Description

Fingolimod oral liquid composition and application thereof

[0001] The present invention claims priority from the prior application filed on May 13, 2024, entitled “A fingolimod oral liquid composition and its application” and application number 202410590969.2. The contents of the above-mentioned prior application are incorporated into this text by reference. Technical Field

[0002] The present application relates to the field of biomedicine, and specifically to a fingolimod oral liquid composition and its application. Background Art

[0003] Fingolimod (CAS No. 162359-55-9, chemical name 2-amino-2-[2-(4-octylphenyl)ethyl]-1,3-propanediol) is an S1P (sphingosine-1-phosphate) receptor modulator that can block the outflow of lymphocytes from secondary lymphoid organs and prevent reactive lymphocytes from entering the central nervous system. Clinically, it can mainly prevent the relapse of multiple sclerosis and the development of physical disability.

[0004] Among the various compositions of fingolimod and its salts, oral solid compositions such as capsules and orally disintegrating tablets have been marketed, but no oral liquid preparation products have been marketed. Therefore, there is a need to develop oral liquid compositions to improve patient compliance.

[0005] However, since oral liquid compositions usually need to be stored for a long time and the storage or transportation environment requirements are generally not strict to facilitate market circulation, the stability of the preparation, especially good long-term stability, is particularly important for the quality assurance of oral liquid compositions.

[0006] In actual pharmaceutical production lines, liquid compositions containing fingolimod or its salts inevitably come into contact with production line equipment (such as stainless steel), which may release iron ions to varying degrees. The inventors surprisingly discovered that, because the alkyl aromatic hydrocarbons in fingolimod are easily oxidized to form ketones, these iron ions catalyze the oxidation of fingolimod, and even trace amounts of iron ions can affect the stability of the fingolimod liquid composition.

[0102] Effect of pH of the composition of Examples 1-5

[0103] Fingolimod hydrochloride pharmaceutical compositions were prepared according to the formulation shown in Table 1. The specific method was as follows: hydroxypropyl-β-cyclodextrin was added to a certain amount of purified water to obtain a solution, fingolimod hydrochloride was then added to the solution to dissolve it, and the other excipients were added in sequence, stirred to dissolve, and finally purified water was added to the volume. Each example composition was sealed in an ampoule and stored at 60°C for 30 days before being tested for impurity content.

[0104] Table 1

[0105] *Reporting limit: 0.05%.

[0106] **For the test data of Example 3, the maximum single impurity and total impurity were 0.05% at day 0, and less than 0.05% at days 10 and 30, which are acceptable errors affected by instrument precision, etc.

[0107] It can be seen that pH has a great influence on the chemical stability of the fingolimod hydrochloride oral solution of the present invention, and the pharmaceutical composition with a pH between 3 and 6 has good high temperature stability.

[0108] Examples 6-10

[0109] Fingolimod hydrochloride pharmaceutical compositions were prepared according to the formulation shown in Table 2, using the same method as in Example 1. The pharmaceutical compositions of each example were sealed in glass bottles and refrigerated at 2-8°C for 5 or 10 days. The bottles were then placed in a dark room under a vertical umbrella light at an illumination of 1000 lx and observed horizontally for the presence of crystals.

[0110] Table 2 Effect of the dosage of hydroxypropyl-β-cyclodextrin

[0111] It can be seen that the weight ratio of hydroxypropyl-β-cyclodextrin to fingolimod hydrochloride has a great influence on the physical stability of the pharmaceutical composition of the present invention. When the above ratio is above 70, the pharmaceutical composition of the present invention has good physical stability.

[0112] Effect of Complexing Agents in Examples 11-14

[0113] In order to simulate the environment of the pharmaceutical composition production line, a trace amount of Fe was added to the formulation. 3+ .

[0114] The fingolimod hydrochloride pharmaceutical composition was prepared according to the prescription in Table 3. The specific method was the same as that in Example 1, except that a specific amount of FeCl3 was added before the volume was fixed. The prepared pharmaceutical composition of each example was sealed in an ampoule and placed under light conditions (illuminance of 4500 lx ± 500 lx, the total illumination of the light source should be not less than 1.2 × 10 6 lux·hr, near-ultraviolet lamp energy is not less than 200W·hr / m 2 ) was placed for 10 days and its related substances were detected.

[0115] It can be seen that the stability of the pharmaceutical composition of the present invention is easily affected by the iron ions leached in the production line, and the addition of a complexing agent can improve the chemical stability of the pharmaceutical composition.

[0116] In the above embodiments, all technical solutions that fall within the scope of the present invention are within the scope of protection of the present invention. It should be noted that for those skilled in the art, several improvements and modifications that do not depart from the principles of the present invention should also be considered within the scope of protection of the present invention.
