A method for purifying a recombinant human type III collagen

Abstract

The application discloses a method for purifying a recombinant human type III collagen, and the amino acid sequence of the collagen is shown as SEQ ID NO:1. The method comprises the following steps: (1) obtaining a fermentation supernatant containing the collagen; (2) loading the supernatant into a heparin affinity chromatography column and eluting with an eluent containing NaCl; (3) diluting the primary eluted protein and loading into a cation chromatography column and eluting with an eluent containing NaCl; and (4) obtaining a collagen stock solution through ultrafiltration and dialysis. The natural affinity and charge characteristics of the collagen and heparin are utilized to realize efficient purification, the purity of the obtained stock solution is greater than or equal to 99%, and the endotoxin is less than or equal to 5 EU / mg, which meets the type III medical instrument standard. The method steps are simple, the cost is low, and the method is easy to scale up, and is suitable for industrial production.
