Description

Technical Field

[0001] The invention relates to a collection kit and a collection method for collecting blood preparations for inspection. Background Art

[0002] Blood products include red blood cell products, plasma products, platelet products, and whole blood products. As for blood products, products containing the components required by patients will be used for blood transfusion. Blood products are stored and transported in medical bags. In order to ensure the safety of blood products, a small amount of samples are sometimes collected for culture inspection. In the culture inspection, the presence of pathogens is detected by collecting the sample in a culture bottle and placing the culture bottle in an environment suitable for bacterial proliferation.

[0003] Among blood products, the inspection of platelet products is also carried out according to the following steps. A small-volume collection bag is connected to a medical bag containing platelet products (hereinafter referred to as a platelet bag). Then, a portion of the platelet products in the platelet bag is transferred to the collection bag. Next, the collection bag is separated from the platelet bag.

[0004] Next, the collection bag is transported to a clean bench. In the clean bench, the platelet preparation in the collection bag is injected into a blood culture bottle. Before the injection, the tube of the collection bag is connected to the tube extending from the sample collection tube. Based on the scale of the sample collection tube, a prescribed amount of the platelet preparation is transferred from the collection bag to the sample collection tube.

[0005] Then, the operator operates the valve of the sample collection tube while visually observing the scale of the sample collection tube, and transfers a specified amount of platelet preparation from the sample collection tube to the blood culture bottle. The culture test performs anaerobic culture and aerobic culture. Therefore, the platelet preparation is divided into a culture bottle for anaerobic culture and a culture bottle for aerobic culture. Then, the culture bottle is set in the culture device and provided for the culture test for a specified period of time.

[0006] For example, US Pat. No. 8,777,921 discloses a collecting tool for collecting a sample from a medical bag. Summary of the invention

[0007] From the viewpoint of further improving the safety of platelet preparations, it is being studied to perform culture inspection on all platelet preparations. Therefore, it is required to improve the working efficiency of culture inspection on platelet preparations. In addition, when all platelet preparations are cultured and inspected, it is desired that the apparatus for dispensing the platelet preparations into culture bottles has a simple structure and can be low-cost.

[0008] An object of the present invention is to solve the above-mentioned problems.

[0009] One viewpoint disclosed below is a collection kit, which comprises: an inflow tube connected to a medical bag containing a collection object; a collection bag connected to the downstream of the inflow tube, having a plurality of storage chambers formed by overlapping and welding two flexible sheets, and containing a predetermined amount of the collection object in each of the storage chambers; a plurality of outflow ports connected to each of the storage chambers to allow the collection object to flow out of the storage chambers; and an adapter connected to the downstream of each of the outflow ports, and connecting the culture bottle to the outflow port by installing a culture bottle, the collection bag having a connecting flow path connecting adjacent storage chambers, the connecting flow path having a tubular portion formed integrally with the storage chambers using the two flexible sheets, and formed into a tubular shape by cutting the surrounding flexible sheets between the adjacent storage chambers.

[0010] In addition, another aspect is a collection method, which uses the collection kit of the above aspect and comprises: a step of connecting the medical bag to the inflow tube; a step of introducing the collection object into the receiving chamber of the collection bag through the inflow tube; a step of separating the medical bag from the inflow tube; a step of closing the tubular portion to prevent the movement of the collection object between the plurality of receiving chambers; and a step of connecting the culture bottle to the adapter and injecting the collection object into the culture bottle.

[0011] The collection kit and collection method of the above-mentioned viewpoint have a simple structure and can therefore be manufactured at low cost, and also have excellent working efficiency. BRIEF DESCRIPTION OF THE DRAWINGS

[0017] (First embodiment)

[0018] Figure 1 The collection kit 10 according to the present embodiment is used in a culture test for confirming the safety of a blood product in a business such as a blood center that manufactures blood products. In the culture test, the culture of anaerobic bacteria and the culture of aerobic bacteria are tested. Therefore, in the culture test, a first culture bottle 90 (see FIG. 1 ) for anaerobic culture is used. Figure 3 ) and the second culture bottle 92 for aerobic culture (see Figure 3 The collection kit 10 is used to collect a liquid sample (platelet preparation) and dispense a predetermined amount into two culture bottles 90 and 92.

[0019] In this embodiment, the example of using a platelet preparation as a blood preparation is described, but there is no particular limitation. Figure 1 As shown, the collection kit 10 includes an inflow tube 12, a collection bag 14, a first sealing member 16, a second sealing member 18, an exhaust valve 20, a first adapter 21a, and a second adapter 21b. In addition, in the collection kit 10, based on the configuration state during use, the direction in which the inflow tube 12 and the exhaust valve 20 are located is referred to as the upper side or the top, and the direction in which the first adapter 21a and the second adapter 21b are located is referred to as the lower side or the bottom. In addition, the direction perpendicular to the up-down direction and along the plane of the collection bag 14 is referred to as the width direction, and the direction perpendicular to the plane of the collection bag 14 is referred to as the thickness direction.

[0020] The inflow tube 12 is a translucent medical tube made of a thermoplastic resin such as vinyl chloride resin. The inflow tube 12 can be connected or disconnected with other medical tubes without exposing the inside to the outside air by, for example, a sterile joint device, a tube seal, etc. The inflow tube 12 has a first end 12a on the upstream side and a second end 12b on the downstream side. The first end 12a is welded and sealed in the initial state. The second end 12b is connected to the upper end of the collection bag 14.

[0021] The collection bag 14 has two flexible sheets 22 overlapping in the thickness direction. The two flexible sheets 22 are welded and joined to each other at a plurality of welded portions 24. The flexible sheet 22 has: a first region 22a for forming one storage chamber (a first storage chamber 26); a second region 22b for forming another storage chamber (a second storage chamber 28); and a connecting region 22c for connecting the first region 22a and the second region 22b. The first region 22a has a rectangular shape of the same size as the second region 22b in a plan view. The connecting region 22c extends in the width direction and integrally connects the first region 22a and the second region 22b. The flexible sheet 22 has a pair of cutouts 22d for cutting the flexible sheet 22 into a rectangular shape on the upper and lower sides of the connecting region 22c. The connecting region 22c is formed between the upper and lower cutouts 22d, and is formed in a manner that the dimension in the vertical direction is narrower than the dimension in the width direction. The connection region 22c is located at the center of the first region 22a and the second region 22b in the vertical direction, but is not particularly limited.

[0022] The collection bag 14 includes a first receiving chamber 26, a second receiving chamber 28, and a connecting flow path 30 defined by a weld portion 24. The first receiving chamber 26 is formed in the first region 22a of the flexible sheet 22. The first receiving chamber 26 receives the platelet preparation introduced into the first culture bottle 90. The first receiving chamber 26 bulges in the thickness direction of the flexible sheet 22 in an initial state (a state when the product is provided). In the first receiving chamber 26, one flexible sheet 22 is separated from another flexible sheet 22 in the thickness direction. The first receiving chamber 26 has a volume for receiving a predetermined amount (for example, about 8 ml or about 10 ml) of the platelet preparation.

[0023] The collection bag 14 has an inflow port 32 connected to the first storage chamber 26, and a first outflow port 34. The inflow port 32 is connected to the upper end of the first storage chamber 26 of the collection bag 14. The inflow port 32 is connected to the second end 12b of the inflow tube 12, so that the inflow tube 12 and the first storage chamber 26 are connected. The first outflow port 34 (outflow port) is connected to the lower end of the first storage chamber 26. The first outflow port 34 is connected to the first sealing member 16. The first adapter 21a is connected to the first outflow port 34 via the first sealing member 16.

[0024] The second storage chamber 28 is formed in the second region 22b of the flexible sheet 22. The second storage chamber 28 stores the platelet product introduced into the second culture bottle 92. The second storage chamber 28 has the same shape as the first storage chamber 26 and stores the platelet product of the same volume.

[0025] The collection bag 14 has a second outflow port 36 and an exhaust port 38 connected to the second storage chamber 28. The second outflow port 36 (outflow port) is connected to the lower end of the second storage chamber 28 of the collection bag 14. The second outflow port 36 is connected to the second sealing member 18. The second adapter 21b is connected to the second outflow port 36 via the second sealing member 18. The exhaust port 38 is connected to the upper end of the second storage chamber 28. The exhaust valve 20 is connected to the exhaust port 38.

[0026] The connecting flow path 30 is a flow path connecting the first storage chamber 26 and the second storage chamber 28. The end 30a on the upstream side of the connecting flow path 30 is connected to the upper part of the first storage chamber 26. The connecting flow path 30 extends obliquely upward from the end 30a, bends at the top 30b and extends downward. The connecting flow path 30 extends downward to the center of the vertical direction of the collection bag 14 on the downstream side of the top 30b, and bends in the width direction at the bending portion 30c. The connecting flow path 30 forms an intermediate portion 30d on the downstream side of the bending portion 30c, and the intermediate portion 30d extends in the width direction in the direction away from the first storage chamber 26. The intermediate portion of the flow path direction of the connecting flow path 30 is located at the center of the width direction of the intermediate portion 30d. The intermediate portion 30d extends along the connecting area 22c of the flexible sheet 22 and extends along the second area 22b. The connecting flow path 30 has a bending portion 30e on the downstream side of the intermediate portion 30d. The connecting flow path 30 on the downstream side of the curved portion 30e changes its direction diagonally upward in a V-shape at the turning portion 30f, and the downstream end 30g located downstream of the turning portion 30f is connected to the lower part of the second storage chamber 28.

[0027] The connecting flow path 30 bulges in the thickness direction in the initial state. In addition, the middle portion 30d of the connecting flow path 30 forms a tubular tubular portion 31, and the portion of the tubular portion 31 passing through the connecting area 22c extends slenderly in the width direction. The tubular portion 31 is sealed by the welded portion 24 narrowed at the upper and lower ends, and the middle portion 30d of the connecting flow path 30 can be sealed by passing through the cutout portion 22d and installing a clamp (klemme) 42. In addition, the tubular portion 31 can be sealed and cut using a tube seal, wherein the tube seal seals and cuts the flow path while heating the tube using high frequency or ultrasonic waves.

[0028] The first sealing member 16 seals the first outflow port 34 in the initial state, preventing the fluid from passing through the first receiving chamber 26 and the first adapter 21a. The first sealing member 16 is a cylindrical member having a breakable plug inside. When the plug is bent in the internal flow path in the initial state, the plug breaks, allowing the internal flow path to be opened. The second sealing member 18 is also a member similar to the first sealing member 16, and seals the second outflow port 36 in the initial state, preventing the fluid from passing through the second receiving chamber 28 and the second adapter 21b.

[0029] The exhaust valve 20 is connected to the second receiving chamber 28 and the first receiving chamber 26 through the exhaust port 38. The exhaust valve 20 has a filter that allows gas to pass through and prevents liquid from passing through. The exhaust valve 20 may have a hydrophilic filter, but is not particularly limited. In the case of the exhaust valve 20 having a hydrophilic filter, after the first receiving chamber 26 and the second receiving chamber 28 are filled with the platelet preparation, when the platelet preparation comes into contact, the pores of the filter are closed by the liquid, and then the exhaust valve 20 prevents the passage of gas and liquid. Such an exhaust valve 20 can prevent air from flowing back to the second receiving chamber 28.

[0030] like Figure 3 As shown, the first adapter 21a receives the neck of the first culture bottle 90. The first adapter 21a penetrates the plug of the first culture bottle 90 through the needle tube inside, so that after the first sealing member 16 is opened, the first culture bottle 90 is communicated with the first receiving chamber 26. The second adapter 21b is the same member as the first adapter 21a, and after the second sealing member 18 is opened, the second culture bottle 92 is communicated with the second receiving chamber 28.

[0031] The collection kit 10 of this embodiment is configured as described above. The collection kit 10 is used as follows.

[0032] In this embodiment, a collection method for dispensing a platelet preparation into two culture bottles 90 and 92 using the collection kit 10 is described. First, the platelet bag 94 is connected to the collection kit 10. Figure 2 As shown, the platelet bag 94 is a medical bag containing a platelet preparation. The platelet bag 94 has a connecting tube 96. The tube 96 is connected to the inflow tube 12. The tube 96 is connected to the inflow tube 12 using a sterile connection device without contact with the outside air.

[0033] Next, the platelet preparation is introduced into the collection bag 14. In this step, the collection kit 10 and the platelet bag 94 are arranged so that the platelet bag 94 is above the collection bag 14. The first sealing member 16 and the second sealing member 18 of the collection kit 10 are maintained in a closed state.

[0034] The platelet preparation flows out of the platelet bag 94 by gravity and flows into the collection bag 14 through the inflow tube 12. The platelet preparation flowing into the collection bag 14 flows into the first storage chamber 26 through the inflow port 32. The platelet preparation is stored in the first storage chamber 26, and the liquid level of the platelet preparation in the first storage chamber 26 rises. The air inside the first storage chamber 26 is squeezed out by the platelet preparation and discharged from the exhaust valve 20 through the connecting flow path 30 and the second storage chamber 28.

[0035] When the first receiving chamber 26 is filled with the platelet preparation, the platelet preparation flows into the end 30a on the upstream side of the connecting flow path 30. Then, the platelet preparation flows from the upstream to the downstream in the connecting flow path 30, and flows into the second receiving chamber 28 from the lower part of the second receiving chamber 28. As the platelet preparation flows in, the liquid level of the platelet preparation inside the second receiving chamber 28 rises. The air in the second receiving chamber 28 is discharged from the exhaust valve 20. The second receiving chamber 28 is filled with the platelet preparation. Furthermore, when the exhaust valve 20 contacts the platelet preparation, the exhaust valve 20 is closed to prevent the inflow of the platelet preparation. Thus, the introduction of the platelet preparation into the collection bag 14 is completed. A predetermined amount (for example, about 8 mL) of platelet preparation is contained in the first receiving chamber 26 and the second receiving chamber 28.

[0036] Then, the platelet bag 94 is separated from the inlet tube 12 by a tube seal (high frequency or ultrasonic seal) or the like. The tube seal separates the inlet tube 12 from the tube 96 of the platelet bag 94 and seals the first end 12a of the inlet tube 12 by welding. The inlet tube 12 and the tube 96 are separated without exposing the internal flow path to the outside air.

[0037] The removed platelet bag 94 is stored until the culture test is completed and then used. In addition, the collection kit 10 filled with the platelet preparation is carried into the clean bench.

[0038] Then, if Figure 3 As shown, the first culture bottle 90 is connected to the first adapter 21a of the collection kit 10, and the second culture bottle 92 is connected to the second adapter 21b. Then, the tubular portion 31 connecting the flow path 30 is closed by the clamp 42. By closing the tubular portion 31, the movement of the fluid is prevented between the first receiving chamber 26 and the second receiving chamber 28. It should be noted that the sealing of the tubular portion 31 can be performed by using a tube seal while cutting. In addition, the sealing of the tubular portion 31 can be performed by the following method: a seal (also called ultrasonic sealing or high-frequency sealing) in which the resin is melted and sealed by heating using ultrasonic waves or high-frequency (electromagnetic waves), thereby fusing the two flexible sheets 22. In this case, there is no need to set the clamp 42 in the circuit in advance.

[0039] Then, the first sealing member 16 is opened, the platelet preparation in the first storage chamber 26 is sucked out by the negative pressure of the first culture bottle 90, and a predetermined amount of the platelet preparation is introduced into the first culture bottle 90. In addition, the second sealing member 18 is opened, the platelet preparation in the second storage chamber 28 is sucked out by the negative pressure of the second culture bottle 92, and a predetermined amount of the platelet preparation is introduced into the second culture bottle 92.

[0040] The first culture bottle 90 is removed from the first adapter 21a, and the second culture bottle 92 is removed from the second adapter 21b. Through the above steps, the collection of the platelet product using the collection kit 10 is completed.

[0041] As described above, the collection kit 10 of the present embodiment is a structure in which the collection bag 14 is integrally formed using two flexible sheets 22, and therefore can be manufactured at a low cost. In addition, the number of times the tubes are joined and separated from each other can be suppressed to a minimum, so the operating efficiency is excellent. Moreover, there is no need to measure and collect the platelet preparation by visual measurement based on the scale line, so the operation becomes easy and the measurement error can be reduced. In addition, in the collection kit 10, the tubular portion 31 of the collection bag 14 is closed by a clamp or the like, thereby being able to reliably separate the first receiving chamber 26 and the second receiving chamber 28, and being able to prevent the platelet preparation from moving between the first receiving chamber 26 and the second receiving chamber 28. Therefore, the collection kit 10 can suppress the deviation in the amount of the platelet preparation introduced into the first culture bottle 90 and the second culture bottle 92.

[0042] (Second embodiment)

[0043] like Figure 4 As shown, the collection kit 10A of this embodiment is different from the above in that it further includes a frame member 50 for holding the collection bag 14. Figure 1 The collection kit 10 shown is different. It should be noted that Figure 4 In the collection kit 10A, the inflow tube 12, the collection bag 14, the exhaust valve 20, and the first adapter 21a and the second adapter 21b are the same as those of the collection kit 10, so their detailed description is omitted.

[0044] like Figure 5A As shown, the collection kit 10A has a frame member 50 for holding the collection bag 14. The frame member 50 has a first frame member 50a and a second frame member 50b. The first frame member 50a is arranged adjacent to one side of the thickness direction of the collection bag 14. The second frame member 50b is arranged adjacent to the other side of the thickness direction of the collection bag 14. The frame member 50 has a structure in which the collection bag 14 is sandwiched from the thickness direction by the first frame member 50a and the second frame member 50b.

[0045] like Figure 5A and Figure 5BAs shown, the first frame member 50a has a contact plate 52, a frame body 54, and a connecting portion 56. The contact plate 52 has: a rectangular first flat plate portion 52a formed in a portion corresponding to the first region 22a; and a second flat plate portion 52b formed in a portion corresponding to the second region 22b. The frame body 54 is formed in the first flat plate portion 52a over three sides except for the side facing the center in the width direction. In addition, the frame body 54 is formed in the second flat plate portion 52b over three sides except for the side facing the center in the width direction. The frame body 54 bulges in the thickness direction relative to the first flat plate portion 52a and the second flat plate portion 52b. The frame body 54 has a groove for passing the piping connected to the inlet pipe 12, the first sealing member 16, the second sealing member 18 and the exhaust valve 20 at a predetermined position. The first flat plate portion 52a and the second flat plate portion 52b are connected to each other via the connecting portion 56. The connection part 56 is located at, for example, the lower end of the first frame member 50a. The connection part 56 is located away from the tubular part 31 of the collecting bag 14 and is arranged at a position that does not hinder the attachment of the clip 42 and the like to the tubular part 31.

[0046] The first frame member 50a has a recessed portion 58 formed in a portion surrounded by the frame body 54. The recessed portion 58 accommodates a portion of the first accommodation chamber 26 and the second accommodation chamber 28 in the thickness direction.

[0047] The second frame member 50b is formed symmetrically with the first frame member 50a in the thickness direction, and has the same abutment plate 52, frame body 54 and connection portion 56 as the first frame member 50a. The second frame member 50b has a recessed portion 58 in a portion surrounded by the frame body 54, which accommodates the first accommodation chamber 26 and the other portion of the second accommodation chamber 28 in the thickness direction.

[0048] The abutment plate 52 of the first frame member 50a and the abutment plate 52 of the second frame member 50b abut against each other in such a manner as to sandwich the first receiving chamber 26 and the second receiving chamber 28 from the thickness direction, thereby preventing the first receiving chamber 26 and the second receiving chamber 28 from expanding (deforming) in the thickness direction. The abutment plate 52 prevents the variation in the amount of platelet preparation introduced by preventing the change in the volume caused by the expansion of the first receiving chamber 26 and the second receiving chamber 28 in the thickness direction.

[0049] The frame body 54 of the first frame member 50a and the frame body 54 of the second frame member 50b sandwich and hold the peripheral edge of the collection bag 14 from the thickness direction. The frame body 54 improves the handling efficiency of the collection kit 10A by maintaining the planar shape of the collection bag 14. In addition, the frame member 50 having the frame body 54 prevents the volume change caused by the deformation of the first storage chamber 26 and the second storage chamber 28, thereby suppressing the variation of the introduced amount of the platelet preparation.

[0050] It should be noted that in the collection kit 10A of this embodiment, a clamp structure for closing the tubular portion 31 may be integrally mounted on the frame member 50. In this case, there is no need to mount a separately prepared clamp 42 on the tubular portion 31, and operability and handling are further improved.

[0051] The above disclosed contents are summarized as follows.

[0052] One aspect is a collection kit (10), comprising: an inflow tube (12) connected to a medical bag (94) containing a collection object; a collection bag (14) connected downstream of the inflow tube, having a plurality of storage chambers (26, 28) formed by overlapping and welding two flexible sheets (22), and containing a predetermined amount of the collection object in each of the storage chambers; a plurality of outflow ports (34, 36) connected to each of the storage chambers to allow the collection object to flow out of the storage chambers; and adapters (21a, 21b) connected downstream of each of the outflow ports, and connecting the culture bottle to the outflow port by installing a culture bottle (90, 92), the collection bag having a connecting flow path (30) connecting adjacent storage chambers, the connecting flow path having a tubular portion (31) formed integrally with the storage chambers using the two flexible sheets and formed into a tubular shape by cutting the surrounding flexible sheets between the adjacent storage chambers.

[0053] The above-mentioned collection kit is a structure that uses two flexible sheets to form a collection bag in one piece, so it can be manufactured at low cost. In addition, the collection kit closes the tubular portion of the collection bag by a clamp, etc., so that adjacent storage chambers can be reliably separated, and the deviation of the introduced amount of the platelet preparation can be suppressed.

[0054] In the above-mentioned collection kit, the plurality of storage chambers include: a first storage chamber (26) connected to the inflow tube at the top; and a second storage chamber (28) arranged in the first storage chamber via the connecting flow path, and an exhaust valve (20) for exhausting the air inside the first storage chamber and the second storage chamber can be connected to the top of the second storage chamber. The collection kit exhausts the air inside the first storage chamber and the second storage chamber through the exhaust valve, thereby enabling smooth introduction of the platelet preparation.

[0055] In the above-mentioned collection kit, the upstream end (30a) of the connecting flow path can be connected to the upper part of the first storage chamber, and the downstream end (30g) of the connecting flow path can be connected to the lower part of the second storage chamber. The collection kit can smoothly introduce the platelet preparation into the first storage chamber and the second storage chamber without residual air.

[0056] In the above-mentioned collection kit, the plurality of outflow ports may include: a first outflow port (34) connected to the lower portion of the first storage chamber; and a second outflow port (36) connected to the lower portion of the second storage chamber. The collection kit can introduce the platelet preparation in the first storage chamber and the platelet preparation in the second storage chamber into respective culture bottles from different outflow ports, thereby eliminating the need to transfer the platelet preparation between different storage chambers, and providing excellent operability.

[0057] The collection kit may further include a frame member (50) that holds the peripheral edge of the flexible sheet and keeps the flexible sheet in a flat shape. The collection kit can keep the collection bag in a flat shape, so there is no need to maintain the collection bag in a flat shape during use, and thus the handling is excellent.

[0058] In the above-mentioned collection kit, the frame member may include: a frame body (54) that holds the peripheral edge of the flexible sheet; and a contact plate (52) that contacts the receiving chamber from the thickness direction of the flexible sheet to prevent the receiving chamber from expanding in the thickness direction. The collection kit can suppress the variation in the amount of platelet preparation introduced by suppressing the expansion of the receiving chamber in the thickness direction.

[0059] Another aspect is a collection method, which uses the above-mentioned collection kit and includes: a step of connecting the medical bag to the inflow tube; a step of introducing the collection object into the storage chamber of the collection bag through the inflow tube; a step of separating the medical bag from the inflow tube; a step of closing the tubular portion to prevent the movement of the collection object between the plurality of storage chambers; and a step of connecting the culture bottle to the adapter and injecting the collection object into the culture bottle. According to this collection method, the work efficiency of the culture inspection of the platelet preparation can be improved.

[0060] In the above-mentioned collecting method, in the step of closing the tubular portion to prevent movement of the collected object between the plurality of storage chambers, the tubular portion may be closed by a clamp or ultrasonic sealing or high-frequency sealing.

[0061] It should be noted that the present invention is not limited to the above-mentioned disclosure, and various configurations can be adopted without departing from the gist of the present invention.
