Description

MICRONEEDLE ASSEMBLY AND WEARABLE SENSING APPARATUS COMPRISING SAID MICRONEEDLE ASSEMBLYTECHNICAL FIELD

[0001] The disclosure relates to a microneedle assembly comprising a needle structure, the needle structure comprising at least one microneedle array. Each microneedle array comprises at least one microneedle. The disclosure furthermore relates to a wearable sensing apparatus comprising the microneedle assembly and a method of manufacturing the microneedle assembly.BACKGROUND

[0002] Wearable sensors enable continuous measurement of different vital parameters such as heart rate and body analytes. A continuous glucose monitor (CGM) is a wearable device that continuously measures blood glucose using a 5 mm long sensing filament that is inserted through the skin of the user. The application of the sensor requires a sharp 10 mm long needle that can cause pain or bleeding. To avoid the pain and potential bleeding sub-millimeter sensing needles called microneedles have been developed.

[0003] In order to penetrate the skin, the sensing microneedles must have a sharp tip that very efficiently penetrates the upper skin layer (epidermis) and that does not undergo plastic deformation. In addition, there should be an accurately patterned sensing area that can interact with different body analytes just below the sharp tip of the microneedle. This requires microneedles with excellent mechanical properties, high dimensional accuracy, and high precision coating which is typically achieved using unconventional fabrication methods that are poorly scalable and high-cost.

[0004] Hence, there is a need for an improved microneedle assembly suitable for wearable sensing devices.SUMMARY

[0005] It is an object to provide an improved microneedle assembly for wearable sensing devices. The foregoing and other objects are achieved by the features of the independent claims. Further implementation forms are apparent from the dependent claims, the description, and the figures.

[0006] According to a first aspect, there is provided a microneedle assembly comprising a needle structure comprising at least one microneedle array, each microneedle array comprising at least one microneedle; and a reinforcement structure comprising at least one perforation array, each perforation of the perforation array (s) being configured to accommodate an individual microneedle of the microneedle array (s) ; each microneedle having a first length and each perforation having a second length, the first length > the second length, the needle structure and the reinforcement structure being configured to abut, when assembled, such that each microneedle extends through, and protrudes from, one of the perforations.

[0007] This allows a solution that is simple to manufacture and which is durable as the reinforcement structure stabilizes the microneedles during skin puncturing and fixes the microneedles during assembly and processing. Furthermore, the reinforcement structure provides electrical insulation between individual microneedles or rows of microneedles.

[0008] In a possible implementation form of the first aspect, the needle structure comprises a planar section, each microneedle being an electrode extending from a surface of the planar section. This facilitates manufacturing the needle structure as a single-piece body.

[0009] In a further possible implementation form of the first aspect, the needle structure comprises conductive material and the planar section is configured to electrically connect the electrodes to an electric device or system, facilitating interconnection between electrodes and other parts such as a sensor system.

[0010] In a further possible implementation form of the first aspect, the reinforcement structure comprises a planar sheet of insulating material, each perforation of the reinforcement structure comprising a throughgoing opening extending through the planar sheet in a direction perpendicular to a main plane of the planar sheet; and a hollow body of the insulating material extending from the planar sheet in the direction perpendicular to the main plane, the throughgoing opening and the hollow body being coaxially aligned to form a channel configured to accommodate one of the microneedles. This fixates the microneedles in the correct position during skin puncturing and sensor fabrication. Furthermore, the hollow bodies insulate the microneedles electrically from each other and from external factors (e.g. sweat) .

[0011] In a further possible implementation form of the first aspect, a free end of the hollow body comprises a rounded or chamfered surface to facilitate skin penetration.

[0012] In a further possible implementation form of the first aspect, a lumen of the hollow body has a tapered shape in a direction from the throughgoing opening towards the free end of the hollow body, in order to self-align the microneedles during the insertion.

[0013] In a further possible implementation form of the first aspect, the microneedles of the microneedle array (s) comprise a conductive material and are arranged in parallel with each other, each microneedle comprising an elongated part and a free end configured to penetrate the skin of a user. This facilitates microneedles which may be conductively interconnected and / or manufactured from one single piece.

[0014] In a further possible implementation form of the first aspect, the free end of the microneedle and / or the elongated part of the microneedle is coated with a functional layer, facilitating functional capabilities of the microneedles.

[0015] In a further possible implementation form of the first aspect, the free end of the microneedle is at least partially coated with a biosensitive material or provided with a  structure made of biosensitive material, facilitating the sensing capabilities of the microneedles.

[0016] In a further possible implementation form of the first aspect, the biosensitive material is coated with a layer of protective material or at least partially surrounded by a structure made of protective material, providing mechanical protection of the biosensitive material.

[0017] In a further possible implementation form of the first aspect, the elongated part of the microneedle is coated with a functional layer of sensing, insulating, adhesive, and / or sealing material, facilitating for example sensing capabilities or other properties.

[0018] In a further possible implementation form of the first aspect, the microneedle comprises at least one recessed area. Such recesses protect the biosensitive material from mechanical forces, e.g. shear force, that occur during skin penetration. When the biosensitive material is deposited in a recess, it does not protrude from the surface of the microneedle, thus the mechanical damage during skin penetration may be reduced.

[0019] In a further possible implementation form of the first aspect, each microneedle array comprises at least one counter electrode, one working electrode, and one reference electrode. This allows the electrodes to be addressed individually.

[0020] In a further possible implementation form of the first aspect, the needle structure comprises at least three microneedle arrays, and each microneedle array comprises only counter electrodes, only working electrodes, or only reference electrodes, electrodes within one microneedle array being electrically interconnected by a portion of the needle structure. This allows several electrodes to be addressed simultaneously.

[0021] In a further possible implementation form of the first aspect, the planar sheet of the reinforcement structure comprises a plurality of grooves, each groove extending across and interconnecting the perforations of one perforation array, and each groove being configured to at least partially accommodate the portion of one microneedle array. This facilitates manufacturing by machining.

[0022] In a further possible implementation form of the first aspect, the microneedle assembly further comprises a slotted substrate, each slot of the slotted substrate being configured to at least partially accommodate the portion of one microneedle array, the portion forming an electrical connector. This allows the interconnection between electrodes and other parts of the system, e.g. a wearable system, to be done at low temperatures such that any sensing layer is not affected by heat.

[0023] According to a second aspect, there is provided a wearable sensing apparatus comprising the microneedle assembly as described above. This facilitates a wearable device that is simple to manufacture as well as durable and reliable.

[0024] According to a third aspect, there is provided a method of manufacturing a microneedle assembly according to the above, comprising the steps of manufacturing the needle structure by machining or etching a conductive material; manufacturing the reinforcement structure by etching, machining, or molding an insulating material; coating an interface between the needle structure and the reinforcement structure with an adhesive material; and inserting the needle structure into the reinforcement structure such that each perforation of the reinforcement structure accommodates a microneedle of the needle structure.

[0025] This method provides an assembly that is simple to manufacture and which is durable as the reinforcement structure stabilizes the microneedles during skin puncturing and fixes the microneedles during assembly and processing. Furthermore, the reinforcement structure provides electrical insulation between individual microneedles or rows of microneedles.

[0026] In a possible implementation form of the third aspect, the method comprises the additional step of removing sections of the needle structure that are not accommodated within the perforations of the reinforcement structure. This allows the microneedles to be  in direct contact with electrical connectors. It also allows the microneedles to be individually addressable.

[0027] In a further possible implementation form of the third aspect, the method comprises the additional step of depositing at least one functional layer on the needle structure before coating the interface between the needle structure and the reinforcement structure. This may for example facilitate the sensing capability of the assembly.

[0028] These and other aspects will be apparent from the embodiments described below.BRIEF DESCRIPTION OF THE DRAWINGS

[0041] The present invention relates to a microneedle assembly 1 comprising a needle structure 2 comprising at least one microneedle array 3, each microneedle array 3 comprising at least one microneedle 4; and a reinforcement structure 5 comprising at least one perforation array 6, each perforation 7 of the perforation arrays 6 being configured to accommodate an individual microneedle 4 of the microneedle arrays 3; each microneedle 4 having a first length L1 and each perforation having a second length L2, the first length L1 > the second length L2, the needle structure 2 and the reinforcement structure 5 being configured to abut, when assembled, such that each microneedle 4 extends through, and protrudes from, one of the perforations 7.

[0042] As illustrated in Fig. 1, the microneedle assembly 1 comprises a needle structure 2 and a reinforcement structure 5.

[0043] The needle structure 2 comprises at least one microneedle array 3. Each microneedle array 3 comprises at least one microneedle 4. Figs. 3 and 4 illustrate three microneedle arrays 3, wherein each microneedle array 3 comprises three microneedles 4. This is merely an example as the microneedle assembly 1 may comprise any suitable number of microneedle arrays 3, and each microneedle array 3 may comprise any suitable number of microneedles 4. One microneedle array 3 is illustrated by a dashed rectangle in Fig. 4.

[0044] The reinforcement structure 5 comprises at least one perforation array 6. Each perforation 7 of the perforation array (s) 6 is configured to accommodate an individual microneedle 4 of the microneedle array (s) 3. The reinforcement structure 5 therefore comprises perforations 7 that correspond in number and layout to the number and layout of the microneedles 4 of the needle structure 2. One perforation array 6 is illustrated by a dashed rectangle in Figs. 4, 5, and 7.

[0045] The needle structure 2 and the reinforcement structure 5 are configured to abut, when assembled, such that each microneedle 4 extends through, and protrudes from, one of the perforations 7. This is illustrated in Figs. 1, 3, and 4 which show the microneedle assembly 1 and the microneedles 4 of the needle structure 2 penetrating the perforations 7 of the reinforcement structure 5.

[0046] As illustrated in Figs. 1 and 11, each microneedle 4 has a first length L1 and each perforation has a second length L2, the first length L1 > the second length L2. In other words, the first length L1 of the microneedle 4 is longer than the second length L2 of the perforation 7, such that the microneedle 4 necessarily protrudes from beyond the perforation 7.

[0047] As shown in Figs. 2 and 8 to 10, the needle structure 2 may comprise a planar section 8. The planar section 8 of the needle structure 2 may, in other words, be substantially two-dimensional or sheet-shaped.

[0048] Each microneedle 4 may be an electrode extending from the surface of the planar section 8. The needle structure 2 may comprise conductive material. The microneedles 4 may comprise a conductive material. The planar section 8 may be configured to electrically connect the electrodes to an electric device or system such as an electric source or electronic amplifier. The electrodes may be electrically separated.

[0049] Each microneedle array 3 may comprise at least one counter electrode 4, one working electrode 4, and one reference electrode 4.

[0050] The needle structure 2 may comprise at least three microneedle arrays 3, and in turn each microneedle array 3 may comprise only counter electrodes 4, only working electrodes 4, or only reference electrodes 4. The electrodes 4 within one microneedle array 3 are electrically interconnected by a portion 19 of the needle structure 2, for example, a portion 19 of the planar section 8 as shown in Fig. 3.

[0051] The needle structure 2 may be a single-piece body comprising the planar section 8 as well as the microneedles 4. The planar section 8 may be a sacrificial part that is used for manipulating multiple microneedles during manufacture and assembly, and which is subsequently removed. Removal of the sacrificial part allows each microneedle to be addressed individually.

[0052] The microneedles 4 may be arranged in parallel with each other, within each microneedle array 3 and / or between microneedle arrays 3.

[0053] Each microneedle 4 may comprise an elongated part 15 and a free end 14 configured to penetrate the skin of the user / wearer, see Figs. 2 and 8 to 10.

[0054] The free end 14 of the microneedle 4 and / or the elongated part 15 of the microneedle may be coated with a functional layer.

[0055] The free end 14 of the microneedle 4 may be at least partially coated with a biosensitive material (not shown) or be provided with at least one structure made of biosensitive material 16 as illustrated in Figs. 9 and 10.

[0056] The biosensitive material may be coated with a layer of protective material (not shown) or at least partially surrounded by a structure made of protective material 17 as illustrated in Fig 10., providing mechanical protection of the biosensitive material.

[0057] As shown in Fig. 2, the elongated part 15 of the microneedle may be coated with a functional layer 22 of sensing, insulating, adhesive, and / or sealing material. This surface below the functionalized free end 14 or tip may be covered with an uncured polymer, e.g. an adhesive, that serves as an insulator and a sealant. Such an insulator, arranged below the functionalized microneedle tips, may replace the insulating function of the hollow body 11.

[0058] The microneedle 4 may comprise at least one recessed area 18, as shown in Figs. 9 and 10. Recesses 18 may be made in the insulating coating, for example by making a pattern using laser ablation. Recesses 18 may also be created by micromaching cavities and / or depressions on the microneedle’s surface. The purpose of such depressions is to further protect the biosensitive material from mechanical forces, e.g. shear force, that occur during skin penetration. When the biosensitive material is deposited in a depression, it does not protrude from the surface of the microneedle 4, thus the mechanical damage during the skin penetration may be reduced.

[0059] The reinforcement structure 5 may comprise a planar sheet 9 of insulating material. Each perforation 7 of the reinforcement structure 5 may comprise a throughgoing opening 10 extending through the planar sheet 9 in a direction perpendicular to a main plane of the planar sheet 9, as shown in Figs. 5 and 7.

[0060] The reinforcement structure 5 may also comprise a hollow body 11, for example, a cylinder, of insulating material that extends from the planar sheet 9 in the direction perpendicular to the main plane, as shown in Fig. 5. The throughgoing opening 10 and the hollow body 11 are coaxially aligned to form a channel configured to accommodate one of the microneedles 4.

[0061] As shown in Fig. 6, a free end 12 of the hollow body 11 may comprise a rounded or chamfered surface to facilitate skin penetration.

[0062] A lumen 13 of the hollow body 11 may have a tapered shape in a direction from the throughgoing opening 10 towards the free end 12 of the hollow body 11 (not shown) .

[0063] The planar sheet 9 of the reinforcement structure 5 may comprise a plurality of grooves 20, each groove 20 extending across and interconnecting the perforations 7 of one perforation array 6, and each groove 20 being configured to at least partially accommodate the portion 19 of one microneedle array 3. Three such grooves 20 are illustrated with dotted lines in Fig. 7.

[0064] As shown in Fig. 2, the microneedle assembly 1 may further comprise a slotted substrate 21. Each slot of the slotted substrate 21 is configured to at least partially accommodate portion 19 of one microneedle array 3, the portion forming an electrical connector. The slotted substrate 21 may be part of a printed circuit board assembly.

[0065] The electrical connection to the remaining system may be performed using low-temperature contacting methods such as slot connectors, spring-loaded pins, or electrically conductive glue.

[0066] The present invention also relates to a wearable sensing apparatus 23 comprising the microneedle assembly 1 as illustrated in Fig. 11. The wearable sensing apparatus 23 may comprise, in addition to the microneedle assembly 1, one or several of an analog frontend  for signal amplification and filtering, an analog-to-digital converter, a microprocessor, a battery, a wireless communication module and antenna, a temperature sensor, and a printed circuit board. The wearable sensing apparatus 23 may be pressed into the skin of the user such that the free ends 14, or tips, of the microneedles 4, which protrude from a bottom side of the wearable sensing apparatus 23, penetrate through the skin by at most 1 mm and at least around 200 μm or at least through the entire epidermis layer.

[0067] The wearable sensing apparatus 23 may be a continuous glucose monitor or any other wearable body analyte monitor that uses the electrochemical sensing principle. The wearable sensing apparatus 23 may be a skin patch, an earring, glasses, a smartwatch, a bracelet, or a necklace.

[0068] The present invention furthermore relates to a method of manufacturing a microneedle assembly 1 according to the above. The method comprises the steps of manufacturing the needle structure 2 by machining or etching a conductive material; manufacturing the reinforcement structure 5 by etching, machining, or molding an insulating material; coating an interface between the needle structure 2 and the reinforcement structure 5 with an adhesive material; and inserting the needle structure 2 into the reinforcement structure 5 such that each perforation 7 of the reinforcement structure 5 accommodates a microneedle 4 of the needle structure 2.

[0069] The needle structure 2 may be made from at least one planar sheet of hard conductive material, for example using laser cutting. The reinforcement structure 5 may for example be manufactured using microinjection molding. The reinforcement structure 5 may also be manufactured by machining (e.g. laser cutting or milling) when comprising grooves 20 as shown in Fig. 7. The insulation may be patterned using laser ablation.

[0070] The method may comprise the additional step of removing sections of the needle structure 2 which are not accommodated within the perforations 7 of the reinforcement structure 5. The removal of one or several sections of the needle structure 2, including the entire planar section 8, may be performed by mechanical or laser cutting, followed by a  polishing step. After the polishing step, the bottom parts of each microneedle become exposed. Another substrate, for example, slotted substrate 21, comprising electrical contacts can be aligned with the bottom parts of the microneedles to establish the electrical connection. The contacts on the substrate may be spring-loaded or coated with a conductive adhesive, thus avoiding any high-temperature processes (e.g. soldering) .

[0071] The method may also comprise the additional step of depositing at least one functional layer on the needle structure 2 before coating the interface between the needle structure 2 and the reinforcement structure 5. For example, the areas of the microneedle below the free end 14 may be functionalized using different metals and biosensitive layers through fabrication methods such as screen printing.

[0072] The various aspects and implementations have been described in conjunction with various embodiments herein. However, other variations to the disclosed embodiments can be understood and effected by those skilled in the art in practicing the claimed subject-matter, from a study of the drawings, the disclosure, and the appended claims. In the claims, the word “comprising” does not exclude other elements or steps, and the indefinite article “a” or “an” does not exclude a plurality. The mere fact that certain measures are recited in mutually different dependent claims does not indicate that a combination of these measures cannot be used to advantage.

[0073] The reference signs used in the claims shall not be construed as limiting the scope. Unless otherwise indicated, the drawings are intended to be read (e.g., cross-hatching, arrangement of parts, proportion, degree, etc. ) together with the specification, and are to be considered a portion of the entire written description of this disclosure. As used in the description, the terms “horizontal” , “vertical” , “left” , “right” , “up” and “down” , as well as adjectival and adverbial derivatives thereof (e.g., “horizontally” , “rightwardly” , “upwardly” , etc. ) , simply refer to the orientation of the illustrated structure as the particular drawing figure faces the reader. Similarly, the terms “inwardly” and “outwardly” generally refer to the orientation of a surface relative to its axis of elongation, or axis of rotation, as appropriate.
