Abstract
Description
[0001] Cosmetic use of iso-urolithin A on sensitive skin
[0002] The present invention relates to the cosmetic use of at least iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates, for preventing and / or treating a skin manifestation on sensitive skin.
[0003] The skin is capable of responding to external physicochemical and mechanical stimuli, giving rise to warning sensations such as stinging and itching. These stimuli are of varied origin.
[0004] In particular, subjects with sensitive skin are more sensitive to such stimuli. Sensitive skin is defined as non-pathological skin, which may display adverse skin manifestations.
[0005] As a general rule, undesirable skin manifestations may be expressed in two aspects, namely sensations of discomfort felt on the skin and / or onset of visible skin signs.
[0006] In the case of sensations of discomfort, these may typically consist of itching, heating, stinging and / or tautness sensations. As a representation of visible skin signs, mention may in particular be made of redness.
[0007] These skin manifestations are more frequent in the most exposed areas of the body, namely the hands, feet, face and scalp.
[0008] This skin reactivity conventionally results in the manifestation of signs of discomfort in response to the subject coming into contact with a triggering element which may have various origins.
[0009] These skin reactions may occur in particular on areas subjected to certain daily or frequently repeated hygiene activities such as shaving, hair removal, cleaning with toiletry products or cleaning products, application of adhesives (dressings, patches, attachment of prosthetics) or in the case of activities associated with sports, occupations or simply lifestyle and the use of clothing, tools or equipment generating localized friction, and the intake of certain foods. They may also be amplified by temperature variations, wind and psychological stress.
[0010] It is furthermore known that the risk of manifestation of these skin reactions is exacerbated in the case of sensitive skins.
[0011] Thus, the onset of signs of discomfort, which appear within minutes following the subject coming into contact with the triggering element, is one of the essential characteristics of sensitive skins. They mainly consist of dysesthetic sensations. The term dysesthetic sensations means more or less painful sensations perceived in a skin area such as stinging, tingling, itching, heating, discomfort, tautness, etc. These subjective signs exist most often in the absence of visible clinical signs such as redness or desquamation.
[0012] The reactivity of sensitive skin is not related to a process resulting in an inflammatory response and does not induce a specific immune and memory response.
[0013] In this respect, it should be completely distinguished from skins displaying inflammatory and allergic reactions such as dermatosis, eczema and / or ichthyosis, and against which a certain number of treatments have already been proposed.
[0014] For obvious reasons, it would be advantageous to have compositions capable of preventing and / or treating these undesirable skin manifestations.
[0015] The aim of the present invention is specifically that of meeting this need.
[0016] The present invention thus relates to the cosmetic use of at least iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates, for preventing and / or treating a skin manifestation on sensitive skin.
[0017] Iso-urolithin A, or 3,9-dihydroxy-6H-dibenzo[c]chromen-6-one, is the compound having the following general formula (I):
[0018] [Chem 1]
[0019] Iso-urolithin A is a microbial metabolite produced by enzyme catabolism in commensal bacteria from the intestine from ellagic acid and low-bioavailability ellagitannins. Ellagic acid and ellagitannins are obtained from edible plants such as berries, tropical fruits such as pomegranate and medicinal plants such as Camellia sinensis.
[0020] Mention may be made for example of iso-urolithin A marketed by Astatech under the sales reference ATE638637155.
[0021] The term "salt" of iso-urolithin A according to the invention means a salt formed by an inorganic or organic base. As examples of basic salts, mention may be made of alkali metal hydroxides such as sodium, potassium and lithium; alkaline earth-metal hydroxides such as calcium and magnesium; hydroxides of other metals, such as aluminum and zinc; ammonia and organic amines such as non-substituted or hydroxy-substituted mono-, di- or tri-alkylamines; dicyclohexylamines; tributyl amines; pyridine; N-methyl-N-ethylamine; diethylamine; triethylamine; mono-, bis- or tris-(2-hydroxy-alkylamines) such as mono-, bis- or tris-(2- hydroxyethyl)amine, 2-hydroxy-tert-butylamine ortris-(hydroxymethyl)methylamine, N, N-di- alkyl-N-(hydroxyalkyl)-amines, such as N,N-dimethyl-N-(2-hydroxyethyl)amine or tri-(2- hydroxyethyl)amine; N-methyl-D-glucamine; and amino acids such as arginine and lysine.
[0022] A salt of iso-urolithin A according to the invention is preferably a basic salt, in particular a sodium or potassium salt.
[0023] In the sense of the invention, the expression "skin manifestation" covers sensations of discomfort, in a subject in particular with sensitive skin, as well as temporary visible and unsightly skin signs capable of affecting said subject. Thus, preferably, the skin manifestation is a sensation of discomfort.
[0024] The subject (or person) is a subject with sensitive skin, preferably a human subject.
[0025] The term "skin" means all the skin of the body, and preferably the skin of the face, scalp, neckline, neck, arms and forearms, or even more preferably, the skin of the face (in particular the forehead, nose, cheeks, chin), neckline and neck.
[0026] As specified above, the sensations of discomfort may be itching, heating, stinging and / or tautness.
[0027] These undesirable manifestations concern most particularly people said to have sensitive skin, who have a lowered reactivity threshold; these skins will therefore display these clinical signs much more quickly and frequently than other types of skins.
[0028] Preferably, the skin manifestation on a sensitive skin according to the invention is expressed by the manifestation of one or more visible skin signs such as redness and desquamation.
[0029] According to another of its aspects, the present invention relates to the cosmetic use of at least iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates as a soothing agent.
[0030] The term "soothing agent" means an agent capable of reducing a skin manifestation, preferably on a normal skin, in particular capable of reducing redness and / or a sensation of discomfort. By normal skin, it is meant a non-pathological and non-sensitive skin. Preferably, the soothing agent is a skin-soothing agent, preferably a topical skin-soothing agent.
[0031] According to another of its aspects, it relates to the cosmetic use of at least isourolithin A and / or one of its salts and / or one of its solvates such as hydrates for preventing and / or treating manifestations of dysesthetic sensations on sensitive skins.
[0032] Therefore, the aim of the invention is in particular that of preventing and / or reducing sensations of skin discomfort, such as for example stinging, tingling, itching, heating, discomfort and / or tautness, in particular in people with sensitive skins.
[0033] Preferably, iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates according to the invention is administered topically.
[0034] According to another of its aspects, the invention relates to a cosmetic treatment process comprising topical application on a subject's sensitive skin of a cosmetic composition comprising at least iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates.
[0035] Such a process is cosmetic, therefore non-therapeutic. The same applies for the different uses concerned in the present application.
[0036] In the sense of the present invention, "prevent" means reducing the risk of manifestation of a phenomenon.
[0037] In the sense of the present invention, the term "treat" means compensate for a physiological dysfunction and more generally reduce, or suppress, an undesirable disorder, the manifestation of which is in particular a consequence of this dysfunction.
[0038] Sensitive skin is different from allergic skin. Its reactivity does not stem from an immunological process and generally only results in dysesthetic sensations.
[0039] For obvious reasons, the absence of visible signs renders the diagnosis of sensitive skin difficult. This diagnosis is most often based on taking history from the subject.
[0040] Consequently, the development of products for "sensitive skins" has required tools for evaluating the sensory reaction of the skin. The first tools were based from their design on the essential characteristics of sensitive skins namely the presence of signs of discomfort induced by a topical application. Thus, the lactic acid "stinging test" was the first test proposed. It is carried out by recording the stinging sensations reported by a volunteer after applying a 10% lactic acid solution on the sides of the nose. Subjects reporting moderate or substantial stinging sensations are referred to as "stingers" and considered as having sensitive skin. On account of this skin sensitivity to topical product application, these subjects are then selected to test so-called sensitive skin products. More recently, novel tests have been proposed which use other discomfort inducers such as capsaicin. Indeed, it has been demonstrated in the literature that this sensitive skin condition may be associated an exacerbated TRPV1 (transient receptor potential vanilloid 1) calcium channel response to capsaicin (specific TRPV1 receptor agonist) (Jourdain, R., et al., Ethnic variations in facial skin neurosensitivity assessed by capsaicin detection thresholds. Contact Dermatitis, 2009. 61(6): p. 325-31). The activation of this receptor results in the genesis of an intracellular calcium flux, the release of neuromediators and the activation of a signaling cascade causing the sensation of discomfort. Thus, reducing this signaling cascade by limiting the intracellular calcium fluxes generated by the direct and / or indirect activation of these receptors on the surface of keratinocytes represents a particularly useful tool for identifying compounds of interest for treating sensitive skins.
[0041] In the sense of the present invention, the expression "sensitive skins" covers reactive skins and intolerant skins.
[0042] An intolerant skin is a skin which reacts at least with sensations of heating, tautness, tingling and / or redness, to various factors such as the application of cosmetic or dermatological products or soap. In general, these signs are associated with hyper- seborrheic skin.
[0043] A reactive skin is a skin which reacts by itching or stinging, to various factors such as the environment, emotions, foods, wind, friction, shaving, hard water with a high limescale concentration, temperature variations, humidity or wool.
[0044] It is understood that these sensations may in parallel be associated with the manifestation of skin signs. Thus, as a general rule, this type of skin may be associated with dry skin or skin presenting with redness. Dry skin is essentially manifested by a sensation of tautness and / or tension. The skin is also rough to the touch and appears to be covered with flakes. When the skin is slightly dry, these flakes are plentiful and difficult to see with the naked eye. They are less and less numerous but increasingly visible with the naked eye when this disorder worsens.
[0045] Within the scope of the present invention, the sensitive skins in question do not exhibit any inflammatory signs. Iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates may be formulated in a composition at a rate of at least 0.00001% by weight, in particular at a rate of 0.0001 to 20% by weight and more particularly at a rate of 0.001 to 2% by weight with respect to the total weight of the composition.
[0046] The compositions according to the invention may be presented in any dosage forms normally available for the selected mode of administration. The substrate may be of various natures according to the type of composition in question.
[0047] Preferably, iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates according to the invention is administered topically, i.e. on the skin surface. As topical compositions, mention may be made of aqueous, hydroalcoholic or oily solutions, dispersions such as lotion or serum, emulsions of liquid or semi-liquid consistency such as milk, suspensions or emulsions such as cream, aqueous or anhydrous gel, microemulsions, microcapsules, microparticles or ionic and / or non-ionic vesicular dispersions. These compositions are prepared using routine methods.
[0048] These compositions may in particular constitute creams for cleansing, protection, treatment or care for the face, hands, feet, for the major anatomical folds or for the body (for example day creams, night creams, cleansing, foundation creams, sun creams), makeup products such as fluid foundations, makeup-removing milks, body milks for protection or care, after-sun milks, lotions, gels or foams for skin care, such as cleansing and disinfection lotions, sun lotions, artificial tanning lotions, bath compositions, deodorant compositions containing a bactericidal agent, aftershave gels or lotions, depilatory creams or compositions against insect bites.
[0049] The compositions according to the invention may also consist of solid preparations constituting soaps or cleansing bars.
[0050] They may also be used for the scalp in the form of solutions, creams, gels, emulsions, mousses, or alternatively in the form of aerosol compositions also containing a propellant under pressure.
[0051] The compositions according to the invention may comprise one or more routine adjuvants in the cosmetic field, such as organic solvents, hydrophilic or lipophilic gelling agents, preservatives, fatty substances such as oils, emulsifiers, antioxidants, solvents, perfumes, fillers, sun filters, odor absorbers, dyestuffs and mixtures thereof. The quantities of these various adjuvants are those conventionally used in the fields in question, and for example from 0.01 to 20% of the total weight of the composition. These adjuvants, depending on their nature, may be introduced into the fatty phase or into the aqueous phase. These adjuvants, as well as their concentrations, must be such that they do not harm the advantageous properties of iso-urolithin A and / or one of its salts and / or one of its solvates.
[0052] By way of organic solvents suitable for use in the composition of the invention, mention may be made of water-miscible organic solvents.
[0053] According the present invention, "water-miscible solvent" means a compound that is liquid at ambient temperature and water-miscible, having in particular a miscibility in water greater than 50% by weight at 25°C and atmospheric pressure.
[0054] The water-miscible organic solvents may be chosen from lower monoalcohols having 1 to 5 carbon atoms such as ethanol and isopropanol, glycols having 2 to 8 carbon atoms such as ethylene glycol, hexylene glycol, propylene glycol, 1 ,3-butylene glycol, pentylene glycol, dipropylene glycol, and glycerin.
[0055] By way of oils suitable for use in the composition of the invention, mention may be made for example of:
[0056] - hydrocarbon oils of animal origin;
[0057] - hydrocarbon oils of plant origin;
[0058] - synthetic esters and ethers, in particular of fatty acids, such as the oils of formulas R1COOR2 and R1OR2 wherein R1 represents the residue of a fatty acid including 8 to 29 carbon atoms, and R2 represents a hydrocarbon chain, branched or not, containing 3 to 30 carbon atoms;
[0059] - linear or branched hydrocarbons, of mineral or synthetic origin;
[0060] - fatty alcohols having from 8 to 26 carbon atoms;
[0061] - partially hydrocarbonated and / or siliconized fluorinated oils;
[0062] - silicone oils;
[0063] - mixtures thereof.
[0064] The term hydrocarbon oil in the list of oils cited above denotes any oil comprising mostly carbon and hydrogen atoms, and optionally ester, ether, fluorinated, carboxylic acid and / or alcohol groups.
[0065] The other fatty substances that could be present in the oily phase are for example fatty acids including 8 to 30 carbon atoms; waxes; silicone resins; and silicone elastomers. These fatty substances may be selected in varied ways by those skilled in the art in order to prepare a composition having the desired properties, for example in terms of consistency or texture.
[0066] According to a particular embodiment of the invention, the composition used within the scope of the invention is anhydrous.
[0067] The term "anhydrous" means a composition comprising less than 5% by weight of water with respect to the total weight of the composition, preferably less than 3% by weight, even more preferably less than 1 % with respect to the total weight of the composition, or even totally free from water (0%).
[0068] According to a particular embodiment of the invention, the composition used within the scope of the invention is a water-in-oil (W / O) or oil-in-water (O / W) emulsion. The proportion of the oily phase of the emulsion may range from 5 to 90% by weight, and preferably from 5 to 60% by weight relative to the total weight of the composition.
[0069] The emulsions generally contain at least one emulsifier selected from amphoteric, anionic, cationic or non-ionic emulsifiers, used alone or in a mixture, and optionally a coemulsifier. The emulsifiers are suitably selected according to the emulsion to be obtained (W / O or O / W). In general, the emulsifier and the co-emulsifier are present, in the composition, in a proportion ranging from 0.3 to 30% by weight, and preferably from 0.5 to 20% by weight with respect to the total weight of the composition.
[0070] By way of emulsifiers and co-emulsifiers suitable for use in the invention, mention may be made for example of O / W emulsifiers in particular non-ionic emulsifiers such as esters of fatty acid and polyethylene glycol, esters of fatty acid and glycerin as well as W / O emulsifiers such as dimethicone copolyols, alkyl-dimethicone copolyols, cross-linked elastomer solid organopolysiloxane including at least one oxyalkylenated group.
[0071] By way of hydrophilic gelling agents, mention may be made of carboxyvinyl polymers (carbomer), acrylic copolymers such as acrylate / alkylacrylate copolymers, polyacrylamides, polysaccharides, natural gums and clays; by way of lipophilic gelling agents, mention may be made of modified clays such as bentones, metal salts of fatty acids, hydrophobic silica and polyethylenes.
[0072] By way of fillers which can be used in the composition of the invention, mention may be made, besides pigments, of silica powder; talc; starch; polyamide particles; polyethylene powders; acrylic copolymer-based microspheres; expanded powders such as hollow microspheres; silicone resin microbeads; and mixtures thereof. These fillers may be present in quantities ranging from 0.1 to 20% by weight and preferably from 1 to 10% by weight with respect to the total weight of the composition or the preparation according to the invention. The invention also relates to a cosmetic treatment process comprising topical application on a subject's sensitive skin of a cosmetic composition comprising at least isourolithin A and / or one of its salts and / or one of its solvates such as hydrates.
[0073] The cosmetic treatment process of the invention may be implemented in particular by administering the cosmetic and / or dermatological compositions as defined above, according to the usual technique of use of these compositions. For example: applications of creams, gels, serums, lotions, makeup-removing milks or after-sun compositions on the skin or dry scalp, application of a hair lotion or shampoo to the wet scalp, regarding topical application.
[0074] The cosmetic treatment process according to the invention may thus be implemented by topical administration, for example daily, of iso-urolithin A and / or one of its salts and / or one of its solvates such as hydrates.
[0075] The process according to the invention may comprise single administration. According to another embodiment, the administration is repeated for example 2 to 3 times daily for one or more days, and generally over a prolonged period of at least 4 weeks, or 4 to 15 weeks, with where applicable one or more break periods.
[0076] The invention is now illustrated by the following non-limiting examples.
[0077] Example 1 : Soothing effect of iso-urolithin A
[0078] Principle of method for measuring the soothing effect
[0079] Monitoring the activation of certain calcium channel type receptors (TRPV1 or transient receptor potential vanilloid 1) (whether direct or indirect) is enabled by measuring calcium influx in the cytoplasm of skin cells.
[0080] A soothing effect is obtained if intracellular calcium flux inhibition is observed.
[0081] Protocol for culturing cells for preparing the intracellular calcium flux measurement
[0082] 1. Corneal epidermal keratinocytic cells
[0083] The cells were isolated from epidermal tissue according to the Mat’Tek protocol described in 2006 (Development of a method for assessing micronucleus induction in a 3D human skin model (Epiderm), Rodger D.Curren and al, Volume 607, Issue 2, September 5, 2006, Pages 192-204). HCE (Human Corneal Epithelial) cells are inoculated in sterile black Advanced TC (Greiner) 96-well plates at the cellular density of 20,000 cells / well / 0.2ml for testing 48h after inoculation, and 10,000 cells / well / 0.2ml for testing 72h after inoculation.
[0084] 2. Intracellular fCa2+1 measurement & Expression of results
[0085] The molecules tested are urolithin A from Sigma Aldrich (SML-1791), and iso-urolithin A from Astatech (ATE638637155). a. Solubility of the molecules to be evaluated
[0086] The solubility of each molecule is preferably tested in HBSS buffer (Gibco-lnvitrogen) supplemented with 20mM of HEPES (Gibco-lnvitrogen) and adjusted to pH 7.4. For non- water-soluble molecules, DMSO (Sigma-Aldrich) is used initially, followed by ethanol (Prolabo). For a molecule soluble in DMSO, the final DMSO concentration administered to the keratinocytes is 2% whereas for a molecule soluble in ethanol, the final ethanol concentration must not exceed 1%. Each molecule is tested in the agonist version of the test in order to ensure the non-activation of the receptors by the molecule before evaluating its soothing properties. The maximum dose tested is determined according to the nonactivation of the receptors but also the solubility of the molecules and / or their cytotoxicity on the cells. At least three concentrations are tested per molecule and are produced by successive dilutions.
[0087] For soluble, non-agonist and non-cytotoxic molecules, the maximum dose tested on the HCE keratinocyte cells is 1 mg / ml. b. Preparation of the fluorescent calcium probe
[0088] The calcium concentration is measured using the 5pM fura-2AM Ester fluorescent probe (Molecular Probes). The probe is previously solubilized in DMSO. An equal volume of 10% Pluronic F-127 surfactant (Molecular Probes) is added for better solubilization of the probe. c. Cell treatment
[0089] After 48 or 72 hours of inoculation, the cells are washed twice in HBSS buffer then incubated for 30 minutes at 37°C in the presence of the previously prepared calcium probe. 2 washes in HBSS buffer are performed before subsequently treating the cells for 30 minutes at ambient temperature and protected from light with the solutions of molecules to be evaluated at the defined concentrations. d. Preparation of the reference agonist solution
[0090] In parallel, the reference TRPV1 pathway agonist solution is prepared extemporaneously then deposited in a 96-well plate enabling automatic injection on the cells in order to perform real-time monitoring of the calcium influx.
[0091] The reference agonist of the TRPV1 pathway is capsaicin (Sigma-Aldrich), soluble in 1% DMSO, used at 300pM. e. Real-time calcium influx monitoring
[0092] The fluorescence is measured on a FlexStation 3 microplate reader (Molecular Devices) at a double excitation wavelength: 340 nm (bound Ca2+), 380nm (free Ca2+) and at an emission length of 510 nm. The measurements are made for 30 seconds - baseline - and for 3 minutes after the automatic injection by the Flexstation of the reference agonist (agonist or HBSS buffer or DMSO or ethanol, volume of 50pl / well). 3 controls are produced in parallel: HBSS control; DMSO and ethanol control if required, and Activation Control. Each condition is replicated on at least 2 independent tests with 6 wells per condition and per test.
[0093] Results
[0094] The results are expressed as a ratio of the fluorescence intensity of 2 wavelengths (340nm / 380nm) and processed as a % variation of the ratio on a kinetics of 10 to 300 seconds.
[0095] The response peak obtained with the molecule to be evaluated in combination with the agonist is compared with that of the agonist alone, and the inhibition percentage is then determined.
[0096] 1. Effect of iso-urolithin A on TRPV1
[0097] For each urolithin, an inhibitory capability in respect of the calcium flux induced by the capsaicin agonist for the TRPV1 pathway is observed at the tested concentrations with respect to the 0 pM control (Table 1). Iso-urolithin A has an inhibiting effect in respect of the calcium flux induced by capsaicin of approximately 45-60% at the tested concentrations, unlike urolithin which has an inhibiting effect in respect of the calcium flux of approximately 30-35% (Table 2). This receptor has particularly been associated with people with sensitive skin, and its targeting is of interest for reducing stinging and heating sensations in skin discomfort. Percentage calcium flux variation in response to capsaicin (TRPV1 agonist) and after adding different urolithin A and iso-urolithin A concentrations
[0098] The percentage calcium flux variation of Table 1 is calculated as follows:
[0099] % Calcium flux variation = ((fluorescence intensity at 340nm for x second(s) I fluorescence intensity at 380nm for x second(s)) - (fluorescence intensity at 340nm to I fluorescence intensity at 380nm tO)) I (fluorescence intensity at 340nm to I fluorescence intensity at 380nm tO).
[0100] [Table 1]
[0101] Percentage calcium flux inhibition in response to capsaicin (TRPV1 agonist) and after adding different urolithin A and iso-urolithin A concentrations
[0102] [Table 2]
[0103] The percentage inhibition of Table 2 is calculated as follows:
[0104] % inhibition = 100 - ((% Variation of calcium flux of compound "X" x 100) / % Variation of calcium flux of TRPV1 receptor agonist, i.e. capsaicin)
[0105] The results given above show a greater inhibition capability in respect of calcium influxes generated after TRPV1 receptor activation for iso-urolithin A (according to the invention) compared to urolithin A (non-invention), suggesting a greater antagonistic effect of isourolithin A on the TRPV1 receptor than that observed with urolithin A.
[0106] Iso-urolithin A therefore represents a molecule exhibiting good efficacy for treating itching and stinging sensations. Iso-urolithin A has a soothing effect and could be used on the following skin conditions: stinging, tautness, discomfort, itching, redness and / or sensation of heating of the skin (including the scalp).
