Abstract
Description
[Technical field]
[0001] The present invention relates to a biofilm disinfectant composition and a method for disinfecting a biofilm. [Background technology]
[0002] Once bacteria attach to a target object, the bacteria grow on the target object and form a biofilm. A biofilm is also called a biofilm or slime, and generally refers to a three-dimensional structure formed by microorganisms such as bacteria attaching to the surface of a material and growing, and enveloping the microorganisms with polymeric substances such as polysaccharides, proteins, and nucleic acids produced by the microorganisms. Biofilms are a problem for ordinary people in various situations. For example, they cause unpleasant slimy dirt and bad odors in household water environments such as the kitchen, sinks, and around the drains in the bathroom. Biofilms form not only in direct water environments, but also in scratches on cutting boards, inside dishwashing sponges, washing machine drums, and even in fabric products such as clothing, dish towels, dusters, and foot mats, and are prone to cause problems in situations where water, moisture, and dirt coexist. In addition, detergents containing various surfactants are widely used for cleaning textile products such as clothing. In recent years, as consumers have become more hygienic, there has been growing interest in bacteria and viruses attached to the surface of textile products. For this reason, consumers take extra time and cost to clean such products by soaking socks and sweaty clothes, which are prone to bacterial growth, in a cleaning solution for a while, or by using disinfectants or bleach in combination with textile cleaning agents.
[0003] US Patent No. 5,399,633 discloses a germicidal light-duty dishwashing detergent composition, either gel or liquid, containing a dioctyldimethylammonium salt and an alkyl ether carboxylate salt. [Prior art documents] [Patent documents]

[0005] Biofilms that adhere to target objects, particularly textile products, are extremely difficult to remove because the polymers that compose the biofilm act as a protective barrier that protects bacteria from various substrates, and it is also extremely difficult to sterilize the bacteria present within the biofilm. The present invention provides a biofilm disinfectant composition that has excellent disinfecting properties against bacteria present in a biofilm attached to a target article, particularly a textile product, and a method for disinfecting a biofilm using the same.
[0006] In the present invention, the biofilm disinfectant composition refers to a composition that has a disinfecting effect on bacteria present in a biofilm. In the present invention, the method for sterilizing a biofilm refers to a method for sterilizing bacteria present in a biofilm. In the present invention, biofilm sterilization refers to sterilization of bacteria present in a biofilm. In the present invention, the viewpoint of biofilm sterilization means the viewpoint of further enhancing the effect of sterilizing bacteria present in a biofilm. [Means for solving the problem]

R 1b -O-[(PO) p / (EO) q ]-SO3M (b1) [In general formula (b1), R 1b is a hydrocarbon group having 8 to 22 carbon atoms, PO is a propyleneoxy group, EO is an ethyleneoxy group, " / " is a symbol indicating that the bonding order of PO and EO does not matter, p is the average number of moles of PO added and is a number of 0 to 10, q is the average number of moles of EO added and is a number of 0 to 25, and M is a hydrogen ion, an alkali metal ion, an alkaline earth metal ion (1 / 2 atom), an ammonium ion, or an organic ammonium ion.
[0010] The present invention also relates to a method for sterilizing a biofilm, which comprises contacting a target article on which a biofilm exists with the biofilm disinfectant composition of the present invention or a treatment liquid obtained by diluting the composition with water.
[0011] The present invention also relates to a method for sterilizing a biofilm, which comprises contacting a treatment liquid containing the above-mentioned components (a) and (b) with an object on which a biofilm exists.
[0012] The present invention also relates to use of a composition containing the above-mentioned components (a) and (b) for disinfecting biofilms attached to textile products or hard articles. Effect of the Invention
[0013] According to the present invention, there is provided a biofilm disinfectant composition having excellent disinfecting properties against bacteria present in a biofilm attached to a target article, particularly a textile product, and a method for disinfecting a biofilm using the same. DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS
[0014] The reason why the biofilm disinfectant composition of the present invention and the biofilm disinfection method using the same have excellent disinfecting properties against bacteria present in biofilms attached to target objects, particularly textile products, is not entirely clear, but is presumed to be as follows. In the present invention, it is believed that the combination of a specific cationic surfactant (component (a)) and a specific anionic surfactant (component (b)) allows the base to penetrate into the biofilm and act to sterilize the biofilm. Specifically, the specific cationic surfactant (component (a)) and the specific anionic surfactant (component (b)) form a loose ion complex, which prevents the bactericidal component (a) from non-specifically adsorbing to the biofilm surface, and the ion complex of the components (a) and (b) allows the component (a) to penetrate into the biofilm. In addition, the penetrated ion complex has a moderate cationicity and therefore acts on bacteria, making sterilization possible. However, the present invention is not limited to the above-mentioned mechanism of action.
[0015] [Biofilm disinfectant composition] <Component (a)> The biofilm disinfectant composition of the present invention contains, as component (a), one or more compounds selected from (a1) a compound represented by the following general formula (a1) (hereinafter referred to as component (a1)) and (a2) a bispyridinium compound (hereinafter referred to as component (a2)).

[0030] The internal olefin sulfonate of component (b2) is a sulfonate obtained by sulfonating, neutralizing and hydrolyzing an internal olefin (an olefin having a double bond inside the olefin chain) having a carbon number of preferably 10 or more, more preferably 14 or more, and preferably 20 or less, more preferably 18 or less as a raw material. Such internal olefins include those containing a trace amount of so-called alpha olefins (hereinafter also referred to as α-olefins) in which the double bond is located at the first position of the carbon chain. When an internal olefin is sulfonated, β-sultone is quantitatively produced, and a part of the β-sultone is converted to γ-sultone and olefin sulfonic acid, which are further converted to hydroxyalkanesulfonate and olefin sulfonate in the neutralization and hydrolysis process (for example, J. Am.Oil Chem. Soc. 69, 39(1992)). Here, the hydroxy group of the obtained hydroxyalkanesulfonate is inside the alkane chain, and the double bond of the olefin sulfonate is inside the olefin chain. Furthermore, the obtained product is mainly a mixture of these, and some of the products may contain trace amounts of hydroxyalkanesulfonates having a hydroxy group at the end of the carbon chain or olefinsulfonates having a double bond at the end of the carbon chain. In this specification, these products and mixtures thereof are collectively referred to as internal olefin sulfonate (component (b2)). Furthermore, hydroxyalkanesulfonate is referred to as hydroxy form of internal olefin sulfonate (hereinafter also referred to as HAS form), and olefin sulfonate is referred to as olefin form of internal olefin sulfonate (hereinafter also referred to as IOS form). The mass ratio of the HAS form and the IOS form of the compound in component (b2) can be measured by a high performance liquid chromatography mass spectrometer (hereinafter abbreviated as HPLC-MS). Specifically, the mass ratio can be calculated from the HPLC-MS peak area of component (b2).

The content of each compound having a sulfonic acid group at a different position in the (b2) component can be measured by HPLC-MS. The content of each compound having a sulfonic acid group at a different position in this specification is determined as a mass ratio based on the HPLC-MS peak area of the compound having a sulfonic acid group at each position in the total HAS body of the (b2) component.
[0035] The content of olefin sulfonate in component (b2) in which a sulfonic acid group is present at position 1 is preferably 10 mass% or less in component (b2) from the viewpoint of biofilm sterilization, more preferably 7 mass% or less, even more preferably 5 mass% or less, and even more preferably 3 mass% or less, and from the viewpoint of reducing production costs and improving productivity, it is preferably 0.01 mass% or more. [0073] [Biofilm bactericidal activity evaluation] (1-1) Preparation of biofilm-formed fabric Glycerol stock solutions of Rhodotorula mucilaginosa (clothing isolate) and Methylobacterium variable (clothing isolate) were smeared on potato dextrose agar medium (Difco Laboratories) and cultured statically at 30°C for 2 days. Glycerol stock solutions of Moraxella osloensis (clothing isolate) were smeared on SCD agar medium (Shioya MS Co., Ltd. SCD agar medium "Daigo", for general bacterial testing) and cultured statically at 37°C for 1 day. After culture, the bacterial cells were scraped off with a disposable loop (As One Co., Ltd. Disposable Loop 10) and suspended in R2A medium (Shioya MS Co., Ltd. R2A medium "Daigo"). The turbidity (OD600) of each bacterial suspension was measured using a spectrophotometer (Model: PD-303S, manufactured by APEL Co., Ltd.), and the bacterial suspension was diluted so that R. mucilaginosa had an OD600 of 0.2, and M. variable and M. osloensis had an OD600 of 0.01. Hiraori cotton cloth (Tanigasashira Shoten, Cotton 2003) cut into 3 x 3 cm pieces was sterilized by pressure in an autoclave (PHC Corporation, high pressure steam sterilizer, Lab Autoclave, Model: MLS-3781). The sterilized Hiraori cotton cloth was placed in a No. 6 screw tube (Maruem Corporation, Screw Tube Bottle No. 6), and 1 mL of each of the above three bacterial strains and 7 mL of R2A medium were added. The tube was then sealed with a lid and cultured at 30°C and 200 rpm for 2 days in a small constant temperature shaking incubator, Bioshaker (TAITEC Corporation, Model: BR-23FP). After the culture, the Hiraori cotton cloth was transferred to a new No. 6 screw tube, and 10 mL of autoclave-sterilized ion-exchanged water was added. The tube was washed by shaking at 25°C and 150 rpm for 3 minutes in a small constant temperature shaking incubator, Bioshaker. After washing, the Hiraori cotton fabric was air-dried in a safety cabinet for 1 hour to prepare a biofilm-formed fabric. Biofilm formation on the fabric was confirmed by loosening the fabric into threads with tweezers, staining the threads, and observing them with a confocal laser scanning microscope (LSM880 ZEN, Carl Zeiss, Inc.). The threads observed were stained for 30 minutes with a live cell staining dye (C375-Cellstain(TM)-CFSE, Dojindo Laboratories, Inc.), a dead cell staining dye (P378-Cellstain(TM)-PI solution, Dojindo Laboratories, Inc.), a cellulose staining dye (Calcofluor White staining solution, Sigma-Aldrich, Inc.), a sugar staining dye (Lectin PNA From Arachis hypogaea (peanut), Alexa Fluor(TM) 647 Conjugate, Thermo Fisher Scientific, Inc.), and a sugar staining dye (Concanavalin Alexa Fluor(TM) 647 Conjugate, Thermo Fisher Scientific, Inc.).
[0074] (1-2) Preparation of biofilm-supported hard surfaces For Example 31 in Table 5, 0.3 mL of each of the above three bacterial species and 2.1 mL of R2A medium were added per well of a polystyrene 6-well plate (Corning Co., Ltd. Falcon Cell Culture 6-well multi-well plate, flat bottom, with lid), the plate was sealed with a lid, and then cultured at 30° C. for 2 days. After culture, the moisture in the upper layer of the 6-well plate was sucked off and air-dried in a safety cabinet for 1 hour to form a hard surface for biofilm formation. The formation of biofilms on hard surfaces was confirmed by observation using a digital microscope (Keyence Corporation, VHX-7000).
[0075] (2) Biofilm sterilization test (Biofilm sterilization evaluation) The prepared biofilm-formed cloth was cut to 1.5 x 1.5 cm, and the sterilized flat-woven cotton cloth was cut to 1.5 x 3 cm. One cut biofilm-formed cloth and two sterilized flat-woven cloths were stacked together, for a total of three pieces, and placed in a 5 mL tube (Eppendorf Tube 5.0 mL, model number: 0030 119.401, manufactured by Eppendorf Co., Ltd.). There, 3 mL of a test treatment solution prepared by diluting each biofilm disinfectant composition in Tables 1 to 7 with model tap water (4 ° dH) 3000 times (in Table 4, for Example 29, the test treatment solution diluted to the concentration shown in the table was used, and for Example 30, the test treatment solution was used without dilution at the concentration shown in the table. For Examples 32, 33, and 34 in Table 6, the hardness of the model tap water for dilution 3000 times was changed to that shown in the table to prepare a test treatment solution.) was added, and shaking washing was performed for 10 minutes at 25 ° C. and 150 rpm in a small constant temperature shaking incubator bioshaker, which was used as a washing step. The washed cloth was transferred to a 5 mL tube containing 3 mL of model tap water, and shaking washing was performed for 3 minutes under the same conditions, which was used as a rinsing step. After alternately repeating this washing and rinsing step four times, only the cloth with the biofilm formed was taken out and air-dried for 30 minutes in a safety cabinet. For the biofilm-forming hard surface, 3 mL of the test treatment solution obtained by diluting the biofilm disinfecting composition of Example 31 in Table 5 30,000 times with model tap water (4 ° dH) was added per well, and shaking washing was performed for 10 minutes at 25 ° C. and 150 rpm in a small constant temperature shaking incubator bioshaker, which was used as the washing step. The test treatment solution in the bioshaker after washing was sucked up with a micropipette (Eppendorf Research (R) plus manufactured by Eppendorf Co., Ltd.), 3 mL of model tap water was added, and shaking washing was performed for 3 minutes under the same conditions, which was used as the rinsing step. After alternately repeating this washing and rinsing step four times, the model tap water in the bioshaker after rinsing was sucked up with a micropipette, and the biofilm-forming hard surface after rinsing was air-dried for 30 minutes in a safety cabinet.
[0076] (Residual viable bacteria count measurement) The biofilm-formed cloth after air drying and the biofilm-formed cloth before contact with the test treatment liquid were each placed in a 1.5 mL tube (Eppendorf Tube 1.5 mL, model number: 0030 125.150, manufactured by Eppendorf Co., Ltd.) containing 1 mL of LP dilution liquid (LP dilution liquid "Daigo" manufactured by Shioya MS Co., Ltd.). With the lid on, the cloth was placed in an ultrasonic cleaner (Emerson Japan Co., Ltd., tabletop ultrasonic cleaner, model number: 2510JDTH) for 15 minutes to disperse the bacteria remaining on the cloth in the LP dilution liquid. After stirring the liquid for 3 seconds with a vortex (Vortex Mixer, model number: VTX-3000L, manufactured by AS ONE Co., Ltd.), the bacteria liquid was diluted with LP dilution liquid for 10 min. The solution was serially diluted 1:1 and dispensed into a petri dish (Aznol Petri dish, 90 mm diameter, 15 mm height, manufactured by AS ONE Corporation) at 100 μL each. Approximately 15 mL of SCD agar medium was added, the lid was put on, the mixture was lightly mixed, and the mixture was left to stand at room temperature until the agar solidified. After one day of static culture at 37°C, the number of remaining viable M. variable bacteria was counted using a colony counter pen (Colony counter pen type, manufactured by AS ONE Corporation, model number: 2-5764-01). For the biofilm-formed hard surfaces, 3 mL of LP dilution solution was placed per well on the air-dried biofilm-formed hard surfaces and on the biofilm-formed hard surfaces before contact with the test treatment solution. The biofilm was detached from the wells by pipetting 20 times with a micropipette and dispersed in the LP dilution solution. This bacterial solution was serially diluted 10-fold with the LP dilution solution, and the number of remaining viable M. variable bacteria was counted using the same method as above. (Method for calculating biofilm bactericidal activity value) The biofilm bactericidal activity value was calculated using the following formula. The results are shown in 1 to 7. The higher the bactericidal activity value, the better the biofilm bactericidal activity. Biofilm bactericidal activity value = LogA-LogB A: Average number of remaining viable bacteria before contact with the test treatment liquid B: Average number of remaining viable bacteria after contact with the test treatment liquid
