Abstract
Description
Technical Field
[0001] The invention belongs to the technical field of drug detection, and specifically relates to a detection method for related substances in cationic lipid YK-009 and application thereof. Background Art
[0002] Since the successful launch of the mRNA vaccine for novel coronavirus infection, lipid nanoparticles (LNP) have become the preferred delivery carrier for nucleic acid products such as mRNA and siRNA, and are widely used in mRNA vaccines and therapeutic drugs. mRNA has a large molecular weight and strong hydrophilicity. Its biological activity is thousands of times that of small molecule drugs, and its industrial production is relatively easy. However, its single-stranded structure makes it extremely unstable and easily degraded. It also carries a negative charge and has to pass through the cell membrane with the same negative charge on the surface, making its delivery difficult. LNP is a multi-component system that can encapsulate mRNA in the internal cavity of the structure. This structure can improve the stability of mRNA in vivo and protect it from degradation, thereby exerting its effect.
[0003] Lipid nanoparticles (LNP) are lipid vesicles with a uniform lipid core, which are widely used in the delivery of RNA vaccines and nucleic acid drugs. LNP encapsulation can protect mRNA from extracellular RNases and assist in the intracellular delivery of mRNA. LNP has become the most widely used non-viral vector due to its simple operation, high biosafety, good repeatability, high transfection efficiency and low cytotoxicity. The mRNA novel coronavirus infection vaccines of the three giants BioNTech, Moderna and CureVac all use LNP delivery technology.
[0004] The LNP delivery system is mainly composed of ionized cationic lipids (SM102 or ALC-0315 or YK-009), phospholipids (DSPC or DOPE), cholesterol and auxiliary lipids (DMG-PEG2000) to prepare lipid prescriptions. The four lipids usually have a molar ratio of 50:10:38.5:1.5, and the ratio of ionizable cationic lipids to RNA is maintained at 15:1 (w / w), so the amount of cationic lipids used in each unit preparation is very large. Ionizable lipids are protonated under acidic conditions, carry positive charges, and electrostatically bind to the negatively charged nucleic acid phosphate backbone, resulting in the RNA drug substance being encapsulated, thereby forming lipid nanoparticles to ensure the stability of mRNA. LNPs will be taken up by cells through receptor-mediated endocytosis. After being taken up by cells, LNPs will enter the endosomes. Ionizable lipids are protonated in the acidic environment of the endosomes and carry positive charges, which helps to destroy the endosomal membrane and enable LNPs to escape into the cytoplasm. After escaping from the endosomal structure, mRNA is released from the LNP and enters the cytoplasm, while a large number of ionizable cationic lipids remain in the cell. These fragments can interact with cellular components and cause various adverse effects in the cell: the positive charge of the cationic lipids can react with the negative charge of the cellular components, resulting in membrane porosity and calcium ion influx, which may lead to cell death; cationic lipids can affect hepatocytes (most LNPs are currently targeted to the liver) by interfering with amino acid metabolism, energy metabolism, lipid metabolism, and increasing oxidative stress; the interaction of cationic lipids with the negative charge of the mitochondrial membrane may destroy its integrity, leading to the release of reactive oxygen species (ROS) in the cell, which may trigger exponential cell damage; if the cationic lipids reach certain areas of the brain, they can cause serious neurological effects.
[0005] Since cationic lipids are used in large amounts and have a long residence time in the body, the presence of relatively small amounts of impurities will lead to an increase in the absolute number of impurities in the body. The structures of these impurities usually contain similar cationic structures and lipophilic genes that are not easily metabolized, which can cause many unpredictable cytotoxic effects. Therefore, it is very necessary and essential to strictly limit the impurity content in the cations and find scientific quality testing methods to strictly control the impurity limits. Unfortunately, in searching the existing literature, no very scientific quality control methods have been found for cationic lipids that have been on the market or have been made public.
[0006] YK-009(C 40 H 79NO5) is the third-generation cationic liposome independently developed by Yuekang Pharmaceutical Group Co., Ltd. Compared with the second-generation LNP made of DLin-MC3-DMA, the LNP made of YK-009 liposome has the advantages of smaller particle size, higher uniformity, higher in vitro cell transfection efficiency, lower cytotoxicity and high in vivo expression, but its synthesis process is relatively complicated, and a series of impurities with similar properties will be produced during the process, including intermediates, degradation impurities, by-products and by-product derivative impurities. Since these impurities have no obvious chromophore groups in the structure of YK-009 and their properties are very similar, it is difficult to separate YK-009 from its impurities (related substances), and no separation and detection methods for related substances in YK-009 or other similar cations have been retrieved.

[0183]
[0184] Table 16
[0185]
[0186] Conclusion: When the column temperature was adjusted within ±5℃ in the chromatographic conditions, the RSD of the main peak area in the system suitability solution was less than 2.0%, and the separation of YK-009 and adjacent impurities was 2.0 and 1.9, respectively, both greater than 1.5, indicating that the system was in good condition. When the pH value of the acetic acid-triethylamine solution was 6.5, the RSD of the main peak area in the system suitability solution was less than 2.0%, and the separation of YK-009 and adjacent impurities was 2.4. When the pH value of the acetic acid-triethylamine solution was 7.5, the RSD of the main peak area in the system suitability solution was less than 2.0%, and the separation of YK-009 and adjacent impurities was 2.3, indicating that the system was in good condition. After adjusting the column temperature and the pH value of the acetic acid-triethylamine solution, there was no significant difference between the results of each impurity and the normal conditions. Based on the above results, the column temperature and the pH value of the acetic acid-triethylamine solution in the chromatographic conditions had good durability.
[0187] Example 2 Selection of chromatographic column
[0188] Based on the chromatographic conditions of Example 1, the type of chromatographic column was changed, and other chromatographic conditions remained unchanged, and YK-009 and its 10 known impurities were detected. Accurately measure 5 μL of the mixed impurity reference solution prepared in Example 1, inject it into the high performance liquid chromatograph respectively, and record the results. Figure 2-3 .
[0189] Table 17
[0190]
[0191] Conclusion: The separation of YK-009 and its impurities by HPLC analysis using the two different chromatographic columns mentioned above revealed that the chromatographic column used in method 1 (Waters ACQUITY UPLC Oligonucleotide BEH C18) yielded a spectrum ( Figure 2 ) has a wide peak shape for YK-009 and its impurities, of which the symmetry factor of YK-009 is 2.91, impurities 5 and 9 are not baseline separated, impurities 3 and 6 are wrapped by the main peak, and the separation degree between adjacent impurities and YK-009 is 0.51, indicating poor impurity separation. Using the ordinary C18 column (Agilent Poroshell 120 EC-C18) of method 2, the spectrum obtained is ( Figure 3 ) has a wider peak shape of YK-009, which is triangular and has a symmetry factor of 4.43. The peak shapes of impurities 5, 9 and 4 are also wider, which reduces the sensitivity of impurities. The tail of the main peak YK-009 can clearly be seen to contain impurities, and the impurity separation effect is not good. In contrast, the chromatographic column (Waters ACQUITY UPLC CSH Phenyl-Hexyl) in Example 1 is used to obtain the spectrum ( Figure 1) It can be clearly seen that the peak shapes of the 10 impurities and YK-009 are good, the impurity symmetry factors are between 0.9 and 1.1, the YK-009 symmetry factor is 0.74, and the separation degree between the main peak YK-009 and the adjacent impurities is 2.72, which achieves good separation of YK-009 and the 10 impurities and meets the detection requirements of related impurities in YK-009.
[0192] Example 3 Selection of mobile phase solvent
[0193] Based on the chromatographic conditions of Example 1, the mobile phase B solvent was changed, and other chromatographic conditions remained unchanged, and YK-009 and its 10 known impurities were detected. Accurately measure 5 μL of the test solution prepared in Example 1 and inject them into the high performance liquid chromatograph respectively, and record the results. Figure 4 .
[0194] Table 18
[0195]
[0196] Conclusion: The separation of YK-009 and its impurities by HPLC analysis using the two different solvents as mobile phases found that the spectrum obtained by using acetonitrile as the solvent in Example 1 ( Figure 1 ) has a good peak shape, the symmetry factors of each impurity are between 0.9 and 1.1, the symmetry factor of YK-009 is 0.74, all impurities can be well separated from YK-009, and the separation degree between adjacent impurities and YK-009 is greater than 1.5, which can achieve the separation and detection of 10 known impurities in YK-009. In contrast, when methanol in the comparative example is used as the solvent, the spectrum obtained ( Figure 4 ) The main peak YK-009 has a poor peak shape, severe broadening, and a symmetry factor of 3.61. The separation between the main peak YK-009 and the adjacent impurities is less than 1.0. There is one impurity peak before YK-009, which may be wrapped by the main peak, making it impossible to separate and detect the main peak and 10 impurities.
[0197] Example 4 Selection of the amount of acetic acid-triethylamine added in the mobile phase
[0198] Based on the chromatographic conditions of Example 1, the amount of ion pair reagent (acetic acid-triethylamine) added was changed, and other chromatographic conditions remained unchanged, and YK-009 and its 10 known impurities were detected. Accurately measure 5 μL of the test solution prepared in Example 1 and inject it into the high performance liquid chromatograph respectively, and record the results, see Figure 5 and Figure 6 .
[0199] Table 19
[0200]
[0201] Conclusion: The separation of YK-009 and its impurities was analyzed by HPLC using the mobile phases with different ion pair reagent additions. It was found that when 0.5% acetic acid-triethylamine was added in method 1, the spectrum obtained was ( Figure 5 ) has a poor peak shape, a symmetry factor of 2.14, and a separation degree of 2.5 between YK-009 and adjacent impurities. However, it can be clearly seen from the figure that there is a problem with the elution of the peak tail, and the impurity peak may be wrapped. When 0.7% of acetic acid-triethylamine is added, the peak shape of the main peak is significantly improved, and the separation degree of YK-009 and adjacent impurities is 1.2, which basically meets the separation requirements, but impurities 4 and 9 cannot be baseline separated, and the separation degree is only 0.85. When 1.5% of acetic acid-triethylamine is added, the spectrum obtained is ( Figure 6 ) in which the separation degree of YK-009 from adjacent impurities is greater than 1.5, but impurities 4 and 9 are inseparable, and the peak shape of YK-009 is not much different from that when the addition amount is 1.0%, and both are better than the peak shape when the addition amount is 0.5%. When 1.0% of acetic acid-triethylamine is added in Example 1, the spectrum obtained is ( Figure 1 ) The main peak YK-009 has a good peak shape, the symmetry factor of YK-009 is 0.74, and the separation between YK-009 and adjacent impurities is greater than 1.5, which can achieve the separation and detection of 10 known impurities in YK-009. At the same time, considering the influence of ion pair reagents on separation, instruments and chromatographic columns, a 1.0% addition amount can meet the needs.
[0202] Example 5 Selection of elution gradient
[0203] Based on the chromatographic conditions of Example 1, the elution gradient was changed, and other chromatographic conditions remained unchanged to detect YK-009 and its impurities. Accurately measure 5 μL of the test solution prepared in Example 1 and inject it into the high performance liquid chromatograph respectively, and record the results. Figure 7 and Figure 8 .
[0204] Table 20
[0205]
[0206] Conclusion: The separation of YK-009 and its impurities was analyzed by HPLC using the above different elution gradients. It was found that when the elution gradient of method 1 was used, the spectrum obtained ( Figure 7 ) has a separation degree of 0.49 between YK-009 and adjacent impurities. It can be clearly seen from the figure that the low-proportion organic phase has a poor elution effect on the main peak and impurities, resulting in no impurity peaks in the first 30 minutes of the chromatogram, and the main peak and impurities are concentrated in the back and are dense, with poor separation. When the elution gradient of method 2 is used, the spectrum obtained is ( Figure 8) It can be seen that the impurity separation is improved compared with method 1, but impurities 5 and 9 are encapsulated, and the separation degree between the main peak YK-009 and the subsequent impurities is 0.90, which needs further optimization. When the elution gradient in Example 1 is used, the spectrum obtained is ( Figure 1 ) The main peak YK-009 has a good peak shape, the symmetry factor of YK-009 is 0.74, and the separation degree between YK-009 and adjacent impurities is greater than 1.5, which can realize the separation and detection of 10 known impurities in YK-009.
[0207] Example 6 Selection of mobile phase additives
[0208] Based on the chromatographic conditions of Example 1, the mobile phase additives were changed to detect YK-009 and its impurities. Accurately measure 5 μL of the test solution prepared in Example 1 and inject them into the high performance liquid chromatograph respectively, and record the results. Fig. 9 .
[0209] Table 21
[0210]
[0211] Conclusion: The separation of YK-009 and its impurities by HPLC analysis with the mobile phases of the above-mentioned different additives revealed that the spectrum obtained by using ammonium formate in method 1 ( Fig. 9 ) has a wide main peak shape, a symmetry factor of 3.89, and a separation degree of 2.06 between YK-009 and adjacent impurities. However, it can be seen from the figure that the number of impurity peaks detected is small, and there may be impurity peaks that are wrapped. At the same time, the impurity peaks have a poor peak shape and a tailing phenomenon. When acetic acid-triethylamine in Example 1 is used, the spectrum obtained is ( Figure 1 ) The main peak YK-009 has a good peak shape, the symmetry factor of YK-009 is 0.74, and the separation degree between YK-009 and adjacent impurities is greater than 1.5, which can realize the separation and detection of 10 known impurities in YK-009.
