Description

[Technical field]

[0001] The present invention relates to a dextranase-containing oral composition which provides a high feeling of efficacy in the oral cavity and is effective for preventing or inhibiting dental caries. [Background technology]

[0002] Dextranase has the effect of chemically removing plaque by breaking down the water-insoluble glucan that forms plaque, and also has the effect of inhibiting plaque formation, so it has been incorporated into oral compositions as an ingredient for preventing or inhibiting dental caries (Patent Documents 1 to 4; JP 2011-132139 A, JP 2002-187829 A, JP 2001-163747 A, JP 2015-20970 A).

[0003] However, the above-mentioned action and effect of dextranase is difficult to recognize by visual inspection, and it is difficult to feel the effect of use, so that the user's motivation to use may not be maintained. In order to maximize the action and effect of dextranase, it is important to maintain the user's motivation to use and encourage continued use. For this purpose, it is considered effective to make the user feel that it is working in the oral cavity every time it is used, for example, by feeling a refreshing plaque removal, thereby increasing the user's motivation. [0004] [Patent Document 1] JP 2011-132139 A [Patent Document 2] JP 2002-187829 A [Patent Document 3] JP 2001-163747 A [Patent Document 4] JP 2015-20970 A Summary of the Invention [Problem to be solved by the invention]

[0005] The present invention has been made in consideration of the above circumstances, and aims to provide an oral composition containing dextranase that provides a high sense of efficacy in the oral cavity and is effective for preventing or inhibiting dental caries. [Means for solving the problem]

[0006] As a result of intensive research by the present inventors to achieve the above object, they found that when a water-soluble tin salt and a cationized cellulose and / or an α-olefin sulfonate are combined in an oral composition containing dextranase, a unique and long-lasting remarkable refreshing feeling is imparted to the oral cavity, and thus not only the actual effect of removing plaque and inhibiting plaque formation can be imparted to the oral cavity with each use, but also a high level of effectiveness due to the refreshing feeling can be imparted and a good taste can be ensured. That is, in the present invention, it was found that an oral composition containing (A) dextranase, (B) a water-soluble tin salt, and (C) one or more selected from cationic cellulose (C1) and α-olefin sulfonate (C2) imparts a high level of effectiveness in the oral cavity even with a single use, has a good taste when used, and is effective for preventing or inhibiting dental caries, and thus the present invention was completed.

[0007] More specifically, in the present invention, by using the component (A) in combination with the component (B) in the oral cavity composition, a unique and long-lasting remarkable fresh feeling in the oral cavity (a distinctive sensation that the oral cavity is cleaned and continues to remain even after time has passed since use, which is sometimes abbreviated as "refreshing feeling in the oral cavity") can be imparted, and a high sense of effectiveness can be imparted. The fresh feeling in the oral cavity is a unique sensation that is specifically obtained by using the component (A) in combination with the component (B), and is different from the refreshing feeling obtained by, for example, a general oral flavor such as menthol or ethanol. Furthermore, when the component (A) is used in combination with the component (B), the bad aftertaste derived from the component (B) stands out as an unpleasant taste, and even if a general oral flavor is added, the aftertaste becomes unpleasant. However, by further adding the component (C), the unpleasant taste is suppressed without causing an unpleasant aftertaste as described above, and a good taste is ensured, and a high sense of effectiveness can be imparted by the refreshing feeling in the oral cavity. As shown in the comparative examples below, when the oral composition contains the (A) component but not the (B) and (C) components, the effect (refreshing feeling in the oral cavity, the refreshing feeling in the oral cavity immediately after use and 10 minutes after use) is low (Comparative Example 1), and when the oral composition contains the (A) and (B) components but not the (C) component, the lack of unpleasant taste after use is poor (Comparative Example 2). In contrast, the oral composition of the present invention containing the (A), (B) and (C) components shown in the examples had a high effect (refreshing feeling in the oral cavity) and a good lack of unpleasant taste after use. Patent Documents 1 and 2 show an improvement in the retention of dextranase in the oral cavity by a cationic polymer, Patent Document 3 shows an improvement in the inhibitory effect of dextranase on plaque reattachment by a cationic polymer, and Patent Document 4 shows oral biofilm removal by a combination of α-olefin sulfonic acid, dextranase, and sugar alcohol. In contrast, the present invention combines components (A), (B), and (C) to impart a unique, long-lasting, and remarkable feeling of freshness in the oral cavity, improving the perceived effect of component (A) and also improving the unpleasant taste.

[0008] Accordingly, the present invention provides the following oral compositions. [1] (A) dextranase, (B) Water-soluble tin salts and (C) one or more selected from cationic cellulose (C1) and α-olefin sulfonate (C2) An oral composition comprising: [2] (B) The oral composition according to [1], wherein the water-soluble tin salt is one or more selected from tin chloride and tin fluoride. [0009] According to the present invention, it is possible to provide an oral composition that not only provides the actual effects of plaque removal and plaque formation inhibition by component (A), but also gives a high feeling of effectiveness that makes the user feel that component (A) is working, and also has a good taste. The oral composition of the present invention effectively exhibits the effects of plaque removal and plaque formation inhibition by component (A), and is effective for preventing or inhibiting dental caries. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS

[0010] The present invention will now be described in further detail. The oral composition of the present invention contains (A) dextranase, (B) a water-soluble tin salt, and (C) one or more members selected from cationic cellulose (C1) and α-olefin sulfonate (C2).

[0011] (A) Dextranase has the effect of chemically removing plaque by breaking down the water-insoluble glucan that forms plaque, and exerts the effects of removing plaque and inhibiting plaque formation. Dextranase having a concentration of 10,000 to 14,000 units / g is usually used, and when a dextranase having a concentration of 13,000 units / g is used, the amount of dextranase to be blended is preferably 0.0077 to 1.54% (mass%, the same applies below) of the entire composition, and more preferably 0.015 to 0.38%. The greater the amount of dextranase blended, the greater the effect of removing plaque and the effect of inhibiting plaque formation, but if the amount is too high, it may affect palatability, and a blending amount of 1.54% or less ensures a satisfactory taste. Furthermore, the content of the dextranase bulk substance in the composition is preferably 1 to 200 units / g, more preferably 2 to 20 units / g. Here, one unit of dextranase is the amount of dextranase that produces free reducing sugars equivalent to 1 μmol of glucose per minute when reacted with dextran as a substrate.

[0012] (B) Water-soluble tin salt, when used in combination with component (A), provides a unique, long-lasting and remarkable feeling of freshness in the oral cavity, and this freshness has the effect of imparting a sense of efficacy. (B) Examples of water-soluble tin salts include tin chloride, tin fluoride, tin acetate, tin gluconate, tin oxalate, tin sulfate, tin lactate, tin tartrate, tin citrate, tin malate, tin phosphate, tin pyrophosphate, and tin metaphosphate. Of these, tin chloride and tin fluoride are preferred. As the tin chloride, tin(II) chloride (stannous chloride) can be used, and it may be a hydrate such as tin(II) chloride dihydrate. As the tin fluoride, tin(II) fluoride (stannous fluoride) or tin(IV) fluoride (stannic fluoride) can be used. In particular, tin(II) fluoride (stannous fluoride) is preferred because it can also impart a caries prevention effect. These may be used alone or in combination of two or more.

[0013] The amount of the water-soluble tin salt (B) is preferably 0.02 to 1% of the total composition in terms of tin ions, more preferably 0.1 to 0.65%. If the tin ion content is 0.02% or more, a sufficiently refreshing feeling in the oral cavity is obtained, and if it is 1% or less, the unpleasant taste improving effect of the (C) component is fully exerted and the appearance stability is ensured.

[0014] The component (C) is at least one selected from the group consisting of (C) cationic cellulose (C1) and α-olefin sulfonate (C2), and the cationic cellulose (C1) and α-olefin sulfonate (C2) can also be used. The component (C) has the effect of improving the unpleasant taste caused by the combined use of the components (A) and (B).

[0015] Examples of the cationic cellulose (C1) include cellulose derivatives having dimethyldiallylammonium, 2-hydroxy-3-(trimethylammonio)propyl, etc., added as cationic groups. Examples include hydroxyethylcellulose dimethyldiallylammonium chloride, O-[2-hydroxy-3-(trimethylammonio)propyl]hydroxyethylcellulose chloride, etc., and in particular, hydroxyethylcellulose dimethyldiallylammonium chloride is preferred from the viewpoint of improving unpleasant taste. The nitrogen content is preferably 0.1 to 3%, more preferably 0.5 to 2.5%. These may be used alone or in combination of two or more.

[0016] Specifically, hydroxyethyl cellulose dimethyl diallyl ammonium chloride is obtained by graft polymerization of dimethyl diallyl ammonium salt to hydroxyethyl cellulose, and a 2% aqueous solution having a viscosity of 10 to 3,000 mPa·s (BH type Brookfield viscometer, rotor No. 2, 20 rpm, 21°C) can be used. For example, AkzoNobel's Cellcoat L-200 (2% aqueous solution viscosity: 35 to 350 mPa·s, BH type Brookfield viscometer, rotor No. 2, 20 rpm, 21°C, measurement time 1 minute) and Cellcoat H-100 (2% aqueous solution viscosity: 500 to 2,750 mPa·s, BH type Brookfield viscometer, rotor No. 2, 20 rpm, 21°C, measurement time 1 minute) can be mentioned. In addition, Nouryon Japan's CELQUAT L-200 can also be used. In addition, the viscosity of O-[2-hydroxy-3-(trimethylammonio)propyl]hydroxyethyl cellulose chloride can be measured in a 1% or 2% aqueous solution, and one with a viscosity of 500 to 2,600 mPa s for a 1% aqueous solution (BH Brookfield viscometer, rotor No. 2, 20 rpm, 25°C) or one with a viscosity of 5 to 800 mPa s for a 2% aqueous solution (BL viscometer, rotor No. 2, 30 rpm, 25°C) can be used. Examples include Leoguard KGP (viscosity of 2% aqueous solution: 5 to 50 mPa·s), Leoguard G (viscosity of 2% aqueous solution: 100 to 600 mPa·s), Leoguard GP (viscosity of 2% aqueous solution: 100 to 600 mPa·s), Leoguard LP (viscosity of 2% aqueous solution: 300 to 800 mPa·s), all manufactured by Lion Corporation, as well as Leoguard MGP (viscosity of 1% aqueous solution: 500 to 1,200 mPa·s) and Leoguard MLP (viscosity of 1% aqueous solution: 1,000 to 2,600 mPa·s).

[0017] When cationic cellulose (C1) is blended as component (C), its blending amount is preferably 0.02 to 0.2% of the total composition, more preferably 0.05 to 0.1%. When the blending amount is 0.02% or more, the unpleasant taste improving effect is sufficiently obtained and the unpleasant taste is suppressed. When the blending amount is 0.2% or less, the refreshing feeling in the oral cavity is sufficiently maintained, and the unpleasant taste (bitterness) of the composition itself is prevented from appearing, and the unpleasant taste improving effect is sufficiently maintained.

[0018] As the α-olefin sulfonate (C2), an alkali metal salt such as a sodium salt or potassium salt of an α-olefin sulfonic acid having 14 to 16 carbon atoms can be used, and preferably an α-olefin sulfonate having 14 carbon atoms, particularly a sodium salt (generic name: sodium tetradecene sulfonate). These are commercially available products that can be used in oral preparations, and for example, the product name "K Lipolan PJ-400CJ" manufactured by Lion Specialty Chemicals Co., Ltd. can be used. When α-olefin sulfonate (C2) is blended as component (C), the blending amount is preferably 0.1 to 3% of the total composition, more preferably 0.4 to 2.5%, and even more preferably 0.4 to 2%. When the blending amount is 0.1% or more, the unpleasant taste improvement effect is sufficiently obtained and the unpleasant taste is suppressed. When the blending amount is 3% or less, the refreshing feeling in the oral cavity is sufficiently maintained, and the unpleasant taste (bitterness) of the composition itself is prevented from appearing, and the unpleasant taste improvement effect is sufficiently maintained.

[0019] In the present invention, the ratio of the blend amount of component (B) (as tin ions) to the blend amount of component (C) (components (C1) and / or (C2)) is preferably in the following range. When cationic cellulose (C1) is blended as component (C), the mass ratio of (B) / (C1) is preferably 0.3 to 30, more preferably 1.5 to 10. When the mass ratio of (B) / (C1) is within the above range, the oral cavity has a more excellent fresh feeling and the unpleasant taste improving effect is also more excellent. When the mass ratio of (B) / (C1) is less than 0.3, the stickiness of component (C1) adversely affects the oral cavity's fresh feeling, and the oral cavity may become less fresh, while when it exceeds 30, the unpleasant taste improving effect may not be obtained sufficiently. In addition, when α-olefin sulfonate (C2) is blended as component (C), the mass ratio of (B) / (C2) is preferably 0.02 to 5, more preferably 0.05 to 1.5. When the mass ratio of (B) / (C2) is within the above range, the fresh feeling in the oral cavity is more excellent and the effect of improving unpleasant taste is also more excellent. When the mass ratio of (B) / (C2) is less than 0.02, the degreasing feeling of component (C2) may adversely affect the fresh feeling in the oral cavity, and the fresh feeling in the oral cavity may be reduced.

[0020] The oral cavity composition of the present invention may further contain a cooling agent (D). The incorporation of component (D) enhances the refreshing feeling in the oral cavity, further improving the feeling of effectiveness. Examples of the (D) component include N-ethyl-p-menthane-3-carboxamide, ethyl-3-(p-menthane-carboxamide) acetate, N-(4-cyanomethylphenyl)-p-menthanecarboxamide, N-(2-(pyridin-2-yl)-3-p-menthanecarboxamide), 2-isopropyl-N,2,3-trimethylbutyramide, menthone glycerol acetal, menthoxypropane-1,2-diol, menthyl lactate, and menthyl succinate. Among these, N-(4-cyanomethylphenyl)-p-menthanecarboxamide and N-(2-(pyridin-2-yl)-3-p-menthanecarboxamide) are preferred. These may be used alone or in combination of two or more.

[0021] Specifically, the following commercially available products can be used. N-Ethyl-p-menthane-3-carboxamide; Symrise, WS-3 Ethyl 3-(p-menthane-carboxamide) acetate; Toyotama Fragrance Co., Ltd., WS-5 N-(4-cyanomethylphenyl)-p-menthanecarboxamide: EVERCOOL 180, manufactured by Givaudan Japan Co., Ltd.
