Abstract
Description
[Background technology]
[0001] The present disclosure relates to compounds that act as aryl hydrocarbon receptor (AHR) activity modulators, pharmaceutical formulations thereof, and methods of using the compounds to treat diseases and disorders, such as inflammatory and autoimmune diseases and disorders.
[0002] The aryl hydrocarbon receptor (AHR) is a ligand-dependent transcription factor that regulates gene expression in various cells, such as epithelial cells and immune cells. Activation of AHR can lead to changes in inflammatory cytokine profiles and antioxidant responses in the cellular environment, facilitating the healing process of diseased tissues. AHR signaling may play a key role in maintaining skin and ocular homeostasis by regulating the skin immune network, keratinocyte differentiation, skin barrier function and pigmentation, retinal epithelial cells, rod photoreceptors, choroidal endothelial cells, choroidal neovascularization, and responses to oxidative stress. Cells found in the skin and eye that express AHR include keratinocytes, sebocytes, fibroblasts, melanocytes, endothelial cells, Langerhans cells, choroidal endothelial cells, retinal pigment epithelial cells, rod photoreceptors, and immune cells present in the skin and eye. Thus, regulation of the AHR signaling pathway is involved in the pathology of various diseases and disorders of the skin and eye.
[0003] AHR signaling is also known to regulate the composition and function of different cell types in the gastrointestinal tract, and thus has a prominent role in maintaining the balance between health and disease. AHR is involved in several physiological processes, including homeostasis and immune regulation in epithelial barriers, such as those formed by intestinal epithelial cells (IECs). A powerful immune response in the body occurs in the intestine, and therefore there is great interest in elucidating the molecular mechanisms underlying the role of AHR in cells of the intestinal mucosa (IECs and various immune cells, including B cells, T cell receptor γδ T cells (TCRγδ), T helper 17 cells (Th17), regulatory T cells (Treg), type 1 regulatory T cells (Tr1), innate lymphoid cells (ILCs), macrophages (MQs), intraepithelial lymphocytes (IELs), dendritic cells (DCs), and neutrophils). Thus, aberrant AHR activity is involved in several intestinal pathologies, such as intestinal inflammation, infection, and cancer.
[0004] AHR signaling is also known to play a major role in bone remodeling by altering the interaction between bone-forming osteoblasts and bone-resorbing osteoclasts. The overall effect of AHR activation in osteoblasts is the suppression of cell differentiation, and AHR agonism has a dose-dependent effect on osteoblasts, with overactivation and underactivation inhibiting and promoting bone formation, respectively. Although the effects of AHR regulation in osteoclasts are less well understood, the AHR pathway has been implicated in both stimulating and impairing osteoclast differentiation. Thus, the AHR pathway is an attractive target for the treatment of various human diseases in which osteoblasts and osteoclasts are involved in the pathogenesis, including bone-destructive diseases such as osteoporosis and cancer. [0074] Sterile injectable forms of the compositions disclosed herein may be aqueous or oleaginous suspensions. These suspensions may be formulated according to techniques known in the art using suitable dispersing or wetting agents and suspending agents. Sterile injectable preparations may also be sterile injectable solutions or suspensions in non-toxic parenterally acceptable diluents or solvents, for example as a solution in 1,3-butanediol. Among the acceptable vehicles and solvents that may be used are water, Ringer's solution, and isotonic sodium chloride solution. In addition, sterile fixed oils are conventionally used as a solvent or suspending medium. For this purpose, any bland fixed oil may be used, including synthetic mono- or di-glycerides.
