[0001] The present invention belongs to the field of medicine, and relates to oxy-amide compounds, preparation methods thereof, pharmaceutical compositions comprising the compounds, and medical uses thereof.Background of the Invention

[0002] The Hippo signaling is an evolutionarily conserved pathway that controls organ size by regulating cell proliferation and apoptosis. Core to the Hippo pathway is a kinase cascade consists of protein kinases Mst1 / 2, LATS1 / 2 and transcription co-activators YAP / TAZ. When Hippo pathway is activated (Hippo-On) , Mst1 / 2 phosphorylate and activate LATS1 / 2. LATS1 / 2 in turn phosphorylate YAP / TAZ and limit YAP / TAZ function by means of cytoplasmic retention and proteasomal degradation. On the other hand, inactive Hippo pathway (Hippo-Off) allows YAP / TAZ to enter the nucleus where YAP / TAZ associate primarily with TEAD family transcription factors to activate expression of pro-proliferation and anti-apoptosis genes.

[0003] Hippo signaling pathway dysregulation, characterized by hyperactivity of YAP / TAZ, has been implicated in different types of cancers. The most prominent cancer related to Hippo pathway dysregulation is malignant pleural mesothelioma (MPM) . In MPM, loss of NF2 and / or LATS1 / 2 function results in Hippo-Off and YAP / TAZ hyperactivation. Moreover, elevated levels of YAP / TAZ have been observed and connected to poor prognosis in breast cancer, lung cancer, ovarian cancer, colorectal cancer, pancreas cancer, prostate cancer, gastric cancer, liver cancer, etc.

[0004] In addition to controlling cell growth and apoptosis, the Hippo pathway is also implicated in drug resistance and immunosurveillance escape of cancer cells. As reported in several genetic and pharmacological studies, YAP / TAZ activation confers resistance to both chemotherapeutic drugs and small molecule drugs targeting KRAS, BRAF, MEK, EGFR, etc. Accumulating evidence has demonstrated the immunomodulatory effects of YAP / TAZ in malignant neoplasms. YAP / TAZ can promote not only the PD-L1 expression in cancer cells, but also the expression of CXCL5 which recruits myeloid derived suppressor cells (MDSCs) to suppress T cells in tumor microenvironment. Association with TEAD family transcription factors is necessary for YAP / TAZ’s function due to the lack of DNA-binding domain in themselves. Therefore, modalities disrupting YAP / TAZ-TEAD interaction have emerged as attractive strategies for YAP / TAZ inhibition in cancer. In this invention, we designed and optimized compounds as TEADs binders that selectively disrupt the YAP / TAZ-TEAD interaction and benefit the treatment of diseases mentioned above.SUMMARY OF THE INVENTION

[0180] The effect of compounds on cell viability was assessed using NCI-H2052 (ATCC, CRL-5915) and NCI-H226 (ATCC, CRL-5826) mesothelioma cells cultured in RPMI1640 medium containing 10%FBS and 1%Penicillin / Streptomycin at 37℃ in a humidified 5%CO2 incubator. Briefly, both cell lines were seeded at 1000 cells / 40uL / well into white 384-well plates with addition of serial diluted compounds described above. After incubation for 72 hours in the incubator, 40uL of CellTiter-Glo solution (Promega, G9241) was added to each well and incubate at room temperature for 10 minutes. Compound-induced cell viability change was determined by measuring the luminescence intensity on VICTOR Nivo multimode plate reader (PerkinElmer) . For data analysis, the background luminescence intensity value determined in wells containing growth medium only was subtracted from all data points. Dose-dependent compound efficacy was expressed as percentage of DMSO-treated control and IC50s was calculated using a four-parametric fit model with Prism GraphPad.

[0190] Balb / c Mouse, SPF grade were administered with the Example compound via IV and oral gavage as a clear and colorless solution. Plasma was collected from mice per time point at 0.083, 0.25, 0.5, 1, 2, 4, 6, 8, 24 h after administration. The concentrations of the test article were measured in plasma by HPLC–MS / MS at each time point. Chromatographic separation was carried out on Waters BEH C18 Column (2.1×50 mm, 1.7 μm) column (MAC-MOD Analytical, Chadds Ford, PA) , using a gradient elution method with water and acetonitrile, both containing 0.1 %formic acid. Mass spectrometric measurements in positive electrospray ionization were directed at quantifying the mass transition with [M + H] + as the precursor ion on API6500, triple quadruple mass spectrometer (Sciex, Framingham, MA) . The relevant pharmacokinetic parameters were estimated using noncompartmental methods using WinNonlin (Enterprise, version 8.2) .
