Abstract
Description
[0001] ANTIVIRAL COMPOSITION COMPRISING CHITOSAN AND XANTHOHUMO-
[0002] LONE
[0003] Technical field
[0004] The present invention relates to the technical field of antiviral compositions. The invention relates in particular to a composition comprising at least chitosan and xanthohumolone, its preparation process and its uses.
[0005] State of the art
[0006] December 2019 marked the beginning of the COVID-19 pandemic, which has since spread worldwide with more than 350 million cases and over 5.5 million deaths.
[0007] With the virus spreading rapidly, many scientific teams around the world have been working to find solutions to limit the spread of the virus.
[0008] According to the World Health Organization (WHO), the virus can spread when small liquid particles are expelled from the mouth or nose when an infected person coughs, sneezes, talks, or breathes deeply. These particles vary in size, from large "respiratory droplets" to smaller "aerosols."
[0009] The Covid-19 virus, like many airborne viruses, is transmitted primarily through respiratory droplets. These droplets, laden with viral particles, could infect a susceptible individual either through direct contact with a mucous membrane (direct transmission) or through contact with an infected surface via the nasal, oral, or conjunctival mucous membranes (indirect transmission). They can be projected several meters away but do not persist in the air.
[0010] Once inhaled, to infect its host, the COVID-19 virus attaches to a protein present on the surface of cells, particularly lung cells: the ACE2 receptor. This attachment then allows it to penetrate the cell and infect its host.
[0011] One of the strategies being considered aims to prevent the virus from entering the cell by acting on the mechanisms necessary for the virus to attach to its receptor.
[0012] In addition to isolation, ventilation and barrier gestures, the most widespread solution is wearing a mask during close contact to avoid the transmission of droplets and aerosols.
[0013] There are three main types of masks on the market to combat the transmission of airborne pathogens.
[0014] FFP (Filtering Face Piece) type masks are classified according to their capacity to filter aerosols and meet the NF EN 149 standard.
[0015] -FFP1 (80% filtration of aerosols)
[0016] -FFP2 (94% filtration of aerosols) -FFP3 (99% filtration of aerosols).
[0017] This type of mask protects the wearer from inhaling airborne particles and especially larger droplets that could contain infectious agents.
[0018] There are also surgical masks that comply with the NF EN 14683 standard. By preventing the projection of droplets emitted by the wearer of the mask, this type of mask limits contamination of the external environment and other people. There are also several types of surgical masks classified according to their robustness.
[0019] And finally, there are so-called "general public" masks, which are textile masks with guaranteed filtration, and are mostly washable and reusable. They are easily recognizable. They are reserved for use outside the healthcare system.
[0020] “General public” masks have filtration properties greater than 90% of 3 micron particles for category 1 or greater than 70% of these same particles for category 2.
[0021] All masks on the market are effective in limiting the spread of airborne viruses, including SARS-CoV-2, which causes COVID-19, but they also all have the same drawbacks, namely:
[0022] -very limited comfort when breathing, especially during prolonged use,
[0023] -a significant amount of waste due to the short lifespan,
[0024] -masks do not have any specific action to act against viruses, it is only a physical barrier.
[0025] With the emergence of pandemics such as COVID-19, there is a great need to find new solutions to limit the spread of viruses, particularly airborne viruses such as COVID-19.
[0026] One of the solutions proposed by the prior art is the combination of an antiviral vaccine with a means of preventing the spread of viruses.
[0027] However, there are currently no effective solutions to limit the spread of airborne viruses while having a protective antiviral action.
[0028] There is therefore a need for a means of protection aimed at combating the spread and infection of viruses, particularly airborne viruses such as covid-19.
[0029] Summary of the invention
[0030] To meet this need, the invention proposes a new composition comprising a mixture comprising at least chitosan and xanthohumolone.
[0031] Chitosan is a natural biopolymer derived from chitin, particularly from crustaceans.
[0032] Xanthohumolone is a flavonoid derived from hops, particularly from the Humulus Lupulus species. Chitosan and xanthohumolone are molecules that have long been used in many fields such as food, cosmetics, and pharmaceuticals. Their safety profile and innocuousness are already known from the prior art.
[0033] The inventors surprisingly discovered that the combination of chitosan and xanthohumolone made it possible to obtain a composition with a synergistic antiviral effect, particularly against airborne viruses such as covid 19.
[0034] Such a composition is particularly useful in the context of the invention and overcomes the drawbacks of the prior art, it is biocompatible and has significant antiviral activity. Furthermore, the residues resulting from the degradation of the composition according to the invention are not toxic or genotoxic and do not induce inflammation, particularly of the skin or mucous membranes.
[0035] The composition according to the invention has a xantohumolone / chitosan ratio of between 0.2 and 0.7, preferably between 0.4 and 0.6, even more preferably 0.5.
[0036] In one embodiment, the invention relates to a composition comprising a mixture of chitosan and xanthohumolone, as well as a biocompatible solvent such as acetic acid.
[0037] The composition according to the invention is suitable for use in human or animal health, preferably for use as an antiviral or antibacterial composition.
[0038] According to another aspect, the composition according to the invention is also suitable for the treatment of medical devices.
[0039] Advantageously, the composition according to the invention makes it possible to disinfect and protect medical devices to prevent the spread of viruses and / or bacteria.
[0040] Also, the invention also relates to a kit comprising a vaccine and a composition comprising the combination of chitosan and xanthohumolone.
[0041] According to a final aspect, the invention also relates to a process for preparing the composition according to the invention comprising in particular a step of dissolving the chitosan, mixing, heating and adding a xanthohumolone solution.
[0042] Other characteristics and advantages will emerge from the detailed description of the invention and from the examples which are purely illustrative and in no way limitative of the scope of the invention.
