Description

Technical Field The present invention belongs to the field of biomedicine, and particularly relates to a product for diagnosing lung cancer. Background Lung cancer is a common malignant tumor. At present, clinical methods for diagnosing lung cancer include imageological examination, sputum cytopathology examination, bronchoendoscopy examination, molecular pathology examination, etc. However, the methods are not easy to operate and prone to false positive. In order to improve the diagnosis and treatment of lung cancer, a number of tumor markers have been applied in the field of lung cancer in recent years. (1) Carcinoembryonic antigen (CEA): CEA is a proteoglycan complex, which mainly exists in digestive tract epithelial tissue, pancreas, liver, biliary tract, lung, mammary gland, etc., and is the most widely used tumor marker at present, with abnormal expression in various cancer tissues. The positive rate of CEA in patients with lung cancer is about 50%, and is higher in patients with adenocarcinoma than in patients with squamous cell carcinoma; and the serum level of CEA is increased with the increase of clinical stage. (2) Cytokeratin 19 fragment (CYFRA21-1): Cytokeratin belongs to the intermediate filament protein family and is widely distributed on the surface of normal tissues, such as lamellar or squamous epithelium. In malignant epithelial cells, activated proteases accelerate cell degradation, resulting in the release of a large amount of cytokeratin fragments into blood. Cytokeratin 19 (CKI9) is expressed in both adenocarcinoma and squamous cell carcinoma in lung cancer, especially in squamous cell carcinoma. Determination of cytokeratin has an important clinical value for the classification and staging of lung cancer, especially for the diagnosis of squamous cell carcinoma in non-small cell lung cancer. (3) Neuron-specific enolase (NSE): NSE is an enolase involved in glycolytic pathway, which exists in nerve tissue and neuroendocrine tissue. It has been found to exist in tumors associated with the origin of neuroendocrine tissue, especially in small cell lung cancer where excessive expression of NSE occurs, resulting in a significant increase of NSE in serum. NSE has a high sensitivity to small cell lung cancer, so NSE is superior to other tumor markers in the diagnosis and treatment of small cell lung cancer. (4) Squanlous cell carcinoma antigen (SCC-Ag): Squamous cell carcinoma associated antigen is a glycoprotein that exists in cytoplasm of squamous cell carcinoma of uterus, cervix, lung, head, neck, etc., especially in cells of non-keratinous carcinoma. Compared with CEA, SCC-Ag has a lower sensitivity but a higher specificity to lung squamous cell carcinoma, but the sensitivity and specificity of SCC-Ag are lower than those of CYFRA21-1. (5) Cancer antigen 125 (CA125): CA125 is a glycoprotein tumor associated antigen that exists in epithelial cells of ovarian tumor. CA125 is initially found to have a significant increase in patients with ovarian cancer, and later found to have a relatively high positive rate in patients with lung cancer. Tumor markers can reflect the existence and change of tumors and provide a guidance for the diagnosis of tumors, and have a simple detection method and a good repeatability, which have become a research hotspot in cancer diagnosis in recent years. Screening out a more reliable biomarker closely related to the diagnosis of lung cancer will provide a new method for the diagnosis of lung cancer.
