[0006] Cytokines belonging to the common γ chain (γc) family are central regulators of the development, proliferation, survival and differentiation of multiple cell lineages of the innate and adaptive immune system, and as such are of high interest for anticancer therapeutic applications.

[0007] Among them, IL-2 is the most comprehensively one. It became the first immunotherapy approved by the United States Food and Drug Administration (FDA) for cancer treatment nearly 30 years ago. However, IL-2 has the effect of promoting the proliferation of regulatory T cells and CD4+ helper T cells. It can also eliminate self-relative T cells via activation-induced cell death (AICD) to prevent autoimmunity. Thus, IL-15 has gradually emerged as an alternative to IL-2 in cancer treatment because of its functional similarity to IL-2 with several added benefits. For example, the physiological / pathological roles and mechanisms of action of IL-15 in immunity have been more thoroughly investigated. IL-15 is essential in the growth, mobilization and activation of immune cells such as natural killer (NK) cells, CD8+ T cells, γδ T cells and natural killer T (NKT) cells. IL-15 can also enhance the anti-tumor activity of immune cells.

[0008] Recombinant IL-15 has been tested as an antitumor agent in various clinical trials, which showed promising anti-tumor activities. However, due to unspecific activation of immune cells by IL-15 during systemic circulation, excessive toxicity has been reported in cancer patients, which limits its clinical application.

[0009] Thus, there is a need to develop cancer therapies targeting the IL-15 pathway with enhanced anti-tumor efficacy and limited toxicities.SUMMARY

[0010] This disclosure relates to engineered IL2Rβ variants, protein constructs, and methods of use thereof. In some embodiments, the variants and protein constructs thereof include one or more non-native histidine residues within wildtype IL2Rβ extracellular region, without interfering the overall structure or residues critical for IL2Rβ / IL-15 interaction. As a result, the engineered IL2Rβ variants or protein constructs thereof can have a comparable or high binding affinity to IL-15 at a neutral pH (e.g., about pH 7.5) and a low binding affinity to IL-15 at an acidic pH (e.g., about pH 6.0) . Because tumor microenvironment usually has an acidic pH, the engineered IL2Rβ variants and protein constructs thereof described herein can be used as a “protective lid” to shield IL-15 from activating immune cells prematurely during systemic circulation. Once the variants or protein constructs reach tumor sites, the lid can be loosened up, which enables IL-15 to activate immune cells within the tumor microenvironment.

[0011] Also provided herein are methods for screening IL2Rβ variants or protein constructs thereof with desired pH-dependent binding ability to an IL2Rβ ligand (e.g., IL-15 or a functional variant thereof) . The methods can be useful for identifying IL2Rβ variants or protein constructs thereof to selectively release the IL2Rβ ligand within tumor microenvironment. In some embodiments, the screened IL2Rβ variants or protein constructs thereof can be linked to a targeting moiety that specifically binds to a tumor-associated antigen, to direct the IL2Rβ variants or protein constructs thereof to tumor sites.

[0219] To determine the binding activity between IL-15 and engineered IL2Rβ variants, ELISA assays were performed. Specifically, His-tagged IL-15 sushi domain (or His-sushi-IL15_wt; SEQ ID NO: 19) and variants was diluted in coating buffer (15 mM Na2CO3, 35 mM NaHCO3, 7.7 mM NaN3 , pH 9.6 in ddH2O) (10 μg / mL) and incubated in the 96 well half area plate (CORNING, Cat#: 3690) at 4℃ overnight. The wells were blocked with 2%BSA (APOLO Biochemical, Cat#: APL-007-500g) in PBS (Gibco, Cat#: 14190-136) at room temperature for 1 hour, and then washed with PBST (0.05% 20 in PBS) . After washing, the wells were incubated with serially diluted IL2Rβ_wt and variants (from 50 nM, 8-fold diluted with 0.5%BSA in PBS, pH 7.5) at room temperature for 2 hours. The plate was washed three times with PBST, then incubated with pH 7.5 and pH 6.0 0.5%BSA in PBS at room temperature for 30 minutes. After washing with PBST, anti-human Fcγ-HRP antibody in dilution solution (1: 3000 dilute with 0.2%BSA in PBS; Jackson ImmunoResearch, Cat#: 109-005-098) was added and incubate at room temperature for 1 hour. Next, the plate was washed four to five times with PBST. Finally, color development was carried out with TMB substrate (Invitrogen, Cat#: 00-4201-56) . The color development was stopped by adding an equal volume of 2N H2SO4 per well as TMB substrate solution. Absorbance at 450 nm in each well was measured using a microplate reader (VarioskanTM LUK, Thermo Scientific, type3020) .
