Abstract
Description
Cosmetic composition for improving skin and pharmaceutical composition for preventing or treating skin damage containing honokiol and polyol
[0001] This patent application claims priority to Republic of Korea Patent Application No. 10-2023-0085480, filed with the Korean Intellectual Property Office on July 3, 2023, the disclosure of which is incorporated herein by reference.
[0002] The present invention relates to a cosmetic composition for improving skin and a pharmaceutical composition for preventing or treating skin damage, comprising honokiol and a polyol.
[0003] Aging is a functional, structural, and biochemical process that occurs continuously from birth to death. It occurs throughout the cells and tissues that make up the human body, and is characterized by a decrease in metabolic rate, an increase in disease, and a decrease in adaptability, ultimately leading to the death of cells and the entire body.
[0004] Among these, skin aging is being studied in two major ways: one is 'internal aging', which refers to the aging phenomenon due to the increase in age, and the other is 'external aging', which refers to the aging phenomenon caused by the external environment, such as ultraviolet rays or pollution.
[0005] There have been many theories proposed regarding skin aging, but the most scientific theory regarding skin aging is the theory of skin aging due to oxidation.
[0006] Human skin is composed of the epidermis, dermis, and connective tissue, including the stratum corneum. Among these, the stratum corneum is a layer of dead cells formed through the differentiation of keratinocytes, the basal cells of the epidermis, and is responsible for protecting the body from the influence of the external environment. In addition, the dermis layer, which exists inside the skin, is composed of collagen and elastin, which provides elasticity to the skin and prevents it from sagging. The antioxidant theory is the theory that collagen and elastin are damaged by free radicals generated by external factors such as ultraviolet rays, which reduces skin elasticity and forms wrinkles.
[0007] Until now, natural antioxidants such as vitamin E, vitamin C, carotenoids, glutathione, and plant extracts have been used to prevent skin aging, especially wrinkles. However, these substances have low effectiveness or cause serious side effects on the skin, such as skin irritation, redness, and inflammation.
[0008] As the trend of retinol products has recently increased, consumer demand has increased for products that combine the powerful wrinkle-improving effects of retinol with low irritation. However, due to limitations in improving side effects such as phototoxicity, the need for natural materials as substitutes for retinol with less irritation and superior efficacy has emerged.
[0009] The present inventors confirmed that honokiol and polyols have excellent skin elasticity enhancement, moisturizing improvement, barrier improvement, inflammation relief, and antioxidant activity.
[0010] Accordingly, the purpose of the present invention is to provide a cosmetic composition for improving skin comprising honokiol and polyol.
[0011] Another object of the present invention is to provide a pharmaceutical composition for preventing or treating skin damage comprising honokiol and polyol.
[0012] Another object of the present invention relates to the use of honokiol and polyol for improving skin and preventing or treating skin photoaging caused by exposure to ultraviolet rays.
[0013] The present invention relates to a cosmetic composition for improving skin and a pharmaceutical composition for preventing or treating skin damage, comprising honokiol, an ingredient derived from magnolia tree bark, and a polyol. According to the present invention, when honokiol is dissolved in a polyol, skin elasticity, moisturizing, barrier improvement, anti-inflammatory, and antioxidant activities are increased.
[0014] The inventors of the present invention confirmed the activity of honokiol and polyol on skin cells and confirmed that they exhibit more effective elasticity, moisturizing, barrier improvement, anti-inflammatory, and antioxidant effects than the known ingredient retinol.

[0059] The present invention relates to a cosmetic composition for improving skin comprising honokiol and polyol.
[0060] Hereinafter, the present invention will be described in more detail with reference to the following examples. However, these examples are only intended to illustrate the present invention, and the scope of the present invention is not limited by these examples.
[0061] Throughout this specification, "%" used to indicate the concentration of a particular substance is (wt / wt)% for solid / solid, (wt / vol)% for solid / liquid, and (vol / vol)% for liquid / liquid, unless otherwise stated.
[0062]
[0063] Example 1: Cytotoxicity Evaluation
[0064] Toxicity evaluation of honokiol on skin epidermal and dermal cells was performed.
[0065] Specifically, HaCaT cells, which are keratinocytes of the skin, and Hs68 cells, which are fibroblasts of the skin dermis, were seeded at 2 X 10 in a 96-well plate. 4 / well and cultured for 24 hours at 37℃, 5% CO2 in a medium containing FBS (Fetal Bovine Serum) and antibiotics (Antibiotic antimycotic). The next day, honokiol was treated at concentrations of 0.1, 0.5, 1, and 5 ppm in a medium containing only antibiotics and further cultured. After 24 hours, the medium was replaced with one containing MTT tetrazolum (Sigma-aldrich, St. Loudos, USA) and incubated at 37℃ for 4 hours to evaluate cell metabolic activity and determine the toxic concentration of honokiol to skin cells.
[0066] As can be seen in Figures 1a and 1b, honokiol did not induce cytotoxicity at concentrations below 5 ppm for both stratum corneum cells and dermal cells.
[0067]
[0068] Example 2: Confirmation of the effect of honokiol
[0069] 2-1. Confirmation of collagen production
[0070] We investigated whether honokiol increases the amount of skin collagen.
[0071] Specifically, 25 mJ / cm was administered to Hs68 cells, which are dermal cells. 2 After exposure to UVB (ultraviolet B) for 24 hours, RA (retinoic acid), EGCG (epigallocatechin gallate), retinol, and honokiol were treated at a concentration of 1 ppm each.
[0072] Afterwards, the mRNA expression levels of COL1A1, a factor related to the promotion of collagen production, and MMP-1, a collagen-degrading enzyme, were measured through qRT-PCR analysis, converted to the level of actin expression, and the relative values for the expression levels in the UVB non-irradiated group were calculated and compared.
[0073] As can be seen in Figure 2a, the honokiol treatment group showed higher collagen production compared to the UVB only irradiation group, and showed the highest collagen production among all UVB irradiation groups.
[0074] As can be seen in Figure 2b, the honokiol-treated group showed lower collagen-degrading enzyme production compared to the UVB-only irradiation group.
[0075] As a result, honokiol was confirmed to have the effect of increasing skin collagen content by promoting collagen production and inhibiting collagen decomposition.
[0076]
[0077] 2-2. Confirmation of elasticity increase effect
[0078] We investigated whether honokiol increases skin elasticity.
[0079] Specifically, 25 mJ / cm was administered to Hs68 cells, which are dermal cells. 2 After exposure to ultraviolet-B (UVB) of the century, RA, EGCG, retinol, and honokiol were treated at 1 ppm each for 24 hours.
[0080] Afterwards, the mRNA expression levels of the ELN1 gene, a factor related to the promotion of elastin production, and the FBN gene, a factor related to the promotion of fibronectin production, were measured through qRT-PCR analysis, and compared by converting them to the actin expression level from the non-UVB irradiated group.
[0081] As can be seen in Figures 3a and 3b, the honokiol treatment group not only showed the highest expression level among the UV irradiated groups, but also showed a higher expression level than the UV non-irradiated group.
[0082] As a result, honokiol was confirmed to increase skin elasticity by promoting the production of elastin and fibronectin, which are skin components responsible for elasticity.
[0083]
[0084] 2-3. Confirmation of moisturizing improvement effect
[0085] We investigated whether honokiol improves skin hydration.
[0086] Specifically, HaCaT cells, which are stratum corneum cells, were treated with retinol and honokiol at 0.1 and 1 ppm, respectively, for 24 hours.
[0087] Afterwards, the mRNA expression levels of AQP3, a factor related to moisture movement in the dermis, and HAS3, a factor related to hyaluronic acid synthesis enzyme, were measured through qRT-PCR analysis, and compared by converting them to the level of β-actin expression.
[0088] As can be seen in Figures 4a and 4b, the honokiol treatment group not only showed higher expression levels overall compared to the retinol treatment group, but also significantly increased the expression levels of related genes, especially when treated at 1 ppm.
[0089] As a result, honokiol was confirmed to improve skin hydration by activating moisture channels and promoting hyaluronic acid synthesis.
[0090]
[0091] 2-4. Confirmation of barrier improvement effect
[0092] We investigated whether honokiol improves the skin barrier.
[0093] Specifically, HaCaT cells, which are stratum corneum cells, were treated with retinol and honokiol at 0.1 and 1 ppm, respectively, for 24 hours.
[0094] Afterwards, the mRNA expression levels of filaggrin (FLG), a moisturizing factor related to keratinocytes forming the skin barrier, and CLDN1, a skin barrier binding factor, were measured through qRT-PCR analysis, and compared by converting them to the β-actin expression level.
[0095] As can be seen in Figure 5a, the honokiol treatment group not only showed higher expression levels overall compared to the retinol treatment group, but also significantly increased the expression level of the FLG gene, especially when treated at 1 ppm.
[0096] As can be seen in Figure 5b, the honokiol treatment group significantly increased the expression level of the CLDN1 gene compared to the control group, especially when treated with 1 ppm.
[0097] As a result, honokiol was confirmed to improve skin hydration by promoting skin barrier differentiation and strengthening the bond between skin barriers.
[0098]
[0099] 2-5. Confirmation of the effect of alleviating epidermal inflammation
[0100] We investigated whether honokiol alleviates inflammation of the skin epidermis.
[0101] Specifically, HaCaT cells, which are stratum corneum cells, were treated (+) with 10 μg / ml of Poly I:C, which is an inflammatory response inducer, and 10 μg / ml of IL-4 protein, and an experimental group was prepared in which 1 μM of dexamethasone (DEXA), 0.1 ppm of retinol, and 1 ppm of honokiol, which are control inflammation-reducing substances, were treated for 4 hours simultaneously with the induction of inflammatory response. An untreated group (-) was prepared as a negative control group.
[0102] Afterwards, the expression level of the TSLP gene, an inflammation-related factor expressed in skin keratinocytes, was measured through qRT-PCR analysis, and the results were converted to actin expression levels for comparison.
[0103] As can be seen in Figure 6, it was confirmed that the honokiol treatment group in cells in which an inflammatory response was induced had the effect of alleviating inflammation of the epidermis by significantly reducing the expression level of the TSLP gene when treated with 1 ppm.
[0104]
[0105] 2-6. Confirmation of antioxidant effect
[0106] Whether honokiol exhibits antioxidant activity was confirmed through the DPPH test.
[0107] Specifically, ascorbic acid (AA), retinol, and honokiol were treated in DPPH solution at 10 and 100 ppm, respectively, at room temperature for 30 minutes. Absorbance was measured at 517 nm, and antioxidant activity was expressed as the percentage decrease in absorbance compared to the control group. At this time, ascorbic acid, an antioxidant, was used as a control group to determine the percentage decrease in absorbance.
[0108] As can be seen in Figure 7, the honokiol treatment group was found to exhibit significantly higher antioxidant activity compared to retinol.
[0109]
[0110] Example 3: Stabilization of high-concentration honokiol
[0111] 3-1. Selection of solvent
[0112] Honokiol is a poorly soluble ingredient with low solubility in water, requiring dissolution in an organic solvent. Therefore, to stabilize a cosmetic composition containing honokiol at a high concentration of 5 wt%, three solvents—water, butylene glycol (BG), and medium-chain triglyceride (MCT) oil—were used and verified.
[0113] 0D color precipitation 5% honokiol (BG) light brown XX 5% honokiol (MCT) light yellow XX
[0114] As can be seen in Table 1 above, when honokiol was dissolved at 5 wt%, its solubility in water was low, but it was confirmed to dissolve well in BG and MCT.
[0115]
[0116] 3-2. Stability and potency tests according to selected solvent application
[0117] Honokiol was dissolved in BG and MCT at a concentration of 5 wt%, and the formation of precipitates was observed under sunlight conditions at 4°C, 25°C, 45°C, and 50°C, and HPLC (High-performance liquid chromatography) was performed for potency analysis.
[0118] 5% Honokiol (BG) 5% Honokiol (MCT) 4℃ 5.19 5.17 5.14 5.13 25℃ 5.12 5.07 45℃ 5.10 5.03 Cycle 5.11 5.03
[0119] As can be seen in Table 2 above, honokiol maintained its potency well even in harsh environments of low temperature, high temperature, and cycles, so it could be judged that the stability of the substance itself was excellent. Next, after manufacturing the same composition, the formation of precipitates was confirmed under sunlight conditions at 4℃, 25℃, 45℃, and 50℃ for 3 months.
[0120] 1M Color precipitation 5% Honokiol (BG) 25℃ light brown XX 4℃ light brown XX 45℃ brown XX 50℃ brown XX light light brown XX 5% Honokiol (MCT) 25℃ light yellow X△ 4℃ light yellow XX 45℃ yellow XO 50℃ yellow XO light light yellow X△
[0121] As can be confirmed in Table 3 above, when BG and MCT were used as solvents, overall stability was observed under all conditions. However, in the case of MCT, precipitation occurred under high temperature conditions, which was different from BG, from 1 month (1M), and signs of precipitation were found under 25℃ and sunlight conditions. This was judged to be because the stability of the honokiol component itself was high, but its dissolution stability in MCT was low. After 3 months, there was no significant difference in the properties compared to the results after 1 month.
[0122] In conclusion, it was confirmed that the poorly soluble honokiol had higher solubility stability in water-soluble polyols than in oil-soluble ones.

[0124] Example 4: Confirmation of the effect of combination of honokiol and solvent
[0125] 4-1. Confirmation of collagen production
[0126] The amount of collagen production was compared in the same manner as in Example 2-1, but the treatment samples were changed to honokiol solution dissolved in RA (retinoic acid), retinol, DMSO (dimethyl sulfoxide), BG (honokiol solution dissolved in BG), and PG (propylene glycol) (honokiol solution dissolved in PG).
[0127] Unlike the above Example 2-1, where a 5% (w / w) concentration solution was prepared as a sample to confirm stability in a high-concentration dissolved state, the honokiol solution for treatment to cells was prepared at a concentration of 1 mg / ml, so that the honokiol single component treated to cells was 1 ppm.
[0128] As can be seen in Figure 8a, the group treated with honokiol solution dissolved in BG showed higher collagen production compared to the UVB only irradiation group, and showed the highest collagen production among all UVB irradiation groups.
[0129] As can be seen in Figure 8b, all honokiol solution-treated groups showed lower collagen-degrading enzyme production compared to the UVB only irradiation group.

[0131] 4-2. Confirmation of elasticity increase effect
[0132] The degree of increase in skin elasticity was compared in the same manner as in Example 2-2 above, but the mRNA expression levels of the ELN1 gene, a factor related to promoting elastin production, and the FLG gene, a factor related to promoting epidermal cell production, were measured and compared by converting them to the actin expression level from the UVB non-irradiated group.
[0133] The treatment samples were prepared in the same manner as in Example 4-1 above, using RA, retinol, a honokiol solution dissolved in DMSO (Dimethyl Sulfoxide), a honokiol solution dissolved in BG, and a honokiol solution dissolved in PG (Propylene Glycol), so that the honokiol component alone treated on the cells was 1 ppm.
[0134] As can be seen in Figures 9a and 9b, the group treated with honokiol solution dissolved in BG not only showed the highest expression level among the UV irradiated groups, but also showed a higher expression level than the non-UV irradiated group, and showed the highest expression level among all UVB irradiated groups.
[0135] As a result, honokiol was confirmed to increase skin elasticity by promoting the production of elastin and fibronectin, which are skin components responsible for elasticity.
[0136] The present invention relates to a cosmetic composition for improving skin and a pharmaceutical composition for preventing or treating skin damage, comprising honokiol and a polyol.
