Abstract
Description
Technical Field
[0001] The present disclosure relates to the field of cosmetics, and in particular to a composition having the function of activating skin cells to endogenously synthesize hyaluronic acid. Background Art
[0002] Hyaluronic acid, also known as hyaluronic acid, is an acidic mucopolysaccharide that is widely present in the skin, joints and other parts of the human body. It has many important functions and effects, including but not limited to:
[0003] (1) Moisturizing and locking in moisture: Hyaluronic acid has a strong ability to absorb water, and can absorb hundreds of times its own weight in water. It is one of the best moisturizing substances in nature. This property makes it an important ingredient in skin care products, which can effectively lock in skin moisture, prevent skin dryness, and keep the skin hydrated and shiny.
[0004] (2) Anti-aging: Hyaluronic acid not only moisturizes, but also promotes the regeneration and repair of skin cells, thus achieving an anti-aging effect. As we age, the amount of hyaluronic acid in the skin gradually decreases, causing the skin to sag and wrinkles to increase. Supplementing hyaluronic acid can increase skin elasticity, reduce wrinkles, and restore the skin to a youthful state.
[0005] (3) Filling and shaping: In the field of medical cosmetology, hyaluronic acid can be used for filling and shaping. By injecting hyaluronic acid into sunken areas or areas that need shaping, such as the bridge of the nose and chin, the filling effect can be quickly achieved, making the facial contour fuller and more three-dimensional.
[0006] (4) Promote wound healing: Hyaluronic acid can also promote wound healing. It can enhance the migration and proliferation of cells, promote angiogenesis and granulation tissue formation, thereby accelerating the wound healing process and reducing scar formation.
[0007] Therefore, supplementing the skin with hyaluronic acid is a common beauty and skin care method, and the supplementation methods usually include skin care product supplementation and medical beauty injection. However, for different skin types and under different environmental conditions, the skin's absorption efficiency of hyaluronic acid varies greatly, and there are many cases of insufficient absorption. The reasons for insufficient absorption include: (1) The integrity of the skin barrier. If the skin barrier is damaged, such as the stratum corneum is too thick or the skin is in a dry state, it may affect the penetration and absorption of hyaluronic acid. (2) The molecular weight of hyaluronic acid. Hyaluronic acid with a larger molecular weight may have difficulty penetrating the deep layers of the skin, while hyaluronic acid with a smaller molecular weight is more easily absorbed by the skin. (3) The method and frequency of use. If the method of use is improper, such as not sufficiently massaging to promote absorption, or the frequency of use is too low, it may lead to insufficient absorption of hyaluronic acid.
[0008] In order to avoid the problem of insufficient absorption of exogenously provided hyaluronic acid, it is urgent to develop a skin care product that can effectively promote skin cells to actively secrete endogenous hyaluronic acid. Summary of the invention
[0009] The purpose of the present disclosure is to provide a composition that can effectively promote the endogenous synthesis and secretion of hyaluronic acid by skin cells, thereby eliminating the absorption process of exogenous hyaluronic acid and solving the problem of low absorption rate of exogenous hyaluronic acid.
[0010] In order to solve the above technical problems and achieve the above objectives, the present disclosure provides the following technical solutions:
[0011] In one aspect of the present disclosure, a composition is provided, which can activate the endogenous synthesis of hyaluronic acid in skin cells, and the composition includes 0.05% to 3% (w / w) of a component inducing hyaluronic acid synthesis and 0.0001% to 2.5% (w / w) of a component activating mitochondrial autophagy; wherein the component inducing hyaluronic acid synthesis includes hydroxypropyl tetrahydropyrantriol and / or sodium polyglutamate, and the mitochondrial autophagy activating composition is selected from one or more of nicotinamide, nicotinamide adenine dinucleotide, carnosine and hexapeptide-11.
[0012] In an optional embodiment, the hyaluronic acid synthesis-inducing component is hydroxypropyl tetrahydropyrantriol and sodium polyglutamate. Optionally, the mass percentage of hydroxypropyl tetrahydropyrantriol in the composition is 1% to 3%, preferably 2%. Optionally, the molecular weight of sodium polyglutamate is 700kDa to 1200kDa, and the mass percentage of sodium polyglutamate in the composition is 0.01% to 1%, preferably 0.05%.
[0013] In an optional embodiment, the components for activating mitochondrial autophagy are nicotinamide, nicotinamide adenine dinucleotide, carnosine and hexapeptide-11. Optionally, the mass percentage of nicotinamide in the composition is 1% to 3%, preferably 2%. Optionally, the mass percentage of nicotinamide adenine dinucleotide in the composition is 0.1% to 1%, preferably 0.11%. Optionally, the mass percentage of carnosine in the composition is 0.1% to 0.5%, preferably 0.2%. Optionally, the mass percentage of hexapeptide-11 in the composition is 0.0001%.
[0014] In an optional embodiment, the composition contains, by mass percentage, 1% to 3% of hydroxypropyl tetrahydropyrantriol, 0.01% to 1% of sodium polyglutamate, 1% to 3% of nicotinamide, 0.1% to 1% of nicotinamide adenine dinucleotide, 0.1% to 0.5% of carnosine and 0.0001% of hexapeptide-11.
[0015] Preferably, the composition contains 2% hydroxypropyl tetrahydropyrantriol, 0.05% sodium polyglutamate, 2% nicotinamide, 0.11% nicotinamide adenine dinucleotide, 0.2% carnosine and 0.0001% hexapeptide-11 by mass percentage.
[0016] In an optional embodiment, the composition further contains one or more of a thickener, a stabilizer, an emulsifier, a humectant, a preservative and a chelating agent.
[0017] A second aspect of the present disclosure provides use of the composition described in any of the aforementioned embodiments in preparing a skin care product.
[0018] The inventors of the present disclosure unexpectedly discovered that a single type of hyaluronic acid synthesis-inducing component has a very limited promoting effect on the synthesis and secretion of hyaluronic acid by skin cells. However, when the components that induce hyaluronic acid synthesis are mixed with components that activate mitochondrial autophagy to form a composition, the activation effect of the synthesis and secretion of hyaluronic acid by skin cells can be significantly improved. The inventors infer that the hyaluronic acid synthesis signal pathway in skin cells should have an intersecting, overlapping or synergistic promotion relationship with the signal pathway that activates mitochondrial autophagy, so that the hyaluronic acid synthesis capacity of skin cells can be significantly improved by simply adding components that activate mitochondrial autophagy to the components that induce hyaluronic acid synthesis. [0025] Unless otherwise specified, the term "HaCaT detection model" used in this disclosure is an experimental model constructed based on HaCaT cells (human immortalized keratinocytes), which is widely used in biomedical research, drug screening, cosmetic testing and other fields. HaCaT cells are a human keratinocyte cell line that has been immortalized and has similar biological properties and functions to normal keratinocytes. These cells can stably proliferate in vitro and maintain high activity and metabolic levels, making them an ideal choice for constructing skin-related detection models. The HaCaT detection model can be used for the safety assessment of cosmetics. By co-culturing cosmetic ingredients with HaCaT cells and observing the response of the cells to the cosmetic ingredients, it is possible to preliminarily determine whether the cosmetics will cause adverse reactions such as skin irritation and allergies, as well as metabolite detection.
[0026] Unless otherwise specified, the term "composition" used in this disclosure refers to a high-concentration skin care product, which is generally thicker than lotions, emulsions, etc., contains rich nutrients and active substances, and is a highly effective skin care product with multiple functions such as deep nourishment, repair of damage, whitening and brightening, and anti-aging. When using, you need to pay attention to the appropriate amount, avoid contact with the eyes, and choose the product that suits you.
[0027] Unless otherwise specified, the term "hyaluronic acid synthesis inducing component" used in the present disclosure refers to any substance or component that can stimulate or promote the synthesis of hyaluronic acid (HA) in cells (especially skin cells, articular chondrocytes, etc.). These components may include but are not limited to: 1. Growth factors: such as fibroblast growth factor (FGF), vascular endothelial growth factor (VEGF), etc., which can promote cell proliferation and differentiation, thereby enhancing the synthesis capacity of hyaluronic acid. 2. Vitamins and coenzymes: Certain vitamins such as vitamin C (ascorbic acid) play a key role in the synthesis of hyaluronic acid, and participate in the biosynthesis of hyaluronic acid precursors - UDP-N-acetylglucosamine (UDP-GlcNAc) and UDP-glucuronic acid (UDP-GlcUA) as coenzyme factors. 3. Minerals: trace elements such as zinc and copper, which are cofactors of various enzyme activities and have an important influence on the synthesis process of hyaluronic acid. 4. Plant extracts: Many active ingredients from plants, such as green tea extract and licorice extract, have been found to promote the synthesis of hyaluronic acid or enhance the skin's moisturizing ability. 5. Amino acids, peptides and their salts: As the basic building blocks of proteins, certain amino acids and peptides (such as collagen peptides) can stimulate the synthesis of extracellular matrix, such as sodium glutamate and its polymers. 6. Other bioactive substances: such as polysaccharides, nucleic acid fragments, etc., may also promote the synthesis of hyaluronic acid or maintain its stability through different mechanisms, such as hydroxypropyl tetrahydropyrantriol.
[0028] Unless otherwise specified, the term "mitochondrial autophagy activation component" used in the present disclosure refers to a specific molecule or compound that can trigger or promote the mitochondrial autophagy process. Mitochondrial autophagy is a process in which cells selectively remove damaged or dysfunctional mitochondria through the autophagy mechanism, which is essential for maintaining cellular homeostasis and energy metabolism balance. There are many types of components that activate mitochondrial autophagy, and they participate in and promote the mitochondrial autophagy process through different mechanisms. The following are some of the main types of components that activate mitochondrial autophagy: 1. Signaling molecules and kinases; for example, PINK1 (PTEN Induced Kinase 1): When the mitochondrial membrane potential is impaired, PINK1 accumulates and activates on the outer membrane of mitochondria, and then recruits and activates Parkin to jointly initiate mitochondrial autophagy. Parkin: An E3 ubiquitin ligase that, after being phosphorylated and activated by PINK1, can ubiquitinate proteins on the outer membrane of mitochondria to promote the recognition and encapsulation of autophagosomes. ULK1 (Unc-51 Like Autophagy Activating Kinase 1): A key kinase for the initiation of autophagy, it participates in the formation of the ULK1 complex, which is activated under conditions such as nutrient deprivation to initiate the autophagy process. In addition, there are hexapeptide-11, etc. 2. Receptor proteins; for example, BNIP3 (Bcl-2 / Adenovirus E1B 19kDa Interacting Protein 3), BNIP3L (BNIP3-Like Protein), FUNDC1 (FUN14 Domain-Containing Protein 1), etc.: These mitochondrial autophagy receptor proteins are mostly located in the outer membrane of mitochondria, and bind to autophagosome membrane proteins such as LC3 through LIR (LC3-interacting region), mediating the recognition and encapsulation of mitochondria by autophagosomes. NDP52 (Nuclear Dot Protein 52), OPTN (Optineurin), etc.: These proteins, as autophagy receptors or adapters, also participate in the process of mitochondrial autophagy, and promote the formation of autophagosomes by recognizing and binding to ubiquitinated mitochondrial outer membrane proteins. 3. Regulatory proteins and molecules; for example, AMPK (AMP-Activated Protein Kinase): As an energy sensor, AMPK is activated under energy stress conditions, and then phosphorylates autophagy-related proteins such as ULK1, promoting the initiation of autophagy. mTOR (Mammalian Target of Rapamycin): A key protein that negatively regulates autophagy. It inhibits the activity of autophagy-related proteins such as ULK1 when nutrients are sufficient. When nutrients are deprived, mTOR activity decreases, relieving the inhibition of autophagy.Other small molecule compounds: Some small molecule compounds discovered in recent years, such as Rapamycin and its derivatives, can promote mitochondrial autophagy by inhibiting mTOR activity or activating other autophagy-related signaling pathways, as well as nicotinamide, nicotinamide adenine dinucleotide, etc. 4. Ubiquitination system; ubiquitination is an important part of the mitochondrial autophagy process. In addition to Parkin, there are other E3 ubiquitin ligases involved in the ubiquitination process of mitochondrial outer membrane proteins, providing markers for the recognition of autophagosomes, such as carnosine.
[0029] Unless otherwise specified, the term "thickener" used in this disclosure refers to an important component used to improve the texture, stability and feel of skin care products. There are many types of thickeners, each with its own unique characteristics and functions, such as xanthan gum, carbomer 940, silica gel, hydroxyethyl cellulose, etc.
[0030] Unless otherwise specified, the term "stabilizer" used in this disclosure refers to a class of chemical substances used to maintain stable product performance. Their main function is to prevent or slow down the changes in active ingredients, colors, fragrances, etc. in skin care products due to physical, chemical or microbiological factors, thereby ensuring the safety and effectiveness of the products. For example, antioxidants: vitamin E, tocopherol, tertiary butyl hydroxybenzoate, etc.; pH adjusters: citric acid, sodium hydroxide, potassium hydroxide, etc.
[0031] Unless otherwise specified, the term "emulsifier" as used in this disclosure refers to an agent in skin care products that can mix water and oil together to form a stable emulsion. This not only contributes to the texture and appearance of the product, but also ensures that the active ingredients are evenly distributed on the skin. Common emulsifiers include polyglyceryl-3 diisostearate, glyceryl oleate, etc.
[0032] Unless otherwise specified, the term "humectant" used in this disclosure refers to a chemical substance that can form a thin film on the skin surface to prevent water evaporation and thus maintain the moisture of the skin. Common moisturizers include glycerin, trehalose, collagen, etc. These moisturizers play a key role in skin care products and can improve problems such as dry and rough skin, making the skin softer, smoother and more elastic.
[0033] Unless otherwise specified, the term "preservative" used in this disclosure is an important component to ensure the stability of skin care products and extend the shelf life. They prevent skin care products from being contaminated and deteriorating during use by inhibiting or killing the growth of microorganisms such as bacteria, molds, and yeasts. For example, parabens (parabens): This is a widely used class of preservatives, including methyl parabens, ethyl parabens, propyl parabens, etc. Phenoxyethanol: Phenoxyethanol is a commonly used alcohol preservative with broad-spectrum antibacterial properties, which can effectively prevent the growth of bacteria, molds, and yeasts. Its use concentration is usually low, and the irritation is relatively small, making it a relatively safe preservative. Benzoic acid and its salts: Benzoic acid and its salts (such as sodium benzoate) are one of the traditional preservatives, which play a preservative role by inhibiting the respiratory enzyme system of microorganisms. Formaldehyde releasers: This type of preservative, such as diazolidinyl urea, DMDM hydantoin, etc., kills microorganisms by slowly releasing formaldehyde. Other new preservatives: such as plant extract preservatives (such as tea tree oil, aloe vera extract, etc.), biological preservatives (such as lysozyme, nisin, etc.), etc.
[0034] Unless otherwise specified, the term "chelating agent" used in the present disclosure is also called a complexing agent or a chelating ligand, which is an inorganic or organic substance that can combine with metal ions to form a stable complex. The chelate formed by this combination has a ring structure, and its stability is higher than that of general non-chelating coordination compounds. The most common coordinating atoms in the chelating agent are oxygen and nitrogen, and its structure is diverse, including straight chain, branched chain and ring structure. Skin care products often contain a variety of ingredients, including metal ions. Chelating agents can combine with these metal ions to prevent them from oxidation, decomposition and other reactions in skin care products, thereby maintaining the overall stability of skin care products. Common chelating agents include polyphosphates, aminocarboxylic acids, hydroxycarboxylic acids, etc. These chelating agents can not only effectively stabilize the skin care formula, but also bring additional skin care effects to skin care products through their unique chemical properties.
