Abstract
Description
Field of the Invention
[0001] The present invention relates to the use of topical compositions for enhancing the nighttime repair-related functions of the skin. Background Art
[0002] Exposure of human skin to UV radiation can produce a range of adverse effects on the skin, both acutely and chronically. Additionally, current lifestyles, especially among younger generations, that often involve delayed and insufficient sleep, overexposure to blue light, and inherent stress / anxiety are causes for concern. These factors impact multiple skin functions, ultimately affecting skin appearance, including pigmentation, aging, and radiance. Skin processes are naturally regulated to provide more protection during the day, while at night, the skin focuses on repair. Because skin function changes from day to night, it would be advantageous if nighttime cosmetics could help skin repair, while daytime products could help skin maximize protection.
[0003] The present inventors found that vitamin B3 compounds or derivatives thereof provide this benefit when looking for solutions to the problem of providing enhanced levels of nighttime repair-related functions to the skin. They also found a synergistic interaction to provide a combination of active substances for this benefit. They found that the combination of vitamin B3 compounds or derivatives thereof and alkyl-substituted dihydroxybenzenes (wherein the alkyl group contains 2 to 10 carbon atoms) provides this synergistic effect. They further found that when the composition comprising the vitamin B3 compound is applied to the skin in the evening (evening) / night (night), for example, from about 5pm to 12am at the end of the day, the nighttime repair function of the skin is enhanced. In addition, they found that when the composition comprising the vitamin B3 compound or derivatives thereof and the alkyl (C2-C10)-substituted dihydroxybenzenes is applied to the skin in the evening / night, for example, from about 5pm to 12am at the end of the day, the nighttime repair function of the skin is enhanced.
[0004] It is therefore an object of the present invention to deliver enhanced nighttime repair related functions to the skin.
[0005] Another object of the present invention is to deliver this benefit through multiple actives that interact synergistically, thereby ensuring that low concentrations of actives are used, which provides the consumer with a low cost product and a product with minimal side effects. [0021]
[0022] Where R1 is C2-C 10 alkyl; and each of R2 to R6 is an OH group or H; and
[0023] wherein at least two of R2 to R6 are OH.
[0024] The more preferred alkyl-substituted dihydroxybenzenes for use in the present invention have the structure
[0025]
[0026] Where R1 is C2-C 10 Alkyl; R2 and R3 are OH.
[0027] Preferably R1 is an alkyl group containing 2 to 6 carbon atoms, most preferably containing 2, 4 or 6 carbon atoms. The two hydroxyl groups are usually located at the 1st and 3rd positions. Such compounds are generally referred to as substituted resorcinols. Therefore, the most preferred compounds are ethylresorcinol, butylresorcinol or hexylresorcinol. Most preferably, 4-substituted resorcinols. Therefore, the resorcinol that can be used is 4-ethylresorcinol (4-ER), 4-butylresorcinol (4-BR) or 4-hexylresorcinol (4-HR). The most preferred compound for the present invention is 4-hexylresorcinol. This compound has the following structure:
[0028]
[0029] This compound is also known by its synonyms 4-hexylbenzene-1,3-diol and 4-hexyl-1,3-benzenediol.
[0030] Hexylresorcinol is known to exhibit antiseptic and local anesthetic properties. It has been used for topical application for minor skin infections, oral solutions for pain relief, and as a first aid disinfectant. It is also used as a skin lightener in skin care. The compound is also known to have antioxidant properties and has therefore been used in skin care compositions to provide an even-toned complexion or in anti-aging products.
[0031] The alkyl-substituted dihydroxybenzene compound is preferably contained in an amount of 0.01 to 1 wt %, more preferably 0.05 to 0.5 wt %, and still more preferably 0.1 to 0.4 wt % of the composition.
[0032] Without wishing to be bound by theory, the inventors believe that the technical effect, namely the enhancement of the nighttime repair function of the skin, occurs due to the following reasons.
[0033] Vitamin B3 compounds or their derivatives, such as nicotinamide, are the main precursors of nicotinamide adenine dinucleotide (NAD+), which is an essential coenzyme for DNA repair, glycolysis, autophagy, oxidative phosphorylation, cellular energy, etc. Nicotinamide also increases the ratio of NAD+ / NADH in skin cells, and also activates SIRT1 and NAD+-dependent deacetylases, which play an important role in the autophagic flow and the circadian transcription of several core clock genes. NADH effectively acts as an energy storage component and provides high-energy electrons to the mitochondrial electron transport chain when needed. Nicotinamide is also known to induce the antioxidant system in skin cells by activating Nrf2 signaling, which plays an important role in regulating oxidative stress in skin cells.
[0034] It is known that the claimed alkyl-substituted dihydroxybenzene compounds, such as hexylresorcinol (HR), can be used as skin-brightening agents. It can also act as an antioxidant that helps to achieve a more even skin tone. As an antioxidant protectant, hexylresorcinol can neutralize free radicals generated in skin cells by ultraviolet light, blue light, pollution, etc. In addition, HR can activate a variety of other skin functions, including Nrf2 activation, antioxidant response element (ARE)-reporter gene activity, and expression of ARE-dependent genes NAD(P)H dehydrogenase (NQO1) and heme oxygenase-1 (HO-1).
[0035] Based on these insights, the inventors hypothesize that the effects of the combination of claimed ingredients can be attributed to the activation of NRF2 signaling, increasing the expression of NADPH dehydrogenase and other antioxidant enzymes by hexylresorcinol, and enhancing the NAD+ / NADH ratio and activating SIRT1 and NAD+-dependent deacetylases by nicotinamide, leading to the observed benefits.
