Description

[Technical field]

[0001] The present invention relates to an assay device configured to perform an assay using a liquid. [Background technology]

[0002] Assay devices including microchannels are used mainly in the fields of biology, chemistry, etc., when tests, experiments, assays, etc. are performed using minute amounts of liquid such as reagents, processing chemicals, etc. on the order of μl (microliter), i.e., about 1 μl or more and less than about 1 ml (milliliter). In recent years, lateral flow type assay devices, flow-through type assay devices, etc. have come to be used for such assay devices in order to improve cost, operability, durability, and liquid control performance.

[0003] In particular, the lateral flow type assay device is configured to move and manipulate liquids by utilizing the capillary phenomenon of hydrophilic porous media such as paper, cellulose membranes, etc., and is therefore simple. Therefore, the lateral flow type assay device can be produced at low cost, does not require an external mechanism such as a pump, does not require complicated operations, and can have improved durability. The lateral flow type assay device is particularly used when detecting or quantifying the concentration of an antibody or antigen contained in a sample by ELISA (Enzyme-Linked Immuno Sorbent Assay), immunochromatography, etc. In such an assay device, it is desirable to improve the liquid control performance.

[0004] An example of an assay device capable of improving the liquid control performance is an assay device comprising a microchannel through which a liquid can flow, a porous medium spaced apart from one end of the microchannel located on one side of the liquid flow direction, a separation space between the one end of the microchannel and the porous medium, and a peripheral wall defining the separation space together with the porous medium, in which an air vent configured to allow air to flow is provided in the peripheral wall, and the liquid supplied to pass through the microchannel can be separated across the separation space into a portion absorbed by the porous medium and another portion retained in the microchannel. In one example of this assay device, the assay device is configured using a laminated structure in which multiple layer members are stacked. (See, for example, Patent Document 1.) [Prior art documents] [Patent documents]

[0005] [Patent Document 1] International Publication No. 2020 / 045551 Summary of the Invention [Problem to be solved by the invention]

[0006] However, in the example of the assay device, the laminated structure is formed by stacking multiple layer members, so it is difficult to improve the shape precision. In particular, when a large number of assay devices are produced, the manufacturing variation of these assay devices becomes large. In addition, in the example of the assay device, the rigidity of the part composed of the layer members is low, so it is easily deformed.

[0007] For example, when a member such as a porous medium of an assay device is supported by a portion formed of a layer member, deformation of this portion may cause the member such as the porous medium to be displaced, particularly in the direction of liquid flow along the microchannel, which may result in a decrease in the positioning accuracy of the member such as the porous medium, and therefore, in one example of the assay device, it may not be possible to maintain a high measurement accuracy of the assay device.

[0008] In view of these circumstances, it is desirable to suppress manufacturing variations in an assay device, maintain high measurement accuracy of the assay device, and improve the liquid control performance. [Means for solving the problem]

[0009] In order to solve the above problem, one embodiment of an assay device comprises a microchannel configured to allow a liquid to flow therethrough, an absorbent porous medium spaced apart from one end of the microchannel located on one side of the liquid flow direction, a separation space located between the one end of the microchannel and the absorbent porous medium, and a storage space connected to the separation space in the flow direction and storing the absorbent porous medium, the assay device further comprises a lower member which is an integrally molded product located on the lower side in the height direction of the assay device and which constitutes a part of the assay device, the lower member defining a lower part in the height direction of the microchannel, a lower part in the height direction of the separation space, and a lower part in the height direction of the storage space, the lower parts of the separation space and the storage space being inclined downward from the other side of the liquid flow direction toward the same side, and the lower member supports the absorbent porous medium at the lower part of the storage space. Effect of the Invention

[0010] In the assay device according to one embodiment, manufacturing variations can be suppressed, the measurement accuracy of the assay device can be maintained high, and the liquid control performance can be improved. [Brief description of the drawings]

DETAILED DESCRIPTION OF THE PREFERRED EMBODIMENTS

[0012] An assay device according to an embodiment will be described. The assay device according to this embodiment is configured to perform an assay using a liquid. There is no particular limitation on the liquid that can be applied to the assay device according to this embodiment, as long as it can flow through the assay device. The liquid that can be applied to the assay device can include not only chemically pure liquids, but also liquids in which gases, other liquids, or solids are dissolved, dispersed, or suspended.

[0013] For example, the liquid may be hydrophilic, and examples of hydrophilic liquids include liquid samples derived from living organisms, such as whole blood, serum, plasma, blood cells, urine, diluted feces, saliva, sweat, tears, nail extracts, skin extracts, hair extracts, and cerebrospinal fluid of humans or animals. In addition, when the liquid is a reagent used in an assay, the liquid may include a buffer solution, general biochemical reagents, immunochemical reagents, antibody-related reagents, peptide solutions, protein and enzyme-related reagents, cell-related reagents, lipid-related reagents, natural products and organic compounds-related reagents, carbohydrate-related reagents, and the like. However, the liquid is not limited to these. In these cases, the assay device may measure specimens that are effective in clinical testing, diagnosis, or analysis in liquid samples for use in in vitro diagnostic drugs, general-use test drugs, POCT (Point of Care Testing), and the like for pregnancy tests, urine tests, stool tests, adult disease tests, allergy tests, infectious disease tests, drug tests, and cancer tests, but the use of the assay device is not particularly limited. In addition, hydrophilic liquids are not limited to biological samples, but may also include, for example, food suspensions, food extracts, washing water for production lines, wiping liquids, drinking water, river water, soil suspensions, etc. In this case, the assay device may measure pathogens in food or drinking water, or may measure contaminants in river water or soil. These hydrophilic liquids may typically be aqueous solutions that use water as a solvent and allow solution exchange by the assay device.

[0014] As used herein, "lateral flow" refers to a flow of liquid that moves due to gravitational sedimentation as a driving force. Liquid movement based on lateral flow refers to liquid movement in which the driving force of liquid due to gravitational sedimentation acts predominantly. In contrast, liquid movement based on capillary force (capillary phenomenon) refers to liquid movement in which interfacial tension acts predominantly. Liquid movement based on lateral flow is different from liquid movement based on capillary force.

[0015] As used herein, "analyte" refers to a compound or composition present in a liquid and to be detected or measured. For example, the analyte may include, but is not limited to, sugars (e.g., glucose), proteins or peptides (e.g., serum proteins, hormones, enzymes, immunomodulators, lymphokines, monokines, cytokines, glycoproteins, vaccine antigens, antibodies, growth factors, or proliferation factors), fats, amino acids, nucleic acids, cells, steroids, vitamins, pathogens or their antigens, natural or synthetic chemicals, pollutants, therapeutic or illegal drugs or poisons, or metabolites or antibodies of these substances. Note that the liquid may not contain the analyte or may not contain a detectable amount of the analyte.

[0016] As used herein, a "reference substance" is a known substance different from the analyte that is added in a known amount to a liquid for detection of the analyte concentration. The reference substance, like the analyte, can be selected from the above options and can be selected in relation to the analyte. In particular, it can be selected from substances that do not interact with the analyte and are stable.

[0017] As used herein, "microchannel" refers to a path configured to flow liquid within an assay device for detecting or measuring an analyte using a minute amount of liquid on the order of μl (microliter), i.e., about 0.1 μl or more and less than about 1 ml (milliliter), or for measuring such a minute amount of liquid.

[0018] In this specification, "film" refers to a film-like or plate-like object having a thickness of about 200 μm (micrometers) or less, and "sheet" refers to a film-like or plate-like object having a thickness of more than about 200 μm.

[0019] As used herein, "plastic" refers to a material that can be polymerized or that has been polymerized or molded to use a polymeric material as an essential component. Plastic also includes polymer alloys that combine two or more polymers.

[0020] In this specification, the term "porous medium" refers to a material having a plurality of micropores and capable of absorbing and passing liquid, or a material capable of capturing or concentrating solid matter, and includes paper, cellulose membrane, nonwoven fabric, glass fiber, polymer gel, plastic, and the like. For example, the porous medium may be hydrophilic when the liquid is hydrophilic, and may be hydrophobic when the liquid is hydrophobic. In particular, the porous medium may be hydrophilic and may be paper, absorbent cotton, and the like, which comprises a large number of fibers. Furthermore, the porous medium may be one or more of cellulose, nitrocellulose, cellulose acetate, filter paper, tissue paper, toilet paper, paper towel, fabric, cotton, or a hydrophilic porous polymer that is permeable to water.

[0021] "Outline of the assay device" The assay device according to this embodiment will be outlined with reference to Figures 1 to 7. That is, the assay device according to this embodiment is generally configured as follows: As shown in Figures 1 to 7, the assay device has at least one assay module 1 configured to perform an assay using a liquid (not shown).

[0022] In particular, in Figures 1, 3, and 5 to 7, an assay device having six assay modules 1 is shown as an example. However, the number of assay modules is not limited to this. The assay device may have one to five assay modules, or seven or more assay modules.

[0023] 4 to 7, the assay module 1 has a microchannel 2 configured to allow a liquid to flow. Hereinafter, the direction along which the liquid flows in such a microchannel 2 will be referred to as the "flow direction."

[0024] 1 to 4, 6, and 7, one side of the flow direction of the liquid is indicated by a single-sided arrow F1, and the other side of the flow direction of the liquid is indicated by a single-sided arrow F2. In this embodiment, the liquid flows from the other side to one side of the microchannel 2. Therefore, in some cases, one side of the flow direction is called the downstream side, and the other side of the flow direction is called the upstream side.

[0025] 4, 6, and 7, the assay module 1 has an absorbent porous medium 3 disposed at a distance from one end 2a of the microchannel 2 located on one side in the liquid flow direction, i.e., the downstream side. In the following, this absorbent porous medium 3 will be referred to as the first absorbent porous medium 3 as necessary.

[0026] The assay module 1 has a separation space 4 disposed between one end 2a of the microchannel 2 and the absorbent porous medium 3. The assay module 1 has a storage space 5 that stores the absorbent porous medium 3. The storage space 5 is connected to the separation space 4 in the flow direction. In the following, this storage space 5 will be referred to as a first storage space 5 as necessary.

[0027] As shown in Figures 1 to 6, the assay device has a lower member 20 that is located at the lower side in the height direction and is a component that constitutes part of the assay device. The lower member 20 is an integrally molded product. In Figures 2 to 5, the upper side in the height direction of the assay device is indicated by a single-sided arrow H1, and the lower side in the height direction of the assay device is indicated by a single-sided arrow H2. In this specification, the height direction refers to the height direction of the assay device, unless otherwise specified.

[0028] 4 to 6, the lower member 20 defines the lower portion 2b in the height direction of the microchannel 2. The lower member 20 defines the lower portion 4a in the height direction of the separation space 4. The lower member 20 defines the lower portion 5a in the height direction of the storage space 5. With reference to FIGS. 3 and 4, the lower portions 4a, 5a of the separation space 4 and the storage space 5 are inclined downward from the other side to the same side in the liquid flow direction. As shown in FIG. 4, the lower member 20 supports the absorbing porous medium 3 at the lower portion 5a of the storage space 5.

[0029] Further, the assay device according to this embodiment can be generally configured as follows. With reference to Figs. 1, 3, 4, 6, and 7, the assay module 1 of the assay device has an inlet 6 that allows a liquid to be injected into the microchannel 2. The inlet 6 is disposed at the other end 2c of the microchannel 2 located on the other side in the flow direction, i.e., the upstream end 2c. The assay module 1 has an inflow channel 7 that communicates the microchannel 2 and the inlet 6 in the flow direction. The lower member 20 defines a peripheral portion 6a of the inlet 6. The inlet channel 7 is formed to penetrate the peripheral portion 6a of the inlet 6.

[0030] As shown in Figs. 5 to 7, the assay module 1 has two side air passages 8 that allow air to circulate. The two side air passages 8 are adjacent to both side edges 2d in the width direction of the microchannel 2 so as to communicate with the microchannel 2. With reference to Figs. 3 to 7, the assay module 1 has two channel side walls 9 that protrude from the peripheral edge 6a of the inlet 6 along parts of both side edges 2d of the microchannel 2 in the flow direction. The lower member 20 defines the two channel side walls 9. The height of the two channel side walls 9 is approximately the same as the height of the microchannel 2.

[0031] The lower member 20 also defines outer side portions 8a in the width direction of the two lateral air passages 8 and both outer side portions 4b in the width direction of the separation space 4. In Fig. 1, Fig. 3, and Fig. 5 to Fig. 7, the width direction of the assay device is indicated by a double-sided arrow W. In this specification, unless otherwise specified, the width direction refers to the width direction of the assay device.

[0032] Assay device details 1 to 7, the assay device according to this embodiment can be specifically configured as follows. As shown in Fig. 1 to 7, the assay device is arranged so that the height direction is oriented vertically when in use. In this case, the upper and lower sides of the assay device face upward and downward in the vertical direction, respectively.

[0033] 3 to 7, in the assay module 1, an assay is performed in a state where the liquid flows in the microchannel 2, or in a state where the liquid is stationary or temporarily stopped in the microchannel 2. Typically, the concentration of a specimen in the liquid becomes detectable. In particular, as shown in Figs. 1, 3, and 5 to 7, when the assay device has a plurality of assay modules 1, the plurality of assay modules 1 are aligned in the width direction.

[0034] 5 to 7, the assay module 1 has a connecting air passage 10 that connects the two side air passages 8 and extends around the injection port 6. The connecting air passage 10 is configured to allow air to flow through it. Air flows through the two side air passages 8 and the connecting air passage 10, which are connected in series.

[0035] 3, 4, and 6, the assay module 1 has an assay region 11 located in a middle portion 2e of the microchannel 2 in the flow direction. A reagent that specifically binds to a sample in an assay is immobilized in the assay region 11. The assay module 1 has a confirmation region 12 arranged to be aligned with the assay region 11 in the flow direction. The confirmation region 12 is located downstream of the assay region 11.

[0036] The assay area 11 and the confirmation area 12 are separated from each other to such an extent that signals generated therein are distinguishable and detectable. The confirmation area 12 is configured to cause a known reaction (second reaction) that can be considered to have the same reaction time as a reaction (first reaction) occurring in the assay area 11. The assay module 1 has an assay window 13 and a confirmation window 14 formed so as to allow the assay area 11 and the confirmation area 12, respectively, to be confirmed from the outside.

[0037] The assay module 1 has a second absorbent porous medium 15 that is in contact with the first absorbent porous medium 3 in the height direction. The assay module 1 has a second storage space 16 capable of storing the second absorbent porous medium 15. The assay module 1 has an air hole 17 formed to allow air to circulate between the second storage space 16 and the outside of the assay device. With reference to Figs. 4 to 7, the microchannel 2, the separation space 4, the first storage space 5, the injection port 6, the inlet channel 7, the lateral air passage 8, the connecting air passage 10, the second storage space 16, and the air hole 17 are each a space defined by the assay device.

[0038] 1 to 5 and 7, the assay device has an upper member 30 which is a component that is located above the lower member 20 in the height direction and that constitutes a part of the assay device. The upper member 30 is an integrally molded product. The upper member 30 overlaps the lower member 20 from above.

[0039] The assay device has a cover member 40 which is a component that is located above the upper member 30 in the height direction and that constitutes a part of the assay device. The cover member 40 is an integrally molded product. The cover member 40 overlaps the upper member 30 from above.

[0040] "Details of the microchannel" 4 to 7, the microchannel 2 can be specifically configured as follows. As shown in FIGS. 4 and 5, the microchannel 2 is defined in the height direction between an upper portion 2f and a lower portion 2b of the microchannel 2 in the height direction. The height of the microchannel 2 is determined so as to generate an interfacial tension of the liquid that prevents the liquid from leaking into the side air passage 8 when the liquid flows through the microchannel 2. As an example, the height of the microchannel 2 is preferably about 1 μm or more and about 1000 μm or less (i.e., about 1 mm (millimeter)). However, the height of the microchannel is not limited to this.

[0041] Furthermore, the surfaces of the upper portion 2f and the lower portion 2b of the microchannel 2 that come into contact with the liquid are preferably hydrophilized. Such hydrophilization is an optical treatment such as plasma, or a treatment using a blocking agent that can prevent nonspecific binders from adsorbing to these surfaces when the liquid contains nonspecific binders, or includes at least one of these treatments. Examples of blocking agents include commercially available blocking agents such as Block Ace, bovine serum albumin, casein, skim milk, gelatin, surfactants, polyvinyl alcohol, globulin, serum (e.g., fetal bovine serum or normal rabbit serum), ethanol, MPC polymer, etc. Such blocking agents can be used alone or in combination of two or more.

[0042] 6 and 7, the microchannel 2 is defined in the width direction between the side edges 2d of the microchannel 2. The downstream end 2a of the microchannel 2 is formed in a tapered shape so that its width decreases from the upstream to the downstream in the flow direction. As an example, the width of the microchannel 2 is preferably not less than about 100 μm and not more than about 10,000 μm (i.e., about 1 cm (centimeter)). However, the width of the microchannel is not limited to this.

[0043] 4, 6, and 7, the microchannel 2 is defined in the flow direction between the separation space 4 and the inlet channel 7. The microchannel 2 extends substantially linearly in the flow direction. However, the microchannel may extend while being curved or bent.

[0044] As an example, the length in the flow direction of the microchannel 2 is preferably about 10 μm or more and about 10 cm or less. As a further example, the volume of the microchannel 2 is preferably about 0.1 μl or more and about 1000 μl or less, and more preferably about 1 μl or more and less than about 500 μl. However, the length in the flow direction and the volume of the microchannel are not limited thereto.

[0045] "Details of separation space" 4 to 7, the separation space 4 can be configured in detail as follows. As shown in Figs. 4, 6, and 7, the separation space 4 is connected to the microchannel 2 and two side air passages 8 located upstream of the separation space 4 in the flow direction. The two outer side portions 4b of the separation space 4 are connected to the outer side portions 8a of the two side air passages 8 in the flow direction, respectively. The downstream end of the separation space 4 located downstream in the flow direction is defined by the first absorbing porous medium 3.

[0046] As shown in Figs. 4 to 7, the separation space 4 has a flow path region 4c connected to the micro flow path 2 in the flow direction. The separation space 4 has two ventilation regions 4d connected to the two side ventilation paths 8 in the flow direction, respectively. The two ventilation regions 4d are adjacent to both side edges of the flow path region 4c in the width direction. The two ventilation regions 4d communicate with the flow path region 4c in the width direction. In the separation space 4, the upper end of the outer side portion 4b is located higher in the height direction than the upstream end of the flow path region 4c in the flow direction. For example, the distance between the upper end of the outer side portion 4b and the upstream end of the flow path region 4c in the height direction is about 5 mm. However, this distance is not limited to about 5 mm.

[0047] 4 to 6, the lower parts of the two ventilation regions 4d in the height direction are located lower in the height direction than the lower part of the flow path region 4c in the height direction. The lower parts of the two ventilation regions 4d are formed so as to be recessed downward in the height direction from the lower part of the flow path region 4c. The lower part 4a of the separation space 4 includes such lower parts of the flow path region 4c and the two ventilation regions 4d. With reference to FIGS. 3 and 4, the lower parts of the flow path region 4c and the two ventilation regions 4d are inclined downward from the upstream to the downstream in the flow direction. The inclination angle of each ventilation region 4d with respect to the horizontal direction is larger than the inclination angle of the flow path region 4c with respect to the horizontal direction. For example, the inclination angle of the flow path region 4c of the separation space 4 with respect to the horizontal direction can be about 5 degrees. However, this inclination angle is not limited to about 5 degrees.

[0048] 4, 5, and 7, the upper parts of the two ventilation regions 4d in the height direction are located higher in the height direction than the upper part of the channel region 4c in the height direction. The upper parts of the two ventilation regions 4d are formed so as to be recessed upward in the height direction from the upper part of the channel region 4c. The upper part 4e in the height direction of the separation space 4 includes such channel region 4c and the upper parts of the two ventilation regions 4d. The volume of the separation space 4 is larger than the volume of the microchannel 2. However, the volume of the separation space can be equal to or smaller than the volume of the microchannel. The upper and lower surfaces of the channel region 4c that come into contact with the liquid are preferably hydrophilically treated in the same manner as the surfaces of the upper part 2f and the lower part 2b of the microchannel 2.

[0049] As an example, the volume of the separation space 4 is preferably about 0.001 μl or more and about 10,000 μl or less. The ratio of the volume of the separation space 4 to the volume of the microchannel 2 is preferably about 0.01 or more. However, the volume of the separation space and the ratio of the volume of the separation space to the volume of the microchannel are not limited thereto.

[0050] "Details of the first and second absorbent porous media and the first and second containment spaces" 4, 6 and 7, the first and second absorbing porous media 3, 15 and the first and second containing spaces 5, 16 can be specifically configured as follows: The first absorbing porous medium 3 is configured to be able to absorb liquid from one end 2a of the microchannel 2. The first absorbing porous medium 3 is compressed between the upper part 5d and the lower part 5a of the first containing space 5. The first absorbing porous medium 3 also abuts against the outer side part 4b of the separation space 4 in the flow direction.

[0051] The second absorbent porous medium 15 is configured to be able to absorb liquid from the first absorbent porous medium 3. As shown in Figure 4, the second absorbent porous medium 15 is located vertically below the first absorbent porous medium 3. However, the second absorbent porous medium can also be located vertically above the first absorbent porous medium.

[0052] As shown in Figures 4, 6, and 7, the second storage space 16 is located downstream in the flow direction relative to the first storage space 5. The second storage space 16 is connected to the first storage space 5 in the flow direction. The first storage space 5 is capable of storing the upstream portion of the first absorbing porous medium 3 in the flow direction. The second storage space 16 is capable of storing the downstream portion of the first absorbing porous medium 3 in the flow direction and the entire second absorbing porous medium 15.

[0053] 4, 6, and 7, the first storage space 5 has a flow path region 5b that is connected in the flow direction to the flow path region 4c of the separation space 4. The first storage space 5 has two ventilation regions 5c that are respectively connected in the flow direction to the two ventilation regions 4d of the separation space 4. The two ventilation regions 5c are adjacent to both side edges in the width direction of the flow path region 5b. The two ventilation regions 5c communicate with the flow path region 5b in the width direction.

[0054] 4 and 6, the lower parts of the two ventilation regions 5c in the height direction are located lower in the height direction than the lower part of the flow path region 5b in the height direction. The lower parts of the two ventilation regions 5c are formed so as to be recessed downward in the height direction from the lower part of the flow path region 5b. The lower part 5a of the first storage space 5 includes the lower parts of such a flow path region 5b and the two ventilation regions 5c. With reference to FIGS. 3 and 4, the lower parts of the flow path region 5b and the two ventilation regions 5c are inclined downward from the upstream to the downstream in the flow direction. The inclination angle of each ventilation region 5c with respect to the horizontal direction is larger than the inclination angle of the flow path region 5b with respect to the horizontal direction. For example, the inclination angle of the flow path region 5b of the first storage space 5 with respect to the horizontal direction can be about 5 degrees. However, this inclination angle is not limited to about 5 degrees.

[0055] 4 and 7, the upper parts of the two ventilation regions 5c in the height direction are located higher in the height direction than the upper part of the flow path region 5b in the height direction. [0065] The immobilized reagent provided in the assay area 11 specifically reacts with the analyte in the liquid and, together with the added reagent, produces a result that is detectable for the analyte. The analyte detectable result may be observable by the naked eye, for example based on a color change, or the analyte detectable result may be detectable only by a spectrometer or other measurement means.

[0066] Furthermore, the immobilized reagent provided in the assay area 11 can be an enzyme, an antibody, an epitope, a nucleic acid, a cell, an aptamer, a peptide, a molecularly imprinted polymer, an adsorbent polymer, an adsorbent gel, a chemical substance such as iron (III) ions that react with a sample to produce a color, a color reagent, or any other substance that produces a detectable result by reacting with a sample. Typically, the immobilized reagent can be an antibody. The immobilized reagent can be immobilized in the assay area 11 by a well-known immobilization technique such as physical adsorption or chemical adsorption.

[0067] Any labeling substance such as a radioisotope, an enzyme, a gold colloid, a coloring agent, a colored molecule such as quantum dots or latex, a dye, an electrochemically reactive substance, a fluorescent substance, or a luminescent substance can be bound to the immobilized reagent in order to analyze or amplify a detection signal. Alternatively, such a labeling substance can be bound to an added reagent used to be added to the microchannel 2 in the assay step. Specifically, the immobilized reagent can be fixed to one or both of the upper portion 2f and the lower portion 2b that define the microchannel 2 in the height direction.

[0068] The confirmation region 12 is provided with an immobilized reagent that specifically binds to the reference substance. The immobilized reagent in the confirmation region 12 can also be an antibody, similar to the immobilized reagent in the assay region 11. Each of the lower member 20, the upper member 30, and the cover member 40 is an injection molded product. However, at least one of the lower member, the upper member, and the cover member can be a product other than an injection molded product. For example, at least one of the lower member, the upper member, and the cover member can be a three-dimensional molded product, a machined product, or the like.

[0071] Each of the lower member 20, the upper member 30, and the cover member 40 is made of plastic. For example, such plastics include polyolefins (PO) such as polyethylene (PE), high density polyethylene (HDPE), and polypropylene (PP), ABS resin (ABS), AS resin (SAN), polyvinylidene chloride (PVDC), polystyrene (PS), polyethylene terephthalate (PET), polyvinyl chloride (PVC), nylon, polymethyl methacrylate (PMMA), cycloolefin copolymer (COC), cycloolefin polymer (COP), polycarbonate (PC), polydimethylsiloxane (PDMS), polyacrylonitrile (PAN), and polylactic acid (PLA), or other biodegradable plastics or other polymers, or combinations thereof. However, at least one of the lower member, the upper member, and the cover member may be made of a material other than plastic, as long as the material is impermeable to fluids, and such a material other than plastic may be resin, glass, metal, or the like.

[0072] Referring to Figures 3 to 6, the lower member 20 defines the lower portions 2b of the microchannels 2 in the multiple assay modules 1, the lower portions 4a and outer side portions 4b of the separation spaces 4, the lower portions 5a of the first storage spaces 5, the peripheral portions 6a and lower portions 6b of the injection ports 6 including the inlet passages 7, the outer side portions 8a and lower portions 8b of the lateral air passages 8, the flow channel side walls 9, the lower portions 10a, inner peripheral portions 10c, and outer peripheral portions 10d of the connecting air passages 10, and the lower portions 16a of the second storage spaces 16, so as to form these continuously.

[0073] Each assay module 1 has a recess 1a that is provided upstream in the flow direction with respect to the second storage space 16 and is formed so as to be recessed from the lower end surface of the lower member 20. The recess 1a is located heightwise below the microchannel 2, separation space 4, first storage space 5, injection port 6, lateral air passage 8, and connecting air passage 10 of each assay module 1. The recesses 1a of the multiple assay modules 1 are formed so as to be connected to each other in the width direction by the lower member 20.

[0074] 3 to 5 and 7, the upper member 30 defines, so as to be continuous with, the upper portions 2f of the microchannels 2 in the multiple assay modules 1, the upper portions 4e of the separation spaces 4, the upper portions 5d of the first storage spaces 5, the upper portions 8c of the lateral air passages 8, the upper portions 10b of the connecting air passages 10, the upper portions 16b of the second storage spaces 16, and the peripheral portions 17a of the air holes 17. The upper member 30 is preferably transparent.

[0075] 1 to 4, the cover member 40, together with the lower member 20, also defines the periphery 6a of the inlet 6 and the periphery 17a of the vent hole 17 in the multiple assay modules 1. The vent hole 17 is formed to penetrate the upper member 30 and the cover member 40. The assay window 13 and the confirmation window 14 are formed to penetrate the cover member 40. The cover member 40 can be a detachable member of the assay device. Specifically, the cover member 40 can be detachably attached to the assembly of the lower member 20 and the upper member 30.

[0076] "Fluid Control of Assay Device" Fluid control of the assay device according to this embodiment will be described with reference to Figures 4 to 7. Here, the liquids applied to the assay device are referred to as first and second liquids (not shown), and these first and second liquids are supplied to the assay device in order. The first and second liquids are different. However, it is also possible for the first and second liquids to be the same.

[0077] Typically, the amount of each liquid (here, the amount of each of the first and second liquids) supplied to the assay device is about 1 μl or more and less than about 1 ml. Furthermore, the amount of each liquid is preferably about 1.5 μl or more, and more preferably about 3.0 μl or more. The upper limit of the amount of each liquid may be, for example, several μl to several hundred μl. Such an amount of each liquid can stabilize the detection sensitivity of the sample, etc., and facilitate the detection of the sample, etc. In this case, the amount of each liquid can be obtained by one drop of liquid.

[0078] Furthermore, the amount of each liquid may be greater than the capacity of the microchannel 2, in which case the liquid can be well separated across the separation space 4 into a portion absorbed by the absorbing porous medium 3 and another portion retained within the microchannel 2. However, the amount of each liquid may also be less than the capacity of the microchannel or substantially the same as the capacity of the microchannel.

[0079] First, a first liquid is supplied to the inlet 6. The first liquid flows into the microchannel 2 through the inlet 7. The first liquid then flows from the upstream side to the downstream side in the flow direction within the microchannel 2. As the first liquid flows through the microchannel 2 in this manner, an assay is carried out in the assay region 11, and a known reaction (second reaction) that can be considered to have the same reaction time as the reaction (first reaction) occurring in the assay region 11 occurs in the confirmation region 12.

[0080] Furthermore, when the supply of the first liquid continues, particularly when an amount of the first liquid exceeding the capacity of the microchannel 2 is supplied, the first liquid flowing through the microchannel 2 reaches the separation space 4. The first liquid passes through the separation space 4 and comes into contact with the absorbing porous medium 3. The first liquid is absorbed by the absorbing porous medium 3 until the supply of the first liquid is stopped. After the supply of the first liquid is stopped, the first liquid is separated, across the separation space 4, into a portion absorbed based on the capillary force of the absorbing porous medium 3 and another portion that is retained in the microchannel 2.

[0081] Next, after stopping the supply of the first liquid, the second liquid is further supplied to the injection port 6. The supplied second liquid flows in the microchannel 2 in the same manner as the first liquid. At this time, the second liquid pushes out the first liquid that was previously filled in the microchannel 2 into the separation space 4. As a result, a solution exchange is performed in the microchannel 2, in which the first liquid is replaced with the second liquid. Note that, when the second liquid flows in the microchannel 2, an assay is performed in the assay region 11, and a known reaction (second reaction) that can be considered to have the same reaction time as the reaction (first reaction) occurring in the assay region 11 occurs in the confirmation region 12.

[0082] Furthermore, when the supply of the second liquid is continued, particularly when an amount of the second liquid exceeding the amount of the first liquid previously filled in the microchannel 2 is supplied, the first liquid pushed out by the second liquid first passes through the separation space 4 and comes into contact with the absorbing porous medium 3, and then the second liquid, following the first liquid, passes through the separation space 4 and comes into contact with the absorbing porous medium 3. The second liquid flows in the same manner as the first liquid, and further, like the first liquid, the second liquid is separated into a portion absorbed based on the capillary force of the absorbing porous medium 3 and another portion retained in the microchannel 2, separated by the separation space 4.

[0083] The above-mentioned solution exchange can facilitate the occurrence of multi-stage antigen-antibody reactions in ELISA etc. In particular, when the amount of the second liquid L2 supplied to the assay device is substantially the same as the amount of the first liquid filling the microchannel 2 or is greater than the amount of the first liquid, solution exchange can be performed reliably.

[0084] In other words, in the assay device of this embodiment, when multiple liquids are supplied in sequence to the injection port 6, a preceding liquid which is one of the multiple liquids is pre-filled into the microchannel 2, the supply of the preceding liquid is stopped, and then another liquid which is one of the multiple liquids and which succeeds the preceding liquid is supplied to the injection port 6, thereby replacing the preceding liquid with the following liquid in the microchannel 2.

[0085] Furthermore, this liquid exchange of replacing the trailing liquid with the preceding liquid can be repeated. In this case, too, the preceding liquid and the following liquid are typically different. However, the preceding liquid and the following liquid can also be the same.

[0086] As described above, the assay device according to this embodiment is an assay device comprising a microchannel 2 configured to allow a liquid to flow therethrough, an absorbent porous medium 3 arranged at a distance from one end 2a of the microchannel 2 located on one side of the liquid flow direction, a separation space 4 arranged between the one end 2a of the microchannel 2 and the absorbent porous medium 3, and a storage space 5 connected to the separation space 4 in the flow direction and storing the absorbent porous medium 3. The assay device further comprises a lower member 20 which is an integrally molded product located on the lower side in the height direction of the assay device and which constitutes a part of the assay device, the lower member 20 defining a lower portion 2b in the height direction of the microchannel 2, a lower portion 4a in the height direction of the separation space 4, and a lower portion 5a in the height direction of the storage space 5, the lower portions 4a, 5a of the separation space 4 and the storage space 5 being inclined downward from the other side of the liquid flow direction toward the same side, and the lower member 20 supports the absorbent porous medium 3 at the lower portion 5a of the storage space 5.

[0087] In the assay device according to this embodiment, the liquid supplied through the microchannel 2 can be separated across the separation space 4 into a portion absorbed by the absorbing porous medium 3 and another portion retained in the microchannel 2, and this separation can particularly improve the measurement accuracy of the liquid retained in the microchannel 2 and further improve the liquid control performance. The integrally molded lower member 20 provided in such an assay device can be stably produced, for example, by injection molding using a mold, and therefore the rigidity of the integrally molded lower member 20 can be increased and the shape variation of the lower member 20 can be suppressed.

[0088] Therefore, the rigidity of the lower parts 2b, 4a, 5a of the microchannel 2, separation space 4, and storage space 5 defined by the lower member 20 can be increased, and as a result, deformation of the microchannel 2, separation space 4, and storage space 5 can be suppressed, and shape variations of the microchannel 2, separation space 4, and storage space 5 can be suppressed. Furthermore, since the absorbing porous medium 3 can be stably supported by the lower part 5a of the storage space 5, which can suppress deformation, positional deviation of the absorbing porous medium 3 can be suppressed, and as a result, the positioning accuracy of the absorbing porous medium 3 can be improved.

[0089] Since deformation of the microchannel 2, separation space 4, and storage space 5 used in the measurement of the assay device can be suppressed and manufacturing variations thereof can be suppressed, the shape accuracy of the microchannel 2, separation space 4, and storage space 5 as well as the measurement accuracy of the assay device performed using these can be maintained high, and further, the liquid control performance can be improved. Therefore, in the assay device according to this embodiment, manufacturing variations can be suppressed, measurement accuracy can be maintained high, and liquid control performance can be improved.

[0090] The assay device of this embodiment is provided with an injection port 6 that is disposed at the other end 2c of the microchannel 2 located on the other side of the flow direction and enables the liquid to be injected into the microchannel 2, and an inflow channel 7 that connects the microchannel 2 and the injection port 6 in the flow direction, and the lower member 20 defines a peripheral portion 6a of the injection port 6, and the lower member 20 defines the inflow channel 7 so as to penetrate the peripheral portion 6a of the injection port 6.

[0091] In such an assay device, the rigidity of the peripheral portion 6a of the injection port 6 defined by the lower member 20 of the integrally molded product can be increased, and as a result, deformation of the injection port 6 and the inflow channel 7 defined by this peripheral portion 6a can be suppressed, and variation in the shapes of the injection port 6 and the inflow channel 7 can be suppressed. Therefore, the shape accuracy of the path from the injection port 6 through the inflow channel 7 to the microchannel 2 can be maintained high, as well as the accuracy of the measurement of the assay device performed using this path, and further the liquid control performance can be improved.

[0092] The assay device of this embodiment comprises two lateral air passages 8 adjacent to both side edges 2d of the microchannel 2 in the width direction so as to communicate with the microchannel 2 and allow air to flow, and two channel side walls 9 protruding from the peripheral portion 6a of the injection port 6 along parts of both side edges 2d of the microchannel 2 in the flow direction, and the lower member 20 defines the two channel side walls 9, and the height of the two channel side walls 9 matches the height of the microchannel 2.

[0093] In the assay device according to this embodiment, the liquid in the microchannel 2 comes into contact with the air in the lateral air passage 8 in the width direction, and therefore the liquid can be prevented from coming into contact with the upper portion 2f and the lower portion 2b that define the microchannel 2 in the width direction. As a result, the possibility of non-specific adsorption of specimens, reagents, impurities, etc. occurring in the upper portion 2f and the lower portion 2b can be reduced, and the risk of impurities from the upper portion 2f and the lower portion 2b being mixed into the liquid can be reduced. In addition, the effects of viscosity and friction between the liquid in the microchannel 2 and the upper portion 2f and the lower portion 2b that define the microchannel 2 in the width direction can be avoided.

[0094] Furthermore, even if an air gap occurs in the liquid in the microchannel 2, the air gap can be released to the side air passage 8. Furthermore, gases such as nitrogen and oxygen in the side air passage 8 can be efficiently supplied to the liquid in the microchannel 2. As a result, the flow accuracy of the liquid can be improved. Therefore, the control performance of the liquid can be improved.

[0095] The two flow channel side walls 9 having high rigidity can increase the rigidity of the two side air passages 8 and the micro flow channel 2 around the injection port 6, which in turn can suppress deformation of the two side air passages 8 and the micro flow channel 2 and suppress variation in the shapes of the two side air passages 8 and the micro flow channel 2. This improves the accuracy of the shapes of the two side air passages 8 and the micro flow channel 2, and also improves the accuracy of the measurement of the assay device using these, and further improves the liquid control performance.

[0096] In the above assay device, the two flow channel side walls 9 can prevent the liquid immediately after flowing out from the inlet 6 into the micro flow channel 2 from flowing out from the micro flow channel 2 into the two lateral air passages 8 due to the momentum of the liquid. This can improve the liquid control performance.

[0097] In the assay device according to this embodiment, the lower member 20 defines outer side portions 8a in the width direction of the two lateral air passages 8 and two outer side portions 4b in the width direction of the separation space 4.

[0098] In such an assay device, the rigidity of the outer side parts 8a of the two side air passages 8 and the two outer side parts 4b of the separation space 4 defined by the lower member of the integrally molded product can be increased, and as a result, deformation of the two side air passages 8 and the separation space 4 can be suppressed, and variation in the shapes of the two side air passages 8 and the separation space 4 can be suppressed. Therefore, it is possible to maintain high accuracy in the shapes of the two side air passages 8 and the separation space 4, as well as the accuracy of the measurement of the assay device performed using these, and further improve the liquid control performance.

[0099] Although the embodiment of the present invention has been described above, the present invention is not limited to the above-mentioned embodiment, and the present invention can be modified and changed based on the technical concept thereof. [Explanation of symbols]
