Verticillium is a genus of fungi in the division Ascomycota, and are an anamorphic form of the family Plectosphaerellaceae. The genus used to include diverse groups comprising saprobes and parasites of higher plants, insects, nematodes, mollusc eggs, and other fungi, thus the genus used to have a wide-ranging group of taxa characterised by simple but ill-defined characters. The genus, currently thought to contain 51 species, may be broadly divided into three ecologically based groups - mycopathogens, entomopathogens, and plant pathogens and related saprotrophs. However, the genus has undergone recent revision into which most entomopathogenic and mycopathogenic isolates fall into a new group called Lecanicillium. The genus now includes the plant-pathogenic species V. dahliae, V. longisporum, V. albo-atrum, V. nubilum, and V. tricorpus.

The invention provides a method for detecting the resistance of cotton plants to greensickness by molecular marker-assisted selection. The method comprises the following steps: (1), soaking seeds of cotton plants to be detected in a soaking solution; (2), planting the soaked seeds in a disease garden; (3), selecting a cotton plant capable of surviving in the disease garden as a preliminary screened cotton plant; and (4), respectively carrying out PCR (Polymerase Chain Reaction) amplification on genome DNA (Desoxvribose Nucleic Acid) of the cotton plant by a first primer pair and a second primer pair to obtain a first amplification product and a second amplification product, wherein the first primer pair is shown as SEQ ID NO:1 and SEQ ID NO:2, the second primer pair is shown as SEQ ID NO:3 and SEQ ID NO:4, and if the first amplification product has nucleic acid shown as SEQ ID NO:5 and the second amplification product has nucleic acid shown as SEQ ID NO:6, the cotton plant to be detected is a greensickness-resistant plant. According to the method, the resistance to greensickness is predicted, the cotton plants are screened to discard plants which do not show symptoms but are susceptible in a disease garden screening process, the waste of manpower and material resources is reduced, and the breeding efficiency is increased.

Bacillus subtilis YB-04 and microorganism preparation thereof, and application of bacillus subtilis YB-04 in straw degradation

The present invention relates to a strain of Bacillus subtilis YB-04 and a preparation method of Bacillus subtilis YB-04 agent, wherein the preservation number of the Bacillus subtilis YB-04 is CGMCC NO.6497. According to the present invention, the Bacillus subtilis YB-04 provides a significant straw degradation effect, provides significant bacterial inhibition effects on wheat root take-all diseases, sheath blight and the like, and presents broad-spectrum bacterial inhibition effects, wherein a degradation rate can be 49% when the Bacillus subtilis YB-04 separately acts on straws, when the Bacillus subtilis YB-04 and fungi mixedly act on straws to achieve moistening and rotten surface of the straw, a degradation rate achieves the maximal value 59%, and a degradation effect of the experiment combination is far superior to a degradation effect of the commercially available bacterial agent; and the Bacillus subtilis YB-04 agent further presents broad-spectrum bacterial inhibition effects, wherein the Bacillus subtilis YB-04 agent provides good bacterial inhibition effects for cotton verticillium dahlia, wheat take-all pathogen, wheat sheath blight and other rhizosphere plant diseases, straws can be degraded, rhizosphere pathogenic bacteria can be inhibited, rhizosphere microorganism community can be regulated, and good development utilization prospects are provided.

Antagonistic actinomycete for preventing and controlling continuously cropped strawberry root rot and preparation method of biocontrol agent of antagonistic actinomycete

The invention discloses antagonistic actinomycete for preventing and controlling continuously cropped strawberry root rot and a preparation method of a biocontrol agent of the antagonistic actinomycete. Bacterial strain B04 belongs to streptomyces hygroscopicus (Streptomyces hygroscopicus), with the strain preservation number of CGMCC NO.9918. The biocontrol strain B04 is separated from a healthy strawberry root, and has a strong inhibiting effect on pathogenic bacteria, such as fusarium, verticillium dahliae, rhizoctonia, phytophthora and the like, causing strawberry soil-borne root diseases. The antagonistic actinomycete prepared by taking wormcast and wheat bran as the main materials of fermentation medium can completely meet the nutritional requirements produced by solid fermentation of biocontrol antinomycetes. Compared with conventional fermentation, the production cost of the preparation method is reduced by 30 to 50 percent, and the preparation method is beneficial for popularization and application of the biocontrol agent of the antagonistic actinomycete. After the biocontrol agent of the antagonistic actinomycete is applied to continuously cropped soil, the biological control rate reaches greater than 70 percent, and has great significance on preventing and controlling the soil-borne diseases of strawberry, improving the quality of agricultural products and protecting the environment.

The invention discloses a method for remarkably improving resistance of cotton to greensickness through verticillium dahlia interference VdRGS1 gene expression and belongs to the field of biological technique application. The invention provides a nucleotide sequence of a VdRGS1 gene and an ORF sequence and an amino acid sequence of cDNA. Function and phenotypic analysis shows that VdRGS1 is capable of regulating and controlling pathogenicity of verticillium dahlia, formation of microsclerotia, growth of spores and hyphae, and the like, and plays a significant role when verticillium dahlia invades and infect a host. By using a transgenic technique, a greensickness bacterium which can express green fluorescent protein (GFP) is created, by using a virus-induced gene silencing (VIGS) technique, a GFP gene of the transgenosis verticillium dahlia can be efficiently silenced in cotton, the result shows that host induced gene silencing (HIGS) is achieved on the basis of the VIGS technique, andinvading verticillium dahlia genes can be successfully silenced in cotton by using the technique. By using an HIGS technique based on VIGS, the verticillium dahlia VdRGS1 is silenced in cotton, and the resistance of cotton to verticillium dahlia is remarkably improved.

Verticillium lecanii quick-release dispersible tablet and preparation method thereof

The invention discloses a verticillium lecanii quick-release dispersible tablet and a preparation method thereof. Verticillium lecanii concentrated fermentation liquor or dry spore powder thereof is used as a main effective component. Synergist, surface active agent, protective agent, disintegrating agent, wetting agent, filler and lubricant are added as well. The preparation method is implementedby the following steps: 2/3 disintegrating agent and other auxiliary agents, together with verticillium lecanii, are added into a mixer for uniform mixing according to a certain proportion; then thewetting agent with an appropriate amount is added for mixing so that the mixture obtained from the previous step is changed into a wet material; the wet material then undergoes 20-mesh screening so asto be prepared into wet grains; after 24-48-hour drying at the normal temperature in a relatively germfree condition, the rest 1/3 disintegrating agent is added into the dry grains, and then the grains are weighed; the lubricant with a weight accounting for 0.5-0.6% of the grain weight is added for full and uniform mixing, and then the grains are sent to a tablet machine to be pressed into largetablets so that the finished product can be obtained. The verticillium lecanii quick-release dispersible tablet has simple production process and wide material source, is easy for industrial production, can be quickly disintegrated in water to release effective components, takes effect quickly, does not cause dust pollution, has a wide range of prevention and cure and can kill insect and bacteriostasis simultaneously.

Method for efficiently screening verticillium dahliae antagonistic bacteria from soil sample

The invention relates to a method for efficiently and rapidly screening verticillium dahliae antagonistic bacteria. The method comprises the following steps: firstly, coating a plate with soil suspensions of different gradients of a sample; directly spraying and inoculating verticillium dahliae spore liquid on the plate after bacteria grow; culturing continually; when verticillium dahliae hyphae grow on the plate fully, observing whether an inhibition zone is formed on the periphery of the bacteria or not, wherein if the inhibition zone is formed, the bacteria in the inhibition zone are antagonistic bacteria; picking out the antagonistic bacteria, and scribing and purifying the antagonistic bacteria; performing a plate antagonistic experiment on the antagonistic bacteria; further verifying the action of the antagonistic bacteria. By adopting the method, all bacteria growing from the coated plate do not need to be picked out one by one, so that the workload is reduced greatly compared with the conventional method, the screening efficiency is increased, the screening of the antagonistic bacteria is facilitated, and the development and utilization of biocontrol bacteria are accelerated; the method has important practical application value.
