[0008] The present invention relates to a composition comprising a recombinant nucleic acid sequence for generating one or more synthetic anybodies, including an Fc- modified DNA-encoded monoclonal antibody targeting PcrV, and functional fragments thereof, and a method of preventing and / or pneumococcal infection by administenng said composition.

[0009] BACKGROUND

[0010] Multidrug-resistant (MDR) Pseudomonas spp. are among the most difficult pathogens to treat. Infections by Pseudomonas spp. are a leading cause of acute pneumonia and chronic lung infections in individuals with cystic fibrosis, and are the most common source of infections of bum wounds or other injuries where they can lead to septic mortality. Pseudomonas spp. are able to attach to the surfaces of medical devices such as medical implants, catheters, and artificial joints and cause multiple problems, for example clogging a catheter or physically damaging an implant. Pseudomonas, as biofilm forming bacteria, are highly resistant to high levels of antibiotics. Currently, therapeutic antibodies are approved for treatment of multiple diseases. Unfortunately, manufacture and delivery of purified antibodies is cost-prohibitive. Furthermore, these antibody therapies must be re-administered weekly-to-monthly - a challenging consideration in treatment of chronic conditions such as prevention or treatment of biofilm formation on a medical implant.

[0011] Thus there is need in the art for improved therapeutics that prevent and / or treat Pseudomonas aeruginosa infection and biofilm formation. The current invention satisfies this need.

[0013] In some aspects, the present invention relates to an anti-PcrV antibody or fragment thereof.

Deposition of C3b onto the surface of Pseudomonas aeruginosa is suggested to be a less effective pathway for clearance of the bacteria (alternative pathway). However, the classical complement pathway involves antibody recognition of surface antigens and binding of Clq to the Fc-region to activate the cascade resulting in pathogen phagocytosis. Literature suggests that IgG hexamerization plays a major role in the activation of the classical complement pathway. Figure 12B depicts V2L2 DMAb variant binding to Clq (50 pg / mL) coated ELISA plates. Purified V2L2 DMAbs were prepared at a concentration of 0.5 mg / mL (diluted 2-fold). Figure 12C depicts purified V2L2 DMAb Variant Fc-binding guinea pig complement and probed with anti-guinea pig C3 HRP secondary antibody compared to Fc-abrogated TM V2L2, Flu DMAb 2-12C control, and IgG Isoty pe Control. Figure 12D depicts in vivo produced V2L2 DMAb C3 deposition was measured utilizing guinea pig complement and probed with anti-guinea pig C3 HRP secondary antibody. Figure 12E and Figure 12F depicts Opsonophagocytic killing (OPK) was assessed by adding differentiated HL60s, baby rabbit complement, Pseudomonas aeruginosa strain PA14, and purified V2L2 DMAbs in triplicate on 96-well plates and incubated for 1 hour. OPK reactions were plated on LB agar plates and colonies were counted the next day. Control wells included PAM, differentiated HL60s, and baby rabbit complement with no antibody. [0036] The present invention relates to compositions comprising a recombinant nucleic acid sequence encoding an antibody, a fragment thereof, a variant thereof, or a combination thereof. The composition can be administered to a subject in need thereof to facilitate in vivo expression and formation of a synthetic antibody.

11. A composition comprising at least one anti-PcrV antibody or fragment thereof of any one of claims 1-3.

12. A composition comprising at least one nucleic acid molecule of any one of claims 4-7.

13. A composition comprising a combination of nucleic acid molecules of any one of claims 8-10.

14. The composition of claim 13, further comprising a pharmaceutically acceptable excipient.

A method of preventing or treating a disease in a subject, the method comprising administering to the subject the antibody or antibody fragment of any one of claims 1-3, the nucleic acid molecule of any one of claims 4-7, the combination of nucleic acid molecules of any one of claims 8-10 or a composition of any one of claims 10-14.

16. The method of claim 15, wherein the disease is a Pseudomonas aeruginosa infection.
