The invention discloses an in-vitro high-
throughput inoculation method of
tea tree colletotrichum gloeosporioides, and belongs to the technical field of
tea tree biology. The method comprises the following steps: preparing a
spore suspension with the
concentration gradient (
absorbance) OD600 of 0.4, 0.6, 0.8 and 1.0 from the
Colletotrichum calamiae, inoculating the
spore suspension to an in-vitro
tea tree leaf in an injection manner, then placing the leaf in a high-
throughput microenvironment culture device, and culturing for 4 days under the conditions of 28 DEG C, 75%
relative humidity and 12h illumination / 12h
darkness, thereby accurately evaluating the infection efficiency of the
pathogenic bacteria. According to the method, a standardized
inoculation process is combined with high-
throughput microenvironment control, so that the problems of long period, non-uniform environment and low efficiency of a traditional method are solved, and the method has the outstanding advantages of short
inoculation period, good experimental result consistency and capability of realizing batch high-throughput screening; the method can be directly used for tea tree
colletotrichum gloeosporioides resistance identification,
pathogen toxicity analysis and bactericide screening.
