Description

Technical Field

[0001] The present invention relates to a crystal form of a compound and a salt thereof, a preparation method and application thereof, and in particular to a protein arginine methyltransferase inhibitor compound and a crystal form of a salt thereof, a preparation method thereof and application thereof in preparing drugs for treating AAK1-mediated related diseases, and belongs to the field of pharmaceutical chemistry. Background Art

[0002] Connexin-associated kinase 1 (AAK1) is a member of the Ark1 / Prk1 family of serine / threonine kinases. AAK1 mRNA exists in two spliced forms, referred to as the short form and the long form. The long form is predominant and is highly expressed in the brain and heart. AAK1 is enriched in synaptosomal preparations and colocalizes with endocytic structures in cultured cells. AAK1 regulates endocytosis of clathrin-coated cells, an important process in synaptic vesicle recycling and receptor-mediated endocytosis. AAK1 binds to the AP2 complex, a heterotetramer that connects receptor cargo to the clathrin coating. Binding of clathrin to AAK1 stimulates AAK1 kinase activity. AAK1 phosphorylates the mu-2 subunit of AP-2, which promotes binding of mu-2 to the tyrosine-containing sorting motif on the cargo receptor. Mu2 phosphorylation is not essential for receptor uptake, but phosphorylation increases the efficiency of internalization.

[0003] AAK1 has been identified as an inhibitor of neuregulin-1 / ErbB4 signaling in PC12 cells. Loss of AAK1 expression via RNA interference-mediated gene silencing or treatment with the kinase inhibitor K252a (which inhibits AAK1 kinase activity) leads to enhanced neuregulin-1-induced neurite outgrowth. These treatments lead to increased ErbB4 expression and increased accumulation of ErbB4 in or near the plasma membrane. NRG1 and ErbB4 are putative schizophrenia susceptibility genes. SNPs in the two genes are associated with a variety of schizophrenia endophenotypes. Neuregulin 1 and ErbB4KO mouse models have shown morphological changes and behavioral phenotypes associated with schizophrenia. In addition, single nucleotide polymorphisms in the introns of the AAK1 gene are associated with the age of onset of Parkinson's disease. These results suggest that inhibition of AAK1 activity may be useful in treating schizophrenia, cognitive deficits in schizophrenia, Parkinson's disease, neuropathic pain, bipolar disorder, and Alzheimer's disease. Administration of therapeutically effective doses is problematic when used as a therapeutic agent in humans and may result in toxic side effects or ineffectiveness of treatment. Therefore, it is important to select a crystalline or salt form that is stable, reproducible, and has physicochemical properties that are favorable for its use as a therapeutic agent.

[0004] WO2023284838A1 discloses an AAK1 inhibitor and its use, which involves a compound of the following formula (I):

[0005]

[0006] When used as a therapeutic agent in humans, administration of therapeutically effective doses is problematic and can result in toxic side effects or therapeutic ineffectiveness. Therefore, it is important to select a crystalline form that is stable, reproducible, and has physicochemical properties that are favorable for its use as a therapeutic agent. [0131] It can be understood that the expressions such as “preferably, ..., its X-ray powder diffraction pattern further has a characteristic diffraction peak at the following 2θ position” or “more preferably, ..., its X-ray powder diffraction pattern further has a characteristic diffraction peak at the following 2θ position” described in the present invention mean that on the basis of having a characteristic diffraction peak at the aforementioned 2θ position, there is further a characteristic diffraction peak at the “following 2θ position”.

[0132] The X-ray powder diffraction or DSC graph, TGA graph disclosed in the present invention and those substantially the same also belong to the scope of the present invention.

[0133] Unless stated to the contrary, the terms used in the specification and claims have the following meanings.

[0134] As used herein, "the crystal of the present invention", "the crystalline form of the present invention", "the polymorph of the present invention" and the like may be used interchangeably.

[0135] The "room temperature" mentioned in the present invention generally refers to 4-30°C, preferably refers to 20±5°C.

[0136] The crystalline structure of the present invention can be analyzed using various analytical techniques known to those of ordinary skill in the art, including but not limited to, X-ray powder diffraction (XRD), differential scanning calorimetry (DSC) and / or thermogravimetric analysis (TGA), also known as thermogravimetry (TG).
