[0003] The liver is the largest glandular organ in the human body and plays a role in participating in the body's metabolism and detoxification. At the same time, it is also vulnerable to the effects of many drugs, chemicals, environmental pollutants and infections.
[0004] Alcohol is one of the essential beverages in life. Alcohol is contained in wine. Excessive drinking can change people's moods and behaviors and cause drunkenness. After getting drunk, there are often headaches, dizziness, poor self-control, thirst, dehydration, uncoordinated movements, incoherent speech, dilated pupils, decreased blood pressure, increased heart rate, urinary and fecal incontinence and other phenomena. The liver becomes the most severely damaged organ because it can metabolize alcohol. Alcoholic Liver Disease (ALD) is a type of liver disease characterized by excessive drinking as the main cause and different clinicopathological changes such as fatty liver, hepatitis, cirrhosis and liver cancer.
[0005] Excessive drinking has become a common public health problem, and the harm to the human body caused by excessive drinking has attracted wide attention in society. Therefore, the research and development of products for relieving discomfort after drinking and protecting the liver from alcohol has become a research hotspot in the medical field.
[0006] Currently, the products for relieving hangover and protecting the liver are mainly products such as silymarin and naloxone. [0090] (1) Bacteria activation:
[0091] Bacterial strain culture: Expansion culture of bacterial strains preserved in -80℃ refrigerator.
[0092] Preparation of culture medium: 20 g L-arabinose, 2 g maltitol, 2 g beef extract powder, 1 g yeast extract powder, 0.5 g sodium acetate, 0.5 mL Tween-80, 0.2 g potassium dihydrogen phosphate, 1000 mL tap water, pH adjusted to 7.0 (alkaline solution adjustment).
[0093] Operation: Add L-arabinose and other raw materials in the above formula into 1000 mL of water in the formula without any particular order, stir for 10 minutes to dissolve, and then divide into test tubes, each test tube contains 5 mL, seal with kraft paper and place in a medical sterilizer for sterilization at 0.1-0.12 MPa for 20 minutes. The culture medium is ready.
[0094] Inoculation: Under sterile conditions (clean bench), open the -80℃ low-temperature freezer to store the frozen tube of bacteria, and dissolve it at room temperature. Then take one sterilized test tube culture medium, add 0.5 mL of the dissolved bacterial solution into the test tube, seal the tube mouth with kraft paper, shake it gently by hand, and place it in a constant temperature incubator at 37℃ for 24 hours. This is called expansion culture medium.
[0095] (2) Preparation of primary bacterial culture liquid:
[0096] Primary seed culture formula: L-arabinose 8 g, yeast peptone 6 g, beef extract powder 1 g, lactose 1.4 g, sodium acetate 0.8 g, Tween-80 1 mL, tap water 1000 mL, pH natural.
[0097] Operation: Add the above raw materials into 1000 mL of tap water without any distinction, start the blender, and stir for 10 minutes to dissolve them all. This is called the primary seed culture solution.
[0098] Add the stirred and dissolved first-level seed culture solution into the conical flask according to the dosage, then wrap the bottle mouth with 6 layers of gauze and one layer of kraft paper, and put it into the sterilizer for sterilization. Before sterilization, first open the exhaust valve on the sterilizer and close it when a small amount of steam is discharged. When the pressure on the sterilizer reaches 0.05 MPa, open the exhaust valve on the sterilizer to exhaust for 6-8 minutes, then close the exhaust valve and continue heating. When the steam pressure in the sterilizer reaches 0.1-0.12 MPa, start timing. The sterilization time is 20 minutes. After completion, leave the heat source and let it cool down naturally. When the pressure gauge on the sterilizer returns to zero, open the sterilizer, take out the sterilized culture medium and put it into the clean bench for natural cooling. When the temperature drops to 37°C, transfer the above cultured expansion culture solution to the sterilized culture solution in the conical flask. The inoculation amount is 0.5%, that is, 100 mL of first-level seed culture solution and 0.5 mL of expansion culture solution. mL. After completion, seal the bottle mouth with the original gauze and kraft paper, shake it slightly by hand to mix the inoculated bacteria and culture solution evenly, and then place it in a constant temperature box and culture it at 37°C for 18 hours. This is called the primary culture solution.
[0099] (3) Preparation of seed tank strains:
[0100] Sterilization of fermentation equipment, pipelines and sterile filtration systems:
[0101] First, open the valves of all inlet and outlet pipelines and sterile air pipelines, and pass 0.12-0.14 MPa steam. Allow the steam to connect with the pipeline valves and discharge a small amount of steam. Ventilate for 40 minutes, then close the valves of all inlet and outlet pipelines and set aside.
[0102] Sterilization of empty seed tanks and fermentation tanks:
[0103] Close all valves, open the drain valve at the bottom of the tank and the drain valve in the tank interlayer, open the direct steam valve to introduce 0.12-0.14 MPa steam into the pipe, start timing when the temperature in the tank reaches 121°C, the sterilization time is 40 min, then close the drain valve at the bottom of the tank and the drain valve in the interlayer, and the temperature in the tank will naturally drop to 37°C for standby use.
[0104] Seed pot culture:
[0105] Formula: L-arabinose 3%, casein peptone 0.9%, dipotassium hydrogen phosphate 0.4%, onion juice powder 0.8%, sodium acetate 0.7%, ginger juice powder 0.8%, Tween-80 0.1%, polyether defoamer 0.1%, tap water 93.2%, natural pH value.
[0106] Specific operation: First, pour the tap water used in the formula into the sterilized seed tank, start the mixer, add the raw materials required in the formula respectively (the order of addition is not important), then stir continuously and pass steam for heating. First, use the steam in the tank interlayer to heat to 95°C, then switch to direct steam heating. When the temperature in the tank reaches 121°C, maintain it for 20 minutes to achieve the sterilization effect, then turn off the steam, and start cooling through the tap water in the tank interlayer. When the temperature in the tank reaches 37°C, it is ready for use. This is called seed culture medium. Then inoculate the above-cultured first-level bacterial culture solution into the seed tank under sterile conditions. The inoculation amount is 3% of the culture medium in the tank. 100 kg of seed tank culture medium can be inoculated with 3 kg of first-level bacterial solution. Then cover the inoculation cap on the tank and start stirring and culturing. The culture conditions are: temperature 37°C, natural pH value, stirring speed 120 r / min, tank pressure 0.05 MPa, and culture time 12 hours to meet the requirements. If the tank body is lowered, sterile air can be introduced to maintain the tank pressure. If the tank pressure is too high, the air release valve on the tank top can be used to adjust it. This is called seed tank culture.
[0107] (4) Lactobacillus reuteri fermentation:
[0108] Culture medium formula: L-arabinose 4%, casein peptone 3%, trehalose 2%, stachyose 1%, yeast extract powder 2.5%, sodium acetate 0.9%, Tween-80 0.2%, kale powder 0.6%, dipotassium hydrogen phosphate 0.9%, polyether defoamer 0.1%, tap water 84.8%.
[0109] Specific operation: First, add the tap water used in the formula into the sterilized fermentation tank, start the mixer, add the raw materials used in the formula into the fermentation tank (the order of addition is not important), and then pass steam for heating. First, use the interlayer steam to heat to 95°C, and then switch to direct steam heating. When the temperature in the tank reaches 121°C, start timing, and keep it for 30 minutes to achieve the sterilization effect, and then cool it down with the tap water in the interlayer. When the temperature in the tank drops to 37°C, start to inoculate the strain. Before inoculation, first increase the tank pressure of the seed tank to 0.1 MPa, and keep the tank pressure of the fermentation tank at 0.05MPa, then open the inoculation pipeline valve, and transport the seeds cultured in the seed tank to the fermentation tank by pressure difference, and then close the inoculation channel valve.
[0110] The inoculation amount is 5 kg of the culture medium in the seed tank for every 100 kg of the fermentation tank. The culture conditions are: culture temperature 37℃, stirring speed 160 r / min, tank pressure maintained at 0.05 MPa, and the pH value is maintained at 6.0 through the acid and alkali automatic control system during the fermentation process. If the tank pressure decreases, sterile air can be introduced to maintain the tank pressure until the fermentation is completed. The total fermentation time is 25 hours, and then the temperature is lowered to 20℃, which is the fermentation liquid. UHT inactivation treatment is carried out at 120℃ for 6 seconds.
[0111] (5) Centrifugation of fermentation broth:
[0112] The fermentation liquid is pumped into a storage tank for centrifugation. Operation: Start the centrifuge, adjust the speed to 10000 r / min, centrifuge at a feed rate of 3kg / min, collect the inactivated wet bacterial sludge metabolites after centrifugation, and freeze-dry the collected inactivated wet bacterial sludge metabolites.
[0113] (6) Freeze-drying of bacterial sludge metabolites:
[0114] Preparation of bacterial suspension: Take 1000 g of inactivated wet bacterial sludge metabolites, add 4000 mL of sterile saline, 600 g of skimmed milk powder, 2000 g of L-arabinose, 30 g of D-pantothenic acid, and 60 g of inositol.
[0115] Operation: First, take 2500 mL of saline in the formula, add L-arabinose and stir to dissolve, then add 1000 g of inactivated wet bacterial sludge metabolites, then add the remaining saline, skim milk powder and auxiliary materials in the formula and stir well, which is called suspension. The suspension is freeze-dried and the moisture content is finally controlled to 4%, which is the inactivated bacterial metabolite freeze-dried powder. According to the test, the inactivated bacterial metabolite freeze-dried powder has an extracellular polysaccharide content of 28%, a polypeptide content of 7%, an amino acid content of 27%, and an inactivated bacterial cell content of 150 billion / g.
[0116] The inactivated bacteria of freeze-dried powder will be lost or broken during the dilution process. Although the bacteria are broken, the effect of the strain will not be weakened.
[0117] The freeze-dried powder can be widely used in food, baked food, solid beverages, liquid beverages, and compressed candies.
[0118] The freeze-dried powder prepared by the above method was subjected to the following effect verification.

[0125] Alanine aminotransferase (ALT) kit (Nanjing Jiancheng Bioengineering Institute)
[0126] (3) Experimental groups
[0127] The experimental mice were randomly divided into 5 groups: control group, model group, and experimental group (divided into 3 groups, experimental group 1, experimental group 2, and experimental group 3), with 10 mice in each group. They were fasted for 12 hours before the experiment, but were not deprived of water.
[0128] (4) Autonomous activity experiment
[0129] The mice were pre-fed for 10 days. The experiment began on the 11th day. Before the experiment, the mice were placed in the spontaneous activity instrument for 10 minutes to adapt to the environment and ensure the accuracy of the experimental data. During the experiment, the experimental group was gavaged with the probiotic solution (0.5 mL) prepared in step (1), and the control group and the model group were gavaged with an equal volume of sterile water. After 30 minutes, the mice in the experimental group and the model group were gavaged with 52% (v / v) Red Star Erguotou at a rate of 0.05 mL / 10 g body weight, and the mice in the control group were gavaged with an equal volume of sterile water. The mice in each group were placed in the spontaneous activity instrument for 10 minutes in turn, and the number of spontaneous activities of the mice within 5 minutes was recorded. The results are shown in the table. Figure 1 .
[0130] (5) Righting experiment
[0131] On the 13th day, the experimental group was gavaged with the probiotic solution (0.5 mL) prepared in step (1), and the control group and the model group were gavaged with an equal volume of sterile water. 30 min later, the experimental group and the model group mice were gavaged with 0.15 mL / 10 g body weight of 52% (v / v) Red Star Erguotou, and the control group mice were gavaged with an equal volume of sterile water. The disappearance time and recovery time of the righting reaction of each group of mice were recorded, and the tolerance time (min, from the injection of wine to the disappearance of the righting reaction) and the duration (min, from the disappearance of the righting reaction to the recovery) were calculated. The results are shown in Figure 2 .
[0132] (6) Drunk sleep experiment
[0133] On the 15th day, the experimental group was gavaged with the probiotic solution (0.5 mL) prepared in step (1), while the control group and the model group were gavaged with an equal volume of sterile water. 30 min later, the mice in the experimental group and the model group were gavaged with 0.15 mL / 10 g body weight of 52% (v / v) Red Star Erguotou, while the mice in the control group were gavaged with an equal volume of sterile water. The activity of the mice was observed, and the indicators of sleep (drunkenness) were unsteady crawling, dragging of the back abdomen on the ground, and laziness with eyes closed, while the indicators of awakening (sobering up) were free movement, flexible limbs, and mental recovery. The time of falling asleep and waking up of the mice in each group were recorded, and the sleep latency (min, the time from gavage to falling asleep) and duration (min, the time from falling asleep to waking up) were calculated. The results are shown in the table below. Figure 3 .
[0134] (7) Biochemical index detection
[0135] Starting from the 17th day, in addition to normal feeding, the mice in the experimental group were gavaged with the probiotic solution (0.5 mL) prepared in step (1) at the same time every morning, and the control group and the model group were gavaged with an equal volume of sterile water. 30 minutes later, the mice in the experimental group and the model group were gavaged with 52% (v / v) Red Star Erguotou at 0.15 mL / 10 g body weight, and the mice in the control group were gavaged with an equal volume of sterile water for 6 consecutive weeks. After fasting for 12 h, the mice in each group were weighed, blood was collected from their eye sockets, and the serum was separated by centrifugation and stored at -80°C for later use.
[0136] The mice in each group were killed by cervical dislocation, and the liver was removed and weighed. The liver coefficient of the mice was calculated according to the following formula. The results are shown in Figure 4 .
[0137] Liver coefficient (%) = liver wet mass (g) / body mass (g) × 100
[0138] A certain amount of liver tissue was taken and prepared into tissue homogenates of different concentrations. The activities of malondialdehyde (MDA), glutathione (GSH), glutamine transaminase (TG), glutathione peroxidase (GSH-Px), superoxide dismutase (T-SOD) and catalase (CAT) were detected according to the operating procedures of the kit. The results are shown in Figure 5 , Figure 6 , Figure 7 , Figure 8 The activities of ALT and AST in mouse serum were measured by aspartate aminotransferase (AST) and alanine aminotransferase (ALT) biochemical detection kits, respectively. The results are shown in Fig. 9According to the instructions of the ELISA kit, the levels of cytokines IL-1β, IL-6, and TNF-α in serum were determined. The results are shown in Fig.10 .
[0139] A small amount of alcohol has a certain stimulating effect on the brain of mice, which can make the brain excited and increase autonomous activity. Figure 1 It can be seen that the number of autonomous activities of mice in the experimental group within a certain period of time was significantly lower than that of mice in the model group, indicating that probiotics have a certain alleviating effect on alcohol stimulation. Figure 2 , Figure 3 It can be seen that compared with the mice in the model group, the righting reaction tolerance time and sleep latency time of the experimental group mice were significantly increased, and the duration of both was significantly reduced, indicating that probiotics have the effect of sobering up and preventing drunkenness under experimental conditions, and the effect is more obvious.
[0140] The liver coefficient can reflect the degree of liver damage. Figure 4 It can be seen that the liver coefficient of the experimental group mice was significantly lower than that of the model group mice, indicating that probiotics can protect the liver; GSH, T-SOD and CAT can reflect the body's antioxidant capacity and indirectly reflect the lipid peroxidation damage to liver cells. Figure 5 , Figure 6 , Figure 7 It can be seen that compared with the blank control group, the GSH content, GSH-Px, T-SOD, and CAT activities in the liver tissue of the model group were significantly reduced, and the MDA content was greatly increased. Compared with the model group, the GSH content in the experimental group was significantly increased, the GSH-Px, T-SOD, and CAT activities were increased, and the MDA content was significantly decreased, indicating that probiotics can reduce the accumulation of peroxidation products and the level of ROS in liver cells, enhance the body's ability to scavenge free radicals, and promote the production of antioxidants in the liver, thereby reducing the oxidative damage caused by alcohol to the liver; when the TG content is too high, it will cause fat to deposit in liver cells, affecting liver lipid metabolism and causing fatty liver and other diseases. According to Figure 8 It can be seen that compared with the model group, the TG content in the liver of the experimental group decreased, which indicates that probiotics can improve the lipid metabolism disorder caused by alcohol and have a good liver protection effect; the levels of ALT and AST in the blood are often used to evaluate the degree of liver damage. Fig. 9 It can be seen that compared with the control group, the ALT and AST activities in the serum of the model group mice were significantly increased. Compared with the model group, the ALT and AST activities in the experimental group were significantly reduced. This result shows that probiotics play a certain protective role against acute liver damage caused by heavy drinking; proinflammatory factors and inflammatory mediators will further lead to liver cell necrosis and apoptosis, damage the liver, and promote liver fibrosis. Fig.10It can be seen that compared with the control group, the levels of IL-1β, IL-6, and TNF-α in the serum of the model group were significantly increased, and compared with the model group, the levels of IL-1β, IL-6, and TNF-α in the experimental group were significantly decreased. This result shows that probiotics can reduce the levels of inflammatory factors TNF-α, IL-6, and IL-1β in the serum of mice in the model group, alleviate the inflammatory response caused by large amounts of alcohol intake, show good anti-inflammatory effects, and thus play a role in protecting the liver.
[0141] 2. Experiment on regulating the function of intestinal flora in mice
[0142] The mice were divided into a control group and an experimental group, with 10 mice in each group. The control group was fed normally, while the experimental group was fed normally. 30 mg of the above-mentioned freeze-dried powder of the inactivated bacterial metabolite was dissolved in 0.5 mL of sterile water to prepare a solution for gavage. After 30 days of feeding, the mouse feces were aseptically collected, weighed, diluted 10 times with sterile diluent to make a uniform suspension, and then diluted 10 times in turn. The appropriate dilution was selected and inoculated on each selective culture medium. The number of bacteria per gram of wet feces was calculated, and the logarithm was taken for statistical processing. 24 hours after the last administration of the probiotic product, rectal feces were taken to detect intestinal flora. The test results are shown in Table 1.
[0143] Table 1 Suitable culture conditions and detection of intestinal flora (x-±SD)
[0144]
[0145] As can be seen from Table 1, probiotics can increase the number of beneficial bacteria (Bifidobacterium and Lactobacillus), while the number of harmful bacteria (Enterobacter, Enterococcus) did not increase but showed a slight downward trend, indicating that probiotics have the function of regulating the intestinal flora of mice.
[0146] 3. Experiment on the effect of probiotics on gastrointestinal motility in mice
[0147] The mice were divided into a control group, a model group and an experimental group, with 10 mice in each group. The control group and the model group were given 0.5 mL of sterile water by gavage once a day; the experimental group was given a solution of a probiotic product (30 mg dissolved in 0.5 mL of sterile water) (i.e., the lyophilized powder prepared in Example 1) by gavage once a day. After 15 days of gavage, the mice in each group fasted for 16 hours. After that, the model group and the experimental group were given compound difenoxol by gavage, and the control group was given distilled water by gavage. After 30 minutes, the mice in each group were given Arabic gum ink by gavage. After 30 minutes, the mice were killed by cervical vertebral dissection, and the abdomen was quickly cut along the midline of the abdomen to separate the mesentery. The intestinal tube was cut with the pylorus as the starting point and the ileocecal part as the end point. The small intestine was gently pulled into a straight line, and the intestinal tube length was measured as the "total length of the small intestine", and the length from the pylorus to the front of the ink was the "ink advancement length". The ink advancement rate was calculated according to the following formula. The ink advancement rate of the small intestine is shown in Table 2.
[0148]
[0149] Table 2 Small intestinal ink propulsion rate (x - ±SD)
[0150]
[0151] As shown in Table 2, the intestinal motility inhibition model of mice was established by administering compound difenocool. Compared with the model group, the small intestinal ink propulsion rate increased in the experimental group, indicating that probiotics can promote small intestinal motility and digestion.
[0152] 4. Metabolomics analysis
[0153] Metabolomics analysis of mouse feces was performed using ultra-performance liquid chromatography-tandem mass spectrometry.
[0154] Experimental plan: The mice were divided into a control group and an experimental group. The control group was fed normally, while the experimental group was fed normally and gavaged with 30 mg of the solution prepared by the freeze-dried powder of the above-mentioned inactivated bacterial metabolites. After feeding for 90 days, fecal samples of the mice in the control group and the experimental group were placed in 400 μL of extraction solution (acetonitrile: methanol = 1: 1), vortexed for 30 s, and extracted with low-temperature ultrasound for 30 min (5℃, 40 KHz), allowed to stand at -20℃ for 30 min, placed in a 4℃ centrifuge, centrifuged at 13000 r / min for 15 min, the supernatant was taken, dried with nitrogen, re-dissolved with 120 μL of the reconstitution solution (acetonitrile: water = 1: 1), extracted with low-temperature ultrasound for 5 min (5℃, 40 KHz), placed in a 4℃ centrifuge, centrifuged at 13000 r / min for 5 min, and an appropriate amount of supernatant was taken for analysis. 10uL of sample supernatant was separated by BEH C18 column (100mm ×2.1mm, 1.8µm) and then entered mass spectrometry detection. The separation flow rate was 0.40mL / min and the column temperature was 40℃. After completion, the LC-MS raw data was transferred to the computer for processing and analysis. The MS mass spectrum information was matched with the public database to obtain the main metabolite information such as Fig.11 As shown, the contents of the main metabolites were determined, and the results are shown in Table 3.
[0155] Among them, the method for detecting the mucopolysaccharide content is spectrophotometry; the method for detecting the ornithine content is high performance liquid chromatography; the method for detecting the lactobacillus content is high performance liquid chromatography; and the method for detecting the lecithin content is high performance liquid chromatography.
[0156] Table 3 Contents of main metabolites in mice
[0157]
[0158] It can be seen from Table 3 that daily administration of 30 mg of a probiotic product containing 4.5 billion inactivated bacteria and metabolites to mice can significantly increase the contents of mucopolysaccharides, ornithine, lactobacillus, and lecithin in the feces of the mice, indicating that the probiotic product provided by the present invention can sustainably provide the endogenously required amount of mucopolysaccharides, ornithine, lactobacillus, and lecithin during the intestinal reproduction and growth process.
