Abstract
Description
Technical Field
[0001] The present invention belongs to the field of biomedicine technology, and specifically relates to a PROTAC compound for targeted degradation of ALK protein and an application thereof. Background Art
[0002] Anaplastic lymphoma kinase (ALK) was first discovered in anaplastic large cell lymphoma (ALCL) and is the product of the NMP-ALK fusion gene, which is caused by the chromosomal translocation t(2;5)(p23;Q35). Subsequent studies have found more ALK gene rearrangements, including EML4-ALK in non-small cell lung cancer (NSCLC), CARS-ALK in inflammatory myofibroblastic tumor (IMT), CLTC-ALK in diffuse large B-cell lymphoma (DLBCL), etc. In the case of full-length ALK, ALK is activated by ligand-induced homodimerization. However, when chromosomal rearrangements occur in the ALK domain, oligomerization of the ALK fusion protein mediated by the oligomerization domain of the fusion partner induces spontaneous activation of the kinase. These oncogenic tyrosine kinases play a key role in the occurrence and development of various cancers by activating downstream signaling pathways such as PI3K / AKT, Ras / ERK, JAK / STAT, etc. ALK is an important target for the treatment of non-small cell lung cancer, and most ALK-positive patients experience significant remission after treatment with ALK inhibitors. A variety of ALK inhibitors have been used clinically, including crizotinib, alectinib, ceritinib, brigatinib, and lorlatinib. Compared with traditional chemotherapy, targeted drugs have significantly improved progression-free survival (PFS) and objective response rate (ORR). However, drug-resistant mutations will inevitably occur during the use of kinase inhibitors, leading to treatment failure. In addition, kinase inhibitors can only inhibit enzyme activity, which is not enough to completely inhibit the biological function of the target. Therefore, it is of great significance to develop new ALK targeted treatment strategies. Summary of the invention
[0003] In view of the above-mentioned deficiencies in the prior art, the present invention provides a PROTAC compound for targeted degradation of ALK protein and its application, which can effectively solve the problem that existing small molecule inhibitors cannot degrade ALK protein, and the compound can inhibit the proliferation of ALK resistant mutant cell lines.

[0048] Test example
[0049] The anti-Karpas299 cell line proliferation activity and ALK degradation activity of the PROTAC compounds targeted for degradation of ALK protein prepared in Examples 1, 2, and 3 were tested respectively. The specific test methods are as follows, and the specific test results are shown in Tables 1-2.
[0050] 1. Anti-Karpas299 cell line proliferation activity test
[0051] Collect cells in logarithmic growth phase, resuspend and count with a cell counter. Take an appropriate amount of cell suspension according to 9000 cells per well of 96-well plate, mix with culture medium and plate. After 72 hours of action of gradient concentration bifunctional PROTAC compounds, add 10 μL of CCK8 to each well, incubate in the dark for 2 to 4 hours, and detect the absorbance value corresponding to each well at a wavelength of 450 nm with an enzyme reader;
[0052] Cell inhibition rate = 100% - (average absorbance of the drug-added group / average absorbance of the blank control group) × 100%, and the half inhibition concentration IC is obtained by analysis and calculation. 50 , the specific results are shown in Table 1-2.

[0054]
[0055] Table 2: Antiproliferative activity of PROTAC compound S1-6F
[0056]
[0057]
[0058] The data in Tables 1 and 2 show that all PROTAC molecules have good anti-proliferation activity against Karpas299 cell lines. Among them, PROTAC molecules 4B, 5A and 6A have the best activity, which is comparable to that of ceritinib (IC 50 <5nM).
[0059] 2. ALK degradation activity test
[0060] Karpas299 cells in the logarithmic growth phase were plated in a 6-well plate and treated with ALK bifunctional PROTAC compounds S6, L1, 4B, 4C, 5A, 5D, 6A, and 6D for 48 hours. The cells were washed three times with pre-cooled PBS, and then collected with a scraper. An appropriate amount of RIPA lysis buffer containing protease inhibitors was added to lyse the cells on ice for 15 minutes. After low-temperature ultrasound, the supernatant was collected by centrifugation (4°C, 13000 rpm, 15 minutes), and the supernatant was mixed with SDS-PAGE protein loading buffer (sodium dodecyl sulfate-polypropylene glycol). After mixing with 1% 2-amino-4-nitropropene amide gel, the mixture was placed in boiling water and boiled for 10 minutes. The protein sample was separated by 10% SDS-PAGE electrophoresis and then transferred to a PVDF (polyvinylidene fluoride) membrane. The membrane was blocked with skim milk at room temperature for one hour. β-actin was used as the internal reference protein. The corresponding primary antibody and PVDF membrane were incubated at 4°C overnight. After further labeling with the corresponding horseradish enzyme-labeled secondary antibody, the immunoblot was developed by chemiluminescence. The activity of the ALK bifunctional PROTAC compound in degrading ALK was determined by comparing the grayscale of the corresponding protein immunoblot. The specific results are shown in Figure 1 .
[0061] Figure 1 The results showed that PROTAC molecules S6, L1, 4B, 4C, 5A, 5D, 6A, and 6D can induce a significant decrease in ALK protein levels, thereby effectively inhibiting ALK protein function and exerting anti-tumor activity.
