Description

[Technical field]

[0001] The present invention relates to the preparation of crystalline forms of DOPA decarboxylase inhibitors. [Background technology]

[0002] Technical level: 2-Amino-3-hydroxy-N'-(2,3,4-trihydroxybenzyl)propanehydrazide hydrochloride is a compound represented by the formula (I) [ka] It is also known as benserazide hydrochloride of formula (I) and is used in combination with L-DOPA to treat Parkinson's disease. Benserazide hydrochloride of formula (I) inhibits the conversion of L-DOPA to dopamine in extracerebral tissues, and thus can suppress the occurrence of extracerebral side effects.

[0003] Benserazide hydrochloride of formula (I) can be prepared starting from DL-serine as described in WO 02 / 04391 (Scheme 1). Reaction of 2,3,4-trihydroxybenzaldehyde with DL-serinoidazide monohydrochloride in water, methanol, ethanol or mixtures thereof gives the dihydrate form of (DL)-2-amino-3-hydroxy-N-[(E)-(2,3,4-trihydroxyphenyl)methyleneamino]propanamide. If the reaction is carried out in an anhydrous solvent such as absolute ethanol, the anhydrous form of the hydrazone is obtained. Subsequent hydrogenation over Pd / C in methanol gives benserazide hydrochloride of formula (I). [ka]

[0004] Commercially available tablet formulations of benserazide hydrochloride of formula (I) contain the compound in crystalline form I. The XRPD of crystalline form I is reported in US Pat. No. 5,399,633. Patent document 2 describes additional crystalline forms of benserazide hydrochloride of formula (I), such as crystalline form VI or a DMF solvate named crystalline form IX, obtained by recrystallization from an aqueous solution of ethanol (90%) and 1-propanol. The DMF solvate crystalline form IX is not a suitable crystalline form for preparing pharmaceutical preparations. According to the ICH guidelines on residual solvents present in active pharmaceutical ingredients, DMF is a class 2 solvent. US Patent No. 5,999,633 also describes the preparation of crystalline Form I by treating the DMF solvate with a mixture of aqueous ethanol (90%) and 1-propanol. The resulting product is washed with 1-propanol until no DMF is detected by HPLC analysis. However, HPLC analysis is not suitable for finding traces of DMF.

[0005] For example, Non-Patent Document 1 developed an HPLC method for measuring DMF and reported a detection limit of 0.019% (Non-Patent Document 1).

[0006] Furthermore, in light of the recently published EU regulation, EU2021 / 2030, legislators are seeking to minimize the use of DMF and there is a need to identify DMF-free methods. As a result, due to the toxicity of DMF, there is a need for alternative methods for the preparation of crystalline form I of benserazide hydrochloride that are efficient, cost-effective, and suitable for industrial-scale production and purification. [Brief description of the drawings]

[0012] Brief description of the drawings and analytical methods: The crystalline forms I, VI and α of benserazide hydrochloride of formula (I) were characterized by X-ray powder diffraction (XRPD). The water content was determined by titration according to the Karl Fischer method. X-ray diffraction spectra (XRPD) were performed using a Bruker D8 Advance diffractometer with a goniometer radius of 280 mm under the following conditions: CuKα radiation was filtered with a nickel filter (λ = 1.54 Å) and scanned in Bragg-Brentano geometry over the angular range 3–40° in 2θ with an angular pitch of 0.02°. The obtained diffractograms were evaluated using Diffrac.Eva version 4.3.0.1 (Bruker AXS).

[0087] The following examples further illustrate the invention.

[0088] Example 1. Preparation of crystalline form I of benserazide hydrochloride of formula (I): 20 g (61 mmol) of 2-amino-3-hydroxy-N-[(E)-(2,3,4-trihydroxyphenyl)methyleneamino]propenamide, which can be prepared by treating 2,3,4-trihydroxybenzaldehyde with DL-serinoidazide monohydrochloride as described in US Pat. No. 5,999,363, and 1.3 g of Pd / C in 460 mL of methanol are placed in a 500 mL four-neck flask equipped with a magnetic stirrer at 25° C. The mixture is then vigorously stirred under a hydrogen atmosphere at ambient conditions for 6 hours. The suspension is then cooled to 5° C. and filtered through Celite. The panel is washed with 20 mL of methanol and the combined solution is concentrated by vacuum distillation until a pale orange oily residue is obtained. 100 mL of ethanol is added to the oily residue at 25 °C and the pale orange mixture is stirred at an internal temperature of 25 °C. After about 30-60 min, an off-white solid is formed. The mixture is stirred protected from light for 6 h and then the solid is filtered. The panel is washed twice with 20 mL of ethanol and the collected white / off-white solid is dried in an oven at 45 °C for 8 h to give 13.4 g (75% yield) of benserazide hydrochloride of formula (I) in crystalline form I. The residual methanol amount measured by GC-HS was 2500 ppm.

[0089] Example 2. No. 5,999,363, and 3 g of Pd / C in 460 mL of methanol are charged at 25° C. in a 1000 mL four-neck flask equipped with a magnetic stirrer. 40 mL of water is added to the residue at 25 °C and 40 mL of isopropanol is added within 30 min. The clear pale yellow solution is stirred at 25 °C for 16 h. Optionally, 0.02 g of seeds of crystalline form VI of benserazide hydrochloride of formula (I) may be added to the solution. The suspension is cooled to 3 °C at a rate of 10 °C per hour and kept at 3 °C for 2 h. After stirring at 20-25 °C for another 4 hours, the product obtained is filtered. The panel is washed twice with 30 mL of isopropanol and the white / off-white solid recovered is dried under vacuum at 45 °C for 16 hours to obtain 24 g (80% yield) of crystalline form α of benserazide hydrochloride of formula (I) with a water content of 0.89% and a purity of 99.24% (HPLC detector at 210 nm) as determined by Karl Fischer analysis. Residual methanol content measured by GC-HS is 4.5 ppm.

[0093] Example 6. Crystalline form α of benserazide hydrochloride of formula (I): Crystalline form α of benserazide hydrochloride of formula (I), prepared as described in Example 4 or 5, does not convert to another polymorph when suspended in a mixture of ethanol and isopropanol (1:1, v:v) or in isopropanol according to the conditions described in Example 3.
