Technical Field

Background Art
[0004] Interleukin 1 receptor kinase 4 (IRAK4) is a serine / threonine-specific protein kinase, a member of the tyrosine kinase (TLK) family, and a key node in the innate immune response involving interleukin-1, 18, 33 receptors and Toll-like receptors. After extracellular signaling molecules bind to interleukin receptors or Toll-like receptors, they recruit to form a MyD88:IRAK4:IRAK1 / 2 multiprotein complex, leading to IRAK1 / 2 phosphorylation, mediating a series of downstream signal transductions, thereby activating the p38, JNK and NF-kB signaling pathways, and ultimately leading to the expression of pro-inflammatory cytokines. Clinical pathological studies have shown that individuals with IRAK4 mutations have a protective effect against chronic lung disease and inflammatory bowel disease. IRAK4 deficiency itself is not lethal, and individuals can survive to adulthood, and the risk of infection decreases with age. Therefore, IRAK4 has become an important therapeutic target and has attracted widespread research and development interest. Summary of the invention
