Description

DESCRIPTIONTRIAZOLE METHANOL AS NITRIFICATION INHIBITOR FOR FERTILIZER APPLICATIONCROSS-REFERENCE TO RELATED APPLICATIONS

[0001] This application claims priority to European Patent Application number 24193839.8 filed August 9, 2024, the contents of which is incorporated into the present application by reference in its entirety.BACKGROUND OF THE INVENTIONI. Field of the Disclosure

[0002] This disclosure generally concerns compositions for maintaining or reducing the loss of nitrogen content in soils.II. Background

[0003] Nitrogen is one of the most important nutrients for plant growth and productivity. Nitrogen-use efficiency (NUE) is defined as the ratio of a crop’s nitrogen uptake to the total input of nitrogen fertilizer. In agricultural crops, NUE is notoriously poor, with about 50% of nitrogenous fertilizer applied to crop systems lost to the environment as ammonia (NH3), nitrate (NO3 ), and nitrous oxide (N2O, a greenhouse gas). Low NUE in the agriculture industry contributes to high agricultural production costs, environmental pollution, and climate change. These losses are driven by nitrification and subsequent denitrification processes catalyzed by soil microorganisms.

[0004] Nitrification is traditionally considered to be a two-step process where ammonia (NH3) is first oxidized to nitrite (NO2 ) by ammonia oxidizers, and subsequently to nitrate (NO3’ ) by nitrite-oxidizing bacteria (NOB). Recent evidence indicates complete nitrification by Nitrospira bacteria that are present in natural soil environments.

[0005] Various governments throughout the world have imposed regulations that obligate the agricultural industry to reduce their environmental impacts. One method for reducing agricultural environmental impact involves the use of nitrification inhibitors. Nitrification inhibitors are compounds that can prevent or retard the nitrification of ammonia. These inhibitors help to reduce losses of nitrogen in soil that would otherwise be used by plants.298896325.1 - 1 -

[0006] Some plants develop nitrification inhibition naturally through root exudates termed Biological Nitrification Inhibitors (BNIs). These natural compounds include 2-hydroxy-5- methoxy-l,4-benzoquinone (Sorgoleone), methyl-3-(4-hydroxyphenyl)propionate, and 1,9- decanediol. Fertilizer research has led to the development of synthetic nitrification inhibitors (NIs), including nitrapyrin, dicyandiamide (DCD), and 3, 4-dimethylpyrazole phosphate (DMPP). The biological and synthetic nitrification inhibitors are associated with a variety of shortcomings, including instability at high temperatures, instability at high or low pH, variable effectiveness, difficulties in application, high cost, environmental pollution, and food safety risks. Therefore, there exist an unmet need to identify novel, non-toxic nitrification inhibitors that can be effective over broad ranges of soil types, pH, and temperature.SUMMARY OF THE INVENTION

[0085] FIG. 1 : Chemical pathway of ammonia to nitrate and N2O.

[0086] FIGS. 2A-2B: Active site of ammonium oxidase enzyme. FIG. 2A is a schematic depicting a purported ammonia oxidase copper metalloenzyme active site with two copper atoms. FIG. 2B is a ball and stick model of ammonia oxidase active site.DETAILED DESCRIPTION OF THE INVENTION

[0087] Human activity has had the single largest influence on the global nitrogen cycle by introducing unprecedented amounts of reactive nitrogen into ecosystems. The agricultural industry, in particular, is the single largest human activity that affects the global nitrogen-cycle. In agricultural systems, rapid and unchecked nitrification results in inefficient nitrogen-use by crops, leading to nitrogen-leakage and environmental pollution.

[0088] Nitrification is the process by which ammonia is converted to nitrites (NO2 ), nitrates (NO3 ), and nitrous oxide (N2O) (FIG. 1). This process naturally occurs in the environment, where it is carried out by specialized bacteria. Most nitrification is performed by two groups of ammonia-oxidizing bacteria, Nitrosomonas sp. and Nitrobacter spp. These bacteria are generally ubiquitous components of soil microbial populations. Bacterial enzymes that are key to the nitrification process include ammonia monooxygenase, hydroxylamine dehydrogenase, and nitrite oxidoreductase. Inhibition of the nitrification enzyme ammonia monooxygenase, the first enzyme in the nitrification process, retards the conversion of NH4+into NH2OH, and slows fertilizer loss due to nitrification.

[0089] Certain aspects of the present disclosure are directed to compositions that inhibit the nitrification process, thereby increasing nitrogen longevity in soil. This is accomplished by applying one of more of the nitrification inhibitor compounds disclosed herein to a plant, soil, or crop. Methods of Using the Nitrification Inhibitor

[0100] The nitrification inhibitor, including nitrification inhibitor granules and compositions containing the nitrification inhibitor, can be used in methods of inhibiting the nitrification of ammonia in soil. Such methods can include applying to the soil an effective amount of a composition and / or blend containing the nitrification inhibitor of the present invention. The method may include increasing the growth and yield of crops, trees, ornamentals, etc., such as, for example, palm, coconut, rice, wheat, corn, barley, oats, and / or soybeans. The method can include applying the nitrification inhibitor of the present invention to at least one of a soil, an organism, a liquid carrier, a liquid solvent, etc. The composition(s) containing the nitrification inhibitor can be applied to plants and / or soil through any suitable method of application.

[0101] Non-limiting examples of plants that can benefit from the nitrification inhibitor of the present invention include vines, trees, shrubs, stalked plants, fems, etc. The plants may include orchard crops, vines, ornamental plants, food crops, timber, and harvested plants. The plants may include Gymnosperms, Angiosperms, and / or Pteridophytes.

[0102] The effectiveness of compositions comprising the nitrification inhibitor of the present invention can be ascertained by analyzing nitrification activity as described below. It is understood that different soils have different characteristics, which can affect the effectiveness of the nitrification inhibitor compositions. The effectiveness of a nitrification inhibitor composition can also be directly compared to other nitrification inhibitor compositions by doing a side-by-side comparison in the same soil under the same conditions.Examples

[0103] The present invention will be described in greater detail by way of specific examples. The following examples are offered for illustrative purposes only, and are not intended to limit the invention in any manner. Those of skill in the art will readily recognize a variety of noncritical parameters which can be changed or modified to yield essentially the same results.Example I Molecular Modeling Study

[0104] A molecular modeling approach was used to predict the relative binding affinity of a compound of Formula I, as compared to nitrification inhibitors dicyandiamide (DCD) and 3,4-dimethylpyrazole phosphate (DMPP) in the active site of modelled ammonia monooxygenase enzyme, also known as nitrification enzyme (FIG. 2A). The crystal structure298896325.1 - 19 -of ammonia monooxygenase is unknown, however, literature reveals information by homology modelling and other indirect approaches, which indicates that it is a copper metalloenzyme. Ammonia monooxygenase Cu site modelling shows two Cu(II) ions separated by 2.55 A. The atoms of the CU2S2 rhombus deviate from the plane by 0.2 A, and the angle between the two CuS2planes is 170.1°. Cui is bound to Met-A, His-A, and Cys-A from both chains in a distorted tetrahedral geometry, while Cu2 is bound to residues, Met-B, His-B, and Cys58-B from both chains, again in a distorted tetrahedral geometry (FIG. 2A). Both copper atoms depicted in FIG. 2A are in +1 -oxidation states. In light of available information of the active site, density functional theory (DFT) modelling was used to examine ammonia monooxygenase active site geometry. The active site geometry was minimized (FIG. 2B) using B3LYP functional as implemented in the Gaussian 09 package in conjunction with 6-31G(d,p) basis sets for the nonmetal elements such as C, H, N, and O, while for metal Cu an effective core pseudopotentials (SDD) and the associated basis sets with a polarization function was used. An analytical frequency calculation verified all the stationary points. The Gibbs energies were computed at 298.15° K and at atmospheric pressure (unless specified otherwise), assuming an ideal gas behavior using unsealed harmonic vibrational frequencies obtained within the rigid rotor approximation.

[0105] The compounds of Formula II, III, IV, V, and VI candidate inhibitors and the commercial inhibitors DCD and DMPP (for benchmarking purpose) were introduced and the ligand receptor complex geometry minimized using DFT methods. The binding energies (BE) of the inhibitors within the active site were estimated using equation (1):Ebinding — Energy (Enzyme-Inhibitor complex) [Energy (Active site) + Energy (Inhibitor)] Eq(1)298896325.1 - 20 -Table 1 - Calculated DFT Binding Energies298896325.1 - 21 -

[0106] During competitive inhibition, an effective inhibitor molecule, with a higher binding energy within the enzyme active site, would have lower IC50 values. BE was estimated for candidate Formulas II to VI and benchmarks as reported in Table 1. The binding energies of Formulas II to VI were within close range of benchmarks, demonstrating that Formulas II to VI have comparable affinity for ammonia monooxygenase.Example II - Nitrification Inhibitor Synthesis

[0107] Synthesis of the compound of Formula III: The chemicals used were obtained from commercial sources. The solvents and reagents used were of reagent grade and used as received without further purification. NaH (1.682 g, 0.0731 mol) was added in portions to 1,2,4-triazole(5.0 g, 0.0723 mol) in tetrahydrofuran (75 mF) while stirred. The reaction produced hydrogen. Once the effervescence ceased, the products were stirred further for another 1 hour. Acetone (5.5 mL, 0.731 mol) was then added dropwise with stirring and the reaction was then refluxed at 75 °C overnight with stirring. The solution turned off-white, and the solvent was vacuumed to dryness. A white solid was produced and then dissolved in methanol. A solid was later precipitated out through cooling at -10 °C in freezer. The solid was filtered off and thoroughly washed with diethyl ether and dried in vacuum over P2O5. The yield was 85%.298896325.1 - 22 -Formula III

[0108] Synthesis of the compound of Formula VI: 1,2,4-triazole (5.0 g, 0.0723 mol) in tetrahydrofuran (70 mL) was added to NaH (1.682 g, 0.0731 mol) in multiple small portions over stirring, which resulted in hydrogen evolution. Once the effervescence ceased, the mixture was stirred further for another 1 hour. To that mixture, benzaldehyde (7.7 mL, 0.731 mol) in 10 mL tetrahydrofuran was added dropwise with stirring. Then it was refluxed at 75 °C overnight with stirring. The solution turned slight yellow, and the solvent was vacced to dryness. The obtained beige solid was dissolved in hot methanol and later precipitated out through cooling at 0 °C in a freezer. The solid formed was filtered off and thoroughly washed with diethyl ether and dried in vacuum over P2O5. Yield, 81%.Formula VI

[0109] The other triazole derivative candidates of the formulas II, IV, and V can also be synthesized by the reaction of the triazole with NaH and corresponding aldehydes / ketones in good yields.Example III - Nitrification Activity Analysis

[0110] Crop soil and water can be combined to form a sludge. The freshly-acquired sludge may contain a high amount of nitrate nitrogen, therefore, the sludge can be pretreated to reduce298896325.1 - 23 -the chance of denitrification during analysis. Ethylene glycol will be added to the sludge and the resulting sludge will be purged with nitrogen gas for 24 hours to eliminate the nitrate nitrogen. After 24 hours, the sludge will be aerated to eliminate the residual chemical oxygen demand (COD) which could be present. COD refers to the amount of oxygen required to degrade all oxidizable compounds in a measured sample.

[0111] A nitrification batch test will be performed under nitrifying conditions. The activity of nitrification inhibitor Formula II to VI compounds will be compared to that of the nitrification inhibitor dicyandiamide (DCD). COD and NH4-N will be added to the sludge with any one of the compounds of Formula I or DCD. COD, NH4-N, and NO3-N concentrations will be monitored over time (t = 0, 2, 4, 6, 8, 24 and 29 hours). The COD and NH4-N removal will be calculated from the analysis results. Glycol will be included as a reference in the test.

[0112] The pH and oxygen content will also be monitored during the tests. The pH level will be kept between 7 and 8 at the beginning of the tests by adding IN HC1 or IN NaOH. Oxygen will be constantly purged into the sludge solutions throughout the test to get maximum aeration.

[0113] It is expected that the compounds of Formula I will show inhibition of nitrification.* * *
