TEST METHOD AND TEST DEVICE FOR FLOW CYTOMETER, AND FLOW CYTOMETERFIELD

[0001] The present disclosure relates to the technical field of flow cytometry, and in particular to a test method and a test device for a flow cytometer, and a flow cytometer.BACKGROUND

[0002] This section provides background information related to the present disclosure, which is not necessarily prior art.

[0003] In a flow cytometer designed with colinear lasers, there is crosstalk between fluorescence signals excited by different lasers, and the crosstalk signal would interfere with performance of an original signal. For example, a 405nm purple laser has a higher energy density than a 488nm blue laser, and a fluorescence signal generated by the 405nm laser dominates a fluorescence signal generated by the 488nm laser. A signal result of a 488nm fluorescence channel obtained from this cannot represent true performance of the 488nm fluorescence channel. Therefore, a more suitable method for testing performance of a fluorescent channel is needed for the flow cytometer.SUMMARY

[0024] As shown in Figure 1, the test method according to an embodiment of the present disclosure may start at step S110 and end at step S120. The test method according to an embodiment of the present disclosure may be applied in a quality control (QC) process. The quality control process aims to demonstrate whether a flow cytometer is qualified. Typically, key functions and performance of the cytometer are checked during the quality control process.

[0028] In a flow cytometer designed with spatially separated lasers, all lasers are turned on during the quality control process. Since different lasers irradiate cells in the sample with different optical paths, there is not crosstalk among generated fluorescence signals. However, in the flow cytometer 100 designed with colinear lasers, as shown in Figure 2, when all lasers are turned on, due to a fact that lasers generated by different lasers irradiate cells in the sample through a same optical path, there is crosstalk between fluorescence signals excited by different lasers, and crosstalk signal may interfere with performance of an original signal.

[0029] Therefore, when testing performance of a fluorescence channel, step S110 and step S120 are performed. In step S110, one of the plurality of lasers is turned on each time, while the remaining lasers are turned off. Then, in step S120, the performance of the fluorescence channel at a laser wavelength emitted by the laser is tested. The plurality of lasers are turned on one by one in sequence until performances of fluorescence channels of all the lasers are tested. In Figure 3A, only the first laser 111 is turned on to test performance of a fluorescence channel of the first laser 111. In Figure 3B, only the second laser 112 is turned on to test performance of a fluorescence channel of the second laser 112. In Figure 3C, only the third laser 113 is turned on to test performance of a fluorescence channel of the third laser 113. In Figure 3D, only the fourth laser 114 is turned on, to test performance of a fluorescence channel of the fourth laser 114.

[0030] Specifically, testing a performance of a fluorescence channel may include: adjusting a gain of a signal of the fluorescence channel after one of the plurality of lasers is turned on, such that a median of a fluorescence signal is a target median; and determining whether the median of the fluorescence signal subjected to gain adjustment is within a predetermined range, and / or determining whether a coefficient of variation (CV) or robust coefficient of variation (rCV) is less than a predetermined value, and / or determining whether a deviation between an adjusted gain and the gain before the adjustment is within a predetermined range. Here, the determining whether the median is within a predetermined range refers to a further confirmation of a result of a gain adjustment. As an example, it may be determined that the performance of the fluorescence channel meets requirements in a case where the three conditions are all met.

[0031] Therefore, the test method according to the embodiment of the present disclosure can eliminate the crosstalk between different sensors in the colinear laser design, so that the performance of the fluorescence channel of each laser can be understood accurately. In addition, due to the ability to accurately measure the performance of the fluorescence channel of each laser, by controlling the performance within predetermined requirements in the quality control process, it can be ensured that the performance of the fluorescence channel of the flow cytometer is well utilized for an actual sample test.

[0036] Next, a background noise is tested in test S203. In the present disclosure, the tested background noise includes at least one of a photoelectric threshold, sheath noise, and sample noise. The photoelectric threshold represents a noise caused by photoelectric elements. The sheath noise represents a noise caused by sheath (such as particles, impurities, and the like, in the sheath) . The sample noise represents a noise caused by a sample pipeline (such as particles, impurities, and the like, in the sample pipeline) .
