[0005] The pancreas is an organ in the human body that has both exocrine and endocrine functions. The exocrine part includes the acinus, which can secrete a variety of digestive enzymes, and the endocrine part includes the islets of Langerhans, which can secrete hormones such as insulin, which plays a very important role in maintaining blood sugar levels. Fat accumulation in the pancreas may affect pancreatic function, further causing pancreatic disease, diabetes or other related diseases. Specifically, diabetes is a disease of abnormal glucose metabolism. There are two different types of diabetes, including non-insulin-dependent or adult-onset, also known as type II diabetes; and insulin-dependent or juvenile-onset, also known as type I diabetes. In general, type II diabetes usually occurs in adults and is highly correlated with obesity. It can be controlled through proper exercise, diet and medication; type I diabetes patients are more common in children and adolescents. The patient's body cannot produce enough insulin and must be injected with insulin to maintain life.

[0035] Examples
[0036] 1. Materials and Methods
[0037] 1.1 Experimental animals
[0038] The experimental animals used were B6 mice. The number of experimental animals in each group was ≥4 in the preliminary test, and ≥12 in the confirmation test. Male mice were raised in an animal room with a normal light-dark cycle (7:00 am to 7:00 pm is the light period, and the rest is the dark period), a temperature of 23±2℃, and a relative humidity of 55±15%, and weighed 18-23g. They were purchased from Lesco Biotechnology Co., Ltd., and animal experiments were conducted in accordance with the experimental guidelines. First, feed 3-5g of general feed per day, raise for 1-2 weeks to observe the health status, and water is supplied unlimitedly. The body weight is recorded once a week.
[0039] 1.2 Animal groups
[0040] The experimental animals were randomly divided into blank control group (Blank), high-fat control group (HFD), positive control group (Positive control, PS) and experimental group. The blank control group was given normal diet; the high-fat control group was given high-fat diet; the positive control group was given high-fat diet and tube-fed silymarin (silymarin, 5mg / kg / day) (Iranian Journal of Pharmaceutical Research (2016), 15(3): 493-500; Life Sciences 75(2004) 2167–2180; and Pharm Biol. 2016 Jul 8: 1-6); and the experimental group was given high-fat diet and tube-fed with individual experimental ingredients.
[0041] 1.3 Feeding method
[0042] The blank control group was fed with normal feed for 12 weeks, while the high-fat control group, positive control group and experimental group were fed with high-fat feed for 12 weeks. After 8 weeks of feeding, the blank control group and high-fat control group were fed with deionized water once a day; the positive control group was fed with silymarin once a day; and the experimental group was fed with the test compound once a day for 4 weeks.
[0043] 1.4 Blood collection and various analyses
[0044] Blood was collected from the cheek or heart before the start of the experiment and at 8 and 12 weeks after the start of the experiment. Finally, all mice were weighed and sacrificed at the end of the experiment, and blood was collected from the cheek or heart. The mouse blood samples were placed at room temperature for 1 hour to allow them to coagulate. Then, they were centrifuged at 15,700xg for 5 minutes at 4°C in a refrigerated centrifuge to separate the serum.
[0045] 1.4.1 Analysis of triglycerides (TG) and total cholesterol (TCHO, TC) in serum
[0046] The triglyceride (TG) and total cholesterol (TCHO, TC) in serum were measured by blood automatic biochemical analyzer.
[0047] 1.4.2 Pancreatic biochemical function analysis: blood glucose analysis (GLU)
[0048] Blood glucose analysis was performed using a Roche ACCU-CHEK Active blood glucose meter. The mice were fasted overnight the day before blood collection. The code card of the blood glucose test strip was inserted into the blood glucose meter, and then the blood glucose test strip was inserted. When a blood drop pattern appeared on the machine, a drop of blood was collected from the cheek vein of the mouse using a cheek needle and dripped into the blood glucose meter test strip, and the blood glucose data displayed by the blood glucose meter was recorded.
[0049] 1.4.3 Pancreatic biochemical function analysis: intraperitoneal glucose tolerance test (IPGTT)
[0050] Move the mice to a clean cage and fast overnight. There is no food or feces in the feeding funnel or bottom of the clean cage. Make sure the animals have access to drinking water at all times. Measure the weight of the mice, and calculate the volume of glucose solution required for intraperitoneally (IP) injection (the concentration is 250 mg / mL, 2 g glucose / mouse body weight, kg, the required volume (μl) is 8x body weight (g)). Move the mice out, inject the appropriate amount of glucose solution into the mice intraperitoneally, and record the injection time. At 0, 15, 30, 60, 120 and 150 minutes after the injection of glucose solution, blood is collected from the tail of the mice to measure the blood glucose concentration, and blood glucose analysis is performed using a Roche ACCU-CHEK Active blood glucose meter. 1.4.4 Pancreatic biochemical function analysis: fasting insulin concentration analysis
[0051] At the end of the experiment (12 weeks), the mice were fasted and blood was collected according to the instructions of the Mouse ultrasensitive insulin ELISA kit (ALPCO).
[0052] 1.4.5 Analysis of pancreatic biochemical function: homeostasis model assessment of insulin resistance (HOMA-IR)
[0053] At the end of the experiment (12 weeks), the mice were fasted and blood was collected to measure blood glucose and insulin concentrations, which were calculated according to the following formula:
[0054]
[0055] 1.4.6 Analysis of triglycerides (TG) and total cholesterol (TCHO, TC) in pancreatic tissue
[0056] After the mice were sacrificed, the pancreas samples were removed by laparotomy and weighed to compare the pancreas weight / body weight ratio. The remaining pancreas was frozen and stored to detect the triglyceride and total cholesterol content in the pancreas.
[0057] 1.5 Statistical analysis
[0058] All data are expressed as mean ± standard deviation (SD). The experimental results were analyzed by one-way analysis of variance (ANOVA) to determine whether they were statistically significant. The Statistical Package of the Social Science program (Version 13, SPSS Inc.) was used for calculation. Post hoc test with the least significant difference method was then used for multiple comparisons to confirm the significant differences between the groups. The significant difference between the group means was set at p < 0.05.
[0059] 2. Results
[0060] 2.1 Animal experiments
[0061] In the animal experiment, except for the blank group which was given normal feed, the other animals were given high-fat feed. After 8 weeks, the animals in each group were given different treatments for 4 weeks in addition to maintaining the original feed. The blank control group and the high-fat control group were given deionized water, the positive control group was given silymarin, and the test group was given different test compounds, including puerarin, phloridzin, eriodictyol, sucralose, mannitol, saccharin, orange peel, menthol, or a combination of some test compounds. There was no statistically significant difference in the weight gain and daily feed intake of the animals among the groups (p>0.05).
[0062] In addition, after the test groups were administered with different test compounds, the animals did not show any abnormal symptoms, no animals died during the test period, and no lesions or clinical symptoms caused by the test compounds were observed when the animals were sacrificed and autopsied after the test. Therefore, the test compounds are safe.
[0063] 2.2 The test compound has the effect of reducing pancreatic lipids
[0064] The results of the animal experiment showed that the various test compounds showed the effect of reducing pancreatic lipid in animals after a 4-week administration period. The results are shown in Table 1.
[0065] Table 1: Test compounds reduce pancreatic lipids in animals (during 4 weeks of dosing).
[0066]
[0067] The results showed that pancreatic triglycerides and total cholesterol increased in mice fed a high-fat diet. When a single test compound was administered, eriodictyol, mannitol, saccharin, sucralose, hesperidin, puerarin, and menthol significantly reduced pancreatic triglycerides by about 33%-87% compared to the high-fat control group. In particular, eriodictyol had an excellent effect after 4 weeks of treatment, reducing pancreatic triglyceride content by about 87% (p<0.005). In addition, eriodictyol, mannitol, saccharin, sucralose, hesperidin, puerarin, menthol, and phlorizin significantly reduced pancreatic total cholesterol by about 28%-51%; in particular, saccharin had an excellent effect after 4 weeks of treatment, reducing pancreatic total cholesterol content by about 51% (p<0.005).
[0068] When two or three compounds are administered in combination, for example, a combination of menthol and mannitol, or a combination of menthol, mannitol and eriodictyol, wherein the dosage of each component is maintained the same as the dosage used alone or the dosage of each component is lower than the dosage used alone (for example, about one-third of the dosage used alone), a similar or better triglyceride lowering effect can be achieved. Therefore, according to the present invention, the combined use of a specific test compound combination can reduce the dosage, but maintain similar or better efficacy, resulting in a synergistic effect.
[0069] 2.3 The test compound has the effect of regulating pancreatic function and treating diabetic symptoms
[0070] The results of animal experiments show that a variety of test compounds can regulate pancreatic function and reduce pancreatic dysfunction in animals during a 4-week administration period, especially for the function of sugar metabolism. The results are shown in Table 2-1.
[0071] Table 2-1: Test compounds can regulate pancreatic function and reduce pancreatic function damage in animals (drug administration period of 4 weeks)
[0072]
[0073]
[0074] Table 2-2: Test compounds can regulate pancreatic function and reduce pancreatic function damage in animals (drug administration period of 4 weeks)
[0075]
[0076]
[0077] This study examines whether the test compound can modulate pancreatic function and reduce the level of pancreatic dysfunction in animals, especially in the glucose metabolism function.
[0078] The results showed that pancreatic carbohydrate metabolism was impaired in mice fed a high-fat diet (GLU, IPGTT, fasting insulin concentration, and HOMA-IR values increased). When a single test compound was administered, sucralose, menthol, mannitol, and saccharin significantly reduced GLU; eriodictyol, sucralose, menthol, mannitol, saccharin, and puerarin significantly reduced glucose tolerance; phloridzin, eriodictyol, sucralose, menthol, mannitol, saccharin, and puerarin significantly reduced fasting insulin concentration and insulin resistance.
[0079] When two or three compounds are administered in combination, for example, a combination of menthol and mannitol, or a combination of menthol, mannitol and eriodictyol, wherein the dosage of each component is lower than the dosage used alone (for example, about one-third of the dosage used alone), similar or better effects of reducing fasting insulin concentration and insulin resistance (significantly reducing fasting insulin concentration, HOMA-IR value) can be achieved. Therefore, according to the present invention, it is shown that the combined use of a specific test compound combination can reduce the dosage of the drug, but maintain similar or better efficacy, with a synergistic effect.
[0080] 2.4 The test compound has the effect of lowering plasma triglycerides
[0081] The results of the animal experiment showed that a variety of test compounds could reduce triglycerides in the plasma of animals during the 4-week administration period. The results are shown in Table 3.
[0082] Table 3: Test compounds can reduce triglycerides in the plasma of animals (drug administration period of 4 weeks)
[0083]
[0084]
[0085] The results showed that high-fat diet induced an increase in plasma triglycerides in mice. When administered with a single test compound, eriodictyol and phlorizin significantly reduced plasma triglycerides, with the highest reduction being about 27% compared to the high-fat control group.
[0086] When the two test compounds were administered in combination, the combination of menthol and mannitol or the combination of eriodictyol and sucralose significantly reduced plasma triglycerides.
[0087] When the three test compounds were administered in combination, the combination of menthol, mannitol and eriodictyol was effective in lowering plasma triglycerides at low doses.
[0088] The compounds provided by the present invention can reduce pancreatic fat content and improve related symptoms in individuals. These compounds are low-molecular natural plant phenolic compounds, which are widely found in fruits and vegetables, grains, rhizomes, flowers, tea and red wine. They have been confirmed to be safe through animal experiments and have the potential to be developed into health foods or drugs for reducing pancreatic fat, preventing or treating fatty pancreas, improving pancreatic lesions caused by fatty pancreas, diabetes or other related symptoms, regulating pancreatic function, and especially improving insulin resistance.
