Synthetic disease-resistant gene nlp27 and its application

This invention discloses the artificial synthesis of the disease-resistant gene NLP27 and its application, belonging to the field of plant genetic engineering technology. This invention selects a conserved 27-amino acid fragment from Verticillium dahliae NLP1, modifies its nucleotide sequence, and synthesizes the NLP27 gene, which is then overexpressed in potato plants through genetic transformation. Inoculation analysis shows that transgenic plants with the NLP27 gene exhibit high resistance to potato late blight and scab. Quantitative PCR analysis shows that excessively high NLP27 transcript levels significantly inhibit potato plant growth. After screening for expression levels and disease resistance, three transgenic lines with moderate NLP27 expression levels (such as PT1, PT6, and PT7) were obtained. These lines significantly enhanced resistance to potato late blight and scab without sacrificing yield.

The invention discloses a construction method of a novel composite biocontrol flora for preventing and treating cotton verticillium wilt, and relates to the technical field of biological prevention and treatment of agricultural plant diseases. The bacillus polymyxa and the bacillus cereus with the optimal antagonistic activity are respectively screened out through plate confrontation and buckling tests, and the bacillus pseudomycoides is obtained through screening by taking the growth promoting capability of dissolving phosphorus and producing indoleacetic acid as indexes. Through pairwise cross culture verification, the three strains are completely compatible and have no mutual inhibition effect. The method comprises the following steps: respectively and independently activating the three strains, carrying out enlarged culture, preparing single bacterial liquid with the OD600 value of 1.0, and uniformly mixing according to a volume ratio, so as to obtain the novel composite biocontrol flora for preventing and treating the cotton verticillium wilt. According to the invention, antagonistic bacillus and growth-promoting bacillus with complementary functions are screened, single bacteria are independently cultured and then are compounded according to a ratio of 1: 1: 1 to construct a composite flora, so that the synergistic interaction of efficient prevention and control of cotton verticillium wilt, cotton growth promotion and soil phosphorus solubilization is realized.
