[0002] Diagnosis of a variety of diseases involves conducting analysis of a sample from a patient. Fecal sample is used in assay device to determine if the certain analyte, such as, H. Pylori, fecal occult blood, rotavirus, adenovirus, etc. are present in the fecal sample. Fecal occult blood test (FOBT) is a lab test used to check stool samples for hidden (occult) blood that is not readily apparent from a visual inspection. Fecal occult detection is useful for early detection of possible colorectal cancer which may be confirmed by further testing, such as, conducting colonoscopy and obtaining biopsy samples. Colorectal cancer morbidity and mortality rates are reduced by testing for feces occult blood every year. Feces occult blood detection has been recommended as a screening technology for colorectal cancer of the populations by multiple authorities in many countries.

[0071] An exemplary embodiment of sample testing device 100 is shown in FIGS. 1A-1C. The exploded view of device 100 in Fig. 1B shows the individual components of the device 100. The outer chamber 110 is configured for housing testing chamber 120. The outer chamber 110 may include a closed bottom end 110a and an open top end 110b that is sealing engaged with a sealing member 121 of the testing chamber 120. The sealing member 121 engages the open end 110b thereby closing the interior of the outer chamber 110. Testing chamber 120 includes an interior chamber sized to at least partially house the sample collection tube 130. In this embodiment, the sealing member 121 is positioned towards the top end of the testing chamber. The position of the sealing member 121 defines a first portion of the interior chamber 122 that extends above the sealing member 121 and also above the outer chamber 110 and an interior region of the interior chamber that extends below the sealing member 121 and is positioned within the outer chamber 110. Testing chamber 120 includes an open top end 123 and at least a partially open bottom end 124 and the interior chamber 122 extends from the open top end to the bottom end of the testing chamber. The partially open bottom end 124 supports a piercing member 150 visible in FIG. 1C.

[0072] In this embodiment, interior chamber 122 extends from the open top 123 to the bottom of the testing chamber. Testing chamber 120 includes exterior region 125 configured to house at least one test strip. In the embodiment depicted in FIG. 1B, two test strips 126 are positioned in the exterior region 125. Testing chamber 120 includes a substantially cylindrical region 127 that forms at least part of the first portion of the interior chamber and is sized to allow insertion of collection tube 130. Cylindrical region 127 includes means for holding the collection tube 130 in a first position and a second position. The means can be a pair of inverted L-shaped tracks. The L-shaped tracks are positioned on the same horizontal plane and are mirror images of each other. A partial view of one inverted L-shaped track 128 is shown in the cylindrical region 127. The collection tube 130 includes a pair of engagement members 131a and 131b that are positionable in the horizontal parts of the inverted L-shaped tracks where the collection tube 130 is held in a first position. Twisting of the collection tube 130 aligns the engagement members 131a  and 131b with the vertical parts of the inverted L-shaped tracks and downward pressure on the collection tube moves the collection tube into a second position. In the embodiment depicted in FIG. 1B, the collection tube includes a bottom end in a closed configuration having disposed thereon a pierceable foil 132. Sample collection rod 140 includes threads 141 that engage with threads disposed in the interior of the top end of collection tube 130. Cap 142 of the sample collection rod 140 may be used to first screw-on the sample collection rod 140 to close the open end 133 of the sample collection tube and to optionally turn the sample collection tube also such that the engagement members 131a and 131b move across the horizontal parts of the inverted L-shaped tracks and are positioned at the top of the vertical parts of the inverted L-shaped tracks. After the sample has been incubated in the sample collection tube, the cap may be subjected to turning motion (if not already turned) and to downward pressure which moves the collection tube down the vertical parts of the inverted L-shaped tracks and into a second position in which the foil seal is pierced resulting in at least a partial opening of the bottom end of the collection tube. The buffer collection tube may contain a buffer. The collection rod 140 fluidically seals the top open end 133 of the collection tube such that fluid (e.g., buffer with or without the sample) cannot egress from the top end. An annular member 134 is disposed on the exterior of collection tube 130. The annular member 134 is sized to extend to the interior walls of the testing chamber thereby fluidically sealing the interior of the testing chamber which prevents egress of buffer released after piercing of the foil seal. In certain embodiments, the sealing member 121 of the testing chamber 120 is presealed to the open end 110b of the outer chamber such that the testing chamber 120 cannot be removed from the outer chamber 110 without breaking the seal between the sealing member 121 and the outer chamber 110. In certain embodiments, the region 115 positionable over the exterior region 125 configured to house at least one test strip may be substantially transparent to allow inspection of test strip (s) positioned in the exterior region 125. In certain cases, the exterior region includes a slot (s) extending from the sealing member 121 towards the bottom end of the testing chamber.

[0073] FIG. 1C shows a cut-out view of the interior of the outer chamber, testing chamber, and the collection tube. Visible in this view is a piercing member 150. The piercing member 150 may include a channel that is opened upon application of downward  pressure from the collection rod. During use, the piercing member 150 may pierce the bottom end of the collection tube and the bottom end of the collection rod 140 may push open the piercing member such that the channel present in an interior region of the piercing member 150 is connected to the inner region of the collection tube and buffer may flow from the collection tube into the outer chamber. Other configurations of the piercing member are also applicable. In step 1, the collection tube is placed in the first position in the testing chamber. In step 2, the collection tube is pushed down into the testing chamber such that the pierceable bottom end of the collection tube is pierced. At this point, the buffer may directly flow into the outer chamber from a bottom end of the testing chamber or step 3 may be conducted. In step 3, the collection rod may open the top end of the piercing member 150 and channel present in the piercing member may fluidically connect the collection tube to the outer chamber and the buffer may flow into the outer chamber from a bottom end of the testing chamber via the channel. In some embodiments, steps 2 and 3 may occur simultaneously.

Claims

1.A sample testing device, comprising:a sample collection tube comprising:(i) an interior defined between a closed bottom end and an open top end, wherein the closed bottom end is configured for conversion into at least a partially open state and(ii) a sample collection rod configured to sealingly close the open top end;a testing chamber comprising:(i) a substantially cylindrical structure sized to support the sample collection tube in a first position and a second position, wherein a top end of the substantially cylindrical structure is open and the bottom end of the substantially cylindrical structure is open;(ii) a substantially planar structure configured to house at least one test strip;(iii) a sealing member disposed outside the substantially cylindrical structure; andan outer chamber configured to house the testing chamber, wherein the outer chamber comprises an interior defined between a closed bottom end and an open top end, wherein the sealing member is configured to close the open top end,whereinthe sample collection tube comprises a buffer and is positioned in the testing chamber, the testing chamber is positioned in the outer chamber, the sealing member is sealing engaged with the open top end of the outer chamber, and at least one test strip is positioned in the planar structure,when the sample collection tube is in the first position, the closed bottom end of the sample collection tube is in a closed state such that the buffer is contained in the sample collection tube; andwhen the sample collection tube is in the second position, the closed bottom end is converted into at least a partially open state such that the buffer flows to the test strip, andwherein the outer chamber and the sealing member prevent the buffer from exiting the sample testing device.2.The sample testing device of claim 1, wherein the closed bottom end of the collection tube comprises a pierceable material.3.The sample testing device of claim 1, wherein the closed bottom end of the collection tube comprises a detachable cap.4.The sample testing device of claim 3, wherein the detachable cap is configured to detach in response to a downward force.5.The sample testing device of claim 3, wherein the detachable cap comprises a thin wall at the center of the detachable cap and wherein the thin wall is pierceable by application of pressure on the thin wall.6.The sample testing device of any one of claims 1-5, wherein the outer chamber comprises a substantially transparent region positioned adjacent the test strip to allow for viewing of the test strip.7.The sample testing device of any one of claims 1-6, wherein the substantially cylindrical structure comprises means for holding the collection tube in a first position and a second position.8.The sample testing device of claim 7, wherein the means holding the collection tube in a first position and a second position comprise a pair of substantially L-shaped tracks presented in an inverted orientation adjacent to the open top end, each of the substantially L-shaped tracks comprising a horizontal section for holding the collection tube in the first position and a vertical section for allowing the collection tube to move into the second position.9.The sample testing device of any one of claims 1-8, wherein the substantially planar structure configured to house at least one test strip extends from the sealing member towards the bottom end of the outer chamber and terminates adjacent the bottom end of the outer chamber.10.The sample testing device of any one of claims 1-9, wherein the at least one test strip comprises a wicking material.11.The sample testing device of any one of claims 1-10, wherein the collection tube comprises means for holding the collection tube in the first position.12.The sample testing device of claim 11, wherein the means for holding the collection tube in the first position comprises a pair of protruding members.13.The sample testing device of claim 11, wherein the cylindrical structure of the testing chamber comprises a pair of substantially L-shaped tracks presented in an inverted orientation adjacent to the open top end, each of the substantially L-shaped tracks comprising a horizontal section for holding the collection tube in the first position and a vertical section for allowing the collection tube to move into the second position and wherein the pair of protruding members are sized to slidably fit in the horizontal and vertical sections.14.The sample testing device of any one of claims 1-13, wherein the sealing member is disposed adjacent to the top end of the substantially cylindrical structure of the testing chamber.15.The sample testing device of any one of claims 1-13, wherein the sealing member is disposed at the top end of the substantially cylindrical structure of the testing chamber.16.The sample testing device of any one of claims 1-13, wherein the sealing member is disposed at the bottom end of the substantially cylindrical structure of the testing chamber.17.The sample testing device of any one of claims 1-16, wherein the collection rod comprises, in order, from a top end to a bottom end:a cap, a cylindrical element, and a sampling member, wherein the cap has an outer diameter substantially identical to the outer diameter of the top open end of the collection tube and sealing closes the top open end of the collection tube.18.The sample testing device of claim 17, wherein the cylindrical element comprises threads disposed thereon that match threads disposed in an interior region of the collection tube adjacent to the top open end of the collection tube.19.A method for testing a sample using the sample testing device of any one of claims 1-18, the method comprising:disengaging the sample collection rod from the sample collection tube, wherein the sample collection tube is present in the first position;contacting the sample with the sample collection rod;inserting the sample collection rod into the sample collection tube and closing the sample collection tube;incubating the sample in the buffer present in the sample collection tube;moving the sample collection tube into the second position thereby converting the closed bottom end of the sample collection tube into at least a partially open state thereby allowing the buffer to contact the test strip; andanalyzing the test strip.20.The method of claim 19, wherein moving the sample collection tube into the second position comprises applying a downward pressure onto the sample collection tube.21.The method of claim 19 or 20, wherein moving the sample collection tube into the second position comprises applying a turning force to the sample collection tube.22.The method of any one of claims 19-21, wherein incubating the sample in the buffer present in the sample collection tube comprises mixing the sample into the buffer.23.The method of any one of claims 19-22, wherein incubating the sample in the buffer present in the sample collection tube comprises allowing the sample to mix with the buffer for a period of time ranging from 1 min to 30 min, 5 min to 30 min, 10 min to 30 min, 1 min to 20 min, 5 min to 20 min, or 5 min to 15 min.24.The method of any one of claims 19-23, wherein the incubating is performed at ambient temperature.25.The method of claim 24, wherein the ambient temperature ranges from 4℃ to 70℃, 10℃ to 65℃, or 20℃ to 50℃.26.A system for testing a sample, the system comprising:a test device; anda collection tube,the collection tube comprising:an interior defined between a closed bottom end and an open top end, wherein the closed bottom end is configured for conversion into at least a partially open state and a sample collection rod configured to sealingly close the open top end;the test device comprising:a testing chamber comprising:a substantially cylindrical structure sized to support the sample collection tube in a first position and a second position, wherein a top end of the substantially cylindrical structure is open and the bottom end of the substantially cylindrical structure is open;a substantially planar structure configured to house at least one test strip;a sealing member disposed outside the substantially cylindrical structure; andan outer chamber configured to house the testing chamber, wherein the outer chamber comprises an interior defined between a closed bottom end and an open top end, wherein the sealing member is configured to close the open top end,whereinthe sample collection tube comprises a buffer and is positionable in the testing chamber, the testing chamber is positioned in the outer chamber, the sealing member is sealing engaged with the open top end of the outer chamber, and at least one test strip is positioned in the planar structure,when the sample collection tube is in the first position, the closed bottom end of the sample collection tube is in a closed state such that the buffer is contained in the sample collection tube; andwhen the sample collection tube is in the second position, the closed bottom end is converted into at least a partially open state such that the buffer flows to the test strip, andwherein the outer chamber and the sealing member prevents the buffer fromexiting the sample testing device.27.The system of claim 26, wherein the testing chamber comprises a substantially cylindrical element adjacent to the open top end, wherein the cylindrical element expands to allow the collection tube to move into the second position.28.The system of claim 27, wherein the closed bottom end of the collection tube comprises a pierceable material.29.The system of claim 27, wherein the closed bottom end of the collection tube comprises a detachable cap.30.The system of claim 29, wherein the detachable cap is configured to detach in response to a downward force.31.The system of claim 29, wherein the detachable cap comprises a thin wall at the center of the detachable cap and wherein the thin wall is pierceable by application of pressure on the thin wall.32.The system of any one of claims 26-31, wherein the outer chamber comprises a substantially transparent region positioned adjacent the test strip to allow for viewing of the test strip.33.The system of any one of claims 26-32, wherein the substantially planar structure configured to house at least one test strip extends from the sealing member towards the bottom end of the outer chamber and terminates adjacent the bottom end of the outer chamber.34.The system of any one of claims 26-32, wherein the at least one test strip comprises a wicking material.35.The system of any one of claims 26-34, wherein the sealing member is disposed adjacent the top end of the substantially cylindrical structure of the testing chamber.36.The system of any one of claims 26-34, wherein the sealing member is disposed at the top end of the substantially cylindrical structure of the testing chamber.37.The system of any one of claims 26-34, wherein the sealing member is disposed at the bottom end of the substantially cylindrical structure of the testing chamber.38.The system of any one of claims 26-37, wherein the collection rod comprises from a top end to a bottom end: a cap, a cylindrical element, and a sampling member, wherein the cap has an outer diameter substantially identical to the outer diameter of the top open end of the collection tube and sealing closes the top open end of the collection tube.39.The system of claim 38, wherein the cylindrical element comprises threads disposed thereon that match threads disposed in an interior region adjacent to the top open end of the collection tube.40.A method for testing a sample using the system of any one of claims 26-39, the method comprising:disengaging the sample collection rod from the sample collection tube;contacting the sample with the sample collection rod;inserting the sample collection rod into the sample collection tube and closing the sample collection tube;incubating the sample in the buffer present in the sample collection tube;placing the sample collection tube into the testing chamber in the first position;applying downward pressure on the cap of the collection rod thereby converting the closed bottom end of the sample collection tube into at least a partially open state thereby allowing the buffer to contact the test strip; andanalyzing the test strip.41.The method of claim 40, wherein moving the sample collection tube into the second position comprises applying a downward pressure onto the sample collection tube.42.The method of claim 40 or 41, wherein moving the sample collection tube into the second position comprises applying a turning force to the sample collection tube.43.The method of any one of claims 40-42, wherein incubating the sample in the buffer present in the sample collection tube comprises mixing the sample into the buffer.44.The method of any one of claims 40-43, wherein incubating the sample in the buffer present in the sample collection tube comprises allowing the sample to mix  with the buffer for a period of time ranging from 1 min to 30 min, 5 min to 30 min, 10 min to 30 min, 1 min to 20 min, 5 min to 20 min, or 5 min to 15 min.45.The method of any one of claims 40-44, wherein the incubating is performed at ambient temperature.46.The method of claim 45, wherein the ambient temperature ranges from 4℃ to 70℃, 10℃ to 65℃, or 20℃ to 50℃.
