Description

Technical Field

[0001] The invention relates to the technical field of drug synthesis and analysis, and in particular to a detection method and a preparation method of nitisinone impurities. Background Art

[0002] Nitisinone is a synthetic reversible inhibitor of 4-hydroxyphenylpyruvate dioxygenase, mainly used for the treatment of rare pediatric hereditary tyrosinemia type I (HT-1). By inhibiting the normal metabolism of tyrosine in HT-1 patients, it can prevent the accumulation of highly toxic maleylacetoacetate, fumarylacetoacetate and its bypass metabolite succinylacetone, thereby alleviating liver and kidney damage, relieving symptoms, significantly improving long-term prognosis, increasing survival rate, and reducing the incidence of hepatocellular carcinoma. It is suitable for hereditary tyrosinemia type I (HT-1) and can assist in dietary restriction of tyrosine and phenylalanine.

[0003] Any substance that affects the purity of a drug is called an impurity. Drugs may produce different impurity spectra under different process conditions. The control of impurities in drugs is an important aspect of drug research and development, and is also an important indicator for clinical toxicity research. By detecting impurities, we can find out the source, nature, detection method and limit of impurities in drugs, optimize the drug manufacturing process, and then avoid the generation of impurities or reduce impurities to a minimum, thus ensuring and improving drug quality from many aspects.

[0004] Nitisinone APIs will produce byproducts or other degradation products during the synthesis process, storage and transportation. The types and contents of impurities have an important impact on the quality of Nitisinone. Therefore, the research on the preparation and detection methods of impurities in Nitisinone is of great significance. The impurity products prepared by the preparation method can be used for the qualitative and quantitative analysis of impurities in the production of Nitisinone APIs and preparations. The detection method can be used to clarify the source, nature, and limit of impurities in Nitisinone, thereby improving the quality standards of Nitisinone APIs and preparations, and providing guidance for people's drug safety.

[0005] At present, there are very few reports on the detection of impurities in nitisinone. [0168] In the system suitability spectrum of the comparative method, the separation degree between Nitisinone and impurity D is 0.7, which does not reach the baseline separation, the theoretical plate number is not high, the column efficiency is low, the response values of each impurity and the main peak are low, especially the response value of impurity F is lower than the detection limit. It is very likely that it will not be detected in the actual test, resulting in a false negative result, and the sensitivity is low.

[0169] Compared with the system applicability results of the comparative example, the method for detecting related substances of nitisinone provided by the present invention has the advantages of reliability, strong specificity, strong practicability, and each impurity can be effectively detected. Moreover, nitisinone and each impurity are well separated, and a variety of impurities can be detected.
