The invention discloses a single
spore isolation method of
alternaria alternata, which comprises the following steps: collecting
disease tissues infected by the
alternaria alternata, and culturing the
alternaria alternata; preparing a culture medium for growth of
alternaria alternata; melting the end part of the clean glass capillary tube, and stretching the glass capillary tube into a sharp
needle shape; under a
microscope, a
glass needle is used for picking single
alternaria alternata spores from the
disease tissue subjected to moisturizing culture, the single
alternaria alternata spores are transferred into a culture medium to be cultured, and a
pure culture is obtained and serves as a basis for purification and identification. According to the method, the
borosilicate glass capillary tube is adopted and can be repeatedly burnt, melted and drawn to be manufactured into a key tool
glass needle required by single
spore separation, materials are effectively saved, meanwhile, the burning and melting process is the
sterilization process, and the operation process is saved. By optimizing preparation of a single
spore separation tool, the separation efficiency of alternaria alternata can be remarkably improved, the experimental period is shortened, the experimental cost is saved, the success rate of single spore separation is increased, and the single spore
separation process of fungi is quicker, more efficient and more accurate.
