane, BH3-tetrahydrofuran, Red-Al, diisobutylaluminum hydride or LiAlH4.13.The method of claim 8, wherein the step (iii) further comprises reacting the compound of formula (IV) with an oxidizing agent in the polar solution under ultrasound to obtain a compound of formula (V) , 14.The method of claim 13, wherein the oxidizing agent comprises sodium periodate, potassium periodate, o-iodoxybenzoic acid, Dess-Martin reagent or lead tetraacetate.
