AI Technical Summary
Current anti-dandruff treatments, while effective, often rely on non-renewable and energy-intensive materials, and there is a need for sustainable alternatives that can effectively reduce yeast fungi colonization on the scalp, particularly Malassezia, without causing skin irritation.
The use of specific medium-chain glycerol ethers as sole anti-dandruff active ingredients in cosmetic compositions, which are biodegradable, easily incorporated, and provide long-term stability, reducing Malassezia colonization and soothing sensitive scalps.
Significant reduction in Malassezia yeast amounts leads to effective dandruff treatment, with additional benefits of maintaining scalp barrier function and calming effects on sensitive scalps, making them a suitable sustainable alternative.

Abstract
Description
[0001] Use of branched medium-chain glycerol ethers as anti-dandruff agents
[0002] The present invention relates to the cosmetic use of specific glycerol ethers as anti-dandruff active ingredients and as skin-soothing active ingredients, in particular in the cosmetic treatment of dandruff conditions associated with the colonization and proliferation of yeast fungi of the genus Malassezia on hairy skin surfaces.
[0003] Even mild flaking of the scalp or other hairy areas of the body is perceived as a sign of poor care and hygiene. Dandruff is usually accompanied by excruciating itching, which can trigger scratching and lead to injury to the affected skin areas. Skin injuries, in turn, often form the basis for infections and pathogens.
[0004] Dandruff formation can be caused by various factors, such as hereditary predisposition, a tendency toward excessive sebum production (seborrhea), hormonal fluctuations, stress, climatic conditions, or improper hair care. However, the primary contributor to dandruff formation is the colonization of the scalp with yeasts of the genus Malassezia. Therefore, dandruff control has always aimed to reduce and / or eliminate the Malassezia bacterial colonization of the scalp.
[0005] For this purpose, numerous cosmetically acceptable and effective anti-dandruff agents are known from the state of the art, such as piroctone olamine, climbazole, zinc pyrithione or other zinc salts, sulfur and / or selenium sulfide.
[0006] The effect of these anti-dandruff agents is mainly based on their fungicidal properties.
[0007] However, in light of the increasingly important and more sustainable use of raw materials and energy, there is a growing effort to use only those cosmetic raw materials in cosmetic products that can be obtained from renewable resources with the least possible use of energy. However, a reduction in quantity or even a complete replacement of undesirable or problematic substances can only be achieved if alternatives are available that also possess the desired properties for the intended purpose. This also applies to anti-dandruff active ingredients.
[0008] Various plant extracts are known as natural anti-dandruff agents, such as arnica, birch, burdock root, beard lichen, poplar, nettle, myrtle, or hazel root extracts. However, in terms of their immediate and / or long-lasting effectiveness, these extracts cannot completely replace the classic anti-dandruff agents mentioned above.
[0009] There is therefore still a need for compounds with good anti-dandruff efficacy that are available from natural sources. Specific bio-based glycerol ethers have been found to be ideally suited for this purpose.
[0010] WO 2021 / 237207A1 describes the production of bio-based glycerol ethers that can be produced from sustainable, renewable, plant-based raw materials and that – like other branched glycerol ethers, for example ethylhexylglycerol – are excellently suited as preservatives in cosmetic or pharmaceutical compositions or as odor control agents in deodorants.
[0011] Furthermore, according to EP 3944852, some glycerol ethers - such as ethylhexylglycerol - are suitable as boosters to enhance the anti-dandruff efficacy of piroctone olamine in cosmetic compositions.
[0012] However, it is not known to use medium-chain glycerol ethers as an anti-dandruff active ingredient, especially as the sole anti-dandruff active ingredient, in cosmetic compositions for the treatment of dandruff conditions.
[0013] The object of the present invention was to find active ingredients that are easily accessible from natural sources, biodegradable and well tolerated by the skin, which can be easily incorporated into a variety of cosmetic products and are stable over the long term, and which reduce, prevent and / or eliminate excessive colonization of hairy body surfaces, in particular the scalp, by fungi of the genus Malassezia.
[0014] It has surprisingly been found that the cosmetic use of certain medium-chain glycerol ethers enables effective treatment of dandruff conditions, particularly those associated with colonization and / or proliferation of yeasts of the genus Malassezia.
[0015] The inventive use of at least one specific medium-chain glycerol ether as the sole anti-dandruff active ingredient in cosmetic compositions resulted in a significant reduction in the amount of yeasts of the genus Malazessia and consequently in a reduction in dandruff. Furthermore, it was found that the inventive use of at least one specific medium-chain glycerol ether provides additional benefits in maintaining the body's own barrier, particularly on sensitive scalps (where a compromised barrier and increased sebum production are present). The inventive use of at least one specific medium-chain glycerol ether has a balancing and soothing effect on sensitive scalps.
[0016] The present invention is therefore characterized by the following statements:

[0078] According to a further preferred embodiment, the cosmetic formulation containing at least one compound of formula (I) remains on the hair until the next hair cleansing.
[0079] Examples: a) The following cosmetic formulations were prepared using at least one compound according to formula (I) as an antidandruff active ingredient. The amounts given in the tables refer to wt.%.
[0080] 1) Shampoo
[0081] 2) Hair conditioner
[0082] 3) Hair tonic b) The anti-dandruff and skin-soothing efficacy of compounds according to formula (I) was investigated:
[0083] (i) Efficacy against dandruff was investigated using a suspension test. Various formulations were incubated for a certain period of time (< 60 min) with the yeast Malassezia furfur, which had a cell count of 105 - 106 CFU / ml, at 37°C in a buffer solution. After incubation, the yeast cell count was determined using cultural methods (growth on Sabouraud agar). The difference between this value and the respective baseline value is a measure of the microbicidal efficacy of the respective antidandruff compound.
[0084] One cause of dandruff formation is, among other things, the excessive growth of various microorganisms on the scalp. Yeasts of the genus Malassezia play a particularly important role. The growth-inhibiting effect of various anti-dandruff compounds in this experiment thus indicates their anti-dandruff effectiveness (Fig. 1).
[0085] (ii) A healthy scalp forms a protective barrier against external influences, prevents dehydration, and serves as the basis for optimal hair growth. The formation of dandruff is largely promoted by fungal colonization, whose metabolic products negatively affect the scalp's barrier and lead to inflammatory reactions.
[0086] In vitro experiments on artificial skin models have shown that common anti-dandruff agents such as octopirox (piroctone olamine) negatively alter barrier function. The gene expression of all barrier markers examined (transglutaminsel, filaggrin, decorin, involucrin, and loricrin) was reduced by treatment. Treatment with methylheptylglycerol, in contrast, did not lead to a significant decrease in gene expression, even at higher concentrations; some genes could even be stimulated (Fig. 2). Furthermore, it was found that methylheptylglycerol (just like octopirox) has no negative effect on the release of inflammatory markers. The release of interleukin 8 is within the range of the untreated control (Fig. 3).
[0087] In summary, methylheptylglycerin can be considered a milder alternative to the anti-dandruff ingredient octopirox. In addition to killing the Malassezia fungus, it provides additional benefits for maintaining the body's natural barrier. MHG is particularly recommended for consumers with sensitive scalps (where a compromised barrier and increased sebum production are present), as it can contribute to scalp normalization.
[0088] The use of methylheptylglycerin not only works against dandruff-causing fungi, but also has a balancing and soothing effect on the scalp.
